    Assessing the current status of Hexanchus griseus in the Mediterranean Sea using local ecological knowledge

    Fishermen from 9 countries distributed throughout the Mediterranean Sea were interviewed between May and December 2019 with the aim of compiling information about the current impact of fisheries on a large deepwatershark species, the bluntnose sixgill shark (Hexanchus griseus). A total of 382 professional fishermenbelonging to 6 different gears (bottom trawling, bottom longline, drifting longline, trammel nets, gillnets andpolyvalent) took part in the study. Bottom trawlers were the most interviewed fishermen (n = 148) and the bestfleet coverage was obtained for bottom longline (38.89%). Results showed most captures of H. griseus occur in theWestern and Central Mediterranean Sea, particularly during the warm months of the year and most commonly bybottom trawlers and bottom longliners. At-vessel mortality (AVM) was rather low in all gears but a slightlyhigher degree of individual mortality is suggested in trammel and gillnets. The population trend of H. griseus inthe Mediterranean Sea could not be inferred from the interviews as answers were highly variable, but the overalltrend in some countries may suggest this species is showing signs of population decrease. The results of this studyare mostly aligned with the latest IUCN assessment but also recommend reviewing the current status of H. griseusin the Mediterranean basin. Further empirical research on post-release mortality would also be advisable toimplement measures that help reduce this source of mortality

    MEDLEM database, a data collection on large elasmobranchs in the Mediterranean and Black Seas

    The Mediterranean Large Elasmobranchs Monitoring (MEDLEM) database contains more than 3,000 records (with more than 4,000 individuals) of large elasmobranch species from 21 different countries around the Mediterranean and Black seas, observed from 1666 to 2017. The principal species included in the archive are the devil ray (1,868 individuals), the basking shark (935 individuals), the blue shark (622 individuals), and the great white shark (342 individuals). In the last decades, other species such as the thresher shark (187 individuals), the shortfin mako (180 individuals), and the spiny butterfly ray (138) were reported with increasing frequency. This was possibly due to increased public awareness on the conservation status of sharks, and the consequent development of new monitoring programs. MEDLEM does not have homogeneous reporting coverage throughout the Mediterranean and Black seas and it should be considered as a database of observed species presence. Scientific monitoring efforts in the south-eastern Mediterranean and Black seas are generally lower than in the northern sectors and the absence of some species in our database does not imply their actual absence in these regions.     Kinase inhibitors for the treatment of inflammatory and autoimmune disorders

    Drugs targeting inhibition of kinases for the treatment of inflammation and autoimmune disorders have become a major focus in the pharmaceutical and biotech industry. Multiple kinases from different pathways have been the targets of interest in this endeavor. This review describes some of the recent developments in the search for inhibitors of IKK2, Syk, Lck, and JAK3 kinases. It is anticipated that some of these compounds or newer inhibitors of these kinases will be approved for the treatment of rheumatoid arthritis, psoriasis, organ transplantation, and other autoimmune diseases

    A global synthesis reveals biodiversity-mediated benefits for crop production

    Human land use threatens global biodiversity and compromises multiple ecosystem functions critical to food production. Whether crop yield-related ecosystem services can be maintained by a few dominant species or rely on high richness remains unclear. Using a global database from 89 studies (with 1475 locations), we partition the relative importance of species richness, abundance, and dominance for pollination; biological pest control; and final yields in the context of ongoing land-use change. Pollinator and enemy richness directly supported ecosystem services in addition to and independent of abundance and dominance. Up to 50% of the negative effects of landscape simplification on ecosystem services was due to richness losses of service-providing organisms, with negative consequences for crop yields. Maintaining the biodiversity of ecosystem service providers is therefore vital to sustain the flow of key agroecosystem benefits to society

    Crop pests and predators exhibit inconsistent responses to surrounding landscape composition

    The idea that noncrop habitat enhances pest control and represents a win–win opportunity to conserve biodiversity and bolster yields has emerged as an agroecological paradigm. However, while noncrop habitat in landscapes surrounding farms sometimes benefits pest predators, natural enemy responses remain heterogeneous across studies and effects on pests are inconclusive. The observed heterogeneity in species responses to noncrop habitat may be biological in origin or could result from variation in how habitat and biocontrol are measured. Here, we use a pest-control database encompassing 132 studies and 6,759 sites worldwide to model natural enemy and pest abundances, predation rates, and crop damage as a function of landscape composition. Our results showed that although landscape composition explained significant variation within studies, pest and enemy abundances, predation rates, crop damage, and yields each exhibited different responses across studies, sometimes increasing and sometimes decreasing in landscapes with more noncrop habitat but overall showing no consistent trend. Thus, models that used landscape-composition variables to predict pest-control dynamics demonstrated little potential to explain variation across studies, though prediction did improve when comparing studies with similar crop and landscape features. Overall, our work shows that surrounding noncrop habitat does not consistently improve pest management, meaning habitat conservation may bolster production in some systems and depress yields in others. Future efforts to develop tools that inform farmers when habitat conservation truly represents a win–win would benefit from increased understanding of how landscape effects are modulated by local farm management and the biology of pests and their enemies

    Retardation or acceleration of diabetes in NOD/Lt mice mediated by intrathymic administration of candidate beta-cell antigens.

