    The details of the orbiter altitude attainable at the approach and landing tests (ALT) interface determined by the approach and landing shuttle engineering simulation (manned) were documented. The analysis culminated in the verification of the trends observed in a similar analysis performed previously on the space vehicle dynamics simulation (unmanned). Altitude variations attributable to pilot steering variability ranged between 492 ft higher to 383 ft lower. The requirement for this parametric analysis is first elaborated. The specifications, assumptions, and analytical approach used to determine the orbiter altitude at the ALT interface are then presented, followed by the results of the analytical approach and the conclusions and recommendations

    The identification of fire hazards at the San Pablo Reservoir Test Site in California using ERTS-1 data is discussed. It is stated that the two primary fire hazards in the area are caused by wild oat plants and eucalyptus trees. The types of imagery used in conducting the study are reported.     Image products and numeric data were extracted from both TM and MSS data in an effort to evaluate the quality of these data for interpreting major agricultural resources and conditions in California's Central Valley. The utility of TM data appears excellent for meeting most of the inventory objectives of the agricultural resource specialist. These data should be extremely valuable for crop type and area proportion estimation, for updating agricultural land use survey maps at 1:24,000-scale and smaller, for field boundary definition, and for determining the size and location of individual farmsteads

    A seven step procedure developed for evaluating the geometric properties of MSS and TM film produces is being implemented. Some 476 control points were selected of which 238 are being tested and edited for digitization and scaling errors. Tables show statistics established for assessing the spectral characteristics and variability, as well as the spatial resolution and radiometric sensitivity of TM data for a forest environment in an effort to determine the extent to which major forest cover type can be detected and identified on TM digital and image products. Results thus far show that the high quality obtained are more than sufficient for meeting most of the inventory objectives of the renewable resource specialist. The TM data should be extremely valuable for: (1) estimating forest cover types; (2) updating land use survey maps; and (3) determining the size and shape and location of individual forest clearings and water resources

    The geometric quality of TM film and digital products is evaluated by making selective photomeasurements and by measuring the coordinates of known features on both the TM products and map products. These paired observations are related using a standard linear least squares regression approach. Using regression equations and coefficients developed from 225 (TM film product) and 20 (TM digital product) control points, map coordinates of test points are predicted. The residual error vectors and analysis of variance (ANOVA) were performed on the east and north residual using nine image segments (blocks) as treatments. Based on the root mean square error of the 223 (TM film product) and 22 (TM digital product) test points, users of TM data expect the planimetric accuracy of mapped points to be within 91 meters and within 117 meters for the film products, and to be within 12 meters and within 14 meters for the digital products

    The surface of the reusable solid rocket boosters (SRB), which are jettisoned from the Shuttle Orbiter to parachute in the sea, are studied for colonization by marine life. Techniques for monitoring the marine microbial fouling of SRB materials are presented. An assessment of the nature and degree of the biofouling expected on the SRB materials in the recovery zone is reported. A determination of the degree and the effects of seasonal variation occurring on microbial fouling in the retrieval zone waters is made. The susceptibility of the SRB parachute recovery system to microbial fouling and biodeterioration is investigated. The development of scanning electron microscopy and epifluorescence microscopic observation techniques for rapid assessment of microbial fouling is discussed

    Protein sectors: statistical coupling analysis versus conservation

    Statistical coupling analysis (SCA) is a method for analyzing multiple sequence alignments that was used to identify groups of coevolving residues termed "sectors". The method applies spectral analysis to a matrix obtained by combining correlation information with sequence conservation. It has been asserted that the protein sectors identified by SCA are functionally significant, with different sectors controlling different biochemical properties of the protein. Here we reconsider the available experimental data and note that it involves almost exclusively proteins with a single sector. We show that in this case sequence conservation is the dominating factor in SCA, and can alone be used to make statistically equivalent functional predictions. Therefore, we suggest shifting the experimental focus to proteins for which SCA identifies several sectors. Correlations in protein alignments, which have been shown to be informative in a number of independent studies, would then be less dominated by sequence conservation.Comment: 36 pages, 17 figure

    The Circadian Clock Gene Period1 Connects the Molecular Clock to Neural Activity in the Suprachiasmatic Nucleus.

    The neural activity patterns of suprachiasmatic nucleus (SCN) neurons are dynamically regulated throughout the circadian cycle with highest levels of spontaneous action potentials during the day. These rhythms in electrical activity are critical for the function of the circadian timing system and yet the mechanisms by which the molecular clockwork drives changes in the membrane are not well understood. In this study, we sought to examine how the clock gene Period1 (Per1) regulates the electrical activity in the mouse SCN by transiently and selectively decreasing levels of PER1 through use of an antisense oligodeoxynucleotide. We found that this treatment effectively reduced SCN neural activity. Direct current injection to restore the normal membrane potential partially, but not completely, returned firing rate to normal levels. The antisense treatment also reduced baseline [Ca(2+)]i levels as measured by Fura2 imaging technique. Whole cell patch clamp recording techniques were used to examine which specific potassium currents were altered by the treatment. These recordings revealed that the large conductance [Ca(2+)]i-activated potassium currents were reduced in antisense-treated neurons and that blocking this current mimicked the effects of the anti-sense on SCN firing rate. These results indicate that the circadian clock gene Per1 alters firing rate in SCN neurons and raise the possibility that the large conductance [Ca(2+)]i-activated channel is one of the targets
