    Family-friendly backlash—Fact or fiction? The case of organizations\u27 on-site child care centers

    Employer offerings of on-site child care benefits have grown tremendously in the past few decades; both beneficial and detrimental effects on worker attitudes and behaviors have been noted. Some research suggests that offering on-site child care benefits can cause resentment among childless workers and/or workers with children who do not use the center. In a field sample of 271 employees (mean age 34.5 yrs), current and past use of the on-site child care center, as well as anticipated future use of the on-site child care center, were related to more positive proximal reactions such as attitudes closely related to the on-site child care center, but not to more general attitudes or behaviors. Results indicate that any family-friendly backlash may be limited to proximal reactions. These findings are discussed in light of organizational justice theories

    Galaxy Cluster Pressure Profiles as Determined by Sunyaev Zel'dovich Effect Observations with MUSTANG and Bolocam I: Joint Analysis Technique

    We present a technique to constrain galaxy cluster pressure profiles by jointly fitting Sunyaev-Zel'dovich effect (SZE) data obtained with MUSTANG and Bolocam for the clusters Abell 1835 and MACS0647. Bolocam and MUSTANG probe different angular scales and are thus highly complementary. We find that the addition of the high resolution MUSTANG data can improve constraints on pressure profile parameters relative to those derived solely from Bolocam. In Abell 1835 and MACS0647, we find gNFW inner slopes of γ=0.36−0.21+0.33\gamma = 0.36_{-0.21}^{+0.33} and γ=0.38−0.25+0.20\gamma = 0.38_{-0.25}^{+0.20}, respectively when α\alpha and β\beta are constrained to 0.86 and 4.67 respectively.     Shear thickening and jamming in densely packed suspensions of different particle shapes

    We investigated the effects of particle shape on shear thickening in densely packed suspensions. Rods of different aspect ratios and non-convex hooked rods were fabricated. Viscosity curves and normal stresses were measured using a rheometer for a wide range of packing fractions for each shape. Suspensions of each shape exhibit qualitatively similar Discontinuous Shear Thickening. The logarithmic slope of the stress/shear-rate relation increases dramatically with packing fraction and diverges at a critical packing fraction phi_c which depends on particle shape. The packing fraction dependence of the viscosity curves for different convex shapes can be collapsed when the packing fraction is normalized by phi_c. Intriguingly, viscosity curves for non-convex particles do not collapse on the same set as convex particles, showing strong shear thickening over a wider range of packing fraction. The value of phi_c is found to coincide with the onset of a yield stress at the jamming transition, suggesting the jamming transition also controls shear thickening. The yield stress is found to correspond with trapped air in the suspensions, and the scale of the stress can be attributed to interfacial tension forces which dramatically increase above phi_c due to the geometric constraints of jamming. The relationship between shear and normal stresses is found to be linear in both the shear thickening and jammed regimes, indicating that the shear stresses come from friction. In the limit of zero shear rate, normal stresses pull the rheometer plates together due to the surface tension of the liquid below phi_c, but push the rheometer plates apart due to jamming above phi_c.Comment: 13 pages, 13 figures. published in Physical Review

    A multi-centred sequential trial comparing PEGASUS, an intervention to promote shared decision making about breast reconstruction with usual care

    Decision making about breast reconstruction (BR) is complex. The Patients’ Expectations and Goals: Assisting Shared Understanding of Surgery (PEGASUS) intervention aims to support shared decision making by helping women and clinicians clarify and discuss their expectations around reconstructive surgery. We conducted a multi-centred sequential trial comparing PEGASUS (n = 52) with usual care (UC) (n = 86) in women considering reconstruction, who completed outcome measures at baseline, and 3, 6 and 12 months post-surgery. The primary outcome was BR-specific quality of life (Breast-Q) 6 months post-intervention. Secondary outcomes were health-related quality of life (EQ-5D-5L), capabilities (ICECAP-A) and decisional regret, compared using t-tests and Cohen's d. Comparative analyses revealed no significant differences between groups in Breast-Q scores at any time point, except for a favourable effect for UC on psychological well-being at 3 months (t = -2.41, p = .019, d= -0.59). Intervention participants reported significantly higher, therefore improved, ICECAP-A (t = -2.13, p = .037, d = -0.45) and EQ VAS (t = -2.28, p = .026, d = -0.49) scores at 12 months compared to UC. Decisional regret was significantly lower in the PEGASUS group compared to the UC group at 6 months (t =2.06, p = .044, d= -0.51), but this was not sustained at 12 months. In conclusion, the PEGASUS intervention offers some benefits to women considering BR. At times, women experienced less decisional regret, improved health-related quality of life and capability well-being. Findings are discussed in the light of fidelity testing and embedding PEGASUS into practice

