    The use of strontium ferrite in chemical looping systems

    This work reports a detailed chemical looping investigation of strontium ferrite (SrFeO3-δ), a material with the perovskite structure type able to donate oxygen and stay in a nonstoichiometric form over a broad range of oxygen partial pressures, starting at temperatures as low as 250°C (reduction in CO, measured in TGA). SrFeO3-δ is an economically attractive, simple, but remarkably stable material that can withstand repeated phase transitions during redox cycling. Mechanical mixing and calcination of iron oxide and strontium carbonate was evaluated as an effective way to obtain pure SrFeO3-δ. In situ XRD was performed to analyse structure transformations during reduction and reoxidation. Our work reports that much deeper reduction, from SrFeO3-δ to SrO and Fe, is reversible and results in oxygen release at a chemical potential suitable for hydrogen production. Thermogravimetric experiments with different gas compositions were applied to characterize the material and evaluate its available oxygen capacity. In both TGA and in-situ XRD experiments the material was reduced below δ=0.5 followed by reoxidation either with CO2 or air, to study phase segregation and reversibility of crystal structure transitions. As revealed by in-situ XRD, even deeply reduced material regenerates at 900°C to SrFeO3 δ with a cubic structure. To investigate the catalytic behaviour of SrFeO3-δ in methane combustion, experiments were performed in a fluidized bed rig. These showed SrFeO3-δ donates O2 into the gas phase but also assists with CH4 combustion by supplying lattice oxygen. To test the material for combustion and hydrogen production, long cycling experiments in a fluidized bed rig were also performed. SrFeO3-δ showed stability over 30 redox cycles, both in experiments with a 2-step oxidation performed in CO2 followed by air, as well as a single step oxidation in CO2 alone. Finally, the influence of CO/CO2 mixtures on material performance was tested; a fast and deep reduction in elevated pCO2 makes the material susceptible to carbonation, but the process can be reversed by increasing the temperature or lowering pCO2.EPSRC grant no.     Scoping review to identify potential non-antimicrobial interventions to mitigate antimicrobial resistance in commensal enteric bacteria in North American cattle production systems

    A scoping review was conducted to identify modifiable non-antimicrobial factors to reduce the occurrence of antimicrobial resistance in cattle populations. Searches were developed to retrieve peer-reviewed published studies in animal, human and in vitro microbial populations. Citations were retained when modifiable non-antimicrobial factors or interventions potentially associated with antimicrobial resistance were described. Studies described resistance in five bacterial genera, species or types, and 40 antimicrobials. Modifiable non-antimicrobial factors or interventions ranged widely in type, and the depth of evidence in animal populations was shallow. Specific associations between a factor or intervention with antimicrobial resistance in a population (e.g. associations between organic systems and tetracycline susceptibility in E. coli from cattle) were reported in a maximum of three studies. The identified non-antimicrobial factors or interventions were classified into 16 themes. Most reported associations between the non-antimicrobial modifiable factors or interventions and antimicrobial resistance were not statistically significant (P > 0·05 and a confidence interval including 1), but when significant, the results were not consistent in direction (increase or decrease in antimicrobial resistance) or magnitude. Research is needed to better understand the impacts of promising modifiable factors or interventions on the occurrence of antimicrobial resistance before any recommendations can be offered or adopted

    UK Regions, the European Union and Manufacturing Exports

    In advance of the referendum on the UK’s membership of the European Union (EU), this Brief considers the differences in how the UK’s constituent regions and nations interact with other EU countries, focusing upon trade in goods. The regional dimension of the UK/EU relationship has received relatively little attention in the public debate around the implications of ‘Brexit’, yet the evidence examined here suggests that the notion of whether the UK will be ‘better off’ or ‘worse off’ as a result of voting to exit or remain within the EU is too simplistic. An examination of the different trade relationships that different UK regions and nations have with the EU helps us to consider both how the UK will be affected by Brexit, and who will be most affected. In particular, the Brief reflects upon the possible implications of Brexit for the policy agendas around ‘rebalancing’ and ‘the Northern Powerhouse’, which are predicated upon a resurgence of manufacturing industries in the North of England

    In advance of the referendum on the UK’s membership of the European Union (EU), this Brief considers the distribution of EU ‘structural and investment funds’ to the UK’s constituent regions and nations. The regional dimension of the UK/EU relationship has received relatively little attention in the public debate around the implications of ‘Brexit’ with much of the attention focusing on whether the UK as a whole will be ‘better off’ or ‘worse off’ as a result of voting to exit or remain within the EU. Further, whilst the UK’s financial contribution to the EU has been widely debated there has been considerably less attention on the funding that the UK receives from the EU, and how that funding is distributed. Analysis of the regional distribution of EU structural and investment funds suggests that the possible implications of Brexit would vary across the UK’s constituent regions and nations, which experience significantly different economic and social inequalities and have a high degree of variation in their economic performance. The Brief highlights a series of key questions relating to the future of European structural and investment funds that require examination in advance of the referendum, and that policymakers would need to swiftly address should the UK vote to leave the European Union on June 23rd

    Prescribing practices of primary-care veterinary practitioners in dogs diagnosed with bacterial pyoderma

    Concern has been raised regarding the potential contributions of veterinary antimicrobial use to increasing levels of resistance in bacteria critically important to human health. Canine pyoderma is a frequent, often recurrent diagnosis in pet dogs, usually attributable to secondary bacterial infection of the skin. Lesions can range in severity based on the location, total area and depth of tissue affected and antimicrobial therapy is recommended for resolution. This study aimed to describe patient signalment, disease characteristics and treatment prescribed in a large number of UK, primary-care canine pyoderma cases and to estimate pyoderma prevalence in the UK vet-visiting canine population

    Comparison of the CDC Backpack aspirator and the Prokopack aspirator for sampling indoor- and outdoor-resting mosquitoes in southern Tanzania.

