However, the scope of exercise biology extends well beyond energy expenditure and has emerged as a great ‘polypill’, which is safe, reliable and cost-effective not only in disease prevention but also treatment. Biological mechanisms by which exercise influences homeostasis are becoming clearer and involve multi-organ systemic adaptations. Most of the elements of a modern lifestyle influence the indigenous microbiota but few studies have explored the effect of increased physical activity. While dietary responses to exercise obscure the influence of exercise alone on gut microbiota, professional athletes operating at the extremes of performance provide informative data.     Controllability analysis of multi objective control systems

    The performance requirements stated in project specifications often comprise conflicting objectives. These objectives may further be a complex mix of steady state and dynamic performance. Control devices such as solenoid actuators are often chosen purely on steady state force characteristics, due to the difficulty of appraising the conflicting and generally non-linear nature of the performance objectives. This can have ramifications in terms not only of the actuator performance, but also in the overall controllability of the system when closed-loop control is implemented. An example automotive application examining the multi objective controllability of electronically actuated valves is presented. Multi objective evolutionary techniques are utilised to derive the optimal force-displacement characteristics and also dynamic characteristics of the desired actuator under the constraint of design performance criteria. The selected actuator is then assessed for its controllability and dynamic performance

    Kynurenine pathway metabolism and the microbiota-gut-brain axis

    It has become increasingly clear that the gut microbiota influences not only gastrointestinal physiology but also central nervous system (CNS) function by modulating signalling pathways of the microbiota-gut-brain axis. Understanding the neurobiological mechanisms underpinning the influence exerted by the gut microbiota on brain function and behaviour has become a key research priority. Microbial regulation of tryptophan metabolism has become a focal point in this regard, with dual emphasis on the regulation of serotonin synthesis and the control of kynurenine pathway metabolism. Here, we focus in detail on the latter pathway and begin by outlining the structural and functional dynamics of the gut microbiota and the signalling pathways of the brain-gut axis. We summarise preclinical and clinical investigations demonstrating that the gut microbiota influences CNS physiology, anxiety, depression, social behaviour, cognition and visceral pain. Pertinent studies are drawn from neurogastroenterology demonstrating the importance of tryptophan and its metabolites in CNS and gastrointestinal function. We outline how kynurenine pathway metabolism may be regulated by microbial control of neuroendocrine function and components of the immune system. Finally, preclinical evidence demonstrating direct and indirect mechanisms by which the gut microbiota can regulate tryptophan availability for kynurenine pathway metabolism, with downstream effects on CNS function, is reviewed. Targeting the gut microbiota represents a tractable target to modulate kynurenine pathway metabolism. Efforts to develop this approach will markedly increase our understanding of how the gut microbiota shapes brain and behaviour and provide new insights towards successful translation of microbiota-gut-brain axis research from bench to bedside

    The measurement of response shift in patients with advanced prostate cancer and their partners

    BACKGROUND: There is increasing evidence to support the phenomenon of response shift (RS) in quality of life (QoL) studies, with many current QoL measures failing to allow for this. If significant response shift occurs amongst prostate cancer patients, it will be necessary to allow for this in the design of future clinical research and to reassess the conclusions of previous studies that have not allowed for this source of bias. This study therefore aimed to assess the presence of RS and psychosocial morbidity in patients with advanced prostate cancer and their partners. METHODS: 55 consecutive advanced prostate cancer patients and their partners completed the Prostate Cancer Patient & Partner questionnaire (PPP), shortly after diagnosis and again at 3 months and 6 months. At the follow-up visits, both patients and partners also completed a then-test in order to assess RS. RESULTS: Partners consistently showed greater psychological morbidity than patients in relation to the prostate cancer. This was most marked on the General Cancer Distress (GCD) subscale (p < 0.001, paired t-test), and regarding worries about treatment (p = 0.01). Significant RS was identified in partners and patients by the use of the then-test technique, particularly on the GCD subscale, the concerns about treatment and the concerns about urinary symptoms items. CONCLUSION: These results suggest the presence of RS in patients with advanced prostate cancer and their partners, with higher levels of psychosocial morbidity noted amongst partners. This is the first study to identify RS in partners and calls into question the interpretation of all studies assessing changes in QoL that fail to allow for this phenomenon

    Changing situational contexts present a constant challenge to software developers.

