    Feverish fictions: William T. Vollmann and American literary history after postmodernism

    The rise of the New Sincerity in contemporary American fiction has largely been read on terms provided by a handful of early proponents. This article contends that more complex formulations of the notion are necessary if it is to remain useful as a descriptor for important qualities of recent texts. Among issues in need of greater attention are the implications of the New Sincerity for historical awareness, a topic I pursue via consideration of William T. Vollmann’s grappling with literary history. I argue that Vollmann’s works offer more than the pastiche that allegedly defined historical consciousness in postmodernist fiction. His intertextual engagements with Edgar Allan Poe are especially valuable for modelling some of the ways recent fictions bypass postmodernist ahistoricism in favor of connections to a usable past, and, especially, a usable literary tradition. Readings of Vollmann’s ‘The Grave of Lost Stories’ (in several editions) and ‘The Cemetery of the World’ show how he employs research in the textual archive as a figuration of relations to the past that can serve the ends of renewal and recovery.Accepted manuscrip

    The Sentence Is Most Important: Styles of Engagement in William T. Vollmann’s Fictions

    William T. Vollmann frequently asserts that his ideal reader will appreciate the functionality and beauty of his sentences. This article begins by taking such claims seriously, and draws on both literary and rhetorical stylistics to explore some of the many ways that his texts answer to his intention to find “the right sentence for the right job.” In particular, this article argues that Vollmann’s stylistic decisions are most notable when they most directly satisfy his effort to produce texts that foster empathetic knowledge, serve truth, resist abusive power, and encourage charitable action. Extended close analyses of passages from an early and from a mid-career text (The Rainbow Storiesand Europe Central) illustrate Vollmann’s consistency across two decades of his career regarding choices in the areas of figuration (including schemes and tropes of comparison, repetition, balance, naming, and amplification), grammar, deixis, allusion, and other compositional strategies. Particular attention is paid to passages that display the stylistic mechanisms underlying Vollmann’s negotiation of his texts’ moral qualities, including both the moral content of the worlds represented in the texts, and the moral responsibility the texts bear with regard to their audience. The results of my analyses demonstrate that Vollmann typically prioritizes openness, critique, and dialogue not only in terms of incident and character, but also on the scale of the phrase, clause, and sentence. Ultimately, this article shows how Vollmann’s sentences serve his declared intentions and allow readers to recognize compatibilities between Vollmann’s works and the characteristic features of post-postmodernist writing in general

Impact is produced by the Center for Interdisciplinary Teaching & Learning at the College of General Studies, Boston University (www.bu.edu/cgs/citl).In this issue of Impact you will find a humanities scholar deeply engaged with the arcing out of a new territory: the interdisciplinary study of the Grateful Dead. Impact’s own Christopher Coffman’s review essay should be required reading for scholars of popular music, performance studies and history. Peter Wakefield encourages us to see The Great Gatsby anew, in particular in the context of American racism and White supremacy.     The dynamics of the entanglement for coherent excitonic states in the system of two coupled large semiconductor quantum dots (R/aB1R/a_{B}\gg 1) mediated by a single-mode cavity field is investigated. Maximally entangled coherent excitonic states can be generated by cavity field initially prepared in odd coherent state. The entanglement of the excitonic coherent states between two dots reaches maximum when no photon is detected in the cavity.     Isotopic measurements in water vapor, precipitation, and seawater during EUREC4^4A

