    Early epilepsy in children with Zika-related microcephaly in a cohort in Recife, Brazil: Characteristics, electroencephalographic findings, and treatment response.

    OBJECTIVE: To estimate the incidence of epilepsy in children with Zika-related microcephaly in the first 24 months of life; to characterize the associated clinical and electrographic findings; and to summarize the treatment responses. METHODS: We followed a cohort of children, born during the 2015-2016 Zika virus (ZIKV) epidemic in Brazil, with congenital microcephaly and evidence of congenital ZIKV infection on neuroimaging and/or laboratory testing. Neurological assessments were performed at ≤3, 6, 12, 15, 18, 21, and 24 months of life. Serial electroencephalograms were performed over the first 24 months. RESULTS: We evaluated 91 children, of whom 48 were female. In this study sample, the cumulative incidence of epilepsy was 71.4% in the first 24 months, and the main type of seizure was infantile spasms (83.1%). The highest incidence of seizures occurred between 3 and 9 months of age, and the risk remained high until 15 months of age. The incidence of infantile spasms peaked between 4 and 7 months and was followed by an increased incidence of focal epilepsy cases after 12 months of age. Neuroimaging results were available for all children, and 100% were abnormal. Cortical abnormalities were identified in 78.4% of the 74 children evaluated by computed tomography and 100% of the 53 children evaluated by magnetic resonance imaging. Overall, only 46.1% of the 65 children with epilepsy responded to treatment. The most commonly used medication was sodium valproate with or without benzodiazepines, levetiracetam, phenobarbital, and vigabatrin. SIGNIFICANCE: Zika-related microcephaly was associated with high risk of early epilepsy. Seizures typically began after the third month of life, usually as infantile spasms, with atypical electroencephalographic abnormalities. The seizure control rate was low. The onset of seizures in the second year was less frequent and, when it occurred, presented as focal epilepsy

    Cytotoxic, antitumor and toxicological profile of Passiflora alata leaf extract

    Passiflora alata or passion fruit is a native flowering plant from Amazon, geographically spread from Peru to Brazil. The plant has long been used in folks medicine for its pharmacological properties and is included in the Brazilian Pharmacopoeia since 1929. The aim of this study was to evaluate the potential cytotoxic and antitumor activities of Passiflora alata leaf extract (PaLE) in S180-tumor bearing mice. The percentage of cell proliferation inhibition (% CPI) and IC50 in relation to 4 tumor cell lines were determined in PC3, K-562, HepG2 and S180 cell lines using the MTT assay. PaLE showed a CPI > 75% and greater potency (IC50 < 30 µg/mL) against PC3 and S180 cell lines. PaLE showed antitumor activity in treatments intraperitoneally (36.75% and 44.99% at doses of 100 and 150 mg/kg/day, respectively). Toxicological changes were shown in the reduced body mass associated with reduced food consumption, increased spleen mass associated with histopathological increase in the white pulp of the spleen and increased number of total leukocytes with changes in the percentage relationship between lymphocytes and neutrophils.     Antiproliferative activity of synthetic fatty acid amides from renewable resources

    AbstractIn the work, the in vitro antiproliferative activity of a series of synthetic fatty acid amides were investigated in seven cancer cell lines. The study revealed that most of the compounds showed antiproliferative activity against tested tumor cell lines, mainly on human glioma cells (U251) and human ovarian cancer cells with a multiple drug-resistant phenotype (NCI-ADR/RES). In addition, the fatty methyl benzylamide derived from ricinoleic acid (with the fatty acid obtained from castor oil, a renewable resource) showed a high selectivity with potent growth inhibition and cell death for the glioma cell line—the most aggressive CNS cancer

    Brazilian red propolis: extracts production, physicochemical characterization, and cytotoxicity profile for antitumor activity

