    Search for Anti-EA(D) Antibodies in Subjects with an “Isolated VCA IgG” Pattern

    The presence of an “isolated viral capsid antigen (VCA) IgG” pattern in serum is not easy to interpret without the aid of further tests, such as specific immunoblotting or a virus genome search, that often give rise to organisational and economic problems. However, one alternative is to use an enzyme-linked immunosorbent assay (ELISA) to detect anti-early antigen (EA) antibodies, which can be found in about 85% of subjects with acute Epstein-Barr virus (EBV) infections. The purpose of this work was to search for anti-EA(D) antibodies in 130 samples with an isolated VCA IgG pattern at ELISA screening and classified as being indicative of past (102 cases) or acute (28 cases) infection on the basis of the immunoblotting results. Thirty-seven samples (28.5%) were positive for anti-EA(D), of which 25 (89.3%) had been classified by immunoblotting as indicating acute and 12 (11.8%) past EBV infection. This difference was statistically significant (P < .01). The results of our search for anti-EA(D) antibodies correctly identified nearly 90% of acute (presence) or past EBV infections (absence). When other tests are not available, the search for anti-EA antibodies may therefore be helpful in diagnosing patients with an isolated VCA IgG pattern at screening tests

    Innovative approaches to evaluate sugar beet responses to changes in sulfate availability

    In this study, a system based on omics profiling was set-up for sugar beet (Beta vulgaris L. subsp. vulgaris) evaluation after changes in sulfate availability. Seedlings were grown on sulfate-deprived Hoagland solution. Six days after germination, 100 \ub5M MgSO4was added to the solution. Root samples were collected 36 h after treatments. WinRHIZO root-scanning approach was used for the automated image analysis of plant root morphology. Inductively Coupled Plasma Spectrometry (ICP-OES) and quadrupole-time-of-flight mass spectrometry (Q-TOF) were used for ionomic and metabolic analysis, respectively. Nanofluidic real-time PCR (OpenArray system) was used for molecular profiling. OpenArray chips were designed with TaqMan probes for 53 sugar beet genes putatively involved in sulfate nutrition. At morphological level treated seedlings showed significantly higher values (P < 0.01) than untreated plants for root traits related to soil exploration and nutrient uptake, such as total root length, fine roots length and root tips number. ICP-OES, Q-TOF and transcriptomic data revealed changes due to sulfate availability in sugar beet samples. Two key results are highlighted in sulfate-supplied roots and leaves. Firstly, high expression levels of auxin efflux carrier component 1 (PIN) and 5-phosphoribosyl-anthranilate, precursor of tryptophan and auxin synthesis, were observed in roots. Secondly, high levels of 2-Cys peroxiredoxin BAS1, chloroplastic, thioredoxin reductase (NADPH) and cysteine synthase, chloroplastic/chromoplastic, O-acetylserine sulfhydrylase, involved in protection against oxidative stress and cysteine synthase activity, respectively, were observed in leaves. Based on our findings, the combination of evaluated omics approaches could become a key system for the evaluation of the nutritional status of sugar beet under different nutrient availability conditions

    The association of indwelling urinary catheter with delirium in hospitalized patients and nursing home residents: an explorative analysis from the "Delirium Day 2015"

    Backround: Use of indwelling urinary catheter (IUC) in older adults has negative consequences, including delirium. Aim: This analysis, from the "Delirium Day 2015", a nationwide multicenter prevalence study, aim to evaluate the association of IUC with delirium in hospitalized and Nursing Homes (NHs) patients. Methods: Patients underwent a comprehensive geriatric assessment, including the presence of IUC; inclusion criteria were age &gt; 65 years, being Italian speaker and providing informed consent; exclusion criteria were coma, aphasia, end-of-life status. Delirium was assessed using the 4AT test (score ≥ 4: possible delirium; scores 1-3: possible cognitive impairment). Results: Among 1867 hospitalized patients (mean age 82.0 ± 7.5 years, 58% female), 539 (28.9%) had IUC, 429 (22.9%) delirium and 675 (36.1%) cognitive impairment. IUC was significantly associated with cognitive impairment (OR 1.60, 95% CI 1.19-2.16) and delirium (2.45, 95% CI 1.73-3.47), this latter being significant also in the subset of patients without dementia (OR 2.28, 95% CI 1.52-3.43). Inattention and impaired alertness were also independently associated with IUC. Among 1454 NHs residents (mean age 84.4 ± 7.4 years, 70.% female), 63 (4.3%) had IUC, 535 (36.8%) a 4AT score ≥ 4, and 653 (44.9%) a 4AT score 1-3. The multivariate logistic regression analysis did not show a significant association between 4AT test or its specific items with IUC, neither in the subset of patients without dementia. Discussion: We confirmed a significant association between IUC and delirium in hospitalized patients but not in NHs residents. Conclusion: Environmental and clinical factors of acute setting might contribute to IUC-associated delirium occurrence

