    Liver-specific insulin receptor isoform A expression enhances hepatic glucose uptake and ameliorates liver steatosis in a mouse model of diet-induced obesity

    Among the main complications associated with obesity are insulin resistance and altered glucose and lipid metabolism within the liver. It has previously been described that insulin receptor isoform A (IRA) favors glucose uptake and glycogen storage in hepatocytes compared with isoform B (IRB), improving glucose homeostasis in mice lacking liver insulin receptor. Thus, we hypothesized that IRA could also improve glucose and lipid metabolism in a mouse model of high-fatdiet-induced obesity. We addressed the role of insulin receptor isoforms in glucose and lipid metabolism in vivo. We expressed IRA or IRB specifically in the liver by using adeno-associated viruses (AAVs) in a mouse model of diet-induced insulin resistance and obesity. IRA, but not IRB, expression induced increased glucose uptake in the liver and muscle, improving insulin tolerance. Regarding lipid metabolism, we found that AAV-mediated IRA expression also ameliorated hepatic steatosis by decreasing the expression of Fasn, Pgc1a, Acaca and Dgat2 and increasing Scd-1 expression. Taken together, our results further unravel the role of insulin receptor isoforms in hepatic glucose and lipid metabolism in an insulin-resistant scenario. Our data strongly suggest that IRA is more efficient than IRB at favoring hepatic glucose uptake, improving insulin tolerance and ameliorating hepatic steatosis. Therefore, we conclude that a gene therapy approach for hepatic IRA expression could be a safe and promising tool for the regulation of hepatic glucose consumption and lipid metabolism, two key processes in the development of non-alcoholic fatty liver disease associated with obesity

    Infected Dendritic Cells Facilitate Systemic Dissemination and Transplacental Passage of the Obligate Intracellular Parasite Neospora caninum in Mice

    The obligate intracellular parasite Neospora caninum disseminates across the placenta and the blood-brain barrier, to reach sites where it causes severe pathology or establishes chronic persistent infections. The mechanisms used by N. caninum to breach restrictive biological barriers remain elusive. To examine the cellular basis of these processes, migration of different N. caninum isolates (Nc-1, Nc-Liverpool, Nc-SweB1 and the Spanish isolates: Nc-Spain 3H, Nc-Spain 4H, Nc-Spain 6, Nc-Spain 7 and Nc-Spain 9) was studied in an in vitro model based on a placental trophoblast-derived BeWo cell line. Here, we describe that infection of dendritic cells (DC) by N. caninum tachyzoites potentiated translocation of parasites across polarized cellular monolayers. In addition, powered by the parasite's own gliding motility, extracellular N. caninum tachyzoites were able to transmigrate across cellular monolayers. Altogether, the presented data provides evidence of two putative complementary pathways utilized by N. caninum, in an isolate-specific fashion, for passage of restrictive cellular barriers. Interestingly, adoptive transfer of tachyzoite-infected DC in mice resulted in increased parasitic loads in various organs, e.g. the central nervous system, compared to infections with free parasites. Inoculation of pregnant mice with infected DC resulted in an accentuated vertical transmission to the offspring with increased parasitic loads and neonatal mortality. These findings reveal that N. caninum exploits the natural cell trafficking pathways in the host to cross cellular barriers and disseminate to deep tissues. The findings are indicative of conserved dissemination strategies among coccidian apicomplexan parasites

    Interpopulation Variability in Dietary Traits of Invasive Bleak Alburnus alburnus (Actinopterygii, Cyprinidae) Across the Iberian Peninsula

This cyprinid fish is a successful invader in the Iberian Peninsula. No studies exist on its foraging strategies on a large scale for this ecoregion. The aim of the present study was to compare dietary traits of invasive bleak among the main Iberian rivers and a ‘reference’ native bleak population from France. Bleak were sampled during May–June 2019 from the Iberian Rivers Ebro, Tagus, Guadiana, Segura and Guadalquivir and the River Saône (France). Diptera larvae and zooplankton were common food categories in the River Saône. Insect nymphs were more important in the River Ebro. The intake of plant material was higher in the River Tagus. Flying insects were more consumed in the River Guadiana. Nektonic insects were important in the River Guadalquivir. Detritus was a frequent food category for all populations, in terms of occurrence and mass. Dietary parameters followed a unimodal response in relation to the latitudinal gradient, with the maximum values for the Tagus and Guadiana populations.     Evaluation of the Use of Autonomous Reef Monitoring Structures (ARMS) for Describing the Species Diversity of Two Coral Reefs in the Yucatan Peninsula, Mexico

    Autonomous reef monitoring structures (ARMS) have been proposed as a standardized, passive, nondestructive sampling tool. This study assessed the ability of ARMS to capture the cryptic species diversity of two coral reefs by recording species richness and taxonomic representativeness using conventional taxonomy. The capacity of ARMS, as artificial substrates, to favor the establishment of nonindigenous species over native species was also evaluated. The use of ARMS allowed the detection of 370 species morphotypes from nine phyla, yielding 13 new records of geographic distribution expansion, one exotic species for the Gulf of México and the Caribbean Sea, and six newly described species. It was also possible to make spatial comparisons of species richness between both reefs. ARMS captured cryptic diversity exceptionally well, with the exception of echinoderms. Furthermore, these artificial structures did not hinder the colonization ability of native species; in fact, the colonization patterns on the structures themselves represented the spatial differences in the structure of benthic assemblages. This study represents the first effort to make a conventional taxonomic description of the cryptic fauna of the Yucatan Peninsula using ARMS. It is recommended to assess coral reef species diversity, but more taxonomists specialized in marine invertebrates are needed

    Educational nurse-led telephone intervention shortly before colonoscopy as a salvage strategy after previous bowel preparation failure: a multicenter randomized trial

    Background: The most important predictor of unsuccessful bowel preparation is previous failure. For those patients with previous failure, we hypothesized that a nurse-led educational intervention by telephone shortly before the colonoscopy appointment could improve cleansing efficacy. Methods: We performed a multicenter, endoscopist-blinded, randomized controlled trial. Consecutive outpatients with previous inadequate bowel preparation were enrolled. Both groups received the same standard bowel preparation protocol. The intervention group also received reinforced education by telephone within 48 hours before the colonoscopy. The primary outcome was effective bowel preparation according to the Boston Bowel Preparation Scale. Intention-to-treat (ITT) analysis included all randomized patients. Per-protocol analysis included patients who could be contacted by telephone and the control cases. Results: 657 participants were recruited by 11 Spanish hospitals. In the ITT analysis, there was no significant difference between the intervention and control groups in the rate of successful bowel preparation (77.3 % vs. 72 %; P = 0.12). In the intervention group, 267 patients (82.9 %) were contacted by telephone. Per-protocol analysis revealed significantly improved bowel preparation in the intervention group (83.5 % vs. 72.0 %; P = 0.001). Conclusion: Among all patients with previous inadequate bowel preparation, nurse-led telephone education did not result in a significant improvement in bowel cleansing. However, in the 83 % of patients who could be contacted, bowel preparation was substantially improved. Phone education may therefore be a useful tool for improving the quality of bowel preparation in those cases
