    Vertex-primitive groups and graphs of order twice the product of two distinct odd primes

    A non-Cayley number is an integer n for which there exists a vertex-transitive graph on n vertices which is not a Cayley graph. In this paper, we complete the determination of the non-Cayley numbers of the form 2pq, where p, q are distinct odd primes. Earlier work of Miller and the second author had dealt with all such numbers corresponding to vertex-transitive graphs admitting an imprimitive subgroup of automorphisms. This paper deals with the primitive case. First the primitive permutation groups of degree 2pq are classified. This depends on the finite simple group classification. Then each of these groups G is examined to determine whether there are any non-Cayley graphs which admit G as a vertex-primitive subgroup of automorphisms, and admit no imprimitive subgroups.     Biochemical characterization of predicted Precambrian RuBisCO

    The antiquity and global abundance of the enzyme, RuBisCO, attests to the crucial and longstanding role it has played in the biogeochemical cycles of Earth over billions of years. The counterproductive oxygenase activity of RuBisCO has persisted over billions of years of evolution, despite its competition with the carboxylase activity necessary for carbon fixation, yet hypotheses regarding the selective pressures governing RuBisCO evolution have been limited to speculation. Here we report the resurrection and biochemical characterization of ancestral RuBisCOs, dating back to over one billion years ago (Gyr ago). Our findings provide an ancient point of reference revealing divergent evolutionary paths taken by eukaryotic homologues towards improved specificity for CO2, versus the evolutionary emphasis on increased rates of carboxylation observed in bacterial homologues. Consistent with these distinctions, in vivo analysis reveals the propensity of ancestral RuBisCO to be encapsulated into modern-day carboxysomes, bacterial organelles central to the cyanobacterial CO2 concentrating mechanism

Through attention to tracing journeys, located moments and listening to the presence of ghosts (Ogborn, 2005), this report seeks to highlight the range of different ways in which historical geographers have explored lives, deaths, and their transient traces through varied biographical terrains. Continuing to draw attention in historical geography to the darkest of histories, this piece will pivot on moments of discovering the dead to showcase the nuanced ways in which historical geography is opening doors into uncharted lives and unspoken histories

    Estimates of volcanic mercury emissions from Redoubt Volcano, Augustine Volcano, and Mount Spurr eruption ash

    Ash is a potential sink of volcanically sourced atmospheric mercury (Hg), and the concentration of particle-bound Hg may provide constraints on Hg emissions during eruptions. We analyze Hg concentrations in 227 bulk ash samples from the Mount Spurr (1992), Redoubt Volcano (2009), and Augustine Volcano (2006) volcanic eruptions to investigate large-scale spatial, temporal, and volcanic-source trends. We find no significant difference in Hg concentrations in bulk ash by distance or discrete eruptive events at each volcano, suggesting that in-plume reactions converting gaseous Hg0 to adsorbed Hg2+ are happening on shorter timescales than considered in this study (minutes) and any additional in-plume controls are not discernable within intra-volcanic sample variability. However, we do find a significant difference in Hg concentration of ash among volcanic sources, which indicates that volcanoes may emit comparatively high or low quantities of Hg. We combine our Hg findings with total mass estimates of ashfall deposits to calculate minimum, first-order Hg emissions of 8.23 t Hg for Mount Spurr (1992), 1.25 t Hg for Redoubt Volcano (2009), and 0.16 t Hg for Augustine Volcano (2006). In particular, we find that Mount Spurr is a high Hg emitting volcano, and that its 1992 particulate Hg emissions likely contributed substantially to the global eruptive volcanic Hg budget for that year. Based on our findings, previous approaches that use long-term Hg/SO2 mass ratios to estimate eruptive total Hg under-account for Hg emitted in explosive events, and global volcanogenic Total Hg estimates need revisiting

    Amphibian and reptile diversity along a ridge-to-reef elevational gradient on a small isolated oceanic island of the central Philippines

    Despite multiple recent field studies, herpetological species diversity of the Romblon Island Group in the central Philippines—particularly Sibuyan Island—has remained underestimated. Recently, we investigated the diversity of the herpetofauna of Mount Guiting-Guiting Natural Park, based on an elevational transect (10–1557 m a.s.l.). Our surveys resulted in a total of 47 species of amphibians and reptiles, including 14 new island records and one atypical occurrence of a snake species recorded for the first time from a high elevation (939 m a.s.l). These new records constitute a notable increase (21%) in Sibuyan’s herpetological species diversity as compared to surveys from a decade ago. We also provide updates of the taxonomy and identification of species endemic to this island (e.g., members of the genera Platymantis Günther, 1858, Brachymeles Duméril & Gibron, 1839, and Pseudogekko Taylor, 1922), and discuss the importance of continued surveys and field-derived data to inform conservation status assessments of Sibuyan’s unique assemblage of amphibians and reptiles

    Altered Lipidome Composition Is Related to Markers of Monocyte and Immune Activation in Antiretroviral Therapy Treated Human Immunodeficiency Virus (HIV) Infection and in Uninfected Persons

