    Diversity of seed mineral composition of Phaseolus vulgaris L. germplasm

    A collection of 155 accessions of ancient Portuguese common beans (Phaseolus vulgaris L.) was evaluated in relation to the content of 8minerals (Zn, Cu, Fe, Mn, Ca, Mg, P and K) important for human nutrition. A high degree of variability for P, Fe, Zn, Cu, Mn and Ca was observed in the collection. Total correlation matrix analysis revealed the existence of two important sets of strong positive correlations (P 0.0001), one involving P, Fe, Zn, Cu and protein, and the other Ca and Mn. The principal component analysis showed that Zn, Fe and Cu are highly correlated to the first component (27% of variability) and Mn and Ca to the second component (22% of the variability). The high mineral variability observed in the seeds of this common bean collection could be useful for the selection of cultivars with higher nutrition value and for the improvement of seed nutrition quality traits

    Survey of mycotoxins in Southern Brazilian wheat and evaluation of immunoassay methods

    One hundred commercial wheat grain samples were collected during the 2015 sea-son across 78 municipalities in the states of Paraná (PR), Rio Grande do Sul (RS), and São Paulo (SP), Brazil. Separate subsamples were analyzed for the concentration of deoxynivalenol (DON), zearalenona (ZEA) and ochratoxin A (OTA) mycotoxins using two methods: UHPLC-MS/MS (reference method) and a commercial enzyme-linked immunosorbent assay (ELISA) (AgraQuant®). The OTA mycotoxin was not found in the samples by both methods. DON and ZEA were detected in 55 % and 39 % of the samples by the reference method, with overall mean levels of 795.2 μg kg−1 and 79.78 μg kg−1, respectively. There was a significant and positive correlation (Spearman rank) between DON and ZEA estimates by the reference method (r = 0.77, p < 0.001). The DON levels estimated by the immunoassay agreed poorly with the reference, being largely overestimated. Based on a cut-off level of 1000 μg kg−1, the immunoassay correctly classified 57 samples as true negatives and 15 as true positives. Only 28 were classified as false positives. For ZEA, the levels estimated by the two methods were in better agreement than for DON. Using the cut-off level of 200 μg kg−1, 96 % of the samples were classified correctly as true positives and only one sample was classified as false positive. The levels for both mycotoxins were mostly acceptable for human consumption. Further studies should focus on multi-toxin methods compared with immunoassays to understand the reasons of overestimation and the role of immunoassays as a cost-effective solution for fast screening of mycotoxins in the food chain

    Studies of the Cabbibo-Suppressed Decays D+π0+νD^+ \to \pi^0 \ell^+ \nu and D+ηe+νeD^+ \to \eta e^+ \nu_e

    Using 4.8 fb1^{-1} of data taken with the CLEO II detector, the branching fraction for the Cabibbo-suppressed decay D+π0+νD^+\to\pi^0\ell^+\nu measured relative to the Cabibbo favored decay D+K0ˉ+νD^+\to\bar{K^0}\ell^+\nu is found to be 0.046±0.014±0.0170.046\pm 0.014\pm 0.017. Using VcsV_{cs} and VcdV_{cd} from unitarity constraints, we determine f+π(0)/f+K(0)2=0.9±0.3±0.3| f_+^{\pi}(0)/f_+^K(0)|^2=0.9\pm 0.3\pm 0.3 We also present a 90% confidence level upper limit for the branching ratio of the decay D+ηe+νeD^+ \to \eta e^+\nu_e relative to that for D+π0e+νeD^+ \to \pi^0 e^+\nu_e of 1.5.Comment: 10 page postscript file, postscript file also available through http://w4.lns.cornell.edu/public/CLN

    Molecular biogeography of planktonic and benthic diatoms in the Yangtze River

    Background: Diatoms are of great significance to primary productivity in oceans, yet little is known about their biogeographic distribution in oligotrophic rivers. Results: With the help of metabarcoding analysis of 279 samples from the Yangtze River, we provided the first integral biogeographic pattern of planktonic and benthic diatoms over a 6030 km continuum along the world's third largest river. Our study revealed spatial dissimilarity of diatoms under varying landforms, including plateau, mountain, foothill, basin, foothill-mountain, and plain regions, from the river source to the estuary. Environmental drivers of diatom communities were interpreted in terms of photosynthetically active radiation, temperature, channel slope and nutrients, and human interference. Typical benthic diatoms, such as Pinnularia, Paralia, and Aulacoseira, experienced considerable reduction in relative abundance downstream of the Three Gorges Dam and the Xiluodu Dam, two of the world's largest dams. Conclusions: Our study revealed that benthic diatoms are of particular significance in characterizing motile guild in riverine environments, which provides insights into diatom biogeography and biogeochemical cycles in large river ecosystems

    RESUMO O trato urinário normalmente é estéril, no entanto, pode ser contaminado por agentes etiológicos provenientes da microbiota intestinal, dentre os mais comuns pode-se destacar a Escherichia coli. Os microrganismos estão se tornando cada vez mais resistentes a múltiplos antimicrobianos. É notória, portanto, a necessidade de encontrar novas substâncias com propriedades antimicrobianas. Portanto, foram avaliados diferentes extratos de Phyllanthus sp, frente a microrganismos causadores de infecções no trato urinário e comparadas as técnicas de hole-plate e disco difusão. Para ambas as técnicas avaliadas, o extrato de 72 horas mostrou melhor atividade antimicrobiana, na técnica de disco difusão, a bactéria mais sensível foi o Staphylococcus saprophyticcus, que apresentou CMI (Concentração Mínima Inibitória) de 15,84 mg/mL. Com a utilização da técnica de hole-plate, a bactéria mais sensível observada foi Staphylococcus aureus, com valor de CMI igual ao reportado anteriormente. Foi observado que os extratos alcoólicos obtiveram maior eficiência em relação às infusões, que a técnica de hole-plate revelou ser mais eficiente que a técnica de disco difusão e que os cocos Gram positivos foram mais susceptíveis quando comparadas aos bacilos Gram negativos e fungos

