    Transcriptional analysis of late ripening stages of grapevine berry

    * Background: The composition of grapevine berry at harvest is a major determinant of wine quality. Optimal oenological maturity of berries is characterized by a high sugar/acidity ratio, high anthocyanin content in the skin, and low astringency. However, harvest time is still mostly determined empirically, based on crude biochemical composition and berry tasting. In this context, it is interesting to identify genes that are expressed/repressed specifically at the late stages of ripening and which may be used as indicators of maturity. * Results: Whole bunches and berries sorted by density were collected in vineyard on Chardonnay (white cultivar) grapevines for two consecutive years at three stages of ripening (7-days before harvest (TH-7), harvest (TH), and 10-days after harvest (TH+10)). Microvinification and sensory analysis indicate that the quality of the wines made from the whole bunches collected at TH-7, TH and TH+10 differed, TH providing the highest quality wines. In parallel, gene expression was studied with Qiagen/Operon microarrays using two types of samples, i.e. whole bunches and berries sorted by density. Only 12 genes were consistently up-or down-regulated in whole bunches and density sorted berries for the two years studied in Chardonnay. 52 genes were differentially expressed between the TH-7 and TH samples. In order to determine whether these genes followed a similar pattern of expression during the late stages of berry ripening in a red cultivar, nine genes were selected for RT-PCR analysis with Cabernet Sauvignon grown under two different temperature regimes affecting the precocity of ripening. The expression profiles and their relationship to ripening were confirmed in Cabernet Sauvignon for seven genes, encoding a carotenoid cleavage dioxygenase, a galactinol synthase, a late embryogenesis abundant protein, a dirigent-like protein, a histidine kinase receptor, a valencene synthase and a putative S-adenosyl-L-methionine: salicylic acid carboxyl methyltransferase. * Conclusions: This set of up-and down-regulated genes characterize the late stages of berry ripening in the two cultivars studied, and are indirectly linked to wine quality. They might be used directly or indirectly to design immunological, biochemical or molecular tools aimed at the determination of optimal ripening in these cultivars

    Gene expression profiling in susceptible interaction of grapevine with its fungal pathogen Eutypa lata: Extending MapMan ontology for grapevine

    <p>Abstract</p> <p>Background</p> <p>Whole genome transcriptomics analysis is a very powerful approach because it gives an overview of the activity of genes in certain cells or tissue types. However, biological interpretation of such results can be rather tedious. MapMan is a software tool that displays large datasets (e.g. gene expression data) onto diagrams of metabolic pathways or other processes and thus enables easier interpretation of results. The grapevine (<it>Vitis vinifera</it>) genome sequence has recently become available bringing a new dimension into associated research. Two microarray platforms were designed based on the TIGR Gene Index database and used in several physiological studies.</p> <p>Results</p> <p>To enable easy and effective visualization of those and further experiments, annotation of <it>Vitis vinifera </it>Gene Index (VvGI version 5) to MapMan ontology was set up. Due to specificities of grape physiology, we have created new pictorial representations focusing on three selected pathways: carotenoid pathway, terpenoid pathway and phenylpropanoid pathway, the products of these pathways being important for wine aroma, flavour and colour, as well as plant defence against pathogens. This new tool was validated on Affymetrix microarrays data obtained during berry ripening and it allowed the discovery of new aspects in process regulation. We here also present results on transcriptional profiling of grape plantlets after exposal to the fungal pathogen <it>Eutypa lata </it>using Operon microarrays including visualization of results with MapMan. The data show that the genes induced in infected plants, encode pathogenesis related proteins and enzymes of the flavonoid metabolism, which are well known as being responsive to fungal infection.</p> <p>Conclusion</p> <p>The extension of MapMan ontology to grapevine together with the newly constructed pictorial representations for carotenoid, terpenoid and phenylpropanoid metabolism provide an alternative approach to the analysis of grapevine gene expression experiments performed with Affymetrix or Operon microarrays. MapMan was first validated on an already published dataset and later used to obtain an overview of transcriptional changes in a susceptible grapevine – <it>Eutypa lata </it>interaction at the time of symptoms development, where we showed that the responsive genes belong to families known to be involved in the plant defence towards fungal infection (PR-proteins, enzymes of the phenylpropanoid pathway).</p

