    Mechanistic modelling of electrochemical ageing reactions at the graphite anode of lithium-ion batteries

    Lithium-Ionen-Batterien spielen eine wichtige Rolle in einer Gesellschaft, die immer mehr von den Auswirkungen des Klimawandels betroffen ist. Daher ist es notwendig, die CO2-Emissionen und den Verbrauch fossiler Brennstoffe zu reduzieren. Gegenwärtig scheinen Lithium-Ionen-Batterien die idealen Kandidaten für diese Herausforderung zu sein, aber es bedarf weiterer Forschung und Entwicklung, um ihr Verhalten zu verstehen, ihre Grenzen zu kennen und dadurch ihre Leistung zu verbessern. Hierbei haben sich mathematische Modelle und numerische Simulation als Standardtechniken in der Forschung und Entwicklung von Lithium-Ionen-Batterien etabliert und als sehr nützlich erwiesen, um experimentelle Arbeiten zu unterstützen und die Genauigkeit von Modellen zur Lebenserwartungsvorhersage zu erhöhen. Diese Arbeit konzentriert sich auf die elektrochemischen Alterungsreaktionen in der Anode, insbesondere auf das Thema Lithium-Plating und dessen Wechselwirkung mit dem Solid-Electrolyte-Interface (SEI). Ziel dieser Arbeit ist ein tieferes Verständnis dieser Degradationsprozesse durch die Verwendung verfeinerter Modellierungsansätze und der Analyse von Simulationen über einen weiten Bereich von Betriebsbedingungen. Die zugrunde liegenden Gleichungen sind im hauseigenen multiphysikalischen Softwarepaket DENIS implementiert, für die elektrochemische Modellbeschreibung wird der Open Source Code für chemische Kinetik CANTERA verwendet. Die Entwicklung, Parametrierung und experimentelle Validierung eines umfassenden pseudo-dreidimensionalen Multiphysik-Modells einer kommerziellen Lithium-Ionen-Zelle mit Mischkathode und Graphitanode wird vorgestellt. Dieses Modell ist in der Lage, sowohl den Wärme- und Massentransport auf mehreren Skalen, als auch komplexe elektrochemische Reaktionsmechanismen zu beschreiben und zu simulieren, einschließlich der Fähigkeit, eine Mischelektrode zu simulieren, in der mehrere Aktivmaterialien einer Interkalations-/Deinterkalations-Reaktion ausgesetzt sind. Es folgt eine Erweiterung, um den reversiblen Lithium-Plating Vorgang darstellen zu können und die Vorhersage des Alterungsverhaltens über einen weiten Bereich von Bedingungen vorher sagen zu können, wobei der Schwerpunkt auf hohen Strömen und niedrigen Temperaturen liegt, die insbesondere im Feld der Schnellladung interessant sind. Dieses erweiterte Modell wird durch Vergleich mit veröffentlichten experimentellen Ergebnissen überprüft, die ein Spannungsplateau und einen Spannungsabfall als Plating-Indikatoren zeigen, und beinhaltet optional eine explizite Reinterkalationsreaktion, die makroskopische Hinweise auf Plating im speziellen Fall einer Zelle, die keine offensichtlichen Plattierungszeichen zeigt, unterdrückt. Dieses Modell wird verwendet, um Degradationskarten über einen weiten Bereich von Bedingungen und eine eingehende raum-zeitliche Analyse des Anodenverhaltens auf der mesoskopischen und mikroskopischen Skala zu erstellen, um die dynamische und nichtlineare Wechselwirkung zwischen der Interkalations-Reaktion und den Plating-Reaktionen zu demonstrieren. Es wird ein vertiefender Ausblick auf die SEI-Bildung und das SEI-Wachstum gegeben, zusammen mit der qualitativen Beschreibung von drei verschiedenen 1D-Modellen mit abnehmendem Detaillierungsgrad, die mit dem Ziel entwickelt wurden, in Zukunft idealerweise in umfassendere Multiskalen-Modelle einbezogen zu werden. Schließlich wird das erweiterte Modell erfolgreich mit einem zuvor entwickelten SEI-Modell gekoppelt, so dass ein umfassendes Modellgerüst entsteht, das in der Lage ist, sowohl Degradationsprozesse als auch deren kontinuierliche positive Rückkopplung zu simulieren

