    Contrasting Geographical Distributions as a Result of Thermal Tolerance and Long-Distance Dispersal in Two Allegedly Widespread Tropical Brown Algae

    BackgroundMany tropical marine macroalgae are reported from all three ocean basins, though these very wide distributions may simply be an artifact resulting from inadequate taxonomy that fails to take into account cryptic diversity. Alternatively, pantropical distributions challenge the belief of limited intrinsic dispersal capacity of marine seaweeds and the effectiveness of the north-south oriented continents as dispersal barriers. We aimed to re-assess the distribution of two allegedly circumtropical brown algae, Dictyota ciliolata and D. crenulata, and interpret the realized geographical range of the respective species in relation to their thermal tolerance and major tectonic and climatic events during the Cenozoic.Methodology/Principal FindingsSpecies delimitation was based on 184 chloroplast encoded psbA sequences, using a Generalized Mixed Yule Coalescent method. Phylogenetic relationships were inferred by analyzing a six-gene dataset. Divergence times were estimated using relaxed molecular clock methods and published calibration data. Distribution ranges of the species were inferred from DNA-confirmed records, complemented with credible literature data and herbarium vouchers. Temperature tolerances of the species were determined by correlating distribution records with local SST values. We found considerable conflict between traditional and DNA-based species definitions. Dictyota crenulata consists of several pseudocryptic species, which have restricted distributions in the Atlantic Ocean and Pacific Central America. In contrast, the pantropical distribution of D. ciliolata is confirmed and linked to its significantly wider temperature tolerance.Conclusions/SignificanceTectonically driven rearrangements of physical barriers left an unequivocal imprint on the current diversity patterns of marine macroalgae, as witnessed by the D. crenulata–complex. The nearly circumglobal tropical distribution of D. ciliolata, however, demonstrates that the north-south oriented continents do not present absolute dispersal barriers for species characterized by wide temperature tolerances

    Invasion and MMP expression profile in desmoid tumours

    Desmoid tumours are locally invasive soft tissue tumours in which beta-catenin mediated TCF-dependent transcription is activated. The role of soluble factors secreted by the myofibroblastic desmoid tumour, which could stimulate tumour invasiveness, was investigated. Using collagen gel invasion assays, the presence of factors stimulating invasion in desmoid conditioned media (CM) could be established. Since matrix metalloproteinases (MMPs) have been implicated in the process of tumoral invasion, the expression levels of the MMP family members were evaluated. Quantitative reverse transcription-PCR was used to determine the expression levels of MMP1, MMP2, MMP3, MMP7, MMP11, MMP12, MMP13, MMP14 and the inhibitors TIMP1, TIMP2 and TIMP3. Besides overexpression of MMP7, a known TCF-dependent target gene, a striking upregulation of the expression levels of MMP1, MMP3, MMP11, MMP12 and MMP13 in desmoid tumours, compared to unaffected fibroblasts from the same patients, was found. Treating the CM of desmoids with a synthetic and a physiologic MMP inhibitor reduced the invasion-stimulating capacity of the desmoid CM by approximately 50%. These results suggest the involvement of soluble factors, released by the desmoid cells, in stimulating invasion and implicate the MMPs as facilitators of invasion

    Determinants of methotrexate adherence in rheumatoid arthritis patients

    Abstract: In rheumatoid arthritis (RA) patients, weekly intake of methotrexate (MTX) is the basic drug treatment. This observational study aims to investigate how many RA patients are adherent in terms of MTX intake and to identify determinants of non-adherence. Intake of MTX (orally or via injection) was recorded in 129 RA patients with an electronic monitoring system (MEMS\uae) during 16 weeks. In addition, two adherence questionnaires, the Medication Adherence Report Scale (MARS-5) and the Compliance-Questionnaire-Rheumatology (CQR) as well as a visual analogue scale (VAS) measuring MTX adherence, were administered to these patients. As possible determinants of adherence, data on demographics, disease and treatment characteristics, depression, illness cognitions, motivation, and social support were collected. Of all participants, 58 % were fully adherent and 75 % skipped at most one dose during 16 weeks. A better mental health status and suffering from comorbidities had a positive effect on adherence, while living alone had a negative effect. These three predictors explained 30 % of the variance in MTX adherence. Of the three self-report medication adherence measures, the VAS correlated the highest with the results of the electronic monitoring system (r\u2009=\u20090.552, p\u2009=\u20090.01). A relatively high adherence rate was observed in RA patients treated with MTX. The determinants identified by this study could be used to screen patients at risk for non-adherence. A simple VAS scale seems to be an acceptable way for a preliminary screening of MTX adherence

