    Faecal corticosterone metabolite assessment in socially housed male and female wistar rats

    Knowledge of animals’ hormonal status is important for conservation studies in wild or semi-free-ranging conditions as well as for behavioural and clinical experiments conducted in laboratory research, mostly performed on rats and mice. Faecal sampling is a useful non-invasive method to obtain steroid hormone assessments. Nevertheless, in laboratory studies, unlike other contexts, faecal sampling is less utilised. One of the issues raised is the necessity to collect samples belonging to different animals, separately. Usually, researchers using faecal sampling solve this problem through the isolation of animals or taking the cage rather than single animal as unit of study. These solutions though, could lead to unreliable measurements, and cannot be applied in many studies. Our aim was to show the biological reliability of individual faecal corticosterone metabolite (FCM) assessments in socially housed male and female Wistar rats. We analytically validated the enzyme immunoassay kit used for FCM assessments. Then, we exposed the animals to two different stress stimuli that are known to activate the hypothalamus–pituitary–adrenal axis and the following release of corticosterone to biologically validate the EIA kit: environmental enrichment and predator odour. Individual faecal sampling from social animals was collected through short-time handling. The results demonstrated that both the stimuli increased FCM levels in male and female rats showing the reliability of EIA kit assessment and the applicability of our sampling method. We also found a diurnal rhythm in FCM levels. These results could help to increase the use of faecal hormone metabolite determinations in studies conducted on rats

    Sensible heat flux and boundary layer depth measurements by Doppler SODAR and sonic anemometer data

    A validation of a simple mixed-layer similarity relationship, firstly proposed by Panofsky and McCormick (1960), is presented for wind speeds up to 7 ms21 and over an uneven terrain. The surface heat flux and the Planetary Boundary Layer depth, zi, are retrieved from this relationship, by using SODAR measurements of the vertical velocity variance s 2 w, under the hypothesis that the heat flux linearly decreases with height. All the measurements are relative to days characterized by high-pressure conditions, during periods of well-developed convection. The values of the surface heat flux obtained from such a method are compared with those obtained by applying the eddy correlation technique to the vertical wind velocities and virtual temperatures measured by a sonic anemometer. The values of zi obtained from the same relationship are compared with the height of the lowest inversion layer estimated from the facsimile record of the echoes received by the vertical antenna of the SODAR. The spectral behavior of vertical and longitudinal wind velocity from the anemometer and the SODAR is examined, too. In such a way an independent estimate of zi is obtained from the position of the spectral maximum

    A combined Rayleigh-Raman lidar for measurements of tropospheric water vapour and aerosol profiles

    The receiver of the Differential Absorption Lidar system of the University of L’Aquila (Italy) has been upgraded for the detection of Raman scattering from nitrogen and water vapour induced by XeCl and XeF excimer laser lines. In this configuration, only the XeF source is activated, so we can measure the tropospheric water vapour mixing ratio profiles with a height resolution of 300 m and 10 min in time. The lower limit sensitivity for the mixing ratio of water vapour is about 2 Q1024 and the precision ranges between 5% at 2 km and 50% at 9 km. The aerosol back-scattering ratio profiles can be measured with the same altitude and time resolution up to the lower stratosphere, the relative error is below 5% in the troposphere and about 30% at the highest altitudes. Comparisons with coincident PTU balloon-sondes show that the performances of the system in measuring the tropospheric water vapour are well calibrated for studying the water vapour evolution and cloud formation in the troposphere

    Spatial behaviour of yellow-necked wood mouse Apodemus flavicollis in two sub-Mediterranean oak coppice stands

    Strong mutual relationships exist between rodents and ecosystems. By modifying the structure and functioning of ecosystems, human activity can affect rodent behaviour and ecology. The yellow-necked mouse Apodemus flavicollis is widespread in Europe and its spatial ecology has been studied in various habitats, but studies are lacking for Mediterranean forests often altered by forest management practices. We investigated the spatial behaviour of A. flavicollis in a sub-Mediterranean deciduous oak forest of central Italy subject to forest management. We radio-tracked 27 individuals in two different coppice stands, i.e. a recently cut area and a high forest, differing in terms of species-specific habitat quality and rodent population density. We analysed the size of home ranges in relation to habitat type and sex. Our results revealed that home range and core area size did not differ between habitat types or sexes. The spatial behaviour of A. flavicollis thus did not appear to be influenced by population density and habitat quality. The lack of sex-related differences confirms the current knowledge on the species’ spatial ecology. Our findings provide the first useful information on the spatial behaviour of A. flavicollis in sub-Mediterranean deciduous oak forests, whose ecological processes may be markedly influenced by this key-species at several trophic levels.acceptedVersio

    Topic Strategies and the Internal Structure of Nominal Arguments in Greek and Italian

