    Development of a computer technique for the prediction of transport aircraft flight profile sonic boom signatures

    A new computer technique for the analysis of transport aircraft sonic boom signature characteristics was developed. This new technique, based on linear theory methods, combines the previously separate equivalent area and F function development with a signature propagation method using a single geometry description. The new technique was implemented in a stand-alone computer program and was incorporated into an aircraft performance analysis program. Through these implementations, both configuration designers and performance analysts are given new capabilities to rapidly analyze an aircraft's sonic boom characteristics throughout the flight envelope

    Integrating aerodynamics and structures in the minimum weight design of a supersonic transport wing

    An approach is presented for determining the minimum weight design of aircraft wing models which takes into consideration aerodynamics-structure coupling when calculating both zeroth order information needed for analysis and first order information needed for optimization. When performing sensitivity analysis, coupling is accounted for by using a generalized sensitivity formulation. The results presented show that the aeroelastic effects are calculated properly and noticeably reduce constraint approximation errors. However, for the particular example selected, the error introduced by ignoring aeroelastic effects are not sufficient to significantly affect the convergence of the optimization process. Trade studies are reported that consider different structural materials, internal spar layouts, and panel buckling lengths. For the formulation, model and materials used in this study, an advanced aluminum material produced the lightest design while satisfying the problem constraints. Also, shorter panel buckling lengths resulted in lower weights by permitting smaller panel thicknesses and generally, by unloading the wing skins and loading the spar caps. Finally, straight spars required slightly lower wing weights than angled spars

    Performance potential of an advanced technology Mach 3 turbojet engine installed on a conceptual high-speed civil transport

    The performance of an advanced technology conceptual turbojet optimized for a high-speed civil aircraft is presented. This information represents an estimate of performance of a Mach 3 Brayton (gas turbine) cycle engine optimized for minimum fuel burned at supersonic cruise. This conceptual engine had no noise or environmental constraints imposed upon it. The purpose of this data is to define an upper boundary on the propulsion performance for a conceptual commercial Mach 3 transport design. A comparison is presented demonstrating the impact of the technology proposed for this conceptual engine on the weight and other characteristics of a proposed high-speed civil transport. This comparison indicates that the advanced technology turbojet described could reduce the gross weight of a hypothetical Mach 3 high-speed civil transport design from about 714,000 pounds to about 545,000 pounds.     Sonic Boom: Six Decades of Research

    Sonic booms generated by aircraft traveling at supersonic speeds have been the subject of extensive aeronautics research for over 60 years. Hundreds of papers have been published that document the experimental and analytical research conducted during this time period. The purpose of this publication is to assess and summarize this work and establish the state-of-the-art for researchers just entering the field, or for those interested in a particular aspect of the subject. This publication consists of ten chapters that cover the experimental and analytical aspects of sonic boom generation, propagation and prediction with summary remarks provided at the end of each chapter. Aircraft maneuvers, sonic boom minimization, simulation techniques and devices as well as human, structural, and other responses to sonic booms are also discussed. The geometry and boom characteristics of various low-boom concepts, both large civil transports and smaller business-jet concepts, are included. The final chapter presents an assessment of civilian supersonic overland flight and highlights the need for continued research and a low-boom demonstrator vehicle. Summary remarks are provided at the end of each chapter. The studies referenced in this publication have been drawn from over 500 references

    Decreased CXCR1 and CXCR2 expression on neutrophils in anti-neutrophil cytoplasmic autoantibody-associated vasculitides potentially increases neutrophil adhesion and impairs migration

    Introduction: In anti-neutrophil cytoplasmic autoantibody (ANCA)-associated vasculitides (AAV), persistent inflammation within the vessel wall suggests perturbed neutrophil trafficking leading to accumulation of activated neutrophils in the microvascular compartment. CXCR1 and CXCR2, being major chemokine receptors on neutrophils, are largely responsible for neutrophil recruitment. We speculate that down-regulated expression of CXCR1/2 retains neutrophils within the vessel wall and, consequently, leads to vessel damage.Methods: Membrane expression of CXCR1/2 on neutrophils was assessed by flow cytometry. Serum levels of interleukin-8 (IL-8), tumor necrosis factor alpha (TNF-α), angiopoietin 1 and angiopoietin 2 from quiescent and active AAV patients and healthy controls (HC) were quantified by ELISA. Adhesion and transendothelial migration of isolated neutrophils were analyzed using adhesion assays and Transwell systems, respectively.Results: Expression of CXCR1 and CXCR2 on neutrophils was significantly decreased in AAV patients compared to HC. Levels of IL-8, which, as TNFα, dose-dependently down-regulated CXCR1 and CXCR2 expression on neutrophils in vitro, were significantly increased in the serum of patients with active AAV and correlated negatively with CXCR1/CXCR2 expression on neutrophils, even in quiescent patients. Blocking CXCR1 and CXCR2 with repertaxin increased neutrophil adhesion and inhibited migration through a glomerular endothelial cell layer.Conclusions: Expression of CXCR1 and CXCR2 is decreased in AAV, potentially induced by circulating proinflammatory cytokines such as IL-8. Down-regulation of these chemokine receptors could increase neutrophil adhesion and impair its migration through the glomerular endothelium, contributing to neutrophil accumulation and, in concert with ANCA, persistent inflammation within the vessel wall. © 2012 Hu et al.; licensee BioMed Central Ltd.link_to_subscribed_fulltex

