    The effect of credibility assessment techniques on consistency and subsequent memory for the truth

    Repeated interviews are common during an investigation, and perceived consistency between multiple statements is associated with an interviewee’s credibility. Furthermore, research has shown that the act of lying can affect a person’s memory for what truthfully occurred. The current study assessed the influence of lying on memory during initial and repeated interviews, as well as how an interviewer’s approach might affect between-statement consistency for true and false statements. Participants performed a scavenger hunt at two sets of buildings on a university campus and then were either dismissed or interviewed (with a Reverse Order instruction or a Structured Interview) about their activities. Participants chose one set to tell the truth about and then created a lie about activities in another area of campus that had not been visited. One week later, all participants provided a second free recall statement about their activities during the scavenger hunt, and then a final truthful description of both areas that were visited during the scavenger hunt. Truthfully rehearsed experiences were associated with more accurate recall of information learned during the scavenger hunt as well as more consistent and more detailed statements. The Structured Interview led to initially more detailed statements, but more inconsistencies in the form of omissions

    On the Nature of Black Holes in Loop Quantum Gravity

    A genuine notion of black holes can only be obtained in the fundamental framework of quantum gravity resolving the curvature singularities and giving an account of the statistical mechanical, microscopic degrees of freedom able to explain the black hole thermodynamical properties. As for all quantum systems, a quantum realization of black holes requires an operator algebra of the fundamental observables of the theory which is introduced in this study based on aspects of loop quantum gravity. From the eigenvalue spectra of the quantum operators for the black hole area, charge and angular momentum, it is demonstrated that a strict bound on the extensive parameters, different from the relation arising in classical general relativity, holds, implying that the extremal black hole state can neither be measured nor can its existence be proven. This is, as turns out, a result of the specific form of the chosen angular momentum operator and the corresponding eigenvalue spectrum, or rather the quantum measurement process of angular momentum. Quantum mechanical considerations and the lowest, non-zero eigenvalue of the loop quantum gravity black hole mass spectrum indicate, on the one hand, a physical Planck scale cutoff of the Hawking temperature law and, on the other hand, give upper and lower bounds on the numerical value of the Immirzi parameter. This analysis provides an approximative description of the behavior and the nature of quantum black holes

    Automation of large scale transient protein expression in mammalian cells

    Traditional mammalian expression systems rely on the time-consuming generation of stable cell lines; this is difficult to accommodate within a modern structural biology pipeline. Transient transfections are a fast, cost-effective solution, but require skilled cell culture scientists, making man-power a limiting factor in a setting where numerous samples are processed in parallel. Here we report a strategy employing a customised CompacT SelecT cell culture robot allowing the large-scale expression of multiple protein constructs in a transient format. Successful protocols have been designed for automated transient transfection of human embryonic kidney (HEK) 293T and 293S GnTI⁻ cells in various flask formats. Protein yields obtained by this method were similar to those produced manually, with the added benefit of reproducibility, regardless of user. Automation of cell maintenance and transient transfection allows the expression of high quality recombinant protein in a completely sterile environment with limited support from a cell culture scientist. The reduction in human input has the added benefit of enabling continuous cell maintenance and protein production, features of particular importance to structural biology laboratories, which typically use large quantities of pure recombinant proteins, and often require rapid characterisation of a series of modified constructs. This automated method for large scale transient transfection is now offered as a Europe-wide service via the P-cube initiative

    Protein trafficking through the endosomal system prepares intracellular parasites for a home invasion

    Toxoplasma (toxoplasmosis) and Plasmodium (malaria) use unique secretory organelles for migration, cell invasion, manipulation of host cell functions, and cell egress. In particular, the apical secretory micronemes and rhoptries of apicomplexan parasites are essential for successful host infection. New findings reveal that the contents of these organelles, which are transported through the endoplasmic reticulum (ER) and Golgi, also require the parasite endosome-like system to access their respective organelles. In this review, we discuss recent findings that demonstrate that these parasites reduced their endosomal system and modified classical regulators of this pathway for the biogenesis of apical organelles

    Follow-up of the GHSG HD16 trial of PET-guided treatment in early-stage favorable Hodgkin lymphoma.

