    Efeito da claritromicina na celularidade do lavado broncoalveolar em camundongos com doença pulmonar neutrofílica induzida

    OBJECTIVE: Macrolide antibiotics have anti-inflammatory properties in lung diseases. The aim of this study was to investigate the effect of clarithromycin in pulmonary cellular inflammatory response in mice. METHOD: Eight adult Swiss mice were studied. All animals received an intranasal challenge (80 µL) with dead Pseudomonas aeruginosa (1.0 x 10(12) CFU/mL). Bronchoalveolar lavage was performed 2 days later, with total cell count and differential cell analysis. The study group (n = 4) received clarithromycin treatment (50 mg/kg/day, intraperitoneal) for 5 days. Treatment was initiated 2 days before intranasal challenge. RESULTS: There was no significant difference in total cell count between the groups (mean: 2.0 x 10(6) and 1.3 x 10(6), respectively). In both groups, there was a predominance of neutrophils. However, the study group had a higher percentage of lymphocytes in the bronchoalveolar lavage than the control group (median of 19% vs 2.5%, P = .029). CONCLUSION: Clarithromycin alters the cytological pattern of bronchoalveolar lavage of Swiss mice with neutrophil pulmonary inflammation, significantly increasing the percentage of lymphocytes.OBJETIVO: Os antibióticos macrolídeos podem apresentar um efeito antiinflamatório em doenças pulmonares. O objetivo deste estudo é investigar o efeito da claritromicina na resposta inflamatória celular pulmonar em camundongos Swiss. MÉTODO: Foram utilizados 8 camundongos Swiss adultos (6-8 semanas). Todos os animais receberam um desafio intranasal (80 µL) com Pseudomonas aeruginosa mortas (1 x 10(12) UFC/mL). Dois dias após o desafio, foi realizado lavado broncoalveolar (LBA) com contagem total de células (CTC) e exame citológico diferencial. O grupo em estudo (n=4) recebeu tratamento com claritromicina (50mg/kg/dia, intraperitoneal) por 5 dias, sendo iniciado o tratamento 2 dias antes do desafio intranasal. O grupo controle (n=4) não recebeu tratamento com claritromicina. RESULTADOS: Não houve diferença significativa na CTC entre os grupos (média de 2x10(6) e 1,3x10(6), respectivamente). Em ambos os grupos, houve predomínio absoluto de neutrófilos. Contudo, o grupo tratado com claritromicina, apresentou um número percentual significativamente maior de linfócitos no LBA (mediana de 2,5% vs 19%, p=0,029). CONCLUSÃO: O uso de claritromicina altera o exame citológico diferencial do lavado bronco-alveolar de camundongos Swiss com inflamação pulmonar neutrofílica, aumentando significativamente o número percentual de linfócitos

    Histochemistry of Equine Damaged Tendons, Ligaments and Articular Cartilage

    Background: The injury repair process in tendons and ligaments includes different phases such as inflammation, neovascularization, fibroblast proliferation and fibrosis. Collagen type and tissue characteristics of tendon and ligament repair are described such as type collagen differentiation and properties of the scars tissue. The degeneration of articular cartilage when, characterized by loss of the articular layers associated of the decreased of proteoglycans. The aim of this study is to describe by histochemistry techniques the characteristics of tissue scar, collagen type in the repair process of tendons and ligaments, as well as articular cartilage degeneration.Materials, Methods & Results: Tissue samples of equine tendons, ligaments and articular cartilage of the metacarpophalangeal joint region were evaluated by ultrasonography, macroscopically and prepared for routine histopathology (H&E staining). The inclusion criterion of the samples in this study was based on the presence of lesions characterized in H&E stain as fibroplasia, neovascularization, collagenolysis, chondroid metaplasia in tendons and ligaments and fibrillation and cartilaginous eburnation lesions in the articular cartilage samples. The Masson’s trichrome, Picrosirius red and Alcian blue staining techniques were also performed in addition to H&E. Pathologic findings in the tendons and ligaments included fibroplasia, collagenolysis, chondroid metaplasia and lymphohistioplasmacytic inflammation. Tendons and ligaments scars were composed of type III collagen but there was also some type I collagen. Fiber alignment of tendons and ligaments in the reorganization tissue was not flawless and the fiber appearance was characterized by a lack of the fiber crimp and parallelism. The fibroplasia was characterized by endotendinous tickening areas associated with the presence of loose connective tissue. In the areas of loose connective tissue substitution, collagen type fibers are intercalated to a lesser extent by type-III collagen fibers. In the Alcian blue stained samples of articular cartilage observed the surface layer and the matrix zone of calcified cartilage were weakly stained in blue.Discussion: Three special stains were utilized in this study along with the H&E evaluation elucidating the behavior tendons, ligaments and articular cartilage injury. The important observation in this study was fibroplasia in tendons and ligaments seems to be composed by abundant of loose connective tissue, chondrocytes and intermingled collagen type I and III fibers associated with lack of crimps alignment of the fibers. The fragile structure suggested by the Masson’s trichrome stain results (presence of the loose connective tissue) in this study perhaps make the tendons and ligaments receptive to other lesions. The characteristic blue discoloration of collagen fibers was only observed in the loose connective tissue may be because the dye penetration becomes easier when compared to the dense connective tissue (stained in red). The Masson’s trichrome made possible the differentiated the dense connective tissue of the loose connective tissue. The combined histochemistry staining technics allowed an improved characterization of fiber alignment, collagen type, inflammatory cell infiltration and neovascularization, which happens during the repair process of tendons and ligaments. The fibrillation and eburnation of the articular cartilage were associated with the decrease Alcian Blue staining characterized by degeneration process of articular cartilage

