    Insights into evolving global populations of Phytophthora infestans via new complementary mtDNA haplotype markers and nuclear SSRs

    <div><p>In many parts of the world the damaging potato late blight pathogen, <i>Phytophthora infestans</i>, is spread as a succession of clonal lineages. The discrimination of genetic diversity within such evolving populations provides insights into the processes generating novel lineages and the pathways and drivers of pathogen evolution and dissemination at local and global scales. This knowledge, in turn, helps optimise management practices. Here we combine two key methods for dissecting mitochondrial and nuclear diversity and resolve intra and inter-lineage diversity of over 100 <i>P</i>. <i>infestans</i> isolates representative of key clonal lineages found globally. A novel set of PCR primers that amplify five target regions are provided for mitochondrial DNA sequence analysis. These five loci increased the number of mtDNA haplotypes resolved from four with the PCR RFLP method to 37 (17, 6, 8 and 4 for Ia, Ib, IIa, and IIb haplotypes, respectively, plus 2 Herb-1 haplotypes). As with the PCR RFLP method, two main lineages, I and II were defined. Group I contained 25 mtDNA haplotypes that grouped broadly according to the Ia and Ib types and resolved several sub-clades amongst the global sample. Group II comprised two distinct clusters with four haplotypes corresponding to the RFLP type IIb and eight haplotypes resolved within type IIa. The 12-plex SSR assay revealed 90 multilocus genotypes providing accurate discrimination of dominant clonal lineages and other genetically diverse isolates. Some association of genetic diversity and geographic region of contemporary isolates was observed; US and Mexican isolates formed a loose grouping, distinct from isolates from Europe, South America and other regions. Diversity within clonal lineages was observed that varied according to the age of the clone. In combination, these fine-scale nuclear and maternally inherited mitochondrial markers enabled a greater level of discrimination among isolates than previously available and provided complementary perspectives on evolutionary questions relating to the diversity, phylogeography and the origins and spread of clonal lineages of <i>P</i>. <i>infestans</i>.</p></div

    The structure and evolution of a forming galaxy cluster at z = 1.62

    We present a comprehensive picture of the Cl 0218.3−0510 protocluster at z = 1.623 across 10 comoving Mpc. Using filters that tightly bracket the Balmer and 4000 Å breaks of the protocluster galaxies we obtain precise photometric redshifts resulting in a protocluster galaxy sample that is 89 ± 5 per cent complete and has a contamination of only 12 ± 5 per cent. Both star-forming and quiescent protocluster galaxies are located, which allows us to map the structure of the forming cluster for the first time. The protocluster contains six galaxy groups, the largest of which is the nascent cluster. Only a small minority of the protocluster galaxies are in the nascent cluster (11 per cent) or in the other galaxy groups (22 per cent), as most protocluster galaxies reside between the groups. Unobscured star-forming galaxies predominantly reside between the protocluster’s groups, whereas red galaxies make up a large fraction of the groups’ galactic content, so observing the protocluster through only one of these types of galaxies results in a biased view of the protocluster’s structure. The structure of the protocluster reveals how much mass is available for the future growth of the cluster and we use the Millennium Simulation, scaled to a Planck cosmology, to predict that Cl 0218.3−0510 will evolve into a 2.7+3.9 −1.7 × 1014M cluster by the present day

    Training future generations to deliver evidence‐based conservation and ecosystem management

To be effective, the next generation of conservation practitioners and managers need to be critical thinkers with a deep understanding of how to make evidence-based decisions and of the value of evidence synthesis. 2. If, as educators, we do not make these priorities a core part of what we teach, we are failing to prepare our students to make an effective contribution to conservation practice. 3. To help overcome this problem we have created open access online teaching materials in multiple languages that are stored in Applied Ecology Resources. So far, 117 educators from 23 countries have acknowledged the importance of this and are already teaching or about to teach skills in appraising or using evidence in conservation decision-making. This includes 145 undergraduate, postgraduate or professional development courses. 4. We call for wider teaching of the tools and skills that facilitate evidence-based conservation and also suggest that providing online teaching materials in multiple languages could be beneficial for improving global understanding of other subject areas

    Facing the River Gauntlet: Understanding the Effects of Fisheries Capture and Water Temperature on the Physiology of Coho Salmon

    An improved understanding of bycatch mortality can be achieved by complementing field studies with laboratory experiments that use physiological assessments. This study examined the effects of water temperature and the duration of net entanglement on physiological disturbance and recovery in coho salmon (Oncorhynchus kisutch) after release from a simulated beach seine capture. Heart rate was monitored using implanted electrocardiogram biologgers that allowed fish to swim freely before and after release. A subset of fish was recovered in respirometers to monitor metabolic recovery, and separate groups of fish were sacrificed at different times to assess blood and white muscle biochemistry. One hour after release, fish had elevated lactate in muscle and blood plasma, depleted tissue energy stores, and altered osmoregulatory status, particularly in warmer (15 vs. 10°C) and longer (15 vs. 2 min) capture treatments. A significant effect of entanglement duration on blood and muscle metabolites remained after 4 h. Oxygen consumption rate recovered to baseline within 7–10 h. However, recovery of heart rate to routine levels was longer and more variable, with most fish taking over 10 h, and 33% of fish failing to recover within 24 h. There were no significant treatment effects on either oxygen consumption or heart rate recovery. Our results indicate that fishers should minimize handling time for bycatch and maximize oxygen supply during crowding, especially when temperatures are elevated. Physiological data, such as those presented here, can be used to understand mechanisms that underlie bycatch impairment and mortality, and thus inform best practices that ensure the welfare and conservation of affected species

    NASA's Constellation Program is designing a new generation of human rated launch and space transportation vehicles to first replace the Space Shuttle fleet, then support develop of a permanent human habitat on the Moon, and ultimately prepare for human exploration of Mars. The ambitious first step beyond low Earth orbit is to develop the infrastructure required for conducting missions to a variety of locations on the lunar surface for periods of a week and establishment of a permanent settlement at one of the lunar poles where crews will serve for periods on the order of approx.200 days. We present an overview of the most challenging aspects of the lunar environment that will need to be addressed when developing transport and habitat infrastructure for long term human presence on the Moon including low temperatures and dusty regolith surfaces, radiation environments due to galactic cosmic rays and solar energetic particles, charging of lunar infrastructure when exposed to lunar plasma environments, and secondary meteor environments generated by primary impacts on the lunar surface

    A preliminary study of genetic factors that influence susceptibility to bovine tuberculosis in the British cattle herd

    Associations between specific host genes and susceptibility to Mycobacterial infections such as tuberculosis have been reported in several species. Bovine tuberculosis (bTB) impacts greatly the UK cattle industry, yet genetic predispositions have yet to be identified. We therefore used a candidate gene approach to study 384 cattle of which 160 had reacted positively to an antigenic skin test (‘reactors’). Our approach was unusual in that it used microsatellite markers, embraced high breed diversity and focused particularly on detecting genes showing heterozygote advantage, a mode of action often overlooked in SNP-based studies. A panel of neutral markers was used to control for population substructure and using a general linear model-based approach we were also able to control for age. We found that substructure was surprisingly weak and identified two genomic regions that were strongly associated with reactor status, identified by markers INRA111 and BMS2753. In general the strength of association detected tended to vary depending on whether age was included in the model. At INRA111 a single genotype appears strongly protective with an overall odds ratio of 2.2, the effect being consistent across nine diverse breeds. Our results suggest that breeding strategies could be devised that would appreciably increase genetic resistance of cattle to bTB (strictly, reduce the frequency of incidence of reactors) with implications for the current debate concerning badger-culling
