    Stability of a fluid in a rectangular region heated from below

    Finite-difference methods were used to compute hydrodynamic instability due to natural convection in an enclosed horizontal rectangular region heated from below. Critical Rayleigh numbers were determined for a series of Prandtl numbers and length-to-height ratios. For Prandtl numbers greater than unity excellent agreement was obtained between these calculations and the values predicted by Kurzweg on the basis of a linearized theory. However, for Prandtl numbers less than unity the critical Rayleigh numbers exhibited a dependence on N Pr , which was not predicted by the linearized theory. For Rayleigh numbers greater than the critical, complete temperature and velocity fields were determined. The calculations assumed that the fluid motion is two dimensional. Experiments have indicated that the flow may be two or three dimensional depending on minor perturbations in the boundary conditions. Although a number of metastable two-dimensional circulations are possible for symmetrical initial conditions, the calculation always converged to a single, unique solution for any asymmetric initial condition.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/37346/1/690130115_ftp.pd

    Differing effects of age and starvation on reproductive performance in Drosophila melanogaster

    Successful reproduction requires the completion of many, often condition-dependent, stages, from mate searching and courtship through to sperm transfer, fertilisation and offspring production. Animals can plastically adjust their investment in each stage according to the physical and social environment, their own condition, their future reproductive potential, and the condition of their partner. Here we manipulate age and condition, through a nutritional challenge early or late in life, of both male and female Drosophila melanogaster and measure the effects on courtship, mating, and fitness when paired with a standardized (unmanipulated) partner. Older males were slower to start courting and mating, and courted at a slower rate, but males were indifferent to female age or condition despite older females laying and hatching fewer eggs. Female condition had a substantial effect on mating acceptance rate, which dropped dramatically after starvation, and particularly recent starvation experience. In contrast, male condition had little effect on any of the components of reproductive performance we measured. Intriguingly, we found no evidence for additive or multiplicative effects of ageing and starvation: the only significant interaction between these variables was on male latency to initiate courtship - older males were slower to start courting unless they had experienced starvation early in life. These results indicate that the immediate costs of mating differ between males and females, and that the sexes differ in their perception of the opportunity cost sustained by refusing a mating opportunity. Our results support the idea that ageing has more wide-ranging impact on reproductive behaviours than does nutritional challenge

    <p>Abstract</p> <p>Background</p> <p>Maraviroc (MVC) and other CCR5 antagonists are HIV-1 entry inhibitors that bind to- and alter the conformation of CCR5, such that CCR5 is no longer recognized by the viral gp120 envelope (Env) glycoproteins. Resistance to CCR5 antagonists results from HIV-1 Env acquiring the ability to utilize the drug-bound conformation of CCR5. Selecting for HIV-1 resistance to CCR5-antagonists <it>in vitro </it>is relatively difficult. However, the CCR5-using CC1/85 strain appears to be uniquely predisposed to acquiring resistance to several CCR5 antagonists <it>in vitro </it>including MVC, vicriviroc and AD101.</p> <p>Findings</p> <p>Here, we show that Env derived from the parental CC1/85 strain is inherently capable of a low affinity interaction with MVC-bound CCR5. However, this phenotype was only revealed in 293-Affinofile cells and NP2-CD4/CCR5 cells that express very high levels of CCR5, and was masked in TZM-bl, JC53 and U87-CD4/CCR5 cells as well as PBMC, which express comparatively lower levels of CCR5 and which are more commonly used to detect resistance to CCR5 antagonists.</p> <p>Conclusions</p> <p>Env derived from the CC1/85 strain of HIV-1 is inherently capable of a low-affinity interaction with MVC-bound CCR5, which helps explain the relative ease in which CC1/85 can acquire resistance to CCR5 antagonists <it>in vitro</it>. The detection of similar phenotypes in patients may identify those who could be at higher risk of virological failure on MVC.</p

    Unraveling Diffusion in Fusion Plasma: A Case Study of In Situ Processing and Particle Sorting

