    Human papillomavirus (HPV) contamination of gynaecological equipment.

    OBJECTIVE: The gynaecological environment can become contaminated by human papillomavirus (HPV) from healthcare workers' hands and gloves. This study aimed to assess the presence of HPV on frequently used equipment in gynaecological practice. METHODS: In this cross-sectional study, 179 samples were taken from fomites (glove box, lamp of a gynaecological chair, gel tubes for ultrasound, colposcope and speculum) in two university hospitals and in four gynaecological private practices. Samples were collected with phosphate-buffered saline-humidified polyester swabs according to a standardised pattern, and conducted twice per day for 2 days. The samples were analysed by a semiquantitative real-time PCR. Statistical analysis was performed using Pearson's χ(2) test and multivariate regression analysis. RESULTS: Thirty-two (18%) HPV-positive samples were found. When centres were compared, there was a higher risk of HPV contamination in gynaecological private practices compared with hospitals (OR 2.69, 95% CI 1.06 to 6.86). Overall, there was no difference in the risk of contamination with respect to the time of day (OR 1.79, 95% CI 0.68 to 4.69). When objects were compared, the colposcope had the highest risk of contamination (OR 3.02, 95% CI 0.86 to 10.57). CONCLUSIONS: Gynaecological equipment and surfaces are contaminated by HPV despite routine cleaning. While there is no evidence that contaminated surfaces carry infectious viruses, our results demonstrate the need for strategies to prevent HPV contamination. These strategies, based on health providers' education, should lead to well-established cleaning protocols, adapted to gynaecological rooms, aimed at eliminating HPV material

    Aging is associated with impaired renal function, INF-gamma induced inflammation and with alterations in iron regulatory proteins gene expression

    Our aim was to contribute to a better understanding of the pathophysiology of anemia in elderly, by studying how aging affects renal function, ironmetabolism, erythropoiesis and the inflammatory response, using an experimental animal model. The study was performed in male Wistar, a group of young rats with 2 months age and an old one with 18 months age. Old rats presented a significant higher urea, creatinine, interferon (INF)-gamma, ferritin and soluble transferrin receptor serum levels, as well as increased counts of reticulocytes and RDW. In addition, these rats showed significant lower erythropoietin (EPO) and iron serum levels. Concerning gene expression of iron regulatory proteins, old rats presented significantly higher mRNA levels of hepcidin (Hamp), transferrin (TF), transferrin receptor 2 (TfR2) and hemojuvelin (HJV); divalent metal transporter 1 (DMT1) mRNA levels were significantly higher in duodenal tissue; EPO gene expression was significantly higher in liver and lower in kidney, and the expression of the EPOR was significantly higher in both liver and kidney. Our results showed that aging is associated with impaired renal function, which could be in turn related with the inflammatory process and with a decline in EPO renal production. Moreover, we also propose that aging may be associatedwith INF-gamma-induced inflammation and with alterations upon iron regulatory proteins gene expression

    Circulating DNA: Diagnostic Tool and Predictive Marker for Overall Survival of NSCLC Patients

    PURPOSE: The purpose of our study was to determine whether the amounts of circulating DNA (cDNA) could discriminate between NSCLC patients and healthy individuals and assess its value as a prognostic marker of this disease. METHODS: We conducted a study of 309 individuals and the cDNA levels were assessed through real-time PCR methodology. RESULTS: We found increased cDNA levels in NSCLC patients compared to control individuals. We also found a decreased overall survival time in patients presenting high cDNA levels, when compared to lower cDNA concentrations. CONCLUSIONS: Quantification of cDNA may be a good tool for NSCLC detection with potential for clinical applicability

