FATE MAP OF THE BLASTODERM TO DETERMINE SEGMENTAL FATE IN TRIBOLIUM CASTANEUM

    Get PDF
Segmentation in arthropods has been modeled on the well-defined segmentation patterns found in Drosophila. In Drosophila, segments form simultaneously in the blastoderm where morphogenic gradients spanning the AP axis provide patterning inputs. However, in most arthropods, segments form sequentially from a posterior growth zone. Sequential segmentation in arthropods has recently been demonstrated to use a vertebrate- like segmentation clock (Sarrazin et al. 2012). The vertebrate segmentation clock is a molecular oscillator that regulates periodic somite formation (Gibb 2010). In the red flour beetle, Tribolium castaneum, the segmentation clock is coordinated by traveling waves of expression generated by a pair-rule gene oscillator. For this study, we aimed to identify and describe regulatory controls of the Tribolium clock and construct a fate map of the blastoderm. We particularly focus on whether the output of the clock at the blastoderm stage - prior to the striking rearrangements of germband formation - actually determines cell fate. From preliminary studies we know that the caudal and even-skipped genes are two key genes of the Tribolium segmental clock. To understand how the clock is regulated, we identified and isolated 2 kb upstream of the caudal promoter region. We have cloned and sequenced this fragment to use in building reporter constructs that will be used to identify the cis- regulatory region driving wild type caudal expression. A deletion series of those regions showing enhancer activity will be completed to resolve the cis-regulatory regions to smaller domains (~500 bp-1.5 kb). In efforts to generate a fate map of the blastoderm, we created the T3-Nls-Eos DNA template for later mRNA synthesis. We will perform embryo injections with mRNA encoding for EosFP to determine the degree to which segmental fate is determined at the blastoderm stage of development

A Deweyan Perspective on Communication, Cooperation, and Collaboration Between Elementary and Secondary Educators

    Get PDF
In the field of education, the belief is often expressed among educators and non-educators alike that elementary teachers teach kids; secondary teachers teach subject matter. This sets up an unspoken dichotomy that elementary teachers do not teach subject matter and that secondary teachers do not teach kids. This presents a problem situation. At the heart of this problem situation is the fact that elementary and secondary educators have little, if any, contact with one another within the public school setting. As a result, one has no idea what the other is doing, and this serves to further perpetuate stereotypes. Communication does not take place from one grade level to the next, and this leads to an undesirable society. An undesirable society, in other words, is one which internally and externally sets up barriers to free intercourse and communication of experience (Dewey, 1997, p. 99). Because administrators do not build conjoint inservices into the school calendar, and because teachers get caught up with what is taking place in their own classrooms, to the exclusion of what is taking place in their colleagues\u27 classrooms, barriers exist that prevent educators from engaging in free intercourse and communication of experience. Communication, cooperation, and collaboration are not taking place between elementary and secondary educators, yet there is much to be gained by such an alliance. The result is an undesirable society in America\u27s public schools

The narrative oil paintings in this show illustrate one or two female characters set in domestic interior spaces. The scenes bring with them allusions to suggest that something unusual has just happened or is about to happen by heightening the psychological emotional moments. The subject matter is intimate and painted on small supports to reinforce these feelings. The images go beyond the observed visual aesthetic or representation by shifting the picture plane and using dramatic color palettes and lighting to create deeper interior spaces, adding to the tension

The early catechetical renewal in France and its influence in the teaching of religion

Experience of teaching in France in the late 1950s led to an awareness of excellent methods and resources for religious education. This in turn led to a study of the French catechetical renewal movement. While it became evident that the main movement was led by Fr Joseph Colomb in the years immediately following the Second World War, it was soon apparent that the roots extended much further back in time. The present work is concerned with tracing the roots of the early catechetical renewal, set against the background of the early twentieth century. The influence on it of movements such as Modernism, and of the new educational ideas, are considered as well as others from abroad such as the Munich Movement. The effect of the call for a more gospel based approach in the teaching of religion by Bishop Landrieux of Dijon in 1922 and a moving away from the Catechism is discussed. Among the key figures who emerged in the years between the two world wars were Mme Marie Fargues and Mlle Francoise Derkenne, also Fr André Boyer and Fr Charles Quinet. Their creative activity was particularly intense in the 1930s. Their influence, traced in many instances through unpublished correspondence, is shown to be considerable, not only in France but in the wider international sphere. In particular, there is evidence that their change of approach to the concept of religious education as opposed to that of religious instruction came about sooner in France than elsewhere. It is also suggested that the remarkable tenacity of some of the pioneers in the face of considerable opposition may have helped to bring about the changed attitude to the laity, particularly to women, in certain branches of the Christian Church
