    Modulating signaling networks by CRISPR/Cas9-mediated transposable element insertion

    In a recent past, transposable elements (TEs) were referred to as selfish genetic components only capable of copying themselves with the aim of increasing the odds of being inherited. Nonetheless, TEs have been initially proposed as positive control elements acting in synergy with the host. Nowadays, it is well known that TE movement into host genome comprises an important evolutionary mechanism capable of increasing the adaptive fitness. As insights into TE functioning are increasing day to day, the manipulation of transposition has raised an interesting possibility of setting the host functions, although the lack of appropriate genome engineering tools has unpaved it. Fortunately, the emergence of genome editing technologies based on programmable nucleases, and especially the arrival of a multipurpose RNA-guided Cas9 endonuclease system, has made it possible to reconsider this challenge. For such purpose, a particular type of transposons referred to as miniature inverted-repeat transposable elements (MITEs) has shown a series of interesting characteristics for designing functional drivers. Here, recent insights into MITE elements and versatile RNA-guided CRISPR/Cas9 genome engineering system are given to understand how to deploy the potential of TEs for control of the host transcriptional activity.Fil: Vaschetto, Luis Maria Benjamin.     Abstract Chagas disease is a neglected chronic condition with a high burden of morbidity and mortality. It has considerable psychological, social, and economic impacts. The disease represents a significant public health issue in Brazil, with different regional patterns. This document presents the evidence that resulted in the Brazilian Consensus on Chagas Disease. The objective was to review and standardize strategies for diagnosis, treatment, prevention, and control of Chagas disease in the country, based on the available scientific evidence. The consensus is based on the articulation and strategic contribution of renowned Brazilian experts with knowledge and experience on various aspects of the disease. It is the result of a close collaboration between the Brazilian Society of Tropical Medicine and the Ministry of Health. It is hoped that this document will strengthen the development of integrated actions against Chagas disease in the country, focusing on epidemiology, management, comprehensive care (including families and communities), communication, information, education, and research

    Objetivou-se conhecer as dificuldades e facilidades enfrentadas pelas mĂŁes de crianças com deficiĂȘncia visual (DV) no cuidado. Trata-se de uma pesquisa qualitativa, descritiva, realizada no segundo semestre de 2011, com dez mĂŁes de crianças com deficiĂȘncia visual, de um Centro de Educação para deficientes visuais, no sul do Brasil. Foram realizadasentrevistas semiestruturadas e anĂĄlise de conteĂșdo temĂĄtica das informaçÔes. Foram verificados como dificuldades o desconhecimento acerca da doença e da forma de cuidar da criança, a falta de acesso aos serviços de saĂșde, a sobrecarga pela dependĂȘncia da criança, a falta de apoio e o preconceito na prĂłpria famĂ­lia. Quanto Ă s facilidades, foram referidos a vontade do desenvolvimento saudĂĄvel das crianças, a convivĂȘncia com profissionais qualificados para sua educação e o contato com outras crianças com DV. Faz-se importante a instrumentalização da famĂ­lia para o cuidado Ă  criança com DV como forma de garantir a aquisição de habilidades e competĂȘncias, possibilitando-a um viver com qualidade

    WNT7B promotes bone formation in part through mTORC1

    WNT signaling has been implicated in both embryonic and postnatal bone formation. However, the pertinent WNT ligands and their downstream signaling mechanisms are not well understood. To investigate the osteogenic capacity of WNT7B and WNT5A, both normally expressed in the developing bone, we engineered mouse strains to express either protein in a Cre-dependent manner. Targeted induction of WNT7B, but not WNT5A, in the osteoblast lineage dramatically enhanced bone mass due to increased osteoblast number and activity; this phenotype began in the late-stage embryo and intensified postnatally. Similarly, postnatal induction of WNT7B in Runx2-lineage cells greatly stimulated bone formation. WNT7B activated mTORC1 through PI3K-AKT signaling. Genetic disruption of mTORC1 signaling by deleting Raptor in the osteoblast lineage alleviated the WNT7B-induced high-bone-mass phenotype.