    Il biofeedback preoperatorio migliora il recupero della continenza a seguito di prostatectomia radicale: una revisione sistematica con meta-analisi

    BACKGROUND:  Postoperative urinary incontinence is the overall result of urethral sphincter incompetence and modifications in urethral length after radical prostatectomy. Findings for preoperative interventions targeted at preventing post-prostatectomy incontinence include preoperative pelvic floor muscle training (PFMT) and biodfeedback (BFB), which can be managed by nurses in many countries and have been used for decades to speed up continence recovery after surgery. AIM: to determine the effectiveness of preoperative biofeedback (BFB) for post-prostatectomy urinary incontinence compared to pelvic training without BFB, considering the variability between the results of the available studies.  METHODS: A systematic review and meta-analysis was conducted, analyzing the indications provided by the literature regarding preoperative biofeedback for preventing urinary incontinence after open radical prostatectomy, in terms of treatment regimens, timing for beginning the sessions, number of contraction and relaxation exercises, and scheduled work at home. Literature search on Pubmed, CINAHL, Cochrane Library, Web of Science, Scopus, EMBASE, and PEdro. RESULTS: Despite only three papers being suitable for metanalysis, our results support BFB over written instructions for continence recovery after both 3 and 6 moths from surgery. Implementing progressive programs with many different muscular exercises and including relaxation are the main recommendations. CONCLUSIONS: Preoperative biofeedback leads to improved urinary continence after 3 and 6 months from radical prostatectomy. Future studies should focus on the characteristics and number of pelvic muscle contractions required during biofeedback in order to maximize effectiveness.BACKGROUND: L'incontinenza urinaria postoperatoria è il risultato complessivo dell'incompetenza dello sfintere uretrale e delle modifiche della lunghezza dell'uretra dopo la prostatectomia radicale. I risultati degli interventi preoperatori mirati a prevenire l'incontinenza post-prostatectomia includono l'allenamento preoperatorio dei muscoli del pavimento pelvico (PFMT) e il biodfeedback (BFB), che possono essere gestiti dal personale infermieristico in molti Paesi. OBIETTIVO: determinare l'efficacia del biofeedback preoperatorio (BFB) per l'incontinenza urinaria post-prostatectomia rispetto al training pelvico senza BFB, considerando la variabilità dei risultati degli studi disponibili.  METODI: è stata condotta una revisione sistematica con meta-analisi, analizzando le indicazioni fornite dalla letteratura sul biofeedback preoperatorio per la prevenzione dell'incontinenza urinaria in termini di regimi di trattamento, tempi di inizio delle sessioni, numero di esercizi di contrazione e rilassamento e lavoro programmato a casa. La ricerca della letteratura è stata effettuata su Pubmed, CINAHL, Cochrane Library, Web of Science, Scopus, EMBASE e PEdro. RISULTATI: nonostante solo tre articoli siano stati adatti alla metanalisi, i nostri risultati supportano il BFB rispetto alle istruzioni scritte per il recupero della continenza dopo 3 e 6 mesi dall'intervento. L'implementazione di programmi progressivi con molti esercizi muscolari diversi e l'inclusione del rilassamento sono le principali raccomandazioni. CONCLUSIONI: Il biofeedback preoperatorio porta a un miglioramento della continenza urinaria dopo 3 e 6 mesi dalla prostatectomia radicale. Gli studi futuri dovrebbero concentrarsi sulle caratteristiche e sul numero di contrazioni muscolari pelviche richieste durante il biofeedback per massimizzare l'efficacia.Il biofeedback preoperatorio migliora il recupero della continenza dopo la prostatectomia aperta: una revisione sistematica e una meta-analis

    Deciphering OPA1 mutations pathogenicity by combined analysis of human, mouse and yeast cell models

