Mitochondrial DNA haplogroups influence the risk of incident knee osteoarthritis in OAI and CHECK cohorts: a meta-analysis and functional study
Extended report[Abstract] Objective To evaluate the influence of the mitochondrial DNA (mtDNA) haplogroups in the risk of incident knee osteoarthritis (OA) and to explain the functional consequences of this association to identify potential diagnostic biomarkers and therapeutic targets.
Methods Two prospective cohorts contributed participants. The osteoarthritis initiative (OAI) included 2579 subjects of the incidence subcohort, and the cohort hip and cohort knee (CHECK) included 635, both with 8-year follow-up. The analysis included the association of mtDNA haplogroups with the rate of incident knee OA in subjects from both cohorts followed by a subsequent meta-analysis. Transmitochondrial cybrids harbouring haplogroup J or H were constructed to detect differences between them in relation to physiological features including specific mitochondrial metabolic parameters, reactive oxygen species production, oxidative stress and apoptosis.
Results Compared with H, the haplogroup J associates with decreased risk of incident knee OA in subjects from OAI (HR=0.680; 95% CI 0.470 to 0.968; p<0.05) and CHECK (HR=0.728; 95% CI 0.469 to 0.998; p<0.05). The subsequent meta-analysis including 3214 cases showed that the haplogroup J associates with a lower risk of incident knee OA (HR=0.702; 95% CI 0.541 to 0.912; p=0.008). J cybrids show a lower free radical production, higher cell survival under oxidative stress conditions, lower grade of apoptosis as well as lower expression of the mitochondrially related pro-apoptotic gene BCL2 binding component 3 (BBC3). In addition, J cybrids also show a lower mitochondrial respiration and glycolysis leading to decreased ATP production.
Conclusions The physiological effects of the haplogroup J are beneficial to have a lower rate of incident knee OA over time. Potential drugs to treat OA could focus on emulating the mitochondrial behaviour of this haplogroup.Instituto de Salud Carlos III; CIBERCB06/01/0040-SpainInstituto de Salud Carlos III; RETIC-RIER-RD12/0009/0018Instituto de Salud Carlos III; PI14/01254Instituto de Salud Carlos III; CP12/0319

Compilation of parameterized seismogenic sources in Iberia for the SHARE European-scale seismic source model.
Abstract: SHARE (Seismic Hazard Harmonization in Europe) is an EC-funded project (FP7) that aims to evaluate European seismic hazards using an integrated, standardized approach. In the context of SHARE, we are compiling a fully-parameterized active fault database for Iberia and the nearby offshore region. The principal goal of this initiative is for fault sources in the Iberian region to be represented in SHARE and incorporated into the source model that will be used to produce seismic hazard maps at the European scale. The SHARE project relies heavily on input from many regional experts throughout the Euro-Mediterranean region. At the SHARE regional meeting for Iberia, the 2010 Working Group on Iberian Seismogenic Sources (WGISS) was established; these researchers are contributing to this large effort by providing their data to the Iberian regional integrators in a standardized format. The development of the SHARE Iberian active fault database is occurring in parallel with IBERFAULT, another ongoing effort to compile a database of active faults in the Iberian region.
The SHARE Iberian active fault database synthesizes a wide range of geological and geophysical observations on active seismogenic sources, and incorporates existing compilations (e.g., Cabral, 1995; Silva et al., 2008), original data contributed directly from researchers, data compiled from the literature, parameters estimated using empirical and analytical relationships, and, where necessary, parameters derived using expert judgment. The Iberian seismogenic source model derived for SHARE will be the first regional-scale source model for Iberia that includes fault data and follows an internationally standardized approach (Basili et al., 2008; 2009). This model can be used in both seismic hazard and risk analyses and will be appropriate for use in Iberian- and European-scale assessments
Low-count monoclonal B-cell lymphocytosis persists after seven years of follow up and is associated with a poorer outcome
[EN]Low-count monoclonal B-cell lymphocytosis is defined by the presence
of very low numbers of circulating clonal B cells, usually phenotypically
similar to chronic lymphocytic leukemia cells, whose
biological and clinical significance remains elusive. Herein, we re-evaluated
65/91 low-count monoclonal B-cell lymphocytosis cases (54 chronic
lymphocytic leukemia-like and 11 non-chronic lymphocytic leukemialike)
followed-up for a median of seven years, using high-sensitivity
flow cytometry and interphase fluorescence in situ hybridization.
Overall, the clone size significantly increased in 69% of low-count monoclonal
B-cell lymphocytosis cases, but only one subject progressed to
high-count monoclonal B-cell lymphocytosis. In parallel, the frequency
of cytogenetic alterations increased over time (32% vs. 61% of cases,
respectively). The absolute number of the major T-cell and natural killer
cell populations also increased, but only among chronic lymphocytic
leukemia-like cases with increased clone size vs. age- and sex-matched
controls. Although progression to chronic lymphocytic leukemia was
not observed, the overall survival of low-count monoclonal B-cell lymphocytosis
individuals was significantly reduced vs. non-monoclonal Bcell
lymphocytosis controls (P=0.03) plus the general population from
the same region (P≤0.001), particularly among females (P=0.01); infection
and cancer were the main causes of death in low-count monoclonal
B-cell lymphocytosis. In summary, despite the fact that mid-term progression
from low-count monoclonal B-cell lymphocytosis to high-count
monoclonal B-cell lymphocytosis and chronic lymphocytic leukemia
appears to be unlikely, these clones persist at increased numbers, usually
carrying more genetic alterations, and might thus be a marker of an
impaired immune system indirectly associated with a poorer outcome,
particularly among females
Building and surroundings: thermal coupling
Energy building performance can be different according to outdoor conditions or urban environment, at the same time that this last assess, buildings are also affected by the building envelope, as obvious consequence of the thermal and Aeraulic coupling existing between the indoor and outdoor conditions in buildings. Thus, in this coupling is fundamental to typify the transmission phenomenon through the building envelope. Doing this, it is possible to estimate transmission heating losses and gains and also the superficial temperatures of the envelope. In order to assess the transient behaviour of the building envelope it is necessary to develop a predictive model, precise enough, to be integrated in a simulating tool. Detailed and multidimensional models, based in numerical methods, like Finite Element Method (FEM), has a high precision, but its complexity imply resources consumption and computational time, too high to be integrated in these kind of tools. On the contrary, simplified methods are good enough because they are simple and fast, with an acceptable precision in almost all the situations. The present work is focused: (a) Firstly, to develop a simplified RC-network model. The aim of the model is to characterize and to implement with
precision the behaviour of a wall in a simulating software tool based on urban environment, (b) secondly, to express in form of equivalences, the different indoor and outdoor excitations that can exist in the building envelope, and (c) finally, to calibrate the simplified model through its characteristic parameters. For a homogeneous wall and two types of excitations, it has been obtained the characteristic parameters of the model that represent the better adjustment to the real wall. In a first step, it has been obtained the results of the proposal model and a reference model based on FEM, in terms of wall external surface heat flow. Results of both models have been compared, and the resultant characteristic parameters of the model have been obtained through an optimisation method. Results for the wall and for the excitations under analysis show: (1) Characteristic longitude ec, or capacitive node position, it is determined according to a certain value of Fo equal to 2 for both excitations, this value remains constant in time, (2) useful wall thickness, on the contrary, vary as time function, according to a logarithmic law for both excitations, although this function is different depending on the considered excitation, (3) using a constant excitation, coefficients from the previous logarithmic function depends on the range of the excitation, while these are practically independent of the lineal
excitation gradient