    Platelet-rich plasma and platelet gel preparation using PlateltexÂź

    Background  The platelet gel is made by embedding concentrate platelets within a semisolid (gel) network of polymerized fibrin. It is believed that this blood component will be used more and more in the treatment of several clinical conditions and as an adjunctive material in tissue engineering. Several systems are available to produce platelet‐rich plasma (PRP) for topical therapy. Recently, a new system became commercially available, PlateltexÂź. Here we report the technical performance of this system in comparison with the performance of other commercially available systems: PRGFÂź, PRP‐Landesber, CurasanÂź, PCCSÂź, HarvestÂź, VivostatÂź, RegenÂź and FibrinetÂź.Material and Methods  Both the PRP and the gel were prepared according to the manufacturer's directions. The blood samples of 20 donors were used. The yield, the efficiency, and the amount of platelet‐derived growth factor AB (PDGF‐AB), transforming growth factor ÎČ, vascular endothelial growth factor and fibroblast growth factor were measured in the resulting PRP. The feature of the batroxobin‐induced gelation was evaluated.Results  The yield, the collection efficiency and the growth factor content of PlateltexÂź were comparable to those of most of the other available systems. The gelation time was not dependent on the fibrinogen concentration; however, it was strongly influenced by the contact surface area of the container where the clotting reaction took place (P < 0·0001).Conclusions  PlateltexÂź provided platelet recovery, collection efficiency and PDGF‐AB availability close to those provided by other systems marketed with the same intended use. Batroxobin, the enzyme provided to induce gelation, acts differently from thrombin, which is used by most other systems. Platelets treated with thrombin become activated; they release their growth factors quickly. Furthermore, thrombin–platelet interaction is a physiological mechanism that hastens the clot‐retraction rate. On the contrary, platelets treated with batroxobin do not become activated; they are passively entrapped within the fibrin network, and their growth factor release occurs slowly. In these conditions, the clot retraction takes longer to occur. According to these differences between thrombin and batroxobin, it is expected that batroxobin‐induced PRP activation will tailor slow release of the platelet content, thus, providing longer in loco availability of trophic factors. In selected clinical conditions, this durable anabolic factor availability might be preferable to quick thrombin‐induced growth factor release

    Hypermetamorphosis in a leaf-miner allows insects to cope with a confined nutritional space

    International audienceHypermetamorphosis has been described in several Lepidoptera leaf-miner species (mostly Gracillariidae, Epipyropidae, and Phyllocnistidae) and can be defined as a strong modification of the larval morphology associated with a switch in its feeding mode. Evolution of this larval feeding strategy presumably influences nutritional resources that can be exploited and has strong consequences for plant morphology. The following studyfocuses on Phyllonorycter blancardella (Lepidoptera: Gracillariidae), a leaf-miner developing on Malus domestica. We characterize the morphology of larval mouthparts and the resulting morphological impact on leaf tissues. Our results show that first instars do not strongly affect the leaf anatomy and leave most plant cells intact, while later instars significantly disrupt leaf tissues. Additionally, young larvae are ‘‘fluid-feeders’’ and feed on plant cell fluids resulting from the progression of the larvae through the lower layer of the leaf spongy parenchyma. They occupy a feeding niche clearly distinct from later instars that are ‘‘tissue-feeders’’. Hypermetamorphosis in P. blancardella most likely allows insects to cope with a confined nutritional space by partitioning the limited feeding resources, and may help leaf-miners to optimize their nutritio