    Should my automated car drive as I do? Investigating speed preferences of drivengers in various driving conditions

    International audienceStudies investigating the question of how automated cars (ACs) should drive converge to show that a personalized automated driving-style, i.e., mimicking the driving-style of the human behind the wheel, has a positive influence on various aspects of his experience (e.g., comfort). However, few studies have investigated the fact that these benefits might vary with respect to driver-related variables, such as trust in ACs, and contextual variables of the driving activity, such as weather conditions. Additionally, the context of intermediate levels of automation, such as SAE level 3, remains largely unexplored. The objective of this study was to investigate these points. In a scenario-based experimental protocol, participants were exposed to written scenarios in which a character is driven by a SAE level 3 AC in different combinations of conditions (i.e., types of roads, weather conditions and traffic congestion levels). For each condition, participants were asked to indicate how fast they would prefer their AC to drive and how fast they would manually drive in the same situation. Through analyses of variance and equivalence tests, results showed a tendency for participants to overall prefer a slightly lower AC speed than their own. However, a linear regression analysis showed that while participants with the lowest levels of trust preferred an AC speed lower than theirs, those with the highest levels preferred an AC speed nearly identical to theirs. Overall, the results of this study suggest that it would be more beneficial to implement a personalization approach for the design of automated driving-styles rather than a one for all approach

    Effects of environmental, vehicle and human factors on comfort in partially automated driving: A scenario-based study

    International audienceAlthough it is key to improving acceptability, there is sparse scientific literature on the experience of humans as passengers in partially automated cars. The present study therefore investigated the influence of road type, weather conditions, traffic congestion level, vehicle speed, and human factors (e.g., trust in automated cars) on passenger comfort in an automated car classified as Level 3 according to the Society of Automotive Engineers (SAE). Participants were exposed to scenarios in which a character is driven by an SAE Level 3 automated car in different combinations of conditions (e.g., highway Ă— heavy rain Ă— very congested traffic Ă— vehicle following prescribed speed). They were asked to rate their perceived comfort as if they were the protagonist. Results showed that comfort was negatively affected by driving in downtown (vs. highway), heavy rain, and congested traffic. Interaction analyses showed that reducing the speed of the vehicle improved comfort in these two last conditions, considered either individually or in combination. Cluster analysis revealed four profiles: trusting in automation, averse to speed reduction, risk averse, and mistrusting automation. These profiles were all influenced differently by the driving conditions, and corresponded to varying levels of trust in automated cars. This study suggests that optimizing comfort in automated cars should take account of both driving conditions and human profiles

    Quantification of Condensed Tannins in Sainfoin Powder (Onobrychisviciifolia) by FT-NIR Spectroscopy

    Models based on FT-NIR spectroscopy and PLS-regressions were developed over three harvest years to determine the condensed tannins contents (CT, %(w/w)) in sainfoin powders. The three data sets corresponding to the three harvest years were used as calibration and then validation sets, successively. Finally, a global model gathering all the three years data sets has been developed. The developed models predict CT in a range of 2.06 to 11.28 %(w/w). The accuracy of the models depended on the range of CT-values of the calibration and validation sets. Finally, it was possible to predict the CT with a SEP-value lower than 0.5% (w/w) and R2-value higher than 0.9. In the present study, some of the PLS-parameters such as bias, slope and SEP have been statistically evaluated using the international standard ISO 12099:2017. The final global model was very promising since bias was not significantly different from 0, the slope was not significantly different from 1, and obtained SEP was 0.49% (w/w) while the calculated SEP-limit was 0.47% (w/w). The presently developed model was robust over the three years and the global model presented very interesting values. Such approach would be very useful to develop a new quantitative, rapid and low cost method to assess the CT of sainfoin powders. This method will allow us to free ourselves from the traditional chemical method that consumes time, money and chemicals
