    Get PDF

Diversification of Genes Encoding Granule-Bound Starch Synthase in Monocots and Dicots Is Marked by Multiple Genome-Wide Duplication Events

Starch is one of the major components of cereals, tubers, and fruits. Genes encoding granule-bound starch synthase (GBSS), which is responsible for amylose synthesis, have been extensively studied in cereals but little is known about them in fruits. Due to their low copy gene number, GBSS genes have been used to study plant phylogenetic and evolutionary relationships. In this study, GBSS genes have been isolated and characterized in three fruit trees, including apple, peach, and orange. Moreover, a comprehensive evolutionary study of GBSS genes has also been conducted between both monocots and eudicots. Results have revealed that genomic structures of GBSS genes in plants are conserved, suggesting they all have evolved from a common ancestor. In addition, the GBSS gene in an ancestral angiosperm must have undergone genome duplication ∼251 million years ago (MYA) to generate two families, GBSSI and GBSSII. Both GBSSI and GBSSII are found in monocots; however, GBSSI is absent in eudicots. The ancestral GBSSII must have undergone further divergence when monocots and eudicots split ∼165 MYA. This is consistent with expression profiles of GBSS genes, wherein these profiles are more similar to those of GBSSII in eudicots than to those of GBSSI genes in monocots. In dicots, GBSSII must have undergone further divergence when rosids and asterids split from each other ∼126 MYA. Taken together, these findings suggest that it is GBSSII rather than GBSSI of monocots that have orthologous relationships with GBSS genes of eudicots. Moreover, diversification of GBSS genes is mainly associated with genome-wide duplication events throughout the evolutionary course of history of monocots and eudicots

Screen-printed electrochemical-based sensor for taxifolin determination in edible peanut oils

    Get PDF
An electrochemical method for the determination of Taxifolin (Tx) in peanut oil samples using screen-printed electrodes (SPEs) is reported for the first time. The different electroanalytical parameters such as scan rate, supporting electrolyte, pH and accumulation step were optimized. Interference studies were also explored for the determination of Tx in presence of daidzein, quercetin, trans-resveratrol and caffeic acid. The method developed for the Tx determination exhibits a linear response in the concentration range from 0.05 to 1 μM and a limit of detection of 0.021 μM. The electrochemical detection method proposed provides a novel, quick, low-cost, reliable and portable method for the in-situ quality control for monitoring the protected designation of origin of the “Córdoba, Argentina, Peanuts”

The objective of this study was to determine some geometric, gravimetric, and mechanical properties of lemon seeds and how these were influenced by moisture content under a range of post-storage processing conditions. The skin, pulp, and seed fractions of lemon fruits were determined. The seed fraction had a value of 1.86% with an average hull/seed ratio of 38.92%. Seed moisture content was varied by drying with hot air from the storage condition (8.28% d.b.) to a minimum moisture condition (1.31% d.b.). The drying kinetics were adequately described using a two-term exponential model, which suggested the presence of two distinct internal seed resistances to moisture transfer related to its composition. Furthermore, with the exception of thickness, all geometric properties evaluated tended to decrease as moisture content decreased, especially at the driest level evaluated. Within these, the most important variations were found in theoretical volume (24.20%) and width (22.28%), for a moisture content change of 84%. Gravimetric and mechanical properties exhibited the same tendency, with the exception of true density, which increased, and bulk density and bulk porosity which did not change significantly. It is noteworthy that rupture energy decreased by approximately 50% as the moisture content was reduced from 8.28 to 4.62%. In conclusion, drying lemon seeds generated smaller and more fragile structures that could benefit subsequent processes such as dehulling or milling, among others, but could also negatively affect seed movement through undesirable breaking. Therefore, knowledge of the properties studied in this work will be useful for the design of manufacturing and storage equipment, as well as handling and process operations

Study of the Effect of NaCl on Lipolysis in Parmigiano Reggiano Cheese

    Get PDF
Lipolysis of PDO Parmigiano Reggiano cheese from different dairies was studied to evaluate the effect of sodium chloride (NaCl) reduction on the lipidic fraction. The total and individual free fatty acid and diglyceride contents were determined for two groups of samples, normal NaCl content (1.50% p/p) and reduced NaCl content (1.37% p/p). In addition, fat, water, NaCl, and cholesterol contents were also determined. The NaCl content was 9.01% higher in the normal NaCl group than in the reduced NaCl group. The most abundant fatty acid was palmitic acid, followed by oleic, myristic, and stearic acid, which altogether accounted for approximately 75% of the total content. Cheese with a normal NaCl content presented concentrations of C8:0 and C18:0 higher than those of reduced NaCl samples, while the latter showed a higher proportion of C10:0, C14:1c, and C16:0. The total free fatty acid and diglyceride contents were higher in the reduced NaCl samples, so a 9.01% reduction in the concentration of this component could accelerate the lipolysis process. On the contrary, the free fatty acid composition profile was similar in both groups

The objective of this work was to evaluate the relation between the place where formic acid dispensers are located and falling efficacy against Varroa jacobsoni in subtropical (Tucumán) and temperate (Buenos Aires) climates. In each location forty colonies re-ceived four treatments (two or four formic acid dispensers located over brood chambers or over colony bottom). Mite counting recovered from the bottoms was done after three days intervals. Then, two PVC strips with fluvalinate were introduced to obtain the total number of mites in the colony. Formic acid falling efficacy was calculated. Falling efficacy differed significantly between both locations being 34% higher in subtropical climate. No significant difference was detected between dispenser position in the colony.Fil: Eguaras, Martin Javier. Universidad Nacional de Mar del Plata. Facultad de Ciencias Exactas y Naturales.
