An Assay Method for Determining Extra-Cellular Lipases from Pseudomonas aeruginosa
The applicability of an isotopically labelled assay system to determine the lipase production in Pseudomonas aeruginosa was evaluated. Supernatant from cultures of Pseudomonas aeruginosa grown in a medium containing olive oil was incubated with a substrate containing labelled trioleate. Fatty acids were isolated by means of a liquid-liquid partition system. Enzyme activity was determined by measuring the amounts of free fatty acid by liquid scintillation counting. Findings indicate that the isotopicallylabelled, liquid-liquid partitioning assay is reliable, sensitive and adaptable to rapid assay conditions. It was also determined that different strains of Pseudomonas aeruginosa produce varying amounts of lipase. Partial purification of supernatant by gel filtration produced two protein peaks showing enzymatic activity
Isotopic Tracking of Hanford 300 Area Derived Uranium in the Columbia River
Our objectives in this study are to quantify the discharge rate of uranium (U) to the Columbia River from the Hanford Site's 300 Area, and to follow that U down river to constrain its fate. Uranium from the Hanford Site has variable isotopic composition due to nuclear industrial processes carried out at the site. This characteristic makes it possible to use high-precision isotopic measurements of U in environmental samples to identify even trace levels of contaminant U, determine its sources, and estimate discharge rates. Our data on river water samples indicate that as much as 3.2 kg/day can enter the Columbia River from the 300 Area, which is only a small fraction of the total load of dissolved natural background U carried by the Columbia River. This very low-level of Hanford derived U can be discerned, despite dilution to < 1 percent of natural background U, 350 km downstream from the Hanford Site. These results indicate that isotopic methods can allow the amounts of U from the 300 Area of the Hanford Site entering the Columbia River to be measured accurately to ascertain whether they are an environmental concern, or are insignificant relative to natural uranium background in the Columbia River
Positivity of Spin Foam Amplitudes
The amplitude for a spin foam in the Barrett-Crane model of Riemannian
quantum gravity is given as a product over its vertices, edges and faces, with
one factor of the Riemannian 10j symbols appearing for each vertex, and simpler
factors for the edges and faces. We prove that these amplitudes are always
nonnegative for closed spin foams. As a corollary, all open spin foams going
between a fixed pair of spin networks have real amplitudes of the same sign.
This means one can use the Metropolis algorithm to compute expectation values
of observables in the Riemannian Barrett-Crane model, as in statistical
mechanics, even though this theory is based on a real-time (e^{iS}) rather than
imaginary-time (e^{-S}) path integral. Our proof uses the fact that when the
Riemannian 10j symbols are nonzero, their sign is positive or negative
depending on whether the sum of the ten spins is an integer or half-integer.
For the product of 10j symbols appearing in the amplitude for a closed spin
foam, these signs cancel. We conclude with some numerical evidence suggesting
that the Lorentzian 10j symbols are always nonnegative, which would imply

Helioseismological Implications of Recent Solar Abundance Determinations
We show that standard solar models are in good agreement with the
helioseismologically determined sound speed and density as a function of solar
radius, the depth of the convective zone, and the surface helium abundance, as
long as those models do not incorporate the most recent heavy element abundance
determinations. However, sophisticated new analyses of the solar atmosphere
infer lower abundances of the lighter metals (like C, N, O, Ne, and Ar) than
the previously widely used surface abundances. We show that solar models that
include the lower heavy element abundances disagree with the solar profiles of
sound speed and density as well as the depth of the convective zone and the
helium abundance. The disagreements for models with the new abundances range
from factors of several to many times the quoted uncertainties in the
helioseismological measurements. The disagreements are at temperatures below
what is required for solar interior fusion reactions and therefore do not
significantly affect solar neutrino emission. If errors in thecalculated OPAL
opacities are solely responsible for the disagreements, then the corrections in
the opacity must extend from 2 times 10^6 K (R = 0.7R_Sun)to 5 times 10^6 K (R

