Development of curcumin lipid formulations for food applications: transport, permeability and safety evaluation on a mucus-secreting intestinal epithelial cell model

Curcumin is the main phenolic pigment extracted from turmeric, the powdered rhizome of Curcuma longa. It has been shown to exhibit antioxidant, antimicrobial, anti-inflammatory and anticarcinogenic activities however as health promoting agent, curcumin, is limited by its poor solubility in aqueous solution and its low bioavailability and therefore cannot be widely used in food and pharmaceutical processing industry. The challenge addressed in this work was to produce curcumin formulations to enhance its characteristics and evaluate its permeability, transport and cytotoxicity on a stablished in vitro cell co-culture model that mimics the intestinal epithelium. Solid lipid nanoparticles (SLN) and microparticles (SLM) have been visualised as a promising platform on development of formulations for food applications. Since traditional production methods possess a series of limitations, the processing by “green technologies” like supercritical carbon dioxide (scCO2) has been widely investigated. Through Particles from Gas Saturated Solutions (PGSS®) process, beeswax microparticles loaded with curcumin (9:1 (w/w)) were produced and characterized in terms of physicochemical properties: size, morphology, curcumin content and particles dispersion index. Operation process parameters were optimized and defined via response surface methodology and the best response was achieved at 160 bar, 73°C and 10% curcumin load. Under these conditions, encapsulation efficiency was 89.75 ± 2.23 % with a curcumin load of 8.98 % (w/w). Curcumin formulations underwent a digestive process and were tested for their cytotoxicity in Caco-2 monolayer. A triple co-culture has been established and characterized for use as an in vitro intestinal model. To closely mimic the intestinal epithelium the production of mucus by the HT29-MTX-E12 cell line cultured together with the Caco-2 enterocytes and M-cells like phenotype was observed. The model was used to evaluate the transport and permeability of free and encapsulated curcumin and its permeability was stablished as a value of 1.0 x 10-7 cm/s

The research reported in the present article aims to evaluate the teacher education learning scenarios implemented in two curricular units of the basic education course, discussing, in particular, the role of the integrated curriculum approach and of the used technologies, in the development of statistical thinking of prospective teachers. The study used a methodology within the interpretative paradigm. In order to evaluate the learning scenarios, it was used participant observation, documents' collection and a questionnaire with open-ended items whose responses were subjected to content analysis, and the frequencies of the emergent categories were determined. The results support the validation of the implemented scenarios. The students developed the reasoning about data representation and associations. The meaning of mean was a critical aspect that would deserve further research.O estudo apresentado neste artigo tem como objetivo avaliar cenários de aprendizagem implementados em duas unidades curriculares do Curso de Licenciatura em Educação Básica, discutindo, em particular, o papel da abordagem curricular integrada e das tecnologias no desenvolvimento do pensamento estatístico dos futuros docentes. Foi usada uma metodologia no âmbito do paradigma interpretativo. Para avaliar os cenários de aprendizagem, foi usada a observação participante, a recolha documental e um questionário com itens abertos cujas respostas foram objeto de análise de conteúdo, tendo sido determinadas as frequências das categorias emergentes. Os resultados suportam a validação dos cenários implementados. Os estudantes desenvolveram o raciocínio acerca da representação de dados e das associações. Ferramentas numéricas para análise isogeométrica em regime não-linear