    A software process can take many forms and its optimality demands that it should be harmonised with the needs of the given software development situational context. This theoretical proposition is reasonably clear. However, the finer details of the interaction between the software process and the factors of the situational context are much less obvious. In previously published research, the authors have elaborated a reference framework that identifies the factors of a situational context that affect the software process [1]. In this paper, we report on the application of our reference framework in an examination of the changing nature of software development situational contexts. Our corresponding study of fifteen software development companies indicates that certain factors appear more subject to change than others. This finding is a potentially important insight that can help us with the recurring challenge of adapting the software process to changing circumstances

    Effect of vitamin K2 on postural sway in older people who fall:a randomized controlled trial

    OBJECTIVES: Vitamin K is thought to be involved in both bone health and maintenance of neuromuscular function. We tested the effect of vitamin K2 supplementation on postural sway, falls, healthcare costs, and indices of physical function in older people at risk of falls.DESIGN: Parallel-group double-blind randomized placebo-controlled trial.SETTING: Fourteen primary care practices in Scotland, UK.PARTICIPANTS: A total of 95 community-dwelling participants aged 65 and older with at least two falls, or one injurious fall, in the previous year.INTERVENTION: Once/day placebo, 200 μg or 400 μg of oral vitamin K2 for 1 year.MEASUREMENTS: The primary outcome was anteroposterior sway measured using sway plates at 12 months, adjusted for baseline. Secondary outcomes included the Short Physical Performance Battery, Berg Balance Scale, Timed Up &amp; Go Test, quality of life, health and social care costs, falls, and adverse events.RESULTS: Mean participant age was 75 (standard deviation [SD] = 7) years. Overall, 58 of 95 (61%) were female; 77 of 95 (81%) attended the 12-month visit. No significant effect of either vitamin K2 dose was seen on the primary outcome of anteroposterior sway (200 μg vs placebo: -.19 cm [95% confidence interval [CI] -.68 to .30; P = .44]; 400 μg vs placebo: .17 cm [95% CI -.33 to .66; P = .50]; or 400 μg vs 200 μg: .36 cm [95% CI -.11 to .83; P = .14]). Adjusted falls rates were similar in each group. No significant treatment effects were seen for other measures of sway or secondary outcomes. Costs were higher in both vitamin K2 arms than in the placebo arm.CONCLUSION: Oral vitamin K2 supplementation did not improve postural sway or physical function in older people at risk of falls.</p

    Hilbert-Schmidt Separability Probabilities and Noninformativity of Priors

    The Horodecki family employed the Jaynes maximum-entropy principle, fitting the mean (b_{1}) of the Bell-CHSH observable (B). This model was extended by Rajagopal by incorporating the dispersion (\sigma_{1}^2) of the observable, and by Canosa and Rossignoli, by generalizing the observable (B_{\alpha}). We further extend the Horodecki one-parameter model in both these manners, obtaining a three-parameter (b_{1},\sigma_{1}^2,\alpha) two-qubit model, for which we find a highly interesting/intricate continuum (-\infty < \alpha < \infty) of Hilbert-Schmidt (HS) separability probabilities -- in which, the golden ratio is featured. Our model can be contrasted with the three-parameter (b_{q}, \sigma_{q}^2,q) one of Abe and Rajagopal, which employs a q(Tsallis)-parameter rather than α\alpha, and has simply q-invariant HS separability probabilities of 1/2. Our results emerge in a study initially focused on embedding certain information metrics over the two-level quantum systems into a q-framework. We find evidence that Srednicki's recently-stated biasedness criterion for noninformative priors yields rankings of priors fully consistent with an information-theoretic test of Clarke, previously applied to quantum systems by Slater.Comment: 26 pages, 12 figure

    The assessment of cognition in visually impaired older adults

    Background: visual and cognitive impairments are common in later life. Yet there are very few cognitive screening tests for the visually impaired. Objective: to screen for cognitive impairment in the visually impaired. Methods: case-control study including 150 elderly participants with visual impairment (n=74) and a control group without visual impairment (n=76) using vision-independent cognitive tests and cognitive screening tests (MMSE and clock drawing tests (CDT)) which are in part vision dependent. Results: the scoring of the two groups did not differ in the vision-independent cognitive tests. Visually impaired patients performed poorer than controls in the vision-dependent items of the MMSE (T=7.3; df: 148; P<0.001) and in CDT (T=3.1; df: 145; P=0.003). No group difference was found when vision-independent items were added to MMSE and CDT. The test score gain by the use of vision-independent items correlated with the severity of visual impairment (P<0.002).
