Clades of huge phages from across Earth's ecosystems
Bacteriophages typically have small genomes and depend on their bacterial hosts for replication. Here we sequenced DNA from diverse ecosystems and found hundreds of phage genomes with lengths of more than 200 kilobases (kb), including a genome of 735 kb, which is-to our knowledge-the largest phage genome to be described to date. Thirty-five genomes were manually curated to completion (circular and no gaps). Expanded genetic repertoires include diverse and previously undescribed CRISPR-Cas systems, transfer RNAs (tRNAs), tRNA synthetases, tRNA-modification enzymes, translation-initiation and elongation factors, and ribosomal proteins. The CRISPR-Cas systems of phages have the capacity to silence host transcription factors and translational genes, potentially as part of a larger interaction network that intercepts translation to redirect biosynthesis to phage-encoded functions. In addition, some phages may repurpose bacterial CRISPR-Cas systems to eliminate competing phages. We phylogenetically define the major clades of huge phages from human and other animal microbiomes, as well as from oceans, lakes, sediments, soils and the built environment. We conclude that the large gene inventories of huge phages reflect a conserved biological strategy, and that the phages are distributed across a broad bacterial host range and across Earth's ecosystems
Psychometric performance of the brazilian version of the Mini-cuestionario de calidad de vida en la hipertensión arterial (MINICHAL)
This study aimed to evaluate the feasibility, acceptability, ceiling and floor effects, reliability, and convergent construct validity of the Brazilian version of the Mini Cuestionario de Calidad de Vida en la Hipertensión Arterial (MINICHAL). The study included 200 hypertensive outpatients in a university hospital and a primary healthcare unit. The MINICHAL was applied in 3.0 (± 1.0) minutes with 100% of the items answered. A "ceiling effect" was observed in both dimensions and in the total score, as well as evidence of measurement stability (ICC=0.74). The convergent validity was confirmed by significant positive correlations between similar dimensions of the MINICHAL and the SF-36, and significant negative correlations with the Minnesota Living with Heart Failure Questionnaire - MLHFQ, however, correlations between dissimilar constructs were also observed. It was concluded that the Brazilian version of the MINICHAL presents evidence of reliability and validity when applied to hypertensive outpatientsEste estudio tuvo como objetivo evaluar la practicidad, la aceptabilidad, los efectos techo y suelo, la confiabilidad y la validez de constructo convergente de la versión brasileña del Minicuestionario de Calidad de Vida de la Hipertensión Arterial - MINICHAL. Participaron del estudio 200 pacientes hipertensos en seguimiento en ambulatorio, en hospital universitario y en Unidad Básica de Salud. El MINICHAL fue aplicado en 3,0 (±1,0) minutos, con 100% de los ítems respondidos. Fue observado "efecto techo" en ambas dimensiones y puntaje total, así como evidencias de estabilidad de la medida (CCI=0,74). La validez convergente fue confirmada por correlaciones significativas positivas entre dimensiones similares del MINICHAL y del SF-36, y por correlaciones significativas negativas con el Minnesota Living with Heart Failure Questionnaire - MLHFQ, a pesar de que correlaciones entre constructos no similares hubiesen sido observadas. Supplement with Cystoseira canariensis promotes an increase of resistance to the maximum load in the grastrocnemius muscle of female rats
Was evaluated the maximum load supported by the grastrocnemius muscle of female rats after the ministering of Cystoseira canariensis, either associated or not to swimming. Twenty-eight young Wistar female rats were used, divided into 4 groups: control (C, n=7); supplement (S, n=7); supplement and swimming (SSw, n=7); swimming (Sw, n=7). Each one pertaining to the groups S and SSw received 20 mg of myostatin inhibitor per day. The swimming consisted in an aerobe protocol, three times in a week, during eight weeks. The right grastrocnemius muscle of each animal was removed and a tension test was performed in an Emic testing machine. The results (Mean±SEM) were evaluated through ANOVA and Tukey test (p<0.05). A significant difference for maximum load (in N) was verified among the groups C (35.41±1.06) and S (39.98±1.15); Sw (27.94±2.19) and S (39.98±1.15); Sw (27.94±2.19) and SSw (37.78±1.28). In relation to the stretching at the maximum limit (in x10-3m) at the maximum load, the group SSw obtained a value (20.68±1.19) significantly greater than the groups C (17.15±1.11), S and Sw (16.11±1.60). There was a significant difference for body weight gain among the groups treated with supplement and supplement associated to the swimming, with smaller values for this last. The myostatin inhibitor either, associated or not to the swimming, promotes an increase of resistance to the maximum load in the tension test in grastrocnemius muscle of young female rats

Wavelet-based algorithm to the evaluation of contrasted hepatocellular carcinoma in CT-images after transarterial chemoembolization
Background: Hepatocellular carcinoma is a primary tumor of the liver and involves different treatment modalities according to the tumor stage. After local therapies, the tumor evaluation is based on the mRECIST criteria, which involves the measurement of the maximum diameter of the viable lesion. This paper describes a computed methodology to measure through the contrasted area of the lesions the maximum diameter of the tumor by a computational algorithm.Methods: 63 computed tomography (CT) slices from 23 patients were assessed. Non-contrasted liver and HCC typical nodules were evaluated, and a virtual phantom was developed for this purpose. Optimization of the algorithm detection and quantification was made using the virtual phantom. After that, we compared the algorithm findings of maximum diameter of the target lesions against radiologist measures.Results: Computed results of the maximum diameter are in good agreement with the results obtained by radiologist evaluation, indicating that the algorithm was able to detect properly the tumor limits. A comparison of the estimated maximum diameter by radiologist versus the algorithm revealed differences on the order of 0.25 cm for large-sized tumors (diameter > 5 cm), whereas agreement lesser than 1.0 cm was found for small-sized tumors.Conclusions: Differences between algorithm and radiologist measures were accurate for small-sized tumors with a trend to a small decrease for tumors greater than 5 cm.