Doutoramento em Engenharia MecânicaThe present work deals with the development of robust numerical tools for Isogeometric Analysis suitable for problems of solid mechanics in the nonlinear regime. To that end, a new solid-shell element, based on the Assumed Natural Strain method, is proposed for the analysis of thin shell-like structures. The formulation is extensively validated using a set of well-known benchmark problems available in the literature, in both linear and nonlinear (geometric and material) regimes. It is also proposed an alternative formulation which is focused on the alleviation of the volumetric locking pathology in linear elastic problems. In addition, an introductory study in the field of contact mechanics, in the context of Isogeometric Analysis, is also presented, with special focus on the implementation of a the Point-to-Segment algorithm. All the methodologies presented in the current work were implemented in a in-house code, together with several pre- and post-processing tools. In addition, user subroutines for the commercial software Abaqus were also implemented.O presente trabalho foca-se no desenvolvimento de ferramentas numéricas robustas para problemas não-lineares de mecânica dos sólidos no contexto de Análises Isogeométricas. Com esse intuito, um novo elemento do tipo sólido-casca, baseado no método das Deformações Assumidas, é proposto para a análise de estruturas do tipo casca fina. A formulação proposta é validada recorrendo a um conjunto de problemas-tipo disponíveis na literatura, considerando tanto regimes lineares como não-lineares (geométrico e de material). É ainda apresentada uma formulação alternativa para aliviar o fenómeno de retenção volumétrica para problemas em regime linear elástico. Adicionalmente, é apresentado um estudo introdutório da mecânica Do conta to no contexto de Análises Isogeométricas, dando especial ênfase ao algoritmo de Ponto-para-Segmento. As metodologias apresentadas no presente trabalho foram implementadas num código totalmente desenvolvido durante o de correr do mesmo, juntamente com diversas ferramentas para pré- e pós processamento. Avaliação do desempenho energético e definição de PRE de uma unidade hoteleira

O elevado consumo energético e respetivo custo associado nas indústrias, tem sido uma questão que os gestores têm vindo a refletir cada vez mais. A forma como é consumida a energia é a base de análise para verificar se a sua utilização está a ser feita de forma eficiente. As razões para as empresas apresentarem consumos desnecessários são diversas e requerem um estudo completo às instalações. Neste seguimento, surge a implementação de medidas de racionalização de energia com o objetivo de reduzir a fatura energética das industrias e/ou de ajudar no cumprimento de compromissos nacionais e internacionais. Esta dissertação foi elaborada no decorrer de um estágio curricular realizado na empresa Ecoinside, no âmbito de uma nova auditoria feita pela empresa, com base no SGCIE, a uma indústria consumidora intensiva de energia na área da metalomecânica. Neste contexto, o principal objetivo deste documento é o estudo de medidas de racionalização de energia bem como a respetiva análise de viabilidade económica, com base em indicadores. Também é apresentada uma análise temporal dos consumos energéticos da indústria e o retrato da auditoria feita em 2010, bem como da nova auditoria realizada no decorrer do estágio, sendo esta a base para o estudo das medidas. Deste modo, foram feitas medições aos consumos da indústria, realizados levantamentos termográficos a equipamentos e recolhida informação relativamente à iluminação existente bem como, de todos os dados e informações para a formulação de medidas de racionalização de energia.Nowadays, energetic consumption and the associated high costs claims more attention by factory managers. The way energy is consumed is the basis of analysis to see if it´s use is being efficiently. There are many reasons why factories present unnecessary consumption and, because of that, it is important to do a complete study of the facilities. In this way, the implementation of energy rationalization with the objective of reducing the energy invoice of industries and/or help to satisfy the national and international commitments, is required. This master’s thesis was elaborated in the course of a curricular internship at the company Ecoinside with the objective to realize a new audit, based on the SGCIE, to an intensive energy consumer industry. The main goal of this master’s thesis is the study of energy rationalization measures as well as their economic feasibility analysis, based on indicators. It is also presented a temporal analysis of the energy consumption and the results of the audit done in 2010. The new audit carried out during this internship is the basis for the study of the measures. To provide this relevant information, measurements were made to the consumptions of the industry, thermographic surveys to equipment and was also collected information regarding the existing light as well all data and information for the formulation of rationalization of energy measures

