    In this thesis, I will reflect on issues of nostalgia through a series of work based upon the landscape where I grew up. I discuss how my paintings stage themes and negotiate criticality towards memory. The writing analyses my engagement towards the collaging of documentary and fictional sources: and how my paintings seek to dispel legibility and embrace slippage. Alongside discussions of personal experiences, I talk about the relationship of distancing in my practice, concerning tangible and geographic detachment. The thesis expresses my adoration for the history of British painting, and my efforts to internalize its traditions.     Industrial-Age Doubling of Snow Accumulation in the Alaska Range Linked to Tropical Ocean Warming

    Future precipitation changes in a warming climate depend regionally upon the response of natural climate modes to anthropogenic forcing. North Pacific hydroclimate is dominated by the Aleutian Low, a semi-permanent wintertime feature characterized by frequent low-pressure conditions that is influenced by tropical Pacific Ocean temperatures through the Pacific-North American (PNA) teleconnection pattern. Instrumental records show a recent increase in coastal Alaskan precipitation and Aleutian Low intensification, but are of insufficient length to accurately assess low frequency trends and forcing mechanisms. Here we present a 1200-year seasonally- to annually-resolved ice core record of snow accumulation from Mt. Hunter in the Alaska Range developed using annual layer counting and four ice-flow thinning models. Under a wide range of glacier flow conditions and layer counting uncertainty, our record shows a doubling of precipitation since ~1840 CE, with recent values exceeding the variability observed over the past millennium. The precipitation increase is nearly synchronous with the warming of western tropical Pacific and Indian Ocean sea surface temperatures. While regional 20th Century warming may account for a portion of the observed precipitation increase on Mt. Hunter, the magnitude and seasonality of the precipitation change indicate a long-term strengthening of the Aleutian Low

    Identification of activity-induced Egr3-dependent genes reveals genes associated with DNA damage response and schizophrenia

    Bioinformatics and network studies have identified the immediate early gene transcription factor early growth response 3 (EGR3) as a master regulator of genes differentially expressed in the brains of patients with neuropsychiatric illnesses ranging from schizophrenia and bipolar disorder to Alzheimer\u27s disease. However, few studies have identified and validated Egr3-dependent genes in the mammalian brain. We have previously shown that Egr3 is required for stress-responsive behavior, memory, and hippocampal long-term depression in mice. To identify Egr3-dependent genes that may regulate these processes, we conducted an expression microarray on hippocampi from wildtype (WT) and Egr3-/- mice following electroconvulsive seizure (ECS), a stimulus that induces maximal expression of immediate early genes including Egr3. We identified 69 genes that were differentially expressed between WT and Egr3-/- mice one hour following ECS. Bioinformatic analyses showed that many of these are altered in, or associated with, schizophrenia, including Mef2c and Calb2. Enrichr pathway analysis revealed the GADD45 (growth arrest and DNA-damage-inducible) family (Gadd45b, Gadd45g) as a leading group of differentially expressed genes. Together with differentially expressed genes in the AP-1 transcription factor family genes (Fos, Fosb), and the centromere organization protein Cenpa, these results revealed that Egr3 is required for activity-dependent expression of genes involved in the DNA damage response. Our findings show that EGR3 is critical for the expression of genes that are mis-expressed in schizophrenia and reveal a novel requirement for EGR3 in the expression of genes involved in activity-induced DNA damage response

    An integrated transcriptomics-guided genome-wide promoter analysis and next-generation proteomics approach to mine factor(s) regulating cellular differentiation

    Differential next-generation-omics approaches aid in the visualization of biological processes and pave the way for divulging important events and/or interactions leading to a functional output at cellular or systems level. To this end, we undertook an integrated Nextgen transcriptomics and proteomics approach to divulge differential gene expression of infant and pubertal rat Sertoli cells (Sc).Unlike, pubertal Sc, infant Sc are immature and fail to support spermatogenesis. We found exclusive association of 14 and 19 transcription factor binding sites to infantile and pubertal states of Sc, respectively, using differential transcriptomics-guided genome-wide computational analysis of relevant promoters employing 220 Positional Weight Matrices from the TRANSFAC database. Proteomic SWATH-MS analysis provided extensive quantification of nuclear and cytoplasmic protein fractions revealing 1,670 proteins differentially located between the nucleus and cytoplasm of infant Sc and 890 proteins differentially located within those of pubertal Sc. Based on our multi-omics approach, the transcription factor YY1 was identified as one of the lead candidates regulating differentiation of Sc.YY1 was found to have abundant binding sites on promoters of genes upregulated during puberty. To determine its significance, we generated transgenic rats with Sc specific knockdown of YY1 that led to compromised spermatogenesis

    Senegal, with 196,712 km2 land area, is located at the extreme west of the African continent (Longitudes 11°21W - 17°32N and Latitudes 12°8N - 16°41N). The country’s soils are in general of low fertility, fragile and very susceptible to wind and water erosion. The climate is of Sudano-Sahelian type characterized by alternating dry season (November to May) and rainy season (June to October). The 700 km coastline brings climatic differences between coastal areas and inland zones. Rainfall amount follows a latitudinal variation going from 300 mm in the north semi-desertic areas to 1200 mm in the south. Senegal is divided into 7 agro-ecological zones for management perspectives: River Valley, Niayes, Groundnut Basin (North and South), Silvo-Pastoral zone, Eastern Senegal and Upper Casamance, Lower Casamance (CIAT-BFS/USAID, 2016). The country’s economy is mainly driven by crop and livestock production contributing 17% of the GDP and employing about 70% of the population (NAPA, Republic of Senegal 2006). Like other sub-Saharan African countries, Senegal faces food insecurity as a consequence of climate variability and change combined with other global changes (Zougmoré et al., 2015)
