    Selection of parents and segregating populations of carioca common bean resistant to Fusarium wilt.

    Fusarium wilt (Fusarium oxysporum f. sp. phaseoli) is one of the most common diseases in the common bean (Phaseolus vulgaris) crop, and the most viable alternative for its control is the use of resistant cultivars. However, the number of “carioca” cultivars, the most important commercial group in Brazil, which has a good level of resistance to this disease, is still low. To obtain populations with a higher degree of resistance, seven parents with different levels of resistance to Fusarium and which produce carioca grains were crossed in a complete diallel design. The 21 segregating populations obtained were evaluated in the years/generations 2012/F3, 2013/F4, and 2014/F5 in experiments in Santo Antônio de Goiás, GO, in the winter crop season (sowing in May), together with the check cultivars BRS Notável (resistant) and BRS Cometa (susceptible) in an area naturally infested with Fusarium oxysporum f. sp. phaseoli, under a center pivot irrigation system. Reaction to Fusarium wilt, yield, and 100-seed weight were evaluated. Additive and non-additive effects contributed to control of reaction to Fusarium wilt, yield, and 100-seed weight. The statistically negative general combining ability (ĝ) estimates for BRS Notável (-0.51), CNFC 15872 (-0.20), IPR Juriti (-0.18), and BRSMG Talismã (-0.13) indicate that these parents are recommended for obtaining populations more resistant to Fusarium wilt. Among them, cultivar BRS Notável also showed a statistically positive ĝ estimate for grain yield (87.90). Among the parents recommended for resistance to Fusarium wilt, cultivars IPR Juriti (0.99) and BRS Notável (0.23) are those that most contributed to obtaining populations with larger-sized beans. Cultivar BRS Notável is the most promising parent for participation in new crossing blocks, showing non-zero ĝi estimates for all traits, favoring an increase in resistance to Fusarium wilt, yield, and 100-seed weight. Populations BRS Notável / CNFC 15872, BRS Ametista / BRS Notável, BRS Notável / BRSMG Talismã, and BRS Ametista / BRSMG Talismã showed good mean values for reaction to Fusarium wilt, yield, and 100-seed weight and at least one parent with a good general combining ability estimate for resistance to Fusarium wilt. These four populations are promising for obtaining lines with superior performance for resistance to Fusarium wilt, yield, and larger-sized grains

    Evaluation of mutagenicity and antimutagenicity of different fractions of Pterogyne nitens (Leguminosae), using Tradescantia pallida micronuclei assay

    Pterogyne nitens (Fabaceae-Caesalpinioideae) é uma árvore nativa da América do Sul, onde é empregada na medicina popular para o tratamento da ascaridíase. Recentemente, descrevemos o efeito mutagênico do extrato etanólico das folhas de P. nitens. Dessa forma, o presente estudo teve por objetivo aprofundar a avaliação do potencial mutagênico das frações isoladas das folhas de Pterogyne nitens, acetato de etila (AcOEt), n-butanólica (BuOH) e hidroalcóolica (HA). Quando o efeito mutagênico foi observado somente nas maiores concentrações testadas, o potencial antimutagênico também foi avaliado. Os ensaios mutagênicos e antimutagênicos foram realizados utilizando ensaio de micronúcleo em Trandescantia pallida. Na avaliação de mutagenicidade, observou-se o efeito nas frações AcOEt (0,460 mg/mL), BuOH (0,142, 0,285, 0,570 e 1,14 mg/mL) e HA (0,050, 0,100, 0,200 e 0,400 mg/mL). Considerando que o efeito mutagênico da fração AcOEt foi observado somente na concentração mais elevada (0,460 mg/mL), o potencial antimutagênico da mesma foi avaliado. As concentrações de 0,115 e 0,230 mg/mL da fração AcOEt demonstraram atividade antimutagênica. A partir dos resultados do presente estudo, conclui-se que determinadas frações de P. nitens apresentam mutagenicidade (BuOH e HA), enquanto a fração AcOEt apresentou efeito antimutagênico nas maiores concentrações. Esses resultados tornam o estudo da P. nitens bastante promissor, considerando que esta planta possui distribuição geográfica ampla e tem sido pouco estudada.Pterogyne nitens (Fabaceae-Caesalpinioideae) is a tree native to South American, where it is used in folk treatment of ascaridiasis. Recently, we have been describing the mutagenic effect of the ethanol extract of leaves of P. nitens. Thus, the present study aimed at evaluating the mutagenic potential of the ethyl acetate (EtOAc), n- butanol (BuOH) and hydroalcoholic (HA) fractions. When the mutagenic effect was observed only in the highest tested concentrations, the antimutagenic activity was also evaluated. Both mutagenic and antimutagenic assays were performed using T. pallida micronuclei assay. Mutagenicity was observed between different concentrations of the P nitens fractions, EtOAc (0.460 mg/mL), BuOH (0.142, 0.285, 0.570 and 1.14 mg/mL) and HA (0.050, 0.100, 0.200 and 0.400 mg/mL). Whereas the mutagenic effect of the EtOAc fraction was observed in the highest concentration (0.460 mg/mL), its antimutagenic potential was evaluated. The 0.115 and 0.230 mg/mL concentrations of the EtOAc fraction demonstrated antimutagenic activity. Based on the results of the present study we can conclude that some P. nitens fractions (BuOH and HA) demonstrated mutagenic effects whereas the EtOAc fraction shown low mutagenicity and amtimutagenicity in the two higher concentrations. Those results stimulate the studies with P. nitens, which possess spread geographic distribution and it is still low studied

