    Research encounters, reflexivity and supervision

    Reflexivity in qualitative and ethnographic social science research can provide a rich source of data, especially regarding the affective, performative and relational aspects of interviews with research subjects. This paper explores by means of three case examples different ways of accessing and using such reflexivity. The examples are drawn from an empirical psycho-social study into the identity transitions of first-time mothers in an inner-city multicultural environment. Fieldnotes and supervision were used to engage with researcher subjectivity, to enhance the productive use of reflexivity and to address the emotional work of research. The methodology of the supervision was psychoanalytic, in its use of a boundaried frame and of psychoanalytic forms of noticing oneself, of staying engaged emotionally as well as creating a reflective distance. The examples illustrate how this can enhance the knowledge gained about the research subjects

    An inventory of vertebrate roadkill in the Greater Mapungubwe Transfrontier Conservation Area, South Africa

    Using a standard protocol, we conducted vertebrate roadkill surveys in the Greater Mapungubwe Transfrontier Conservation Area (GMTFCA), South Africa, which is a World Heritage Site. A total of 991 roadkill were recorded on the paved roads and 36 roadkill on the unpaved roads. Identifiable roadkill comprised 162 species from 24 orders and 65 families. Ninety-three roadkill could not be identified to species level. Roadkill counts were strongly influenced by road type and season. More roadkill was recorded on the paved than the unpaved roads. Irrespective of road type, the proportion of roadkill was greatest in the hot/wet season (4.3 paved roadkill/km/day paved and 1.3 roadkill/km/day unpaved) and lowest in the cold/dry season (2.0 roadkill/km/day paved and 0.1 roadkill/km/day unpaved). The high numbers of vertebrates identified as roadkill suggests that road traffic has the potential to directly and negatively affect biodiversity conservation in this part of South Africa. We recommend continued roadkill data collection across South Africa to assist with creating an inventory of species most likely to be at risk from roads.     Optimising the cost of roadkill surveys based on an analysis of carcass persistence

    Reliable estimates of wildlife mortality due to wildlife-vehicle collisions are key to understanding its impact on wildlife populations and developing strategies to prevent or reduce collisions. Standardised approaches for monitoring roadkill are needed to derive robust and unbiased estimates of mortality that are comparable across different study systems and ecological contexts. When designing surveys, there is a trade-off between survey frequency (and hence logistical effort and financial cost) and carcass detection. In this regard, carcass persistence (the period a carcass remains detectable before being removed by decomposition or scavengers) is important; the longer a carcass persists, the greater the likelihood it will be detected with lower survey effort by conducting more infrequent surveys. Using multi-taxon carcass data collected over a month of repeated driven surveys, combined with five covariates (species functional group, body weight, carcass position on road, carcass condition [either flattened or not after impact], and rainfall prior to each survey), we explored the drivers of carcass persistence with the overall aim of providing information to optimise the design of carcass surveys along linear infrastructure. Our methodological approach included a survival analysis to determine carcass persistence, linear regressions to test the effect of covariates, a subsampling analysis (using field data and a simulation exercise) to assess how the proportion of carcasses detected changes according to survey frequency, and an analysis to compare the costs of surveys based on study duration, transect length and survey frequency. Mean overall carcass persistence was 2.7 days and was significantly correlated with position on road and within-functional group body weight. There was no evidence for a significant effect of rainfall, while the effect of carcass condition was weakly non-significant. The proportion of carcasses detected decreased sharply when survey intervals were longer than three days. However, we showed that survey costs can be reduced by up to 80% by conducting non-daily surveys. Expanding on the call for a standardised methodology for roadkill surveys, we propose that carcass persistence be explicitly considered during survey design. By carefully considering the objectives of the survey and characteristics of the focal taxa, researchers can substantially reduce logistical costs. In addition, we developed an R Shiny web app that can be used by practitioners to compare survey costs across a variety of survey characteristics.     Effectiveness of an implementation optimisation intervention aimed at increasing parent engagement in HENRY, a childhood obesity prevention programme - the Optimising Family Engagement in HENRY (OFTEN) trial: study protocol for a randomised controlled trial