    OPA1 is the major gene responsible for Dominant Optic Atrophy (DOA) and the syndromic form DOA “plus”. Over 370 OPA1 mutations have been identified so far, although their pathogenicity is not always clear. We have analyzed one novel and a set of known OPA1 mutations to investigate their impact on protein functions in primary skin fibroblasts and in two “ad hoc” generated cell systems: the MGM1/OPA1 chimera yeast model and the Opa1−/− MEFs model expressing the mutated human OPA1 isoform 1. The yeast model allowed us to confirm the deleterious effects of these mutations and to gain information on their dominance/recessivity. The MEFs model enhanced the phenotypic alteration caused by mutations, nicely correlating with the clinical severity observed in patients, and suggested that the DOA “plus” phenotype could be induced by the combinatorial effect of mitochondrial network fragmentation with variable degrees of mtDNA depletion. Overall, the two models proved to be valuable tools to functionally assess and define the deleterious mechanism and the pathogenicity of novel OPA1 mutations, and useful to testing new therapeutic interventions

    Genetic variants affecting NQO1 protein levels impact the efficacy of idebenone treatment in Leber hereditary optic neuropathy

    Idebenone, the only approved treatment for Leber hereditary optic neuropathy (LHON), promotes recovery of visual function in up to 50% of patients, but we can neither predict nor understand the non -responders. Idebenone is reduced by the cytosolic NAD(P)H oxidoreductase I (NQO1) and directly shuttles electrons to respiratory complex III, bypassing complex I affected in LHON. We show here that two polymorphic variants drastically reduce NQO1 protein levels when homozygous or compound heterozygous. This hampers idebenone reduction. In its oxidized form, idebenone inhibits complex I, decreasing respiratory function in cells. By retrospectively analyzing a large cohort of idebenone-treated LHON patients, classified by their response to therapy, we show that patients with homozygous or compound heterozygous NQO1 variants have the poorest therapy response, particularly if carrying the m.3460G>A/MT-ND1 LHON mutation. These results suggest consideration of patient NQO1 genotype and mitochondrial DNA mutation in the context of idebenone therapy

    Biallelic variants in LIG3 cause a novel mitochondrial neurogastrointestinal encephalomyopathy

    none67si: Abnormal gut motility is a feature of several mitochondrial encephalomyopathies, and mutations in genes such as TYMP and POLG, have been linked to these rare diseases. The human genome encodes three DNA ligases, of which only one, ligase III (LIG3), has a mitochondrial splice variant and is crucial for mitochondrial health. We investigated the effect of reduced LIG3 activity and resulting mitochondrial dysfunction in seven patients from three independent families, who showed the common occurrence of gut dysmotility and neurological manifestations reminiscent of mitochondrial neurogastrointestinal encephalomyopathy. DNA from these patients was subjected to whole exome sequencing. In all patients, compound heterozygous variants in a new disease gene, LIG3, were identified. All variants were predicted to have a damaging effect on the protein. The LIG3 gene encodes the only mitochondrial DNA (mtDNA) ligase and therefore plays a pivotal role in mtDNA repair and replication. In vitro assays in patient-derived cells showed a decrease in LIG3 protein levels and ligase activity. We demonstrated that the LIG3 gene defects affect mtDNA maintenance, leading to mtDNA depletion without the accumulation of multiple deletions as observed in other mitochondrial disorders. This mitochondrial dysfunction is likely to cause the phenotypes observed in these patients. The most prominent and consistent clinical signs were severe gut dysmotility and neurological abnormalities, including leukoencephalopathy, epilepsy, migraine, stroke-like episodes, and neurogenic bladder. A decrease in the number of myenteric neurons, and increased fibrosis and elastin levels were the most prominent changes in the gut. Cytochrome c oxidase (COX) deficient fibres in skeletal muscle were also observed. Disruption of lig3 in zebrafish reproduced the brain alterations and impaired gut transit in vivo. In conclusion, we identified variants in the LIG3 gene that result in a mitochondrial disease characterized by predominant gut dysmotility, encephalopathy, and neuromuscular abnormalities.This work was supported by Telethon Grant GGP15171 to E.B. and R.D.G. and by a donation from Kobe city to the Department of General Pediatrics, Kobe University Graduate School of Medicine (K550003302).     The power of video in science communication: the EDUlab media production

