    Reliable microsatellite genotyping of the Eurasian badger (Meles meles) using faecal DNA

    The potential link between badgers and bovine tuberculosis has made it vital to develop accurate techniques to census badgers. Here we investigate the potential of using genetic profiles obtained from faecal DNA as a basis for population size estimation. After trialling several methods we obtained a high amplification success rate (89%) by storing faeces in 70% ethanol and using the guanidine thiocyanate/silica method for extraction. Using 70% ethanol as a storage agent had the advantage of it being an antiseptic. In order to obtain reliable genotypes with fewer amplification reactions than the standard multiple-tubes approach, we devised a comparative approach in which genetic profiles were compared and replication directed at similar, but not identical, genotypes. This modified method achieved a reduction in polymerase chain reactions comparable with the maximumlikelihood model when just using reliability criteria, and was slightly better when using reliability criteria with the additional proviso that alleles must be observed twice to be considered reliable. Our comparative approach would be best suited for studies that include multiple faeces from each individual. We utilized our approach in a well-studied population of badgers from which individuals had been sampled and reliable genotypes obtained. In a study of 53 faeces sampled from three social groups over 10 days, we found that direct enumeration could not be used to estimate population size, but that the application of mark–recapture models has the potential to provide more accurate results

    Metabolic syndrome among individuals living with hypertension in Accra, Ghana

    Metabolic syndrome (MetS) is a major risk factor to cardiovascular diseases. In this study, we investigate the prevalence and associated risky behaviour of MetS in resource-poor urban communities in Accra, Ghana. We analysed data on 111 persons with hypertension, screened and recruited for a therapeutic lifestyle intervention program in August 2015. MetS was measured using the International Diabetes Federation (IDF) and the World Health Organization definitions. The prevalence of MetS was 58.4% and 16.8% by the IDF and WHO definitions respectively. More women (61.8%) compared to men (31.8%) had MetS (p = 0.011). Approximately 31% of the hypertensive patients were engaged in moderate-intensity physical activity; 9.0% were current smokers, 42.0% consumed excess alcohol over the past month prior to the interview and 41.0% discontinued taking their antihypertensive medications without consulting with a doctor. About 42.0% and 37.0% of respondents always consumed fruits and vegetables respectively at least two times a day. The binary logistic regression showed that compared to women, men had lower odds of consuming two or more servings of vegetable in a day (OR: 0.2; 95% CI; 0.1, 0.8). Increase in age was associated with higher odds of consuming fruits at least twice a day (1.0; 1.0, 1.1) but with lower odds of engaging in moderate intensity physical activity (0.9; 0.8, 1.0). Being married was associated with higher odds of engaging in moderate physical activity (2.8; 1.0, 8.2). Therapeutic methods essential for the management of patients with hypertension and MetS should include non-pharmacological remedies targeting the promotion of medication adherence, Dietary Approaches to Stop Hypertension (DASH) and physical activities; these are vital to changing unhealthy lifestyle which worsens the underlying pathology

    Probing sporadic and familial Alzheimer's disease using induced pluripotent stem cells.

    Our understanding of Alzheimer's disease pathogenesis is currently limited by difficulties in obtaining live neurons from patients and the inability to model the sporadic form of the disease. It may be possible to overcome these challenges by reprogramming primary cells from patients into induced pluripotent stem cells (iPSCs). Here we reprogrammed primary fibroblasts from two patients with familial Alzheimer's disease, both caused by a duplication of the amyloid-β precursor protein gene (APP; termed APP(Dp)), two with sporadic Alzheimer's disease (termed sAD1, sAD2) and two non-demented control individuals into iPSC lines. Neurons from differentiated cultures were purified with fluorescence-activated cell sorting and characterized. Purified cultures contained more than 90% neurons, clustered with fetal brain messenger RNA samples by microarray criteria, and could form functional synaptic contacts. Virtually all cells exhibited normal electrophysiological activity. Relative to controls, iPSC-derived, purified neurons from the two APP(Dp) patients and patient sAD2 exhibited significantly higher levels of the pathological markers amyloid-β(1-40), phospho-tau(Thr 231) and active glycogen synthase kinase-3β (aGSK-3β). Neurons from APP(Dp) and sAD2 patients also accumulated large RAB5-positive early endosomes compared to controls. Treatment of purified neurons with β-secretase inhibitors, but not γ-secretase inhibitors, caused significant reductions in phospho-Tau(Thr 231) and aGSK-3β levels. These results suggest a direct relationship between APP proteolytic processing, but not amyloid-β, in GSK-3β activation and tau phosphorylation in human neurons. Additionally, we observed that neurons with the genome of one sAD patient exhibited the phenotypes seen in familial Alzheimer's disease samples. More generally, we demonstrate that iPSC technology can be used to observe phenotypes relevant to Alzheimer's disease, even though it can take decades for overt disease to manifest in patients