    Background: HIV infection and antiretroviral therapy (ART) have both been linked to dyslipidemia and increased cardiovascular disease (CVD) risk. Alterations in the composition of saturated (SaFA), monounsaturated (MUFA), and polyunsaturated (PUFA) fatty acids are related to inflammation and CVD progression in HIV-uninfected (HIV–) populations. The relationships among the lipidome and markers of monocyte and immune activation in HIV-infected (HIV+) individuals are not well understood.Methods: Concentrations of serum lipids and their fatty acid composition were measured by direct infusion-tandem mass spectrometry in samples from 20 ART-treated HIV+ individuals and 20 HIV– individuals.Results: HIV+ individuals had increased levels of free fatty acids (FFAs) with enrichment of SaFAs, including palmitic acid (16:0) and stearic acid (18:0), and these levels were directly associated with markers of monocyte (CD40, HLA-DR, TLR4, CD36) and serum inflammation (LBP, CRP). PUFA levels were reduced significantly in HIV+ individuals, and many individual PUFA species levels were inversely related to markers of monocyte activation, such as tissue factor, TLR4, CD69, and SR-A. Also in HIV+ individuals, the composition of lysophosphatidylcholine (LPC) was enriched for SaFAs; LPC species containing SaFAs were directly associated with IL-6 levels and monocyte activation. We similarly observed direct relationships between levels of SaFAs and inflammation in HIV uninfected individuals. Further, SaFA exposure altered monocyte subset phenotypes and inflammatory cytokine production in vitro.Conclusions: The lipidome is altered in ART-treated HIV infection, and may contribute to inflammation and CVD progression. Detailed lipidomic analyses may better assess CVD risk in both HIV+ and HIV– individuals than does traditional lipid profiling

    influenza monitoring in sardinia italy identifies h3 subtype in mediterranean wild migratory birds

    Introduction: Wild migratory birds are global distributors of pathogens. Sardinia, Italy, is the second largest Island in the Mediterranean and is a land bridge between Europe and Africa. Methodology: We designed a surveillance protocol to investigate wild migratory birds for presence, frequency, and type of avian influenza viruses. We collected over 4,000 avian samples and compared three sampling methods, fecal, cloacal, and tracheal, to determine the most productive for virus identification. To determine frequency of infection, RNA was extracted and RT-PCRs for avian influenza virus genes were run. Positive samples were cultivated for live virus, sub typed and sequenced. Results: Forty-four samples were positive for influenza nucleoprotein gene. We identified two previously unidentified H3 subtype strains and found cloacae to have the highest rate of virus identification and fecal sampling to provide quality RNA and repeatable results for determination of virus presence. Conclusion: Our investigation provides information on the frequency of Mediterranean avian influenza viruses, and validates the initiation of an avian influenza surveillance protocol. Taken together with global avian influenza findings, these results give insight into infectious disease distributions which is important for viral pandemic monitoring and design of preventative measures

    Results of Antiretroviral Treatment Interruption and Intensification in Advanced Multi-Drug Resistant HIV Infection from the OPTIMA Trial

    BACKGROUND: Guidance is needed on best medical management for advanced HIV disease with multidrug resistance (MDR) and limited retreatment options. We assessed two novel antiretroviral (ARV) treatment approaches in this setting. METHODS AND FINDINGS: We conducted a 2×2 factorial randomized open label controlled trial in patients with a CD4 count≤300 cells/µl who had ARV treatment (ART) failure requiring retreatment, to two options (a) re-treatment with either standard (≤4 ARVs) or intensive (≥5 ARVs) ART and b) either treatment starting immediately or after a 12-week monitored ART interruption. Primary outcome was time to developing a first AIDS-defining event (ADE) or death from any cause. Analysis was by intention to treat. From 2001 to 2006, 368 patients were randomized. At baseline, mean age was 48 years, 2% were women, median CD4 count was 106/µl, mean viral load was 4.74 log(10) copies/ml, and 59% had a prior AIDS diagnosis. Median follow-up was 4.0 years in 1249 person-years of observation. There were no statistically significant differences in the primary composite outcome of ADE or death between re-treatment options of standard versus intensive ART (hazard ratio 1.17; CI 0.86-1.59), or between immediate retreatment initiation versus interruption before re-treatment (hazard ratio 0.93; CI 0.68-1.30), or in the rate of non-HIV associated serious adverse events between re-treatment options. CONCLUSIONS: We did not observe clinical benefit or harm assessed by the primary outcome in this largest and longest trial exploring both ART interruption and intensification in advanced MDR HIV infection with poor retreatment options.     The Glycan Shield of HIV Is Predominantly Oligomannose Independently of Production System or Viral Clade

    The N-linked oligomannose glycans of HIV gp120 are a target for both microbicide and vaccine design. The extent of cross-clade conservation of HIV oligomannose glycans is therefore a critical consideration for the development of HIV prophylaxes. We measured the oligomannose content of virion-associated gp120 from primary virus from PBMCs for a range of viral isolates and showed cross-clade elevation (62–79%) of these glycans relative to recombinant, monomeric gp120 (∼30%). We also confirmed that pseudoviral production systems can give rise to notably elevated gp120 oligomannose levels (∼98%), compared to gp120 derived from a single-plasmid viral system using the HIVLAI backbone (56%).     Mass Mortality of Adult Male Subantarctic Fur Seals: Are Alien Mice the Culprits?

    Background: Mass mortalities of marine mammals due to infectious agents are increasingly reported. However, in contrast to previous die-offs, which were indiscriminate with respect to sex and age, here we report a land-based mass mortality of Subantarctic fur seals with apparent exclusivity to adult males. An infectious agent with a male-predilection is the most plausible explanation for this die-off. Although pathogens with gender-biased transmission and pathologies are unusual, rodents are known sources of male-biased infectious agents and the invasive Mus musculus house mouse, occurs in seal rookeries. Methodology / Principal Findings: Molecular screening for male-biased pathogens in this potential rodent reservoir host revealed the absence of Cardiovirus and Leptospirosis genomes in heart and kidney samples, respectively, but identified a novel Streptococcus species with 30 % prevalence in mouse kidneys. Conclusions / Significance: Inter-species transmission through environmental contamination with this novel bacterium, whose congenerics display male-bias and have links to infirmity in seals and terrestrial mammals (including humans)