    GNSS transpolar earth reflectometry exploriNg system (G-TERN): Mission concept

    The global navigation satellite system (GNSS) Transpolar Earth Reflectometry exploriNg system (G-TERN) was proposed in response to ESA's Earth Explorer 9 revised call by a team of 33 multi-disciplinary scientists. The primary objective of the mission is to quantify at high spatio-temporal resolution crucial characteristics, processes and interactions between sea ice, and other Earth system components in order to advance the understanding and prediction of climate change and its impacts on the environment and society. The objective is articulated through three key questions. 1) In a rapidly changing Arctic regime and under the resilient Antarctic sea ice trend, how will highly dynamic forcings and couplings between the various components of the ocean, atmosphere, and cryosphere modify or influence the processes governing the characteristics of the sea ice cover (ice production, growth, deformation, and melt)? 2) What are the impacts of extreme events and feedback mechanisms on sea ice evolution? 3) What are the effects of the cryosphere behaviors, either rapidly changing or resiliently stable, on the global oceanic and atmospheric circulation and mid-latitude extreme events? To contribute answering these questions, G-TERN will measure key parameters of the sea ice, the oceans, and the atmosphere with frequent and dense coverage over polar areas, becoming a "dynamic mapper" of the ice conditions, the ice production, and the loss in multiple time and space scales, and surrounding environment. Over polar areas, the G-TERN will measure sea ice surface elevation (<10 cm precision), roughness, and polarimetry aspects at 30-km resolution and 3-days full coverage. G-TERN will implement the interferometric GNSS reflectometry concept, from a single satellite in near-polar orbit with capability for 12 simultaneous observations. Unlike currently orbiting GNSS reflectometry missions, the G-TERN uses the full GNSS available bandwidth to improve its ranging measurements. The lifetime would be 2025-2030 or optimally 2025-2035, covering key stages of the transition toward a nearly ice-free Arctic Ocean in summer.     Impaired complex I repair causes recessive Leber's hereditary optic neuropathy

    Leber's hereditary optic neuropathy (LHON) is the most frequent mitochondrial disease and was the first to be genetically defined by a point mutation in mitochondrial DNA (mtDNA). A molecular diagnosis is achieved in up to 95% of cases, the vast majority of which are accounted for by 3 mutations within mitochondrial complex I subunit-encoding genes in the mtDNA (mtLHON). Here, we resolve the enigma of LHON in the absence of pathogenic mtDNA mutations. We describe biallelic mutations in a nuclear encoded gene, DNAJC30, in 33 unsolved patients from 29 families and establish an autosomal recessive mode of inheritance for LHON (arLHON), which to date has been a prime example of a maternally inherited disorder. Remarkably, all hallmarks of mtLHON were recapitulated, including incomplete penetrance, male predominance, and significant idebenone responsivity. Moreover, by tracking protein turnover in patient-derived cell lines and a DNAJC30-knockout cellular model, we measured reduced turnover of specific complex I N-module subunits and a resultant impairment of complex I function. These results demonstrate that DNAJC30 is a chaperone protein needed for the efficient exchange of complex I subunits exposed to reactive oxygen species and integral to a mitochondrial complex I repair mechanism, thereby providing the first example to our knowledge of a disease resulting from impaired exchange of assembled respiratory chain subunits

    A qualitative study of the understanding and use of ‘compassion focused coping strategies’ in people who suffer from serious weight difficulties.

    Abstract Background The physical and psychological health problems associated with obesity are now well documented, as is the urgency for addressing them. In addition, associations between quality of life, depression, self-esteem, self-criticism, and obesity are now established indicating a need for a better understanding of the links between self-evaluation, affect-regulation and eating behaviours. Methods Compassion has now been identified as a major source of resilience, helpful self-relating and affect regulation. Thus this study used semi-structured interviews to explore the understanding and experiences of compassion in 2 overweight men and 10 women seeking help for weight problems. The interviews examined people's understandings of compassion, their recall of experiences of compassion in childhood, their current experiences of receiving compassion from others, being compassionate to others, being self-compassionate, and whether they would be compassionate or self-critical for relapses in overeating. Interviews were transcribed and analysed using thematic analysis (Qual Res Psychol, 3: 77-101, 2006). Results Participants saw compassion as related to ‘caring’ and being ‘listened to’. However, their recall of earlier experiences of compassion was of primarily practical help rather than emotional engagement. Typically their response to their own relapse and setbacks were self-criticism, self-disgust and even self-hatred rather than self-caring or understanding. Self-critical/hating responses tend to be associated with poor weight regulation. Conclusions When people with weight problems relapse, or struggle to control their eating, they can become quite self-critical, even self-hating, which may increase difficulties with emotionally coping and maintaining healthy lifestyles and eating habits. Although turning to others for support and compassion, and becoming self-compassionate are antidotes to self-criticism, and are associated with better coping and mental health, many participants did not utilise compassionate strategies – often the opposite. It is possible that interventions that include mindfulness and compassion training could be helpful for these difficulties.N/