    This research aimed to learn about the flight attendants' work and reflect on actions that prevent and promote these professionals' mental health. To accomplish this, eight professionals from three different airlines were interviewed through individual semi-structured interviews. It was identified that the main tasks of the flight attendants are frequently repeated, such as checking safety and emergency equipment and checking the in-flight service. The positive aspect of the profession most mentioned by the participants is the possibility of getting to know different places and cultures. Among the negative points are the difficulty of being present at moments considered socially important and the physical fatigue. In an attempt to compensate for this tiredness, many flight attendants end up using substances to be able to sleep. The resources offered by companies for mental health care are seen as ineffective. It was concluded that the alternating scales and work routines of the flight attendants affect health conditions and end up generating invisibilities in the mental health care of these professionals. Therefore, it is important to strengthen workers' health actions in this sector given the adversities inherent in the work environment.This research aimed to learn about the flight attendants' work and reflect on actions that prevent and promote these professionals' mental health. Para isso, foram entrevistados oito profissionais de três empresas aéreas diferentes, através de entrevistas individuais semiestruturadas. Identificou-se que as principais tarefas dos(as) comissários(as) se repetem com frequência, como o check de equipamentos de segurança e emergência e conferência de serviço de bordo. O fator mais citado como positivo pelos participantes é a possibilidade de conhecer diversos lugares e culturas. Já entre os pontos negativos da profissão estão a dificuldade de se fazer presente em eventos familiares considerados socialmente importantes e o cansaço físico. Na tentativa de compensar esse cansaço, muitos(as) comissários(as) acabam usando substâncias para poder dormir. Os recursos oferecidos pelas empresas para o cuidado em saúde mental são vistos como ineficazes, ou pouco efetivos. Conclui-se, que as escalas alternadas e rotinas de trabalho dos(as) comissários (as) afeta as condições de saúde e acaba gerando invisibilidades no cuidado em saúde mental destes profissionais. Logo, é importante fortalecer as ações de saúde do trabalhador neste setor em vista das adversidades inerentes ao meio de trabalho

    RESUMO: O objetivo deste estudo foi descrever e caracterizar as estruturas que compõem a articulação metacarpofalangena equina por meio de análise ultrassonográfica, anatômica e histológica. Membros torácicos equinos (=70), obtidos em instalações frigoríficas, foram submetidos a exame ultrassonográfico post mortem. Destes, 30 membros apresentaram imagens ultrassonográficas consideradas sem alterações. Posteriormente foi realizada a dissecação dos mesmos e o estudo anátomo-histológico. Critérios como tamanho, forma, arquitetura e ecogenicidade foram observados a fim de caracterizar as imagens ultrassonográficas, anatômicas e histológicas normais das estruturas ligamentares, tendíneas, capsulares e cartilaginosas da articulação metacarpofalangeana equina

    A utilização de antimicrobianos melhoradores de desempenho na criação de frangos de corte é uma prática comum na indústria avícola e, no Brasil, seu uso deve ser feito de acordo com os limites determinados pelo Ministério da Agricultura Pecuária e Abastecimento (MAPA).Objetivou-se com o presente trabalho determinar o efeito da utilização da bacitracina de zinco, dentro dos níveis recomendados pelo Ministério da Agricultura Pecuária e Abastecimento (MAPA), em dietas para frangos de corte sobre o desempenho de aves e criadas de 1 a 42 dias de idade. Utilizou-se o delineamento inteiramente casualizado, com 5 tratamentos, distribuídos em 10 repetições de 25 pintos machos de corte da linhagem Cobb 500®, totalizando 1250 aves. Os tratamentos experimentais foram constituídos de cinco níveis de inclusão de bacitracina de zinco (0, 10, 25, 40 e 55ppm). As variáveis analisadas foram consumo de ração, ganho de peso, conversão alimentar e avaliação econômica das dietas. Para o período total de criação, avaliou-se também a viabilidade dos frangos de corte. A bacitracina de zinco foi eficiente como beneficiador do desempenho de frangos de corte criados no período de 1 a 21, 1 a 35 e 1 a 42 dias. Para o período de 1 a 21 dias, o uso de 55 e 29,3ppm de bacitracina de zinco resultou em menor consumo de ração e maior ganho de peso, respectivamente. No geral, a melhor conversão alimentar foi obtida quando se adicionou 55ppm de bacitracina de zinco na ração de frangos de corte

    Development of an experimental model of neutrophilic pulmonary response induction in mice

    BACKGROUND: Several lung diseases are characterized by a predominantly neutrophilic inflammation. A better understanding of the mechanisms of action of some drugs on the airway inflammation of such diseases may bring advances to the treatment. OBJECTIVE: To develop a method to induce pulmonary neutrophilic response in mice, without active infection. METHODS: Eight adult Swiss mice were used. The study group (n = 4) received an intranasal challenge with 1 x 10(12) CFU/ml of Pseudomonas aeruginosa (Psa), frozen to death. The control group (n = 4) received an intranasal challenge with saline solution. Two days after the intranasal challenge, a bron­choalveolar lavage (BAL) was performed with total cell and differential cellularity counts. RESULTS: The total cell count was significantly higher in the group with Psa, as compared to the control group (median of 1.17 x 10(6) and 0.08 x 10(6), respectively, p = 0.029). In addition to this, an absolute predominance of neutrophils was found in the differential cellularity of the mice that had received the Psa challenge. CONCLUSIONS: The model of inducing a neutrophilic pulmonary disease using frost-dead bacteria was successfully developed. This neutrophilic inflammatory response induction model in Swiss mice lungs may be an important tool for testing the anti-inflammatory effect of some antimicrobial drugs on the inflammation of the lower airways