    BACKGROUND\ud \ud Resting mosquitoes can easily be collected using an aspirating device. The most commonly used mechanical aspirator is the CDC Backpack aspirator. Recently, a simple, and low-cost aspirator called the Prokopack has been devised and proved to have comparable performance. The following study evaluates the Prokopack aspirator compared to the CDC backpack aspirator when sampling resting mosquitoes in rural Tanzania.\ud \ud METHODS\ud \ud Mosquitoes were sampled in- and outdoors of 48 typical rural African households using both aspirators. The aspirators were rotated between collectors and households in a randomized, Latin Square design. Outdoor collections were performed using artificial resting places (large barrel and car tyre), underneath the outdoor kitchen (kibanda) roof and from a drop-net. Data were analysed with generalized linear models.\ud \ud RESULTS\ud \ud The number of mosquitoes collected using the CDC Backpack and the Prokopack aspirator were not significantly different both in- and outdoors (indoors p = 0.735; large barrel p = 0.867; car tyre p = 0.418; kibanda p = 0.519). The Prokopack was superior for sampling of drop-nets due to its smaller size. The number mosquitoes collected per technician was more consistent when using the Prokopack aspirator. The Prokopack was more user-friendly: technicians preferred using the it over the CDC backpack aspirator as it weighs considerably less, retains its charge for longer and is easier to manoeuvre.\ud \ud CONCLUSIONS\ud \ud The Prokopack proved in the field to be more advantageous than the CDC Backpack aspirator. It can be self assembled using simple, low-cost and easily attainable materials. This device is a useful tool for researchers or vector-control surveillance programs operating in rural Africa, as it is far simpler and quicker than traditional means of sampling resting mosquitoes. Further longitudinal evaluations of the Prokopack aspirator versus the gold standard pyrethrum spray catch for indoor resting catches are recommended

    The UHRF1 protein is a key regulator of retrotransposable elements and innate immune response to viral RNA in human cells

    While epigenetic mechanisms such as DNA methylation and histone modification are known to be important for gene suppression, relatively little is still understood about the interplay between these systems. The UHRF1 protein can interact with both DNA methylation and repressive chromatin marks, but its primary function in humans has been unclear. To determine what that was, we first established stable UHRF1 knockdowns (KD) in normal, immortalized human fibroblasts using targeting shRNA, since CRISPR knockouts (KO) were lethal. Although these showed a loss of DNA methylation across the whole genome, transcriptional changes were dominated by the activation of genes involved in innate immune signalling, consistent with the presence of viral RNA from retrotransposable elements (REs). We confirmed using mechanistic approaches that 1) REs were demethylated and transcriptionally activated; 2) this was accompanied by activation of interferons and interferon-stimulated genes and 3) the pathway was conserved across other adult cell types. Restoring UHRF1 in either transient or stable KD systems could abrogate RE reactivation and the interferon response. Notably, UHRF1 itself could also re-impose RE suppression independent of DNA methylation, but not if the protein contained point mutations affecting histone 3 with trimethylated lysine 9 (H3K9me3) binding. Our results therefore show for the first time that UHRF1 can act as a key regulator of retrotransposon silencing independent of DNA methylation

    Steady-state modulation of voltage-gated K+ channels in rat arterial smooth muscle by cyclic AMP-dependent protein kinase and protein phosphatase 2B

    Voltage-gated potassium channels (Kv) are important regulators of membrane potential in vascular smooth muscle cells, which is integral to controlling intracellular Ca2+ concentration and regulating vascular tone. Previous work indicates that Kv channels can be modulated by receptor-driven alterations of cyclic AMP-dependent protein kinase (PKA) activity. Here, we demonstrate that Kv channel activity is maintained by tonic activity of PKA. Whole-cell recording was used to assess the effect of manipulating PKA signalling on Kv and ATP-dependent K+ channels of rat mesenteric artery smooth muscle cells. Application of PKA inhibitors, KT5720 or H89, caused a significant inhibition of Kv currents. Tonic PKA-mediated activation of Kv appears maximal as application of isoprenaline (a β-adrenoceptor agonist) or dibutyryl-cAMP failed to enhance Kv currents. We also show that this modulation of Kv by PKA can be reversed by protein phosphatase 2B/calcineurin (PP2B). PKA-dependent inhibition of Kv by KT5720 can be abrogated by pre-treatment with the PP2B inhibitor cyclosporin A, or inclusion of a PP2B auto-inhibitory peptide in the pipette solution. Finally, we demonstrate that tonic PKA-mediated modulation of Kv requires intact caveolae. Pre-treatment of the cells with methyl-β-cyclodextrin to deplete cellular cholesterol, or adding caveolin-scaffolding domain peptide to the pipette solution to disrupt caveolae-dependent signalling each attenuated PKA-mediated modulation of the Kv current. These findings highlight a novel, caveolae-dependent, tonic modulatory role of PKA on Kv channels providing new insight into mechanisms and the potential for pharmacological manipulation of vascular tone