    Background: Family-based interventions to prevent childhood obesity depend upon parents’ taking action to improve diet and other lifestyle behaviours in their families. Programmes that attract and retain high numbers of parents provide an enhanced opportunity to improve public health and are also likely to be more cost-effective than those that do not. We have developed a theory-informed optimisation intervention to promote parent engagement within an existing childhood obesity prevention group programme, HENRY (Health Exercise Nutrition for the Really Young). Here, we describe a proposal to evaluate the effectiveness of this optimisation intervention in regard to the engagement of parents and cost-effectiveness. Methods/design: The Optimising Family Engagement in HENRY (OFTEN) trial is a cluster randomised controlled trial being conducted across 24 local authorities (approximately 144 children’s centres) which currently deliver HENRY programmes. The primary outcome will be parental enrolment and attendance at the HENRY programme, assessed using routinely collected process data. Cost-effectiveness will be presented in terms of primary outcomes using acceptability curves and through eliciting the willingness to pay for the optimisation from HENRY commissioners. Secondary outcomes include the longitudinal impact of the optimisation, parent-reported infant intake of fruits and vegetables (as a proxy to compliance) and other parent-reported family habits and lifestyle. Discussion: This innovative trial will provide evidence on the implementation of a theory-informed optimisation intervention to promote parent engagement in HENRY, a community-based childhood obesity prevention programme. The findings will be generalisable to other interventions delivered to parents in other community-based environments. This research meets the expressed needs of commissioners, children’s centres and parents to optimise the potential impact that HENRY has on obesity prevention. A subsequent cluster randomised controlled pilot trial is planned to determine the practicality of undertaking a definitive trial to robustly evaluate the effectiveness and cost-effectiveness of the optimised intervention on childhood obesity prevention

    Prevalence and architecture of de novo mutations in developmental disorders.

    The genomes of individuals with severe, undiagnosed developmental disorders are enriched in damaging de novo mutations (DNMs) in developmentally important genes. Here we have sequenced the exomes of 4,293 families containing individuals with developmental disorders, and meta-analysed these data with data from another 3,287 individuals with similar disorders. We show that the most important factors influencing the diagnostic yield of DNMs are the sex of the affected individual, the relatedness of their parents, whether close relatives are affected and the parental ages. We identified 94 genes enriched in damaging DNMs, including 14 that previously lacked compelling evidence of involvement in developmental disorders. We have also characterized the phenotypic diversity among these disorders. We estimate that 42% of our cohort carry pathogenic DNMs in coding sequences; approximately half of these DNMs disrupt gene function and the remainder result in altered protein function. We estimate that developmental disorders caused by DNMs have an average prevalence of 1 in 213 to 1 in 448 births, depending on parental age. Given current global demographics, this equates to almost 400,000 children born per year

    Bi-allelic Loss-of-Function CACNA1B Mutations in Progressive Epilepsy-Dyskinesia.

    The occurrence of non-epileptic hyperkinetic movements in the context of developmental epileptic encephalopathies is an increasingly recognized phenomenon. Identification of causative mutations provides an important insight into common pathogenic mechanisms that cause both seizures and abnormal motor control. We report bi-allelic loss-of-function CACNA1B variants in six children from three unrelated families whose affected members present with a complex and progressive neurological syndrome. All affected individuals presented with epileptic encephalopathy, severe neurodevelopmental delay (often with regression), and a hyperkinetic movement disorder. Additional neurological features included postnatal microcephaly and hypotonia. Five children died in childhood or adolescence (mean age of death: 9 years), mainly as a result of secondary respiratory complications. CACNA1B encodes the pore-forming subunit of the pre-synaptic neuronal voltage-gated calcium channel Cav2.2/N-type, crucial for SNARE-mediated neurotransmission, particularly in the early postnatal period. Bi-allelic loss-of-function variants in CACNA1B are predicted to cause disruption of Ca2+ influx, leading to impaired synaptic neurotransmission. The resultant effect on neuronal function is likely to be important in the development of involuntary movements and epilepsy.     Wildlife roadkill patterns on a major highway in northern Tanzania