    Galaxy Cluster Pressure Profiles as Determined by Sunyaev Zel’dovich Effect Observations with MUSTANG and Bolocam. II. Joint Analysis of 14 Clusters

    We present pressure profiles of galaxy clusters determined from high resolution Sunyaev-Zel'dovich (SZ) effect observations of fourteen clusters, which span the redshift range 0.25<z<0.89 0.25 < z < 0.89. The procedure simultaneously fits spherical cluster models to MUSTANG and Bolocam data. In this analysis, we adopt the generalized NFW parameterization of pressure profiles to produce our models. Our constraints on ensemble-average pressure profile parameters, in this study γ\gamma, C500C_{500}, and P0P_0, are consistent with those in previous studies, but for individual clusters we find discrepancies with the X-ray derived pressure profiles from the ACCEPT2 database. We investigate potential sources of these discrepancies, especially cluster geometry, electron temperature of the intracluster medium, and substructure. We find that the ensemble mean profile for all clusters in our sample is described by the parameters: [γ,C500,P0]=[0.3−0.1+0.1,1.3−0.1+0.1,8.6−2.4+2.4][\gamma,C_{500},P_0] = [0.3_{-0.1}^{+0.1}, 1.3_{-0.1}^{+0.1}, 8.6_{-2.4}^{+2.4}], for cool core clusters: [γ,C500,P0]=[0.6−0.1+0.1,0.9−0.1+0.1,3.6−1.5+1.5][\gamma,C_{500},P_0] = [0.6_{-0.1}^{+0.1}, 0.9_{-0.1}^{+0.1}, 3.6_{-1.5}^{+1.5}], and for disturbed clusters: [γ,C500,P0]=[0.0−0.0+0.1,1.5−0.2+0.1,13.8−1.6+1.6][\gamma,C_{500},P_0] = [0.0_{-0.0}^{+0.1}, 1.5_{-0.2}^{+0.1},13.8_{-1.6}^{+1.6}].     An African-Specific Variant of TP53 Reveals PADI4 as a Regulator of p53-Mediated Tumor Suppression

    TP53 is the most frequently mutated gene in cancer, yet key target genes for p53-mediated tumor suppression remain unidentified. Here, we characterize a rare, African-specific germline variant of TP53 in the DNA-binding domain Tyr107His (Y107H). Nuclear magnetic resonance and crystal structures reveal that Y107H is structurally similar to wild-type p53. Consistent with this, we find that Y107H can suppress tumor colony formation and is impaired for the transactivation of only a small subset of p53 target genes; this includes the epigenetic modifier PADI4, which deiminates arginine to the nonnatural amino acid citrulline. Surprisingly, we show that Y107H mice develop spontaneous cancers and metastases and that Y107H shows impaired tumor suppression in two other models. We show that PADI4 is itself tumor suppressive and that it requires an intact immune system for tumor suppression. We identify a p53–PADI4 gene signature that is predictive of survival and the efficacy of immune-checkpoint inhibitors. Significance: We analyze the African-centric Y107H hypomorphic variant and show that it confers increased cancer risk; we use Y107H in order to identify PADI4 as a key tumor-suppressive p53 target gene that contributes to an immune modulation signature and that is predictive of cancer survival and the success of immunotherapy

