    Civilized Elders and Isolated Ancestors: The Multiple Histories of Contemporary Amazonia

    In this article I consider the impact of Peter Gow’s writing on indigenous histories as a key area of research on Amazonia. Building on his study of kinship as history on the Bajo Urubamba (1991) he presented a regional perspective on the dynamic social categories by which Amazonian people understand their relations with various “others.” Focusing on indigenous agency and modes of thought, Gow challenged certain lines of historical thinking that dominated anthropology at the time. I explore how his ethnographic approach to history has influenced a generation of regional scholarship, including my own work on memory and social transformation among Waorani people in Amazonian Ecuador. I look specifically at how Gow’s approach can help contextualize expressions of historical continuity and difference in indigenous politics. In contrast to relatively static views of history in a global politics of recognition, the categories of difference and their transformation he described remain central to how some Amazonian people relate the past to their hopes for the future. Understanding this process requires Gow’s sensitivity to radically different ideas of the past, as well as attention to the multiple histories that Amazonian people engage with today

    Lost and Found:Contesting Isolation and Cultivating Contact in Amazonian Ecuador

    In May 2003 a group of Waorani men in Amazonian Ecuador led an attack against their “uncontacted” Taromenani neighbors, resulting in a massacre that has fueled ongoing debates about the rights of indigenous people living in “voluntary isolation.” In this article I consider how Waorani understandings of the attack point to indigenous formulations of alterity that challenge what Lucas Bessire (2012) has described as the contemporary politics of isolation. I draw on recent discussions of kinship as a form of mutual belonging that extend beyond common substance (Sahlins 2013), and consider how, in the aftermath of the killings, many Waorani came to see spatially distant others as kinsmen who became disconnected from Waorani in past times. Understood by Waorani as kin of victims, the Taromenani have become both a source of desired relations and a potent image of indigenous strength and autonomy in the context of social and economic transformation

    Body Weight and Plasma Levels of Ghrelin and Leptin during Treatment with Olanzapine

    Although enhanced appetite and weight gain are potential side effects of treatment with antipsychotic agents, particularly olanzapine and clozapine, the mechanisms underlying these side effects are poorly understood. Leptin and ghrelin were recently identified as hormones that play crucial roles in the regulation of energy balance and glucose metabolism. To elucidate relationships between weight change and plasma levels of ghrelin and leptin, we investigated the circulating ghrelin and leptin levels and body weight during olanzapine treatment. Twenty-four patients with schizophrenia were examined during 6-month administration of olanzapine. Ghrelin, leptin, weight and body mass index (BMI) were measured before and after 2, 4, 8, 12, 16, and 24 weeks of olanzapine treatment. The concentration of glucose and various lipid metabolic parameters were measured at baseline and at 24 weeks. Significant increases in weight, BMI and leptin were observed at week 24. On the other hand, the serum levels of ghrelin decreased significantly after olanzapine treatment. In addition, the level of ghrelin was negatively correlated with the leptin level, BMI and weight. The leptin level was positively correlated with both BMI and weight. Ghrelin is associated with metabolic changes, in combination with leptin, during olanzapine treatment. However, further large-scale and longitudinal studies are warranted to elucidate the metabolic changes involving ghrelin, leptin and insulin during treatment with antipsychotics

    Difference Revised: Gender and Transformation among the Amazonian Runa

    In this paper, I will explore how knowledge practices among the Runa of the Ecuadorian Amazon are informed by the specificity of local gender constructions. I will argue that while men learn to be ‘proper’ persons primarily through the ingestion of substances which penetrate inside their bodies and change them from the interior, women learn to become ‘proper’ Runa through imitating and reproducing specific movements. This difference in learning regimes, I argue, is based upon a priori conceptualisation of men and women as distinct kinds of beings. I argue that the Runa conceptualise as gender difference the way in which exteriority and interiority are played out in male and female persons. Unlike other Amazonian cases, women are understood by the Runa as ‘naturally’ predisposed to exteriority. This has important repercussions in the way cultural change is thought to affect women and men, especially in contrast to other Amazonian people

    HIV-1 Vpr Triggers Mitochondrial Destruction by Impairing Mfn2-Mediated ER-Mitochondria Interaction

    Human immunodeficiency virus 1 (HIV-1) viral protein R (Vpr) has been shown to induce host cell death by increasing the permeability of mitochondrial outer membrane (MOM). The mechanism underlying the damage to the mitochondria by Vpr, however, is not clearly illustrated. In this study, Vpr that is introduced, via transient transfection or lentivirus infection, into the human embryonic kidney cell line HEK293, human CD4+ T lymphoblast cell line SupT1, or human primary CD4+ T cells serves as the model system to study the molecular mechanism of Vpr-mediated HIV-1 pathogenesis. The results show that Vpr injures MOM and causes a loss in membrane potential (MMP) by posttranscriptionally reducing the expression of mitofusin 2 (Mfn2) via VprBP-DDB1-CUL4A ubiquitin ligase complex, gradually weakening MOM, and increasing mitochondrial deformation. Vpr also markedly decreases cytoplasmic levels of dynamin-related protein 1 (DRP1) and increases bulging in mitochondria-associated membranes (MAM), the specific regions of endoplasmic reticulum (ER) which form physical contacts with the mitochondria. Overexpression of Mfn2 and DRP1 significantly decreased the loss of MMP and apoptotic cell death caused by Vpr. Furthermore, by employing time-lapse confocal fluorescence microscopy, we identify the transport of Vpr protein from the ER, via MAM to the mitochondria. Taken together, our results suggest that Vpr-mediated cellular damage may occur on an alternative protein transport pathway from the ER, via MAM to the mitochondria, which are modulated by Mfn2 and DRP1