Identification of polymorphisms and balancing selection in the male infertility candidate gene, ornithine decarboxylase antizyme 3
Abstract Background The antizyme family is a group of small proteins that play a role in cell growth and division by regulating the biosynthesis of polyamines (putrescine, spermidine, spermine). Antizymes regulate polyamine levels primarily through binding ornithine decarboxylase (ODC), an enzyme key to polyamine production, and targeting ODC for destruction by the 26S proteosome. Ornithine decarboxylase antizyme 3 (OAZ3) is a testis-specific antizyme paralog and the only antizyme expressed in the mid to late stages of spermatogenesis. Methods To see if mutations in the OAZ3 gene are responsible for some cases of male infertility, we sequenced and evaluated the genomic DNA of 192 infertile men, 48 men of known paternity, and 34 African aborigines from the Mbuti tribe in the Democratic Republic of the Congo. The coding sequence of OAZ3 was further screened for polymorphisms by SSCP analysis in the infertile group and an additional 250 general population controls. Identified polymorphisms in the OAZ3 gene were further subjected to a haplotype analysis using PHASE 2.02 and Arlequin 2.0 software programs. Results A total of 23 polymorphisms were identified in the promoter, exons or intronic regions of OAZ3. The majority of these fell within a region of less than two kilobases. Two of the polymorphisms, -239 A/G in the promoter and 4280 C/T, a missense polymorphism in exon 5, may show evidence of association with male infertility. Haplotype analysis identified 15 different haplotypes, which can be separated into two divergent clusters. Conclusion Mutations in the OAZ3 gene are not a common cause of male infertility. However, the presence of the two divergent haplotypes at high frequencies in all three of our subsamples (infertile, control, African) suggests that they have been maintained in the genome by balancing selection, which was supported by a test of Tajima's D statistic. Evidence for natural selection in this region implies that these haplotypes may be associated with a trait other than infertility. This trait may be related to another function of OAZ3 or a region in tight linkage disequilibrium to the gene.</p
Uranium isotope fractionation by abiotic reductive precipitation
Significant uranium (U) isotope fractionation has been observed during abiotic reduction of aqueous U, counter to the expectation that uranium isotopes are only fractionated by bioassociated enzymatic reduction. In our experiments, aqueous U is removed from solution by reductive precipitation onto the surfaces of synthetic iron monosulfide. The magnitude of uranium isotopic fractionation increases with decreasing aqueous U removal rate and with increasing amounts of neutrally charged aqueous Ca-U-CO3 species. Our discovery means that abiotic U isotope fractionation likely occurs in any reducing environment with aqueous Ca ≥ 1 mM, and that the magnitude of isotopic fractionation changes in response to changes in aqueous major ion concentrations that affect U speciation. Spin Foam Models of Riemannian Quantum Gravity
Using numerical calculations, we compare three versions of the Barrett-Crane
model of 4-dimensional Riemannian quantum gravity. In the version with face and
edge amplitudes as described by De Pietri, Freidel, Krasnov, and Rovelli, we
show the partition function diverges very rapidly for many triangulated
4-manifolds. In the version with modified face and edge amplitudes due to Perez
and Rovelli, we show the partition function converges so rapidly that the sum
is dominated by spin foams where all the spins labelling faces are zero except
for small, widely separated islands of higher spin. We also describe a new
version which appears to have a convergent partition function without drastic
spin-zero dominance. Finally, after a general discussion of how to extract
physics from spin foam models, we discuss the implications of convergence or

One-loop Quantum Gravity in Schwarzschild Spacetime
The quantum theory of linearized perturbations of the gravitational field of
a Schwarzschild black hole is presented. The fundamental operators are seen to
be the perturbed Weyl scalars and associated with the
Newman-Penrose description of the classical theory. Formulae are obtained for
the expectation values of the modulus squared of these operators in the
Boulware, Unruh and Hartle-Hawking quantum states. Differences between the