    MEASUREMENTS of the SUNYAEV-ZEL'DOVICH EFFECT in MACS J0647.7+7015 and MACS J1206.2-0847 at HIGH ANGULAR RESOLUTION with MUSTANG

    We present high resolution (9?) imaging of the Sunyaev-Zel'dovich Effect (SZE) toward two massive galaxy clusters, MACS J0647.7+7015 (z = 0.591) and MACS J1206.2-0847 (z = 0.439). We compare these 90 GHz measurements, taken with the Multiplexed Squid/TES Array at Ninety Gigahertz (MUSTANG ) receiver on the Green Bank Telescope, with generalized Navarro-Frenk-White (gNFW) models derived from Bolocam 140 GHz SZE data as well as maps of the thermal gas derived from Chandra X-ray observations. We adopt a serial-fitting approach, in which gNFW models are first fit to the Bolocam data and then compared to the MUSTANG data to determine an overall best-fit model. For MACS J0647.7+7015, we find a gNFW profile with core slope parameter ? = 0.9 fits the MUSTANG image with and probability to exceed (PTE) = 0.34. For MACS J1206.2-0847, we find , , and PTE = 0.70. In addition, we find a significant (&gt;3s) residual SZE feature in MACS J1206.2-0847 coincident with a group of galaxies identified in Very Large Telescope data and filamentary structure found in a weak-lensing mass reconstruction. We suggest the detected sub-structure may be the SZE decrement from a low mass foreground group or an infalling group. Giant Metrewave Radio Telescope measurements at 610 MHz reveal diffuse extended radio emission to the west, which we posit is either an active galactic nucleus-driven radio lobe, a bubble expanding away from disturbed gas associated with the SZE signal, or a bubble detached and perhaps re-accelerated by sloshing within the cluster. Using the spectroscopic redshifts available, we find evidence for a foreground (z = 0.423) or infalling group, coincident with the residual SZE feature

    Metabolome and microbiome profiling of a stress-sensitive rat model of gut-brain axis dysfunction

Individual diferences in response to stress have been linked to genetic and environmental factors and more recently, a role for the gut microbiota in the regulation of stress-related changes has been demonstrated. However, the mechanisms by which these factors infuence each other are poorly understood, and there are currently no established robust biomarkers of stress susceptibility. To determine the metabolic and microbial signatures underpinning physiological stress responses, we compared stress-sensitive Wistar Kyoto (WKY) rats to the normoanxious Sprague Dawley (SD) strain. Here we report that acute stress-induced strain-specifc changes in brain lipid metabolites were a prominent feature in WKY rats. The relative abundance of Lactococcus correlated with the relative proportions of many brain lipids. In contrast, plasma lipids were signifcantly elevated in response to stress in SD rats, but not in WKY rats. Supporting these fndings, we found that the greatest diference between the SD and WKY microbiomes were the predicted relative abundance of microbial genes involved in lipid and energy metabolism.     Current ecotoxicity testing needs among selected U.S. federal agencies

    U.S. regulatory and research agencies use ecotoxicity test data to assess the hazards associated with substances that may be released into the environment, including but not limited to industrial chemicals, pharmaceuticals, pesticides, food additives, and color additives. These data are used to conduct hazard assessments and evaluate potential risks to aquatic life (e.g., invertebrates, fish), birds, wildlife species, or the environment. To identify opportunities for regulatory uses of non-animal replacements for ecotoxicity tests, the needs and uses for data from tests utilizing animals must first be clarified. Accordingly, the objective of this review was to identify the ecotoxicity test data relied upon by U.S. federal agencies. The standards, test guidelines, guidance documents, and/or endpoints that are used to address each of the agencies’ regulatory and research needs regarding ecotoxicity testing are described in the context of their application to decision-making. Testing and information use, needs, and/or requirements relevant to the regulatory or programmatic mandates of the agencies taking part in the Interagency Coordinating Committee on the Validation of Alternative Methods Ecotoxicology Workgroup are captured. This information will be useful for coordinating efforts to develop and implement alternative test methods to reduce, refine, or replace animal use in chemical safety evaluations