    A single injection of syngeneic islet cells into the thymus of 4-week-old NOD/Lt female mice strongly retards diabetogenesis. The present study used the intrathymic route of antigen administration to compare the relative efficacy of peptides/proteins derived from two major candidate pancreatic beta-cell autoantigens, insulin and GAD65, to modulate diabetogenesis. Intrathymic administration of insulin B chain or recombinant human GAD65 significantly suppressed diabetogenesis during a 20-week follow-up period, whereas no protection was mediated by either insulin A chain or a synthetic peptide (A2) derived from it. Quite unexpectedly, two GAD65-derived peptides near the COOH-terminus (p34 and p35) accelerated diabetes onset. Semiquantitative reverse transcription-polymerase chain reaction analysis was performed on cDNAs from isolated islets or whole pancreases of NOD/Lt females 4 weeks after intrathymic injections. Protection mediated by intrathymic administration with either intact islet cells or GAD65 were correlated with an upregulation of mRNA for T-helper 2 (Th2)-associated cytokines (interleukin [IL]-4, IL-10), concomitant with downregulation of Th1-associated interferon (IFN) transcripts (all normalized to T-cell receptor Cbeta transcripts) in islet-infiltrating lymphocytes. Protection mediated by the intrathymic administration of insulin B chain, however, correlated only with a modest upregulation of IL-4 and IL-10 transcript levels, and no diminution in IFN-gamma transcripts. In contrast, the diabetes-accelerating GAD65 p34 and p35 peptides were not associated with an immune deviation, expressing levels of IFN-gamma characteristic of islet-infiltrating lymphocytes in vehicle-injected NOD controls. Hence, Th1-to-Th2 immune deviation provides only a partial explanation for peptide immunotherapy of diabetes in NOD mice. The finding that certain peptides can accelerate rather than retard diabetogenesis as a function of route and age of administration adds a cautionary note to this type of therapy

    ADP-ribosylation is a post-translational modification that can alter the physical and chemical properties of target proteins and that controls many important cellular processes. Macrodomains are evolutionarily conserved structural domains that bind ADP-ribose derivatives and are found in proteins with diverse cellular functions. Some proteins from the macrodomain family can hydrolyze ADP-ribosylated substrates and therefore reverse this post-translational modification. Bacteria and Streptomyces, in particular, are known to utilize protein ADP-ribosylation, yet very little is known about their enzymes that synthesize and remove this modification. We have determined the crystal structure and characterized, both biochemically and functionally, the macrodomain protein SCO6735 from Streptomyces coelicolor This protein is a member of an uncharacterized subfamily of macrodomain proteins. Its crystal structure revealed a highly conserved macrodomain fold. We showed that SCO6735 possesses the ability to hydrolyze PARP-dependent protein ADP-ribosylation. Furthermore, we showed that expression of this protein is induced upon DNA damage and that deletion of this protein in S. coelicolor increases antibiotic production. Our results provide the first insights into the molecular basis of its action and impact on Streptomyces metabolism

    The use of fishers' Local Ecological Knowledge to reconstruct fish behavioral traits and fishers' perception of conservation relevance of elasmobranchs in the Mediterranean Sea

    The Mediterranean Sea has a long-lasting history of fishery exploitation that, together with other anthropogenic impacts, has led to declines in several marine organisms. In particular, elasmobranch populations have been severely impacted, with drastic decreases in abundance and species diversity. Based on their experience, fishers can provide information on marine species oc- currence, abundance and behavioural traits on a long-term scale, therefore contributing to research on the poorly studied biolog- ical aspects of elusive or rare elasmobranch species. In this study, for the first time, the Local Ecological Knowledge (LEK) of fishers was applied to study the behavioural traits of sharks, rays and skates in 12 FAO-GFCM geographical sub-areas (GSAs) of the Mediterranean Sea. This study found both new insight and proved the reliability of LEK-based catch seasonality, reflect- ing seasonal movements, by comparing LEK-based findings and available literature on five elasmobranch taxa (Mustelus spp., Squalus acanthias, Raja spp., Myliobatis aquila and Scyliorhinus stellaris) in the Adriatic Sea and 7 taxa (Mustelus spp., Raja spp., Prionace glauca, Scyliorhinus canicula, Torpedo spp., Pteroplatytrygon violacea and Isurus oxyrinchus) in the remaining Mediterranean GSAs. In addition, LEK provided new insights into a novel comprehensive representation of species aggregations (Mustelus spp., S. acanthias, M. aquila and S. canicula) in the sampled GSAs and supplied the first descriptions of the size, num- ber of individuals and sex composition of the aggregations. When the limits and shortcomings of LEK-based research are consid- ered, this methodology can be a complementary and cost-effective tool used to study elasmobranchs in either a data-poor scenario or a scenario in which a baseline is missing. LEK can also be useful for the evaluation and inclusion of fishers’ perceptions of bottom-up management and to provide important evidence for conservation plans
