    On-Line influenza virus quantification for viral production processes thanks to affinity-based surface plasmon resonance biosensor

    Influenza virus seasonal epidemics, associated with the constant threat of new pandemic outbreak, challenge vaccine manufacturers to develop responsive processes that can outreach the limitations of traditional egg-based technology. Recent progress made regarding cell culture bioprocesses allowed for numerous alternative strategies to developed future vaccine candidates, as for example the recombinant HA or Virus—like Particles (VLP) vaccines. However, while cell culture allows for more versatility than ovoculture, regarding process development and monitoring, these alternatives still require optimization to seriously concurrence the traditional process. To drive these developments, WHO and regulatory agencies underlined the need for developing better influenza vaccine potency assays1,2. Actual influenza vaccine formulation and lot release rely on single-radial immunodiffusion (SRID) assay, which requires strain-specific reference sera and antigen reagents. However, the annual preparation of these reagents takes between 2 to 6 months and constitutes a critical bottleneck for the release of vaccine lots3. Additionally, SRID is not implementable for process development as such technique cannot handle in-process low concentrated and non-purified material. We developed an assay for rapid and label-free quantification of influenza hemagglutinin (HA) antigen and influenza virus based on surface plasmon resonance (SPR). The method is based on affinity capture of hemagglutinin antigen by sialic-acid terminated glycans present at the surface of the fetuin-functionalized sensor. Conditions were optimized for the regeneration of the surface, in order to run multiple sequential analyses on a unique sensor. Two types of purified standard were used during the development of the assay. Commercial trivalent inactivated vaccine (“TIV”) has been used for the determination of optimal analytical conditions, while a stock of split inactivated H1N1 virus has been produced and calibrated in our laboratory to study the specific response obtained toward this HA subtype. This assay offers a quantification of influenza hemagglutinin within minutes with a wide dynamic range (30 ng/mL-20 µg/mL). Also, the technique provides a limit of detection (LOD) 100 times lower than SRID, and a better reproducibility than SRID and its potential alternatives recently proposed (1,4,5. Additionally, the applicability of this assay for an on-line vaccine production monitoring has been validated by off-line measurement of influenza H1N1 virus particles derived from cell culture supernatant. Such a test allowed to achieve a LOD of 106 Infectious Viral Particles/mL Thus, our assay provides an innovative tool to evaluate influenza new vaccine bioprocesses, from viral production kinetics in mammalian cell culture to vaccine potency evaluation

    Multi-elastic Datacenters: Auto-scaled Virtual Clusters on Energy-Aware Physical Infrastructures

    [EN] Computer clusters are widely used platforms to execute different computational workloads. Indeed, the advent of virtualization and Cloud computing has paved the way to deploy virtual elastic clusters on top of Cloud infrastructures, which are typically backed by physical computing clusters. In turn, the advances in Green computing have fostered the ability to dynamically power on the nodes of physical clusters as required. Therefore, this paper introduces an open-source framework to deploy elastic virtual clusters running on elastic physical clusters where the computing capabilities of the virtual clusters are dynamically changed to satisfy both the user application's computing requirements and to minimise the amount of energy consumed by the underlying physical cluster that supports an on-premises Cloud. For that, we integrate: i) an elasticity manager both at the infrastructure level (power management) and at the virtual infrastructure level (horizontal elasticity); ii) an automatic Virtual Machine (VM) consolidation agent that reduces the amount of powered on physical nodes using live migration and iii) a vertical elasticity manager to dynamically and transparently change the memory allocated to VMs, thus fostering enhanced consolidation. A case study based on real datasets executed on a production infrastructure is used to validate the proposed solution.     This article presents an optical see-through (OST) Augmented Reality system for the treatment of phobia to small animals. The technical characteristics of the OST system are described, and a comparative study of the sense of presence and anxiety in a nonphobic population (24 participants) using the OST and an equivalent video see-though (VST) system is presented. The results indicate that if all participants are analyzed, the VST system induces greater sense of presence than the OST system. If the participants who had more fear are analyzed, the two systems induce a similar sense of presence.     Objective: The aim of this study was to determine whether hypercholesterolemia increases articular damage in a rabbit model of chronic arthritis. Methods: Hypercholesterolemia was induced in 18 rabbits by administrating a high-fat diet (HFD). Fifteen rabbits were fed normal chow as controls. Chronic antigen-induced arthritis (AIA) was induced in half of the HFD and control rabbits, previously immunized, by intra-articular injections of ovalbumin. After sacrifice, lipid and systemic inflammation markers were analyzed in blood serum. Synovium was analyzed by Krenn score, multinucleated cell counting, immunohistochemistry of RAM11 and CD31, and TNF-a and macrophage chemoattractant protein-1 (MCP-1) gene expression. Active bone resorption was assessed by protein expression of receptor activator of nuclear factor kappa-B ligand (RANKL), osteoprotegerin (OPG) and quantification of cathepsin K, contact surface and the invasive area of pannus into bone. Results: Rabbits receiving the HFD showed higher total serum cholesterol, HDL, triglycerides and CRP levels than rabbits fed a normal diet. Synovitis score was increased in HFD, and particularly in AIA and AIA + HFD groups. AIA + HFD synovium was characterized by a massive infiltration of RAM11+ cells, higher presence of multinucleated foam cells and bigger vascularization than AIA. Cathepsin K+ osteoclasts and the contact surface of bone resorbing pannus were also increased in rabbits with AIA + HFD compared with AIA alone. Synovial TNF-a and MCP-1 gene expression was increased in AIA and HFD rabbits compared with healthy animals. RANKL protein expression in AIA and AIA + HFD groups was higher compared with either HFD or normal groups.     Un cuerpo teórico importante de investigación sugiere que el estilo autoritativo de los padres se asocia con unas mejoras en diversos aspectos de ajuste psicosocial de los adolescentes. Este artículo desarrolla y analiza la tipología de los estilos educativos parentales en el contexto de jóvenes filipinos. A partir de una muestra de 3.601 estudiantes, se muestra que el estilo autoritativo se asocia con mayor autoestima, menor impulsividad, mayor intención de alcanzar metas positivas en el futuro, mejor rendimiento académico, menor prevalencia de experiencia sexual y mayor rechazo de diversas formas de sexismo
