    Nocturnal Hypoxemia and CT Determined Pulmonary Artery Enlargement in Smokers

    Background: Pulmonary artery enlargement (PAE) detected using chest computed tomography (CT) is associated with poor outcomes in chronic obstructive pulmonary disease (COPD). It is unknown whether nocturnal hypoxemia occurring in smokers, with or without COPD, obstructive sleep apnoea (OSA) or their overlap, may be associated with PAE assessed by chest CT. Methods: We analysed data from two prospective cohort studies that enrolled 284 smokers in lung cancer screening programs and completing baseline home sleep studies and chest CT scans. Main pulmonary artery diameter (PAD) and the ratio of the PAD to that of the aorta (PA:Ao ratio) were measured. PAE was defined as a PAD >= 29 mm in men and >= 27 mm in women or as a PA:Ao ratio > 0.9. We evaluated the association of PAE with baseline characteristics using multivariate logistic models. Results: PAE prevalence was 27% as defined by PAD measurements and 11.6% by the PA:Ao ratio. A body mass index >= 30 kg/m(2) (OR 2.01; 95%CI 1.06-3.78), lower % predicted of forced expiratory volume in one second (FEV1) (OR 1.03; 95%CI 1.02-1.05) and higher % of sleep time with O-2 saturation < 90% (T90) (OR 1.02; 95%CI 1.00-1.03), were associated with PAE as determined by PAD. However, only T90 remained significantly associated with PAE as defined by the PA:Ao ratio (OR 1.02; 95%CI 1.01-1.03). In the subset group without OSA, only T90 remains associated with PAE, whether defined by PAD measurement (OR 1.02; 95%CI 1.01-1.03) or PA:Ao ratio (OR 1.04; 95%CI 1.01-1.07). Conclusions: In smokers with or without COPD, nocturnal hypoxemia was associated with PAE independently of OSA coexistence

    A placebo-controlled proof-of-concept study of alirocumab on postprandial lipids and vascular elasticity in insulin-treated patients with type 2 diabetes mellitus

    Aim: Type 2 diabetes mellitus (T2DM) is associated with an increased risk of cardiovascular disease (CVD) linked to atherogenic dyslipidaemia and postprandial hyperlipidaemia. Alirocumab, a proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor, improves CVD risk by reducing the concentration of low-density lipoprotein-cholesterol (LDL-C). However, effects of PCK9 inhibitors on other aspects of diabetic dyslipidaemia, particularly in the postprandial situation, are less clear. Material and Methods: Twelve male patients with T2DM on an intensive insulin regimen completed a 6-week randomized, double-blind, placebo-controlled, proof-of-concept study. Participants received three biweekly dosages of subcutaneous alirocumab (150 mg) or placebo. Before and after the intervention, fasting and postprandial triglyceride (TG) plasma levels, apolipoprotein (apo) B48, lipoprotein composition isolated by ultracentrifugation, vascular function and markers of inflammation were evaluated. Results: Alirocumab treatment reduced fasting plasma TG levels (between group median change −24.7%; P = 0.018) and fasting apoB48 serum levels (−35.9%; P = 0.039) compared with placebo. Alirocumab reduced the plasma TG area under the curve (AUC) (−26.4%; P = 0.006) and apoB48 AUC (−55.7%; P = 0.046), as well as plasma TG incremental AUC (−21.4%; P = 0.04) and apoB48 incremental AUC (−26.8%; P = 0.02). In addition, alirocumab reduced fasting and postprandial TG levels in very low-density lipoprotein (VLDL) and LDL. Alirocumab improved fasting pulse wave velocity, but no changes in postprandial markers of inflammation were observed. Conclusions: In addition to the well-known LDL-C-reducing effects, 6 weeks of alirocumab treatment lowered both fasting and postprandial plasma TG levels by reducing the TG levels in VLDL and LDL and the concentration of intestinal remnants

    Rootstock effects on scion gene expression in maritime pine

    Pines are the dominant conifers in Mediterranean forests. As long-lived sessile organisms that seasonally have to cope with drought periods, they have developed a variety of adaptive responses. However, during last decades, highly intense and long-lasting drought events could have contributed to decay and mortality of the most susceptible trees. Among conifer species, Pinus pinaster Ait. shows remarkable ability to adapt to different environments. Previous molecular analysis of a full-sib family designed to study drought response led us to find active transcriptional activity of stress-responding genes even without water deprivation in tolerant genotypes. To improve our knowledge about communication between above- and below-ground organs of maritime pine, we have analyzed four graft-type constructions using two siblings as rootstocks and their progenitors, Gal 1056 and Oria 6, as scions. Transcriptomic profiles of needles from both scions were modified by the rootstock they were grafted on. However, the most significant differential gene expression was observed in drought-sensitive Gal 1056, while in drought-tolerant Oria 6, differential gene expression was very much lower. Furthermore, both scions grafted onto drought-tolerant rootstocks showed activation of genes involved in tolerance to abiotic stress, and is most remarkable in Oria 6 grafts where higher accumulation of transcripts involved in phytohormone action, transcriptional regulation, photosynthesis and signaling has been found. Additionally, processes, such as those related to secondary metabolism, were mainly associated with the scion genotype.     Wettability and corrosion of [NTf2] anion-based ionic liquids on steel and PVD (TiN, CrN, ZrN) coatings