    In this article, we argue that a set of unexpected contrasts in the interpretation of clitic-left-dislocated indefinites in Greek and Italian derive from structural variation in the nominal syntax of the two languages. Greek resists nonreferential indefinites in clitic left-dislocation, resorting to the topicalization of an often bare noun for nonreferential topics. By contrast, clitic left-dislocation is employed in Italian for topics regardless of their definite/indefinite interpretation. We argue that this contrast is directly linked to the wide availability of bare nouns in Greek, which stems from a structural difference in the nominal syntax of the two languages. In particular, we hypothesize that Greek nominal arguments lack a D layer. Rather, they are Number Phrases. We situate this analysis in the context of Chierchia’s (1998) typology of nominals. We argue that, on a par with Italian nouns, Greek nouns are [−arg, +pred]. However, they do not employ a syntactic head (D) for type-shifting to e . Rather, they resort to covert type-shifting, a hypothesis that is necessary to account for the distribution and interpretations of bare nouns in Greek, vis-à-vis other [−arg, +pred] languages like Italian and French. </jats:p

    Urokinase Plasminogen Activator Inhibits HIV Virion Release from Macrophage-Differentiated Chronically Infected Cells via Activation of RhoA and PKCε

    HIV replication in mononuclear phagocytes is a multi-step process regulated by viral and cellular proteins with the peculiar feature of virion budding and accumulation in intra-cytoplasmic vesicles. Interaction of urokinase-type plasminogen activator (uPA) with its cell surface receptor (uPAR) has been shown to favor virion accumulation in such sub-cellular compartment in primary monocyte-derived macrophages and chronically infected promonocytic U1 cells differentiated into macrophage-like cells by stimulation with phorbol myristate acetate (PMA). By adopting this latter model system, we have here investigated which intracellular signaling pathways were triggered by uPA/uPAR interaction leading the redirection of virion accumulation in intra-cytoplasmic vesicles.uPA induced activation of RhoA, PKCδ and PKCε in PMA-differentiated U1 cells. In the same conditions, RhoA, PKCδ and PKCε modulated uPA-induced cell adhesion and polarization, whereas only RhoA and PKCε were also responsible for the redirection of virions in intracellular vesicles. Distribution of G and F actin revealed that uPA reorganized the cytoskeleton in both adherent and polarized cells. The role of G and F actin isoforms was unveiled by the use of cytochalasin D, a cell-permeable fungal toxin that prevents F actin polymerization. Receptor-independent cytoskeleton remodeling by Cytochalasin D resulted in cell adhesion, polarization and intracellular accumulation of HIV virions similar to the effects gained with uPA.These findings illustrate the potential contribution of the uPA/uPAR system in the generation and/or maintenance of intra-cytoplasmic vesicles that actively accumulate virions, thus sustaining the presence of HIV reservoirs of macrophage origin. In addition, our observations also provide evidences that pathways controlling cytoskeleton remodeling and activation of PKCε bear relevance for the design of new antiviral strategies aimed at interfering with the partitioning of virion budding between intra-cytoplasmic vesicles and plasma membrane in infected human macrophages

    Prescription appropriateness of anti-diabetes drugs in elderly patients hospitalized in a clinical setting: evidence from the REPOSI Register

    Diabetes is an increasing global health burden with the highest prevalence (24.0%) observed in elderly people. Older diabetic adults have a greater risk of hospitalization and several geriatric syndromes than older nondiabetic adults. For these conditions, special care is required in prescribing therapies including anti- diabetes drugs. Aim of this study was to evaluate the appropriateness and the adherence to safety recommendations in the prescriptions of glucose-lowering drugs in hospitalized elderly patients with diabetes. Data for this cross-sectional study were obtained from the REgistro POliterapie-Società Italiana Medicina Interna (REPOSI) that collected clinical information on patients aged ≥ 65 years acutely admitted to Italian internal medicine and geriatric non-intensive care units (ICU) from 2010 up to 2019. Prescription appropriateness was assessed according to the 2019 AGS Beers Criteria and anti-diabetes drug data sheets.Among 5349 patients, 1624 (30.3%) had diagnosis of type 2 diabetes. At admission, 37.7% of diabetic patients received treatment with metformin, 37.3% insulin therapy, 16.4% sulfonylureas, and 11.4% glinides. Surprisingly, only 3.1% of diabetic patients were treated with new classes of anti- diabetes drugs. According to prescription criteria, at admission 15.4% of patients treated with metformin and 2.6% with sulfonylureas received inappropriately these treatments. At discharge, the inappropriateness of metformin therapy decreased (10.2%, P &lt; 0.0001). According to Beers criteria, the inappropriate prescriptions of sulfonylureas raised to 29% both at admission and at discharge. This study shows a poor adherence to current guidelines on diabetes management in hospitalized elderly people with a high prevalence of inappropriate use of sulfonylureas according to the Beers criteria