    &quot;Delirium Day&quot;: A nationwide point prevalence study of delirium in older hospitalized patients using an easy standardized diagnostic tool

    Background: To date, delirium prevalence in adult acute hospital populations has been estimated generally from pooled findings of single-center studies and/or among specific patient populations. Furthermore, the number of participants in these studies has not exceeded a few hundred. To overcome these limitations, we have determined, in a multicenter study, the prevalence of delirium over a single day among a large population of patients admitted to acute and rehabilitation hospital wards in Italy. Methods: This is a point prevalence study (called "Delirium Day") including 1867 older patients (aged 65 years or more) across 108 acute and 12 rehabilitation wards in Italian hospitals. Delirium was assessed on the same day in all patients using the 4AT, a validated and briefly administered tool which does not require training. We also collected data regarding motoric subtypes of delirium, functional and nutritional status, dementia, comorbidity, medications, feeding tubes, peripheral venous and urinary catheters, and physical restraints. Results: The mean sample age was 82.0 ± 7.5 years (58 % female). Overall, 429 patients (22.9 %) had delirium. Hypoactive was the commonest subtype (132/344 patients, 38.5 %), followed by mixed, hyperactive, and nonmotoric delirium. The prevalence was highest in Neurology (28.5 %) and Geriatrics (24.7 %), lowest in Rehabilitation (14.0 %), and intermediate in Orthopedic (20.6 %) and Internal Medicine wards (21.4 %). In a multivariable logistic regression, age (odds ratio [OR] 1.03, 95 % confidence interval [CI] 1.01-1.05), Activities of Daily Living dependence (OR 1.19, 95 % CI 1.12-1.27), dementia (OR 3.25, 95 % CI 2.41-4.38), malnutrition (OR 2.01, 95 % CI 1.29-3.14), and use of antipsychotics (OR 2.03, 95 % CI 1.45-2.82), feeding tubes (OR 2.51, 95 % CI 1.11-5.66), peripheral venous catheters (OR 1.41, 95 % CI 1.06-1.87), urinary catheters (OR 1.73, 95 % CI 1.30-2.29), and physical restraints (OR 1.84, 95 % CI 1.40-2.40) were associated with delirium. Admission to Neurology wards was also associated with delirium (OR 2.00, 95 % CI 1.29-3.14), while admission to other settings was not. Conclusions: Delirium occurred in more than one out of five patients in acute and rehabilitation hospital wards. Prevalence was highest in Neurology and lowest in Rehabilitation divisions. The "Delirium Day" project might become a useful method to assess delirium across hospital settings and a benchmarking platform for future surveys

    Plant-soil relationship in heavy metals polluted soils

    L’attività di ricerca ha previsto il confronto fra due siti inquinati da metalli pesanti e due siti controllo naturali. Contrariamente ai parametri chimico-fisici e ai contenuti in metallo dei suoli, le analisi vegetazionali non hanno evidenziato differenze significative tra i siti. E’ stata necessaria la messa a punto strumentale di tecniche ICP-OES, ED(P)XRF per la calibrazione empirica di un XRF portatile, per le osservazioni puntiformi in situ di suoli e flora ed è stata modificata la metodica ufficiale per le stime della biodisponibilità dei metalli pesanti nei suoli inquinati. 6 specie comuni ai 4 siti sono state valutate per i loro TF e BAF. Viene confermata la specie-specificità del TF e del BAF per il quale è stato elaborato un inedito algoritmo per un corretto confronto tra le specie. Tra le sei specie, Silene Alba ha i migliori TF e BAF. Successivamente, sono state valutate alcune caratteristiche del Pennisetum clandestinum, candidato per la fitodepurazione: tollera le concentrazioni di arseniato tipiche dei suoli inquinati, l’arseniato è un inibitore competitivo del trasporto del fosfato, possiede un’elevata velocità di assorbimento dell’arseniato che accumula nella radice, ma non lo trasloca nella frazione epigea, parte dell’As(V) viene ridotto ad As(III) che viene efflusso dalla radiceThe research has provided a comparison between two heavy metals polluted sites and two natural control sites. Unlike physical-chemical parameters and metal content in the soils, vegetation analysis showed no significant differences between sites. The development and optimization of instrumental techniques ICP-OES and ED(P)XRF was necessary to calibrate empirically a portable XRF for soil and flora point observations in situ and the official method for estimation of bioavailability of heavy metals in polluted soil was changed. Six species, common to the four sites, were evaluated for their TF and BAF. Were confirmed the TF and the BAF species-specificity for which it was developed a novel algorithm to obtain a correct comparison among species. Among the six species, Silene Alba has the best TF and BAF. In the second stage, some characteristics of Pennisetum clandestinum, candidate for the phytoremediation, were assessed: it tolerates arsenate concentrations typical of polluted soil, arsenate is a competitive inhibitor of phosphate transport, it has a high uptake rate of arsenic that accumulates in the root, but it doesn’t move in the aboveground fraction and part of As(V) is reduced to As(III), which is efflux from roots