    Thewetting and corrosion behavior of three bis(trifluoromethylsulfonyl)imide-based ionic liquids: 1-Dodecyl-3- methylimidazolium bis(trifluoromethylsulfonyl)imide [C12MIM][NTf2], tributylmethylammonium bis(trifluoromethylsulfonyl)imide [N4441][NTf2] and methyltrioctylammonium bis(trifluoromethylsulfonyl)imide [N1888][NTf2] are tested in this research. The surface tension was measured for temperatures of 293–353 K resulting in the expected linearly decreasing behavior with temperature increase. In addition, contact angle measurements were made on AISI 52100 steel and three coatings (TiN, CrN and ZrN) obtained by PVD technique, finding the regular behavior in hydrophobic (non-polar) systems: high contact angles led to high surface tensions. Complementary parameters like spreading parameter and polarity fraction were calculated to enhance the wetting evaluation of these ionic liquids. [N1888][NTf2]/TiN resulted as the best IL-surface combination for a good wettability, due to the higher dispersion of the charge on the large size cation in this IL and the higher values of total and polar component of the surface free energy for this coating. Finally, SEM-EDS analysis determined that [N1888][NTf2]/ZrN was the best option in order to avoid corrosion problems. The evaporation of water, present as impurity in the ionic liquids, was found the main reason because of corrosion did not occur in the tests carried out at 100 °C

    Journal Article;Hepatitis B virus (HBV) and Hepatitis C virus (HCV) infections pose major public health problems because of their prevalence worldwide. Consequently, screening for these infections is an important part of routine laboratory activity. Serological and molecular markers are key elements in diagnosis, prognosis and treatment monitoring for HBV and HCV infections. Today, automated chemiluminescence immunoassay (CLIA) analyzers are widely used for virological diagnosis, particularly in high-volume clinical laboratories. Molecular biology techniques are routinely used to detect and quantify viral genomes as well as to analyze their sequence; in order to determine their genotype and detect resistance to antiviral drugs. Real-time PCR, which provides high sensitivity and a broad dynamic range, has gradually replaced other signal and target amplification technologies for the quantification and detection of nucleic acid. The next-generation DNA sequencing techniques are still restricted to research laboratories.The serological and molecular marker methods available for HBV and HCV are discussed in this article, along with their utility and limitations for use in Chronic Hepatitis B (CHB) diagnosis and monitoring.Ye

    Characterization of genetic variation within table grape varieties (Vitis vinifera L.) based on AFLP markers

    AFLPs were used to characterize a collection of 35 table grape varieties. A single AFLP reaction with a single combination of selective primers yielded an average of 50 polymorphic and scorable amplified bands in this collection and was enough to unequivocally identify each analyzed variety. Genetic similarity between different varieties was estimated, based on the results obtained with two primer combinations, to be between 0.65 and 0.90, while sibling varieties derived from the same cross showed a genetic similarity higher than 0.80. The analysis of several samples belonging to the same varieties, yet from different origins, showed stability of the AFLP patterns as to unequivocally and reproducibly identify them. Furthermore, some specific primer combinations detected a low level of variation that could be due to somatic variation and would be useful in clonal identification

    Morphological and molecular characterization of grapevine accessions known as Albillo

    Twenty-eight accessions identified under the name of Albillo or cited as a synonym of this ancient Spanish grapevine variety were characterized using 43 morphological descriptors, four different isozyme systems (CO, ACPH, GOT, and SOD) and two Amplified Fragment Length Polymorphism (AFLP) primer combinations. Genetic similarity data resulting from these analyses allowed three different groups of Albillo to be distinguished that corresponded with geographical and historical origin of representative varieties: Turruntés, Malvasía de El Bierzo, and the classical Albillo de Madrid, related to the oldest Albillo ampelographical descriptions. In addition, homonyms as well as varieties incorrectly cited as Albillo synonyms were identified. To ensure accurate variety identification, association among historical descriptions based on morphological descriptors and molecular fingerprints was required. Isozymes proved to be useful markers for varietal identification. However, only AFLP allowed distinction among closely related varieties as well as the analysis of intravarietal variation

    Genetic relationships among biotypes of Bemisia tabaci (Hemiptera: Aleyrodidae) based on AFLP analysis

    Genetic similarities between 13 samples belonging to nine reference biotypes and two field populations of Bemisia tabaci (Gennadius), one field population of B. medinae Gomez-Menor and another of B. afer Priesner and Hosny, were evaluated using amplified fragment length polymorphism (AFLP) markers. The results indicate that B. tabaci biotypes can be grouped together with a minimum similarity coefficient of 0.32 and separated from the two other species with a similarity coefficient of 0.07. Bemisia tabaci biotypes were grouped in four clusters which comprised: (i) Near East and Indian subcontinent biotypes; (ii) B and Q biotypes plus a Nigerian population from cowpea; (iii) New World A biotype; and (iv) S biotype and a Nigerian population from cassava. These results were consistent with a previous grouping of biotypes based on RAPD-PCR analysis. The AFLP assay allowed the scoring of a total of 354 polymorphic bands in two reaction events with the use of two primer combinations