    O objetivo deste trabalho foi avaliar os teores de lignina e celulose em plantas de cana-de-açúcar após a aplicação de dois maturadores para a colheita. O experimento foi conduzido em uma área de cana-soca, cultivar SP 803280, no município de Igaraçu do Tietê/SP. O delineamento experimental utilizado foi o de blocos casualizados, com quatro repetições. Os tratamentos constituíram-se da aplicação de dois maturadores: sulfometuron-methyl (Curavial) e glyphosate (Roundup original). As doses utilizadas foram: glyphosate a 72 g e.a. ha-1; glyphosate a 144 g e.a. ha-1 ; glyphosate a 72 g e.a. ha-1 + sulfometuron methyl a 10 g p.c. ha-1; glyphosate a 108 g e.a. ha-1 + sulfometuron-methyl a 12 g p.c. ha-1; sulfometuron-methyl a 20 g p.c. ha-1; e a testemunha sem aplicação de maturadores. As análises de lignina e celulose foram realizadas pelo método lignina em detergente ácido modificado. O glyphosate e o sulfometuron-methyl alteraram os níveis de lignina no momento da colheita, e esse efeito foi observado também durante o crescimento da cana-de-açúcar (meses após a aplicação desses produtos). O glyphosate a 72 g e.a. ha-1 promoveu reduções nos teores de lignina, na colheita e durante o crescimento da cana-de-açúcar, quando comparados com os da testemunha, enquanto o sulfometuron-methyl isolado na menor dose (10 g ha-1) promoveu aumento nos teores desse biopolímero na soqueira da cana-de-açúcar.The aim of this study was to evaluate the content of lignin and cellulose in sugarcane after application of ripeners. The experiment was carried out in a sugarcane ratoon area, variety SP803280 in Igaraçu Tietê, São Paulo, Brazil. The experimental design was a randomized complete block with four replications. The treatments consisted of application of two ripeners: sulfometuron methyl (Curavial) and glyphosate (Roundup Original). The doses used were: glyphosate 200 mL pc ha-1; glyphosate at 400 mL p.c. ha-1, glyphosate at 200 mL p.c. ha-1 plus sulfometuron methyl at 10 g p.c. ha-1; glyphosate at 150 mL p.c. ha-1 plus sulfometuron methyl at 12 g p.c. ha-1; sulfometuron methyl at 20 g p.c. ha-1 and the untreated control without ripener application. Lignin and cellulose analyses were made using acid detergent lignin modified. Glyphosate and sulfometuron methyl altered lignin levels at harvest and during sugarcane growth (months after application of the products). In general, glyphosate at 200 mL ha-1 promoted reductions in lignin compared to the control, while sulfometuron methyl isolated at a dose of 10 g ha-1 promoted an increase in the levels of this compound in sugarcane

    Efeitos de períodos de permanência de clomazone + hexazinona no solo e na palha de cana-de-açúcar antes da ocorrência de chuvas na eficácia de controle de plantas daninhas

    O presente trabalho teve por objetivo avaliar a eficácia da mistura formulada de clomazone e hexazinona aplicada sobre o solo e em associação com a palha de cana-de-açúcar, após diferentes períodos de permanência sem a ocorrência de chuvas. O experimento foi conduzido em vasos em casa de vegetação, no município de Botucatu-SP. Os vasos foram preenchidos com solo, e as sementes de plantas daninhas (Brachiaria decumbens, Ipomoea grandifolia, Ipomoea hederifolia e Euphorbia heterophylla) foram semeadas superficialmente e, em seguida, cobertas ou não com palha de cana-de-açúcar, dependendo do tratamento utilizado. O delineamento experimental utilizado foi o inteiramente casualizado, com quatro repetições. Os tratamentos foram dispostos em um esquema fatorial 3 x 6, sendo os fatores seis períodos sem ocorrência de chuvas (0, 3, 7, 15, 30 e 60 dias) e três posicionamentos do produto (sobre o solo sem palha, sobre e sob a palha de cana). Foram realizadas avaliações de controle aos 10, 21, 35 e 42 dias após a ocorrência da chuva. A mistura de clomazone + hexazinona promoveu excelentes resultados de controle para todas as espécies estudadas quando aplicada sobre, sob ou na ausência de palha de cana-de-açúcar. No entanto, observou-se tendência de redução nos níveis de controle para períodos superiores a 60 dias sem ocorrência de chuva.The objective of this work was to evaluate the efficacy of the formulated mixture clomazone and hexazinone applied in soil or combined with sugarcane mulch, after different periods of permanence without the occurrence of rain. The experiment was carried out in vases under greenhouse conditions in Botucatu, São Paulo, Brazil. The weeds Brachiaria decumbens, Ipomoea grandifolia, Ipomoea hederifolia and Euphorbia heterophylla were sown and covered or not with sugarcane mulch. The experiment was arranged in a complete randomized design with four repetitions. The treatments were displaced in a 3x6 factorial scheme, with the factors being six periods without rain ( 0, 3, 7, 15, 30, and 60 days) and three modes of herbicide application( on the soil without sugarcane mulch, on and under sugarcane mulch). Control evaluations were carried out at 10, 21, 35 and 42 days after the occurrence of rain. The clomazone + hexazinone mixture promoted an excellent weed control for all the species studied when applied on, under, or without sugarcane mulch. However, the control levels tended to reduce for periods over 60 days without rain