    STUDY OF ELEMENTS CONCENTRATIONS OF EUROPEAN SEABASS (DICENTRARCHUS LABRAX) FILLETS AFTER COOKING ON STEEL, CAST IRON, TEFLON, ALUMINIUM AND CERAMIC POTS

    Some elements and metals (Al, Ca, Cr, Cu, Fe, Mn, Ni, Pb, Se, Sr, Zn, P) were analyzed in European seabass (Dicentrarchus labrax), by comparing the concentration in raw and in the cooked fish. In particular, the cooking of European seabass was carried out on pots made of five different materials (cast iron, aluminum, steel, teflon and ceramic), commonly used in gastronomy. By measuring mineral elements and metals content of the fish before and after cooking on the 5 materials, there is an increase in elements which is due not only to water loss but also by the release of the materials upon contact with the surface of the pots. Among the analyzed elements the exception is represented by Cr, Se and Fe which decrease with cooking, while Pb remains unvaried. The use of 2 model solutions, made of water and water and vinegar, demonstrates that the used cooking materials are not inert to the leaching/release of elements that could be found in the processed fish

    Meccanismi morfofisiologici di adattamento allo stress idrico-nutrizionale in barbabietola da zucchero

    E\u2019 stata studiata la risposta adattativa del sistema radicale alla carenza nutrizionale in genotipi di barbabietola da zucchero (Beta vulgaris L. subsp. vulgaris, Sugar Beet Group). In un sistema diallelico sono stati presi in esame, su plantule allevate in soluzione idroponica, i seguenti indici morfofisiologici del sistema radicale: lunghezza e superficie radicale totale, numero di apici, velocit\ue0 di accrescimento della radice primaria, contenuto di glucosio e fruttosio nella regione apicale della radice primaria e velocit\ue0 di assorbimento dello ione solfato dopo deprivazione dello ione stesso. L\u2019attitudine combinatoria generale (ACG) \ue8 risultata pi\uf9 alta di quella specifica (ACS) per tutti gli indici esaminati con prevalenza, nel loro controllo, degli effetti additivi. E\u2019 stata dimostrata una significativa e positiva correlazione (P<0.05) tra i sopraccitati indici e la produttivit\ue0 in campo. Il confronto tra due genotipi, caratterizzati rispettivamente da alta e bassa produttivit\ue0, ha dimostrato che quello pi\uf9 produttivo presenta, in pieno campo, una maggiore densit\ue0 radicale distribuita lungo il profilo del terreno e un pi\uf9 alto contenuto idrico relativo fogliare (RWC) e abilit\ue0 competitiva verso le malerbe rispetto a quello meno produttivo. L\u2019analisi comparativa della plasticit\ue0 del sistema radicale in risposta alla variazione di disponibilit\ue0 del fosfato e del solfato fra un genotipo coltivato ad elevata produttivit\ue0 e due popolazioni selvatiche provenienti da siti caratterizzati da bassa e alta fertilit\ue0 del suolo ha messo in evidenza che il genotipo coltivato privilegia l\u2019incremento dei caratteri \u201cconcentrazione di glucosio e fruttosio dell\u2019apice radicale\u201d e \u201cvelocit\ue0 di accrescimento della radice primaria\u201d, la popolazione selvatica proveniente dal sito a bassa fertilit\ue0 privilegia l\u2019incremento dei parametri \u201clunghezza radicale totale\u201d e \u201cnumero di apici\u201d mentre quella selvatica del sito ad alta fertilit\ue0 risulta intermedia per tutti i caratteri considerati. Analisi cDNA-AFLP hanno dimostrato l\u2019esistenza di numerosi frammenti genici differenzialmente espressi tra il genotipo coltivato e la popolazione proveniente dal sito a bassa fertilit\ue0 in risposta alla carenza nutrizionale che verranno successivamente clonati e sequenziati. Ci\uf2 potrebbe favorire l\u2019identificazione di nuovi geni coinvolti nella risposta allo stress nutrizionale