    Despite expanding road networks, there is limited understanding of the effects of roads on wildlife in East Africa. We present a baseline survey and describe the patterns of roadkill in the Tarangire–Manyara ecosystem of Tanzania. A 75 km stretch of the Arusha Highway that passes adjacent to Manyara Ranch and Lake Manyara National Park was studied for 10 consecutive days in November 2013 (the rainy season). Wildlife species killed on the road, roadkill frequency and road characteristics were determined. A total of 101 roadkill were recorded (0.13 roadkill km−1) comprising 37 species from all terrestrial vertebrate groups, of which two species, house cat (Felis catus) and domestic dog (Canis lupus), were domesticated species. Birds were the most frequently killed taxon (50%), followed by mammals (30%), reptiles (17%) and amphibians (3%). Excluding birds, roadkill primarily consisted of nocturnal species (65%) versus diurnal species (35%). Most roadkill (77.3%) were encountered on road stretches adjacent to protected areas of Manyara Ranch and Lake Manyara National Park compared with 22.7% on the road stretches adjacent to non-protected areas. These findings highlight that roads are a potential threat to wildlife in East Africa and serve as a baseline for future comparisons.Keywords: mitigation measures, protected areas, road ecology, wildlife mortalitie

    Testing the Value of Citizen Science for Roadkill Studies: A Case Study from South Africa

    Roads impact wildlife through a range of mechanisms from habitat loss and decreased landscape connectivity to direct mortality through wildlife-vehicle collisions (roadkill). These collisions have been rated amongst the highest modern risks to wildlife. With the development of “citizen science” projects, in which members of the public participate in data collection, it is now possible to monitor the impacts of roads over scales far beyond the limit of traditional studies. However, the reliability of data provided by citizen scientists for roadkill studies remains largely untested. This study used a dataset of 2,666 roadkill reports on national and regional roads in South Africa (total length ~170,000 km) over 3 years. We first compared roadkill data collected from trained road patrols operating on a major highway with data submitted by citizen scientists on the same road section (431 km). We found that despite minor differences, the broad spatial and taxonomic patterns were similar between trained reporters and untrained citizen scientists. We then compared data provided by two groups of citizen scientists across South Africa: (1) those working in the zoology/conservation sector (that we have termed “regular observers,” whose reports were considered to be more accurate due to their knowledge and experience), and (2) occasional observers, whose reports required verification by an expert. Again, there were few differences between the type of roadkill report provided by regular and occasional reporters; both types identified the same area (or cluster) where roadkill was reported most frequently. However, occasional observers tended to report charismatic and easily identifiable species more often than road patrols or regular observers. We conclude that citizen scientists can provide reliable data for roadkill studies when it comes to identifying general patterns and high-risk areas. Thus, citizen science has the potential to be a valuable tool for identifying potential roadkill hotspots and at-risk species across large spatial and temporal scales that are otherwise impractical and expensive when using standard data collection methodologies. This tool allows researchers to extract data and focus their efforts on potential areas and species of concern, with the ultimate goal of implementing effective roadkill-reduction measures

    Effects of culverts and roadside fencing on the rate of roadkill of small terrestrial vertebrates in northern Limpopo, South Africa

    We tested the effectiveness of low-level roadside fencing to direct wildlife towards existing culverts beneath the road (underpasses) in order to reduce road deaths of small terrestrial vertebrates. While our results showed a reduction in roadkill count (from eight to one) along the stretches of road where we installed barriers (from an average of 0.33 roadkill/day/km to 0.04 roadkill/day/km), this decrease was not significant, possibly due to the small number of dead animals detected across all sites. Our trial highlights the challenges in acquiring robust evidence for roadkill reduction interventions and, given the small sample size, we were unable to elicit firm conclusions for this study.