    This work starts an in situ processing capability to study a certain diffusion process in magnetic confinement fusion. This diffusion process involves plasma particles that are likely to escape confinement. Such particles carry a significant amount of energy from the burning plasma inside the tokamak to the diverter and damaging the diverter plate. This study requires in situ processing because of the fast changing nature of the particle diffusion process. However, the in situ processing approach is challenging because the amount of data to be retained for the diffusion calculations increases over time, unlike in other in situ processing cases where the amount of data to be processed is constant over time. Here we report our preliminary efforts to control the memory usage while ensuring the necessary analysis tasks are completed in a timely manner. Compared with an earlier naive attempt to directly computing the same diffusion displacements in the simulation code, this in situ version reduces the memory usage from particle information by nearly 60% and computation time by about 20%

    Quasi-coherent fluctuations limiting the pedestal growth on Alcator C-Mod: experiment and modelling

    Performance predictions for future fusion devices rely on an accurate model of the pedestal structure. The candidate for predictive pedestal structure is EPED, and it is imperative to test the underlying hypotheses to further gain confidence for ITER projections. Here, we present experimental work testing one of the EPED hypotheses, namely the existence of a soft limit set by microinstabilities such as the kinetic ballooning mode. This work extends recent work on Alactor C-Mod (Diallo et al 2014 Phys. Rev. Lett. 112 115001), to include detailed measurements of the edge fluctuations and comparisons of edge simulation codes and experimental observations

    A Search for Time Variation of the Fine Structure Constant

    A method offering an order of magnitude sensitivity gain is described for using quasar spectra to investigate possible time or space variation in the fine structure constant, alpha. Applying the technique to a sample of 30 absorption systems, spanning redshifts 0.5 < z< 1.6, obtained with the Keck I telescope, we derive limits on variations in alpha over a wide range of epochs. For the whole sample Delta(alpha)/alpha = -1.1 +/- 0.4 x 10^{-5}. This deviation is dominated by measurements at z > 1, where Delta(alpha)/alpha = -1.9 +/- 0.5 x 10^{-5}. For z < 1, Delta(alpha)/alpha = -0.2 +/- 0.4 x 10^{-5}, consistent with other known constraints. Whilst these results are consistent with a time-varying alpha, further work is required to explore possible systematic errors in the data, although careful searches have so far not revealed any.Comment: 4 pages, 1 figure, accepted for publication in Physical Review Letter

    A Correlation Between Galaxy Morphology and MgII Halo Absorption Strength

    (Abridged) We compared the quantified morphological properties of 37 intermediate redshift MgII absorption selected galaxies to the properties of the absorbing halo gas, observed in the spectra of background quasars. The galaxy morphologies were measured using GIM2D modeling of Hubble Space Telescope WFPC-2 images and the absorbing gas properties were obtained from HIRES/Keck and UVES/VLT quasar spectra. We found a 3.1 sigma correlation between galaxy morphological asymmetries normalized by the quasar-galaxy projected separations, A/D, and the MgII rest-frame equivalent widths. Saturation effects cause increased scatter in the relationship with increasing W_r(2796). We defined a subsample for which the fraction of saturated pixels in the absorption profiles is f_sat<0.5. The correlation strengthened to 3.3 sigma. We also find a paucity of small morphological asymmetries for galaxies selected by MgII absorption as compared to those of the general population of field galaxies, as measured in the Medium Deep Survey. The K-S probability that the two samples are drawn from the same galaxy population is ruled out at a 99.8% confidence level. The A/D-W_r(2796) correlation suggests a connection between the processes that perturb galaxies and the quantity of gas in their halos, normalized by the impact parameter. Since the perturbations are minor, it is clear that dramatic processes or events are not required for a galaxy to have an extended halo; the galaxies appear "normal". We suggest that common, more mild processes that populate halos with gas, such as satellite galaxy merging, accretion of the local cosmic web, and longer-range galaxy-galaxy interactions, consequently also induce the observed minor perturbations in the galaxies.Comment: Accepted for publication in the Astrophysical Journa

    Visible-light-induced intramolecular charge transfer in the radical spirocyclisation of indole-tethered ynones