Estratégias evolucionárias de optimização de parâmetros reais

Mestrado em Engenharia MecânicaActualmente, existem diversos problemas de engenharia cujas propriedades podem ser expressas através de uma função, denominada função objectivo. Existem diversos métodos que possuem como principal objectivo minimizar a referida função. Os métodos baseados no gradiente são métodos nos quais a direcção e tamanho do passo são calculados a partir do declive da função objectivo. Apesar destes métodos necessitarem de reduzidos tempos de computação, estes podem convergir prematuramente ou ficar estagnados em mínimos locais. Os métodos de optimização baseados na teoria evolucionária são aproximações que possuem como principal desvantagem elevados tempos de computação. No entanto, estes apresentam uma grande flexibilidade na modelação de problemas de engenharia. Neste grupo, os algoritmos mais conhecidos e aplicados em problemas de optimização são os Algoritmos Evolucionários (EA’s), Algoritmos Genéticos (GA’s) e Evolução Diferencial (DE). Existem ainda algoritmos baseados em processos naturais tais como o algoritmo de Optimização por Bandos de Partículas (PSO), que reproduz o comportamento de bandos de animais. Neste trabalho é desenvolvido um algoritmo de optimização de procura directa baseado em métodos diferenciais, evolucionários e no comportamento de animais. O algoritmo desenvolvido é aplicado a problemas de engenharia inversa. Numa primeira fase, o algoritmo desenvolvido é validado e comparado com algoritmos existentes recorrendo a um conjunto de funções compostas especialmente criadas para este fim. Numa segunda fase, o algoritmo desenvolvido é aplicado a problemas de Engenharia Mecânica e Mecânica Computacional. Nesta secção, os problemas das três barras e da cúpula de 120 barras são analisados recorrendo ao Método dos Elementos Finitos (MEF). Seguidamente, os problemas de compressão de um provete cilíndrico e da placa com furo central são analisados. Nestes problemas a função a minimizar é dada por um programa do MEF comercial. Finalmente, o algoritmo é aplicado a um problema de identificação de parâmetros de um modelo constitutivo. O algoritmo desenvolvido apresenta bons resultados e uma boa taxa de convergência. ABSTRACT: Nowadays, there are many inverse engineering problems whose properties can be expressed by a function, called objective function. There are several methods whose main goal is to minimize the value of that function. The gradient-based methods are optimization methods in which the step direction and length are calculated in terms of the objective function's slope. Although these methods require little computation time, they may converge prematurely or get trapped in a local minima. The optimization methods based on the evolutionary theory are approaches that need, as a main disadvantage, high computation times. However, they have a great flexibility in modeling engineering problems. In this class of methods, the ones that are best known and more often applied in optimization problem are the Evolutionary Algorithms (EA's), Genetic Algorithms (GA's) and Differential Evolution (DE). There are also nature-inspired algorithms such as the Particle Swarm Optimization method (PSO) that mimics the behavior of animal swarms. In this work a direct search optimization algorithm based on differential and evolutionary methods as well as in the behavior of animals is developed. This algorithm is applied to inverse engineering problems. In a first stage, the developed algorithm is validated and compared with existing optimization algorithms using a set of composite functions specially design for that purpose. In a second phase, the algorithm is applied to Engineering and Computational Mechanics problems. In this section, the three-truss bar problem and the 120-bar dome truss problem that are solved using the Finite Element Method (FEM) are analyzed. Subsequently, the compression of a cylindrical billet and the plate with a central cut-out problems are analyzed. In these problems, the function to minimize is given by a commercial FEM code. Finally, the algorithm is applied to a constitutive model parameter identification problem. The develop algorithm obtains good results and a good convergence rate

Análise e melhoria de procedimentos e ferramentas de suporte à gestão

Os principais objetivos do meu estágio estavam ligados com a melhoria de processos a nível do Departamento Comercial do mercado externo da empresa Revigrés, nomeadamente, redução de custos, gestão de relacionamento com o cliente, interligação de dados e organização departamental. Juntamente com a equipa em que fui integrado, foi possível superar os desafios que me foram propostos, e ainda redefinir alguns procedimentos, com a consequente resolução de problemas existentes na empresa. Isto foi possível através da redução de desperdícios, obtida através de melhor organização de informação, reforço a nível do sistema de faturação eletrónica, implementação de um quadro de plano de trabalho, desenvolvimento de dashboards e reformulação e analise de alguns processos a nível organizativo. Em resumo, os objetivos traçados para o período de nove meses de estágio foram concretizados, tendo o mesmo permitido um crescimento tanto a nível pessoal como profissionalThe main goals of my internship were related to processes improvement at the level of the International Department of Revigrés, namely cost reduction, customer relationship management, data interconnection and departmental organization. Together with the team in which I was integrated, it was possible to surpass the challenges that were proposed to me, as well as redefine some procedures, with the consequent resolution of existing problems within the company. This was possible thanks to reducing waste through better information organization, strengthening the electronic invoicing system, implementing a work plan framework, developing dashboards and redesigning and analyzing some processes at the organizational level.