    n early 2020, an international team set out to investigate trade-wind cumulus clouds and their coupling to the large-scale circulation through the field campaign EUREC4^4A: ElUcidating the RolE of Clouds-Circulation Coupling in ClimAte. Focused on the western tropical Atlantic near Barbados, EUREC4^4A deployed a number of innovative observational strategies, including a large network of water isotopic measurements collectively known as EUREC4^4A-iso, to study the tropical shallow convective environment. The goal of the isotopic measurements was to elucidate processes that regulate the hydroclimate state – for example, by identifying moisture sources, quantifying mixing between atmospheric layers, characterizing the microphysics that influence the formation and persistence of clouds and precipitation, and providing an extra constraint in the evaluation of numerical simulations. During the field experiment, researchers deployed seven water vapor isotopic analyzers on two aircraft, on three ships, and at the Barbados Cloud Observatory (BCO). Precipitation was collected for isotopic analysis at the BCO and from aboard four ships. In addition, three ships collected seawater for isotopic analysis. All told, the in situ data span the period 5 January–22 February 2020 and cover the approximate area 6 to 16° N and 50 to 60° W, with water vapor isotope ratios measured from a few meters above sea level to the mid-free troposphere and seawater samples spanning the ocean surface to several kilometers depth. This paper describes the full EUREC4^4A isotopic in situ data collection – providing extensive information about sampling strategies and data uncertainties – and also guides readers to complementary remotely sensed water vapor isotope ratios. All field data have been made publicly available even if they are affected by known biases, as is the case for high-altitude aircraft measurements, one of the two BCO ground-based water vapor time series, and select rain and seawater samples from the ships. Publication of these data reflects a desire to promote dialogue around improving water isotope measurement strategies for the future. The remaining, high-quality data create unprecedented opportunities to close water isotopic budgets and evaluate water fluxes and their influence on cloudiness in the trade-wind environment.     Genome modeling system: A knowledge management platform for genomics

    In this work, we present the Genome Modeling System (GMS), an analysis information management system capable of executing automated genome analysis pipelines at a massive scale. The GMS framework provides detailed tracking of samples and data coupled with reliable and repeatable analysis pipelines. The GMS also serves as a platform for bioinformatics development, allowing a large team to collaborate on data analysis, or an individual researcher to leverage the work of others effectively within its data management system. Rather than separating ad-hoc analysis from rigorous, reproducible pipelines, the GMS promotes systematic integration between the two. As a demonstration of the GMS, we performed an integrated analysis of whole genome, exome and transcriptome sequencing data from a breast cancer cell line (HCC1395) and matched lymphoblastoid line (HCC1395BL). These data are available for users to test the software, complete tutorials and develop novel GMS pipeline configurations. The GMS is available at https://github.com/genome/gms

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Brent Wood’s The Tragic Odes of Jerry Garcia and the Grateful Dead: Mystery Dances in the Magic Theater is the latest volume in the Ashgate Popular and Folk Music Series. Like other texts in the series, it undertakes musicological analysis of its subject in the context of interpretations that draw on a wide variety of socio-cultural considerations and scholarly fields. For those who teach with the Grateful Dead, Wood’s text offers much on a number of fronts: its interdisciplinary scope provides fresh insights about the Grateful Dead in relation to topics in literary studies, classics, philosophy, musicology, and history, among other disciplines..

    Transcranial Direct Current Stimulation’s Effect on Novice versus Experienced Learning

    Transcranial direct current stimulation (TDCS) is a non-invasive form of brain stimulation applied via a weak electrical current passed between electrodes on the scalp. In recent studies, TDCS has been shown to improve learning when applied to the prefrontal cortex (e.g., Kincses et al. in Neuropsychologia 42:113–117, 2003; Clark et al. Neuroimage in 2010). The present study examined the effects of TDCS delivered at the beginning of training (novice) or after an hour of training (experienced) on participants’ ability to detect cues indicative of covert threats. Participants completed two 1-h training sessions. During the first 30 min of each training session, either 0.1 mA or 2.0 mA of anodal TDCS was delivered to the participant. The anode was positioned near F8, and the cathode was placed on the upper left arm. Testing trials immediately followed training. Accuracy in classification of images containing and not-containing threat stimuli during the testing sessions indicated: (1) that mastery of threat detection significantly increased with training, (2) that anodal TDCS at 2 mA significantly enhanced learning, and (3) TDCS was significantly more effective in enhancing test performance when applied in novice learners than in experienced learners.