    Indole-tethered ynones form an intramolecular electron donor-acceptor complex that can undergo visible-light-induced charge transfer to promote thiyl radical generation from thiols. This initiates a novel radical chain sequence, based on dearomatising spirocyclisation with concomitant C-S bond formation. Sulfur-containing spirocycles are formed in high yields using this simple and mild synthetic protocol, in which neither transition metal catalysts nor photocatalysts are required. The proposed mechanism is supported by various mechanistic studies, and the unusual radical initiation mode represents only the second report of the use of an intramolecular electron donor-acceptor complex in synthesis

    Poloidal asymmetries in edge transport barriers

    Measurements of impurities in Alcator C-Mod indicate that in the pedestal region, significant poloidal asymmetries can exist in the impurity density, ion temperature, and main ion density. In light of the observation that ion temperature and electrostatic potential are not constant on a flux surface [Theiler et al., Nucl. Fusion 54, 083017 (2014)], a technique based on total pressure conservation to align profiles measured at separate poloidal locations is presented and applied. Gyrokinetic neoclassical simulations with XGCa support the observed large poloidal variations in ion temperature and density, and that the total pressure is approximately constant on a flux surface. With the updated alignment technique, the observed in-out asymmetry in impurity density is reduced from previous publishing [Churchill et al., Nucl. Fusion 53, 122002 (2013)], but remains substantial (nz,H/nz,L∌6). Candidate asymmetry drivers are explored, showing that neither non-uniform impurity sources nor localized fluctuation-driven transport are able to explain satisfactorily the impurity density asymmetry. Since impurity density asymmetries are only present in plasmas with strong electron density gradients, and radial transport timescales become comparable to parallel transport timescales in the pedestal region, it is suggested that global transport effects relating to the strong electron density gradients in the pedestal are the main driver for the pedestal in-out impurity density asymmetry.United States.     Asn 362 in gp120 contributes to enhanced fusogenicity by CCR5-restricted HIV-1 envelope glycoprotein variants from patients with AIDS

    <p>Abstract</p> <p>Background</p> <p>CCR5-restricted (R5) human immunodeficiency virus type 1 (HIV-1) variants cause CD4+ T-cell loss in the majority of individuals who progress to AIDS, but mechanisms underlying the pathogenicity of R5 strains are poorly understood. To better understand envelope glycoprotein (Env) determinants contributing to pathogenicity of R5 viruses, we characterized 37 full-length R5 Envs from cross-sectional and longitudinal R5 viruses isolated from blood of patients with asymptomatic infection or AIDS, referred to as pre-AIDS (PA) and AIDS (A) R5 Envs, respectively.</p> <p>Results</p> <p>Compared to PA-R5 Envs, A-R5 Envs had enhanced fusogenicity in quantitative cell-cell fusion assays, and reduced sensitivity to inhibition by the fusion inhibitor T-20. Sequence analysis identified the presence of Asn 362 (N362), a potential N-linked glycosylation site immediately N-terminal to CD4-binding site (CD4bs) residues in the C3 region of gp120, more frequently in A-R5 Envs than PA-R5 Envs. N362 was associated with enhanced fusogenicity, faster entry kinetics, and increased sensitivity of Env-pseudotyped reporter viruses to neutralization by the CD4bs-directed Env mAb IgG1b12. Mutagenesis studies showed N362 contributes to enhanced fusogenicity of most A-R5 Envs. Molecular models indicate N362 is located adjacent to the CD4 binding loop of gp120, and suggest N362 may enhance fusogenicity by promoting greater exposure of the CD4bs and/or stabilizing the CD4-bound Env structure.</p> <p>Conclusion</p> <p>Enhanced fusogenicity is a phenotype of the A-R5 Envs studied, which was associated with the presence of N362, enhanced HIV-1 entry kinetics and increased CD4bs exposure in gp120. N362 contributes to fusogenicity of R5 Envs in a strain dependent manner. Our studies suggest enhanced fusogenicity of A-R5 Envs may contribute to CD4+ T-cell loss in subjects who progress to AIDS whilst harbouring R5 HIV-1 variants. N362 may contribute to this effect in some individuals.</p
