    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear understanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5,6,7 vast areas of the tropics remain understudied.8,9,10,11 In the American tropics, Amazonia stands out as the world's most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepresented in biodiversity databases.13,14,15 To worsen this situation, human-induced modifications16,17 may eliminate pieces of the Amazon's biodiversity puzzle before we can use them to understand how ecological communities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple organism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region's vulnerability to environmental change. 15%–18% of the most neglected areas in ecological research are expected to experience severe climate or land use changes by 2050. This means that unless we take immediate action, we will not be able to establish their current status, much less monitor how it is changing and what is being lost

    A Big-Data Variational Bayesian Framework for Supporting the Prediction of Functional Outcomes in Wake-Up Stroke Patients

    Prognosis in Wake-up ischemic stroke (WUS) is important for guiding treatment and rehabilitation strategies, in order to improve recovery and minimize disability. For this reason, there is growing interest on models to predict functional recovery after acute ischemic events in order to personalize the therapeutic intervention and improve the final functional outcome. The aim of this preliminary study is to evaluate the possibility to predict a good functional outcome, in terms of modified Rankin Scale (mRS 64 2), in thrombolysis treated WUS patients by Bayesian analysis of clinical, demographic and neuroimaging data at admission. The study was conducted on 54 thrombolysis treated WUS patients. The Variational Bayesian logistic regression with Automatic Relevance Determination (VB-ARD) was used to produce model and select informative features to predict a good functional outcome (mRS 64 2) at discharge. The produced model showed moderately high 10\ua0 7\ua05-fold cross validation accuracy of 71% to predict outcome. The sparse model highlighted the relevance of NIHSS at admission, age, TACI stroke syndrome, ASPECTs, ischemic core CT Perfusion volume, hypertension and diabetes mellitus. In conclusion, in this preliminary study we assess the possibility to model the prognosis in thrombolysis treated WUS patients by using VB ARD. The identified features related to initial neurological deficit, history of diabetes and hypertension, together with necrotic tissue relate ASPECT and CTP core volume neuroimaging features, were able to predict outcome with moderately high accuracy

    Gene expression has been extensively studied in plant science research, mainly for the assessment of plant stress responses. Real-time-quantitative polymerase chain reaction (RT-qPCR) is an important tool for obtaining this information because it is a quick and easy technique to acquire a large amount of molecular data for both model and non-model plants. For a successful RT-qPCR analysis, gene expression should be carefully normalised. Genes involved in essential biological processes that exhibit constitutive expression are commonly selected as internal standards to normalise RT-qPCR experiments. In this study, the transcription profiles of 13 candidate reference genes for RT-qPCR were evaluated in three guarana cultivars (BRS-Amazonas, BRS-Maués and BRS-Luzéia) using different tissues (vegetative and fruit) in varying developmental stages. Two different algorithms, NormFinder and GeNorm, were utilised to assess gene stability. In general, the two algorithms did not select the same pairs of genes for all analysed conditions. For the largest group (the fruits of all cultivars), NormFinder selected the pair EF1A/UBQ, whereas GeNorm chose ACT/GAPDH as the best normalising genes. Thus, we recommend the use of at least four reference genes for the normalisation of gene expression in guarana plant studies. © 2015, Botanical Society of Sao Paulo

    Gonadal development in the freshwater crab Sylviocarcinus pictus (H. Milne Edwards, 1853) (Brachyura: Trichodactylidae) from the Guamá river, state of Pará, Brazil

    The development stages of male and female gonads in the freshwater crab Sylviocarcinus pictus (H. Milne Eduards, 1853) were described through macroscopic and microscopic (histology) examinations. The histological description was based on 40 specimens (20 each sex). Four gonadal development stages were found for females: immature, ripening, mature and spawned. The following female cells were found: ovogonia, oocytes in initial vitellogenesis, oocytes in advanced vitellogenesis, follicular cells and post-ovulatory follicles. Three development stages were found for males: immature, maturing and mature, with the indication of: spermatogonia, spermatocytes, spermatids, spermatozoids and spermatophores. These data suggest the pattern described in the literature. Size at sexual maturity was 32.3 mm of carapace width for males and 31.5 mm for females. The gonadal stages observed macroscopically by volume and color were validated through histological analysis and proved to be useful method for the rapid identification of sexual maturity in the species. The present study offers previously unpublished data on the reproductive biology of Sylviocarcinus pictus.<br>Os estádios de desenvolvimento das gônadas de machos e fêmeas de caranguejos Sylviocarcinus pictus (H. Milne Eduards, 1853) foram descritos por meio de observações macroscópicas e microscópicas (técnica histológica). A descrição histológica foi baseada em 40 espécimes (20 de cada sexo). Foram identificados quatro estádios de desenvolvimento para as fêmeas: imaturo, em maturação, maturo e em reabsorção. As seguintes células foram encontradas: ovogônias, ovócitos em vitelogênese inicial, ovócitos em vitelogênese avançada, células foliculares e folículos pós-ovulatórios. Três estádios de desenvolvimento foram encontrados para os machos: imaturo, em maturação e maturo, com indicação de: espermatogônias, espermatócitos, espermátides, espermatozóides e espermatóforos. Tais dados sugerem o padrão descrito na literatura. O tamanho da maturidade sexual foi de 32,3 mm de largura da carapaça para machos e 31,5 mm para fêmeas. Os estádios gonadais observados macroscopicamente por meio do volume e da coloração das gônadas foram validados pela análise histológica, sendo um critério útil e ágil para a identificação da maturidade sexual para a espécie. O presente estudo oferece informações inéditas sobre a biologia reprodutiva de Sylviocarcinus pictus