    PENDAMPINGAN BLENDED LEARNING BERPUSAT PADA KEMAMPUAN LITERASI DAN NUMERASI BAGI GURU SD

    The problems faced by two private schools that have almost the same characteristics, namely SDK Girisonta and SDK Genuk, are implementing distance learning and limited face-to-face learning during the COVID-19 pandemic and preparing AKM (minimum competency assessment) including literacy and numeracy skills. The solution to overcome these problems is blended learning training and mentoring centered on literacy and numeracy skills. The method of implementing the Participatory Action and Learning System method includes awareness, capacity building, mentoring and institutionalization. Teachers' knowledge of blended learning, literacy and numeracy reaches 78 in the good category so they are ready to plan and implement. Learning tools with blended learning and focusing on literacy and numeracy, teachers design and implement them in the classroom during distance learning. Reflection on the results of implementation assistance shows that it is very successful in blended learning (92%), successful in focusing on literacy (81%) and less successful in focusing on numeracy (47%). The suggestion from this community service activity is that the success of blended learning is influenced by the readiness of schools, teachers, parents and students, so good communication and awareness of all parties are needed. Through a variety of electronic learning resources and online games that focus on literacy and numeracy, students accompanied by teachers and parents can learn optimally to develop their potential.  Permasalahan yang dihadapi dua sekolah swasta dan memiliki karakteristik hampir sama, yaitu SDK Girisonta dan SDK Genuk adalah pelaksanana pembelajaran jarak jauh dan tatap muka terbatas di masa pendemi covid-19 dan mempersiapkan AKM (aseesment kompetensi minimum) meliputi kemampuan literasi dan numerasi.  Solusi untuk mengatasi permasalah tersebut adalah pelatihan dan pendampingan blended learning berpusat pada kemampuan literasi dan numerasi. Metode pelaksanaan metode Parcipatory Action and Learnig System meliputi penyadaran, pengkapasitasan, pendampingan dan pelembagaan. Pengetahuan guru terhadap blended learning, literasi dan numerasi mencapai 78 masuk pada kategori baik sehingga siap untuk membuat perencanaan dan mengimplementasikan. Perangkat pembelajaran dengan blended learning dan berfokus pada literasi dan numerasi, para guru merancang dan mengimpelementasikannya di kelas pada saat pembelajaran jarak jauh. Refleksi hasil pendampingan implementasi menunjukkan sangat berhasil pada blended learning (92%), berhasil untuk focus literasi (81%) dan kurang berhasil untuk focus numerasi (47%). Saran dari kegiatan pengabdian ini adalah keberhasilan blended learning dipengaruhi oleh kesiapan sekolah, guru, orang tua dan siswa, maka diperlukan komunikasi yang baik dan kesadaran semua pihak. Melalui variasi sumber belajar eletronik dan permainan online yang berfokus pada literasi dan numerasi, siswa didampingi guru dan orang tua dapat belajar dengan optimal untuk mengembangkan potensinya

    Transit Photometry as an Exoplanet Discovery Method

    Photometry with the transit method has arguably been the most successful exoplanet discovery method to date. A short overview about the rise of that method to its present status is given. The method's strength is the rich set of parameters that can be obtained from transiting planets, in particular in combination with radial velocity observations; the basic principles of these parameters are given. The method has however also drawbacks, which are the low probability that transits appear in randomly oriented planet systems, and the presence of astrophysical phenomena that may mimic transits and give rise to false detection positives. In the second part we outline the main factors that determine the design of transit surveys, such as the size of the survey sample, the temporal coverage, the detection precision, the sample brightness and the methods to extract transit events from observed light curves. Lastly, an overview over past, current and future transit surveys is given. For these surveys we indicate their basic instrument configuration and their planet catch, including the ranges of planet sizes and stellar magnitudes that were encountered. Current and future transit detection experiments concentrate primarily on bright or special targets, and we expect that the transit method remains a principal driver of exoplanet science, through new discoveries to be made and through the development of new generations of instruments.Comment: Review chapte

    Pneumococcal carriage in sub-Saharan Africa--a systematic review.

    BACKGROUND: Pneumococcal epidemiology varies geographically and few data are available from the African continent. We assess pneumococcal carriage from studies conducted in sub-Saharan Africa (sSA) before and after the pneumococcal conjugate vaccine (PCV) era. METHODS: A search for pneumococcal carriage studies published before 2012 was conducted to describe carriage in sSA. The review also describes pneumococcal serotypes and assesses the impact of vaccination on carriage in this region. RESULTS: Fifty-seven studies were included in this review with the majority (40.3%) from South Africa. There was considerable variability in the prevalence of carriage between studies (I-squared statistic = 99%). Carriage was higher in children and decreased with increasing age, 63.2% (95% CI: 55.6-70.8) in children less than 5 years, 42.6% (95% CI: 29.9-55.4) in children 5-15 years and 28.0% (95% CI: 19.0-37.0) in adults older than 15 years. There was no difference in the prevalence of carriage between males and females in 9/11 studies. Serotypes 19F, 6B, 6A, 14 and 23F were the five most common isolates. A meta-analysis of four randomized trials of PCV vaccination in children aged 9-24 months showed that carriage of vaccine type (VT) serotypes decreased with PCV vaccination; however, overall carriage remained the same because of a concomitant increase in non-vaccine type (NVT) serotypes. CONCLUSION: Pneumococcal carriage is generally high in the African continent, particularly in young children. The five most common serotypes in sSA are among the top seven serotypes that cause invasive pneumococcal disease in children globally. These serotypes are covered by the two PCVs recommended for routine childhood immunization by the WHO. The distribution of serotypes found in the nasopharynx is altered by PCV vaccination