    Avvicinare la scienza e la ricerca alle persone, in ogni aspetto, significa avvicinare il pubblico a temi complicati, inspirando buone norme comportamentali di rispetto e tutela ambientale ma anche avvicinare la scienza a chi ogni giorno è in grado di decidere, attraverso i finanziamenti, quali lavori scientifici supportare. Proprio in coscienza di questo, negli ultimi decenni la scienza ha smesso di essere autoreferenziale, comprendendo che divulgare contenuti altamente scientifici a ad un pubblico non specialistico è fondamentale. Divulgare vuol dire creare quell’anello di congiunzione tra la ricerca scientifica più avanzata e il pubblico, significa saper capire il linguaggio spesso criptico dei ricercatori e di adattarlo senza stravolgimenti in qualcosa di comprensibile alle persone comuni. Con questa mission, nel 2015 nasce ufficialmente il gruppo “EDULab dell’IAS-CNR di Capo Granitola”, ovvero un laboratorio creativo di divulgazione scientifica in cui ricercatori, tecnologi e tecnici di varie discipline che condividono la passione di divulgare la scienza e i risultati da essa prodotti nei propri ambiti di competenza. Nel tempo, l’attività del gruppo si è consolidata, portando a numerosi risultati e ad un numero ragguardevole di progetti e programmi. Tra gli innumerevoli prodotti divulgativi sviluppati, l’uso dei media, ed in particolare dei video, si è rivelato uno strumento eccezionale di “comunicazione empatica” attraverso cui il pubblico riesce ad “immergersi” letteralmente nei contenuti proposti, recependo e facendo propri contenuti anche molto complessi. Nell’era del web e dei social-media, infatti, i video sono ormai lo strumento più veloce ed efficace per veicolare informazioni ad un’ampia fetta di popolazione

    ECMO for COVID-19 patients in Europe and Israel

    Since March 15th, 2020, 177 centres from Europe and Israel have joined the study, routinely reporting on the ECMO support they provide to COVID-19 patients. The mean annual number of cases treated with ECMO in the participating centres before the pandemic (2019) was 55. The number of COVID-19 patients has increased rapidly each week reaching 1531 treated patients as of September 14th. The greatest number of cases has been reported from France (n = 385), UK (n = 193), Germany (n = 176), Spain (n = 166), and Italy (n = 136) .The mean age of treated patients was 52.6 years (range 16–80), 79% were male. The ECMO configuration used was VV in 91% of cases, VA in 5% and other in 4%. The mean PaO2 before ECMO implantation was 65 mmHg. The mean duration of ECMO support thus far has been 18 days and the mean ICU length of stay of these patients was 33 days. As of the 14th September, overall 841 patients have been weaned from ECMO support, 601 died during ECMO support, 71 died after withdrawal of ECMO, 79 are still receiving ECMO support and for 10 patients status n.a. . Our preliminary data suggest that patients placed on ECMO with severe refractory respiratory or cardiac failure secondary to COVID-19 have a reasonable (55%) chance of survival. Further extensive data analysis is expected to provide invaluable information on the demographics, severity of illness, indications and different ECMO management strategies in these patients

    Producing accessible communication based on scientific research usually involves a great deal of effort in translating complex concepts into a non professional oriented information, but it is only through widely accessible data that knowledge dissemination have stronger repercussions in society. In concordance to this statement, the CNR IAMC and the “Accademia di Belle Arti di Palermo” collaborated in the work frame of the "Information, dissemination and communication system of the Biodiversity Observatory of Sicily” project, a synergistic interaction between science and art. The researchers held biodiversity seminars to the Academy students, involving 19 professorships, aiming to increase the knowledge degree and awareness on the biodiversity, thus stimulating their creativity. One of the main activities carried out during this collaboration was the conceptualization and designing of the Biodiversity Observatory brand, with the representation of biodiversity as the main briefing. The winning proposal was chosen between 13 projects by popular vote, with the participation of more than 800 students and CNR researchers. The branding is a representation of the environment with the use of different animal silhouettes as a symbol of biodiversity. The elements are arranged on a spiral grid suggesting movement, a current that raises deep, nutrient-rich waters to the ocean surface. Extremely evocative and appealing, the logotype has become an integral part of the Observatory's identity, and it's been profusely applied over the years on communication and divulgative materials, website, exhibitions, and memorabilia. Involving such a large number of students and their social groups (family, friends, and etcetera) meant giving the citizens a leading role in an important activity. Participation of the public in the creation of the graphic identity of a key structure in their territory resulted in a dramatic increase of sensitization on the biodiversity and environmental cause and participation in divulgation activities in the following years