    19F labelled glycosaminoglycan probes for solution NMR and non-linear (CARS) microscopy

    Studying polysaccharide-protein interactions under physiological conditions by conventional techniques is challenging. Ideally, macromolecules could be followed by both in vitro spectroscopy experiments as well as in tissues using microscopy, to enable a proper comparison of results over these different scales but, often, this is not feasible. The cell surface and extracellular matrix polysaccharides, glycosaminoglycans (GAGs) lack groups that can be detected selectively in the biological milieu. The introduction of 19F labels into GAG polysaccharides is explored and the interaction of a labelled GAG with the heparin-binding protein, antithrombin, employing 19F NMR spectroscopy is followed. Furthermore, the ability of 19F labelled GAGs to be imaged using CARS microscopy is demonstrated. 19F labelled GAGs enable both 19F NMR protein-GAG binding studies in solution at the molecular level and non-linear microscopy at a microscopic scale to be conducted on the same material, essentially free of background signals

    Stres na radu i zdravlje medicinskih sestara u jedinicama intenzivne njege u Srbiji

    The aim of this study was to identify and analyse professional stressors, evaluate the level of stress in nurses in Intensive Care Units (ICU), and assess the correlation between the perception of stress and psychological and somatic symptoms or diseases shown by nurses. The research, designed as a crosssectional study, was carried out in the Intensive Care Units (ICU), in health centres in Serbia. The sample population encompassed 1000 nurses. Expanded Nursing Stress Scale (ENSS) was used as the research instrument. ENSS revealed a valid metric characteristic within our sample population. Nurses from ICUs rated situations involving physical and psychological working environments as the most stressful ones, whereas situations related to social working environment were described as less stressful; however, the differences in the perception of stressfulness of these environments were minor. Socio-demographic determinants of the participants (age, marital status and education level) significantly affected the perception of stress at work. Significant differences in the perception of stressfulness of particular stress factors were observed among nurses with respect to psychological and somatic symptoms (such as headache, insomnia, fatigue, despair, lower back pain, mood swings etc.) and certain diseases (such as hypertension, myocardial infarction, stroke, diabetes mellitus etc). In view of permanent escalation of professional stressors, creating a supportive working environment is essential for positive health outcomes, prevention of job-related diseases and better protection of already ill nurses.Cilj je ovoga rada bio identifi cirati i analizirati profesionalne stresore, procijeniti razinu stresa kod medicinskih sestara u jedinicama intenzivne njege te procijeniti korelaciju između percepcije stresa i prisutnosti psiholoških i somatskih simptoma ili bolesti kod medicinskih sestara. Istraživanje je provedeno u obliku studije presjeka u Jedinicama intenzivne njege u zdravstvenim centrima u Srbiji. Uzorak se sastojao od 1000 medicinskih sestara-tehničara. Za procjenu i analizu profesionalnih stresora korišten je upitnik Expanded Nursing Stress Scale (ENSS), koji je pokazao validne metrijske karakteristike na našoj ispitanoj populaciji. Medicinske sestre u Jedinicama intenzivne njege ocijenile su situacije iz fizičkoga i psihološkoga radnog okruženja kao izrazito opterećujuće, a situacije iz socijalnoga radnog okruženja kao manje opterećujuće. Razlika u percepciji stresogenosti navedenih radnih okruženja nije bila statistički značajna. Sociodemografske determinante ispitanika (dob, bračno stanje i stupanj obrazovanja) značajno utječu na percepciju stresa na radnom mjestu. Utvrđena je statistički značajna razlika u opažanju stresogenosti pojedinih stresnih situacija na radnom mjestu između medicinskih sestara u odnosu na postojanje psihosomatskih simptoma (kao što su glavobolja, nesanica, umor, očaj, bol u leđima, česte promjene raspoloženja) ili određenih bolesti (kao što su povišena hipertenzija, infarkt miokarda, cerebrovaskularni inzult, šećerna bolest). Zbog sve izraženije prisutnosti profesionalnih stresora nužno je poduzeti određene strateške mjere kod medicinskih sestara u Jedinicama intenzivne njege. Strateške mjere podrazumijevaju unaprjeđenje psihosocijalne radne klime, što bi unaprijedilo njihovo zdravlje i spriječilo nastanak bolesti u svezi s radom, ali i omogućilo bolju zaštitu već oboljelim medicinskim sestrama