    Brazilian red propolis has been proposed as a new source of compounds with cytotoxic activity. Red propolis is a resinous material of vegetal origin, synthesized from the bees of the Appis mellifera family, with recognized biological properties. To obtain actives of low polarity and high cytotoxic profile from red propolis, in this work, we proposed a new solvent accelerated extraction method. A complete 23 factorial design was carried out to evaluate the influence of the independent variables or factors (e.g., temperature, number of cycles, and extraction time) on the dependent variable or response (i.e., yield of production). The extracts were analyzed by gas chromatography coupled with mass spectrometry for the identification of chemical compounds. Gas chromatography analysis revealed the presence of hydrocarbons, alcohols, ketones, ethers, and terpenes, such as lupeol, lupenone, and lupeol acetate, in most of the obtained extracts. To evaluate the cytotoxicity profile of the obtained bioactives, the 3-(4,5-dimethyl-2-thiazole)-2,5-diphenyl-2-H-tetrazolium bromide colorimetric assay was performed in different tumor cell lines (HCT116 and PC3). The results show that the extract obtained from 70 °C and one cycle of extraction of 10 min exhibited the highest cytotoxic activity against the tested cell lines.     WATER DEMAND AND WATER USE EFFICIENCY IN ‘PALMER’ MANGO CULTIVATION IN THE LOW-MIDDLE SÃO FRANCISCO VALLEY

    ABSTRACT The study aimed to determine the evapotranspiration (ETc) of the mango cv. ‘Palmer’ over two productive cycles, analyzing water-use efficiency (WUE) and crop water productivity (CWP) to propose average crop coefficient (Kc) values for improving irrigation management under cultivation conditions in the Low-Middle São Francisco Valley. The study was conducted from July 2019 to May 2021 in a commercial ‘Palmer’ mango orchard in Petrolina, State of Pernambuco, Brazil. Micrometeorological data was collected throughout the experimental period. The crop evapotranspiration (ETc) was determined using Bowen ratio energy balance (BREB), and then the Kc, WUE, and CWP were determined. The highest ETc values occurred during the floral induction phase (5.14 ± 0.85 mm day-1), with a Kc of 0.85; however, the lowest values were observed during the fruit maturation phase (3.60 ± 0.73 mm day-1), with a Kc of 0.91. Average water consumption per cycle was 1445 mm, with a daily average of 4.39 mm day-1. WUE and CWP were 16.9 and 24.5 kg ha-1 mm-1, respectively. Average Kc values of 0.81, 0.76, 0.85, 0.90, 0.95, and 0.91 are recommended for the vegetative growth, rest period and shoot maturation, floral induction, flowering, fruit development, and fruit maturation phases, respectively

    Development, cytotoxicity and eye irritation profile of a new sunscreen formulation based on benzophenone-3-poly(-caprolactone) nanocapsules

    The objective of this work was to characterize the toxicological profile of a newly developed sunscreen formulation based on polymeric nanocapsules (NCs) loading benzophenone-3 (BZP3). NCs composed of poly(ε-caprolactone) carrot oil and Pluronic® F68 were produced by emulsification-diffusion method. Their mean particle size (Z-Ave) ranged from 280 to 420 nm, polydispersity index (PDI) was below 0.37, while zeta potential (ZP) reached about |+11 mV|. No cytotoxic effects were observed in L929 fibroblast cell line for the blank (i.e., non-loaded) NCs and BZP3-loaded NCs (BZP3-NCs). The semi-solid sunscreen formulation was stable over time (centrifugation testing) and exhibited non-Newtonian pseudoplastic behavior, which is typical of products for topical application onto the skin. The sun protection factor (SPF) value reached 8.84, when incorporating BZP3-NCs (SPF of 8.64) into the semi-solid formulation. A synergistic effect was also observed when combining the formulation ingredients of nanocapsules, i.e., SPF of carrot oil was 6.82, blank NCs was 6.84, and BZP3-loaded NCs was 8.64. From the hen’s egg-chorioallantoic membrane test (HET-CAM) test, the non-irritation profile of the developed formulations could also be confirmed.     High rates of physical inactivity and cardiovascular risk factors in patients with resistant hypertension