    Higher Plasma Soluble Receptor for Advanced Glycation End Products (sRAGE) Levels Are Associated With Incident Cardiovascular Disease and All-Cause Mortality in Type 1 Diabetes: A 12-Year Follow-Up Study

    To investigate the associations of plasma levels of soluble receptor for advanced glycation end products (sRAGE) with incident cardiovascular disease (CVD) and allcause mortality in type 1 diabetes and the extent to which any such associations could be explained by endothelial and renal dysfunction, low-grade inflammation, arterial stiffness, and advanced glycation end products (AGEs). RESEARCH DESIGN AND METHODS - We prospectively followed 169 individuals with diabetic nephropathy and 170 individuals with persistent normoalbuminuria who were free of CVD at study entry and in whom levels of sRAGE and other biomarkers were measured at baseline. The median follow-up duration was 12.3 years (7.6 -12.5). RESULTS - The incidence of fatal and nonfatal CVD and allcause mortality increased with higher baseline levels of log-transformed sRAGE (Ln-sRAGE) independently of other CVD risk factors: hazard ratio (HR) 1.90 (95% CI 1.13-3.21) and 2.12 (1.26 -3.57) per 1-unit increase in Ln-sRAGE, respectively. Adjustments for estimated glomerular filtration rate (eGFRMDRD), but not or to a smaller extent for markers of endothelial dysfunction, low-grade inflammation, arterial stiffness, and AGEs, attenuated these associations to HR 1.59 (95% CI 0.91-2.77) for fatal and nonfatal CVD events and to 1.90 (1.09 -3.31) for all-cause mortality. In addition, in patients with nephropathy, the rate of decline of GFR was 1.38 ml/min/1.73 m2 per year greater per 1-unit increase of Ln-sRAGE at baseline (P = 0.036). CONCLUSIONS - Higher levels of sRAGE are associated with incident fatal and nonfatal CVD and all-cause mortality in individuals with type 1 diabetes. sRAGE-associated renal dysfunction may partially explain this association

    Design and analysis of low boom concepts at Langley Research Center

    The objective of the sonic boom research in the current High Speed Research Program is to ultimately make possible overland supersonic flight by a high speed civil transport. To accomplish this objective, it is felt that results in four areas must demonstrate that such a vehicle would be acceptable by the general public, by the airframers, and by the airlines. It should be demonstrated: (1) that some waveform shape has the possibility of being acceptable to the general public; (2) that the atmosphere would not totally destroy such a waveform during propagation; (3) that a viable airplane could be built which produces such a waveform; and (4) that any performance penalty suffered by a low boom aircraft would be counteracted by the economic benefit of overland supersonic flight. The work being done at LaRC is in support of the third element listed above--the area of configuration design. The initial part of the paper will give a review of the theory being used for configuration designs and discuss two theory validation models which were built and tested within the past two years. Discussion of the wind tunnel and theoretical results (linear theory and higher order methods) and their implications for future designs will be included

    M2 macrophage is the predominant phenotype in airways inflammatory lesions in patients with granulomatosis with polyangiitis

    Background: Macrophages may present two distinct phenotypes indicated as M1 and M2 under different stimuli. M1 and M2 macrophages have divergent functions that range from enhancement of inflammation for M1 to tissue repair and remodeling for M2 macrophages. The objective of this study was to evaluate the distribution of M1 and M2 macrophage phenotypes in biopsies from the airways of patients with active granulomatosis with polyangiitis (GPA) and to analyze their associations with T and B cells in those biopsies, and with nasal carriage of Staphylococcus aureus, disease parameters and therapy. Methods: Consecutive GPA patients (n = 35) with active airway disease, who underwent respiratory tract biopsy were included. Immunohistochemical evaluation was performed to assess the distribution of macrophages and T and B cells using the markers CD68, CD3 and CD20, respectively. CD86 was used as the M1 marker and CD163 as the M2 marker while Tbet and GATA-3 were used as Th1 and Th2 markers, respectively. At the time of the biopsy patients were assessed for nasal carriage of Staphylococcus aureus and treatment. Results: Percentages of macrophages and T cells were significantly higher than those of B cells in lesional tissue from the respiratory tract in GPA. M2 macrophages and Th2 cells were more frequent than M1 macrophages (p = 0.0007) and Th1 cells (p <0.0001), respectively. Percentages of T cells were higher in nose biopsies than in biopsies from other sites (p = 0.021); macrophages and CD163(+) macrophages were more predominant in biopsy sites other than the nose (p = 0.039 and p = 0.012, respectively). Carriage of Staphylococcus aureus was associated with higher T cell scores (p = 0.014). The frequency of macrophages, especially M2 macrophages, was higher in GPA patients treated with immunosuppressive agents (p = 0.010); daily prednisolone dose was positively correlated with all macrophage markers. However, in multivariate analysis no independent associations were found between disease parameters and therapy with macrophage markers or T cells. Conclusion: In GPA, M2 is the predominant macrophage phenotype in the respiratory tract. Although some associations were observed between macrophages and T cells with therapy and nasal carriage of Staphylococcus aureus, they were not independently significant in multivariate analysis