    The primary analysis of the GHSG HD16 trial indicated a significant loss of tumor control with PET-guided omission of radiotherapy (RT) in patients with early-stage favorable Hodgkin lymphoma (HL). This analysis reports long-term outcomes. Overall, 1150 patients aged 18-75 years with newly diagnosed early-stage favorable HL were randomized between standard combined-modality treatment (CMT) (2x ABVD followed by PET/CT [PET-2] and 20 Gy involved-field RT) and PET-2-guided treatment omitting RT in case of PET-2 negativity (Deauville score [DS] < 3). The study aimed at excluding inferiority of PET-2-guided treatment and assessing the prognostic impact of PET-2 in patients receiving CMT. At a median follow-up of 64 months, PET-2-negative patients had a 5-year progression-free survival (PFS) of 94.2% after CMT (n = 328) and 86.7% after ABVD alone (n = 300; HR = 2.05 [1.20-3.51]; p = 0.0072). 5-year OS was 98.3% and 98.8%, respectively (p = 0.14); 4/12 documented deaths were caused by second primary malignancies and only one by HL. Among patients assigned to CMT, 5-year PFS was better in PET-2-negative (n = 353; 94.0%) than in PET-2-positive patients (n = 340; 90.3%; p = 0.012). The difference was more pronounced when using DS4 as cut-off (DS 1-3: n = 571; 94.0% vs. DS ≥ 4: n = 122; 83.6%; p < 0.0001). Taken together, CMT should be considered standard treatment for early-stage favorable HL irrespective of the PET-2-result

    Lexical access speed and the development of phonological recoding during immediate serial recall

    A recent Registered Replication Report (RRR) of the development of verbal rehearsal during serial recall revealed that children verbalized at younger ages than previously thought, but did not identify sources of individual differences. Here, we use mediation analysis to reanalyze data from the 934 children ranging from 5 to 10 years old from the RRR for that purpose. From ages 5 to 7, the time taken for a child to label pictures (i.e. isolated naming speed) predicted the child’s spontaneous use of labels during a visually presented serial reconstruction task, despite no need for spoken responses. For 6- and 7-year-olds, isolated naming speed also predicted recall. The degree to which verbalization mediated the relation between isolated naming speed and recall changed across development. All relations dissipated by age 10. The same general pattern was observed in an exploratory analysis of delayed recall for which greater demands are placed on rehearsal for item maintenance. Overall, our findings suggest that spontaneous phonological recoding during a standard short-term memory task emerges around age 5, increases in efficiency during the early elementary school years, and is sufficiently automatic by age 10 to support immediate serial recall in most children. Moreover, the findings highlight the need to distinguish between phonological recoding and rehearsal in developmental studies of short-term memory

    Effectiveness of remote care interventions: A systematic review informing the 2022 EULAR Points to Consider for remote care in rheumatic and musculoskeletal diseases

    Objective: To perform a systematic literature review (SLR) on different outcomes of remote care compared with face-to-face (F2F) care, its implementation into clinical practice and to identify drivers and barriers in order to inform a task force formulating the EULAR Points to Consider for remote care in rheumatic and musculoskeletal diseases (RMDs).  Methods: A search strategy was developed and run in Medline (PubMed), Embase and Cochrane Library. Two reviewers independently performed standardised data extraction, synthesis and risk of bias (RoB) assessment.  Results: A total of 2240 references were identified. Forty-seven of them fulfilled the inclusion criteria. Remote monitoring (n=35) was most frequently studied, with telephone/video calls being the most common mode of delivery (n=30). Of the 34 studies investigating outcomes of remote care, the majority addressed efficacy and user perception; 34% and 21% of them, respectively, reported a superiority of remote care as compared with F2F care. Time and cost savings were reported as major benefits, technical aspects as major drawback in the 13 studies that investigated drivers and barriers of remote care. No study addressed remote care implementation. The main limitation of the studies identified was the heterogeneity of outcomes and methods, as well as a substantial RoB (50% of studies with high RoB).  Conclusions: Remote care leads to similar or better results compared with F2F treatment concerning efficacy, safety, adherence and user perception outcomes, with the limitation of heterogeneity and considerable RoB of the available studies