    Background: Hypertension.is a multifactorial disease of high prevalence in Brazil and the world’s population and is the major treatable risk factor for cardiovascular disease. Objectives: To investigate the prevalence, according to sex, of risk behaviors and comorbidities associated with Hypertension.in the treated patients in Hiperdia Center Viçosa, MG. Methods: Cross-sectional study that evaluated 172 medical records of hypertensive patients greater than 18 years, nondiabetic, referred to the center of Viçosa Hiperdia. Among the data evaluated from analysis of medical records are the factors and behaviors of cardiovascular risk as overweight / obesity, dyslipidemia, smoking, drinking and physical inactivity, as well as medical conditions or comorbidities associated with hypertension. For data analysis were used the Kolmogorov-Smirnov test, Mann-Whitney test and the Pearson correlation test. The level of significance was 5%. Results: There was a higher prevalence of men among the patients and rates of alcoholism and smoking were significantly higher in this group. Women had a higher rate of obesity. Physical inactivity and dyslipidemia were present in 77% and 44% of patients, respectively, with no difference between sexes. Resistant Hypertension.was found in 71% of patients. Lower education was also very present among the hypertensive. Among the clinical conditions related to hypertension, there was a predominance of left ventricular hypertrophy, followed by kidney disease and cerebrovascular disease. Conclusions: The study showed that 71% of patients were classified as resistant Hypertension.and that, beyond this serious condition, they still showed a combination of behaviors and risk factors that confer a high risk of cardiovascular complicationsFundamento: A hipertensão arterial sistêmica é uma doença multifatorial, de alta prevalência na população brasileira e mundial e constitui o principal fator de risco tratável para as doenças cardiovasculares. Objetivos: Verificar a prevalência, de acordo com o sexo, dos comportamentos de risco e das comorbidades associadas à hipertensão nos pacientes atendidos no Centro Hiperdia de Viçosa, MG Métodos: Estudo transversal que avaliou 172 prontuários de hipertensos maiores de 18 anos, não diabéticos, encaminhados ao Centro Hiperdia de Viçosa. Entre os dados avaliados a partir de análise de prontuários estão os fatores e comportamentos de risco cardiovascular como sobrepeso/obesidade, dislipidemia, tabagismo, etilismo e sedentarismo, bem como condições clínicas ou comorbidades associadas à HAS. Para análise dos dados foram empregados os testes de Kolmogorov-Smirnov, o teste de Mann-Whitney e o teste de correlação de Pearson. O nível de significância adotado foi de 5%. Resultados: Observou-se uma prevalência maior de homens entre os hipertensos analisados e as taxas de etilismo e tabagismo foram significativamente maiores neste grupo. As mulheres apresentaram uma taxa maior de obesidade. O sedentarismo e a dislipidemia estiveram presentes em 77% e 44% dos pacientes, respectivamente, sem diferença entre os sexos. Hipertensão arterial resistente foi encontrada em 71% dos pacientes. Dentre as condições clínicas relacionadas à hipertensão, houve um predomínio da hipertrofia do ventrículo esquerdo, seguida pela doença renal e pela doença cerebrovascular. Conclusões: O estudo mostrou que 71 % dos pacientes eram classificados como hipertensos resistentes e que, além desta séria condição, os mesmos ainda apresentavam uma combinação de comportamentos e fatores de risco que conferem um alto risco de complicações cardiovasculare

    Telephone follow-up of patients after radical prostatectomy : a systematic review

    Objective: to assess and summarize the best scientific evidence from randomized controlled clinical trials about telephone follow-up of patients after radical prostatectomy, based on information about how the phone calls are made and the clinical and psychological effects for the individuals who received this intervention. Method: the search was undertaken in the electronic databases Medline, Web of Science, Embase, Cinahl, Lilacs and Cochrane. Among the 368 references found, five were selected. Results: two studies tested interventions focused on psychological support and three tested interventions focused on the physical effects of treatment. The psychoeducative intervention to manage the uncertainty about the disease and the treatment revealed statistically significant evidences and reduced the level of uncertainty and anguish it causes. Conclusion: the beneficial effects of telephone follow-up could be determined, as a useful tool for the monitoring of post-prostatectomy patients.Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)
