    An optimized strategy to measure protein stability highlights differences between cold and hot unfolded states

    Macromolecular crowding ought to stabilize folded forms of proteins, through an excluded volume effect. This explanation has been questioned and observed effects attributed to weak interactions with other cell components. Here we show conclusively that protein stability is affected by volume exclusion and that the effect is more pronounced when the crowder's size is closer to that of the protein under study. Accurate evaluation of the volume exclusion effect is made possible by the choice of yeast frataxin, a protein that undergoes cold denaturation above zero degrees, because the unfolded form at low temperature is more expanded than the corresponding one at high temperature. To achieve optimum sensitivity to changes in stability we introduce an empirical parameter derived from the stability curve. The large effect of PEG 20 on cold denaturation can be explained by a change in water activity, according to Privalov's interpretation of cold denaturation

    Experimental and numerical assessment of fibre bridging toughening effects on the compressive behaviour of delaminated composite plates

    Increasing the Mode I inter-laminar fracture toughness of composite laminates can contribute to slowing down delamination growth phenomena, which can be considered one of the most critical damage mechanisms in composite structures. Actually, the Mode I interlaminar fracture toughness (GIc) in fibre-reinforced composite materials has been found to considerably increase with the crack length when the fibre bridging phenomenon takes place. Hence, in this paper, the fibre bridging phenomenon has been considered as a natural toughening mechanism able to replace embedded metallic or composite reinforcements, currently used to increase tolerance to inter-laminar damage. An experimental/numerical study on the influence of delamination growth on the compressive behaviour of fibre-reinforced composites characterised by high sensitivity to the fibre bridging phenomenon has been performed. Coupons, made of material systems characterised by a variable toughness related to a high sensitivity to the fibre bridging phenomenon and containing artificial through-the-width delaminations, were subjected to a compressive mechanical test and compared to coupons made of standard material system with constant toughness. Out-of-plane displacements and strains were monitored during the compression test by means of strain gauges and digital image correlation to assess the influence of fibre bridging on delamination buckling, delamination growth and on the global buckling of the specimens, including buckling shape changes. Experimental data were combined with a numerical study, performed by means of a virtual crack closure technique based procedure, named SMart Time XB-Fibre Bridging (SMXB-FB), able to mimic the crack bridging effect on the toughness properties of the material system. The combination of numerical results and experimental data has allowed the deformations and the buckling shape changes to be correlated to the onset and evolution of damage and, hence, contributes to improving the knowledge on the interaction of the failure mechanisms in the investigated composite specimens

    Microbioreactor (micro-Matrix) potential in aerobic and anaerobic conditions with different industrially relevant microbial strains

    Microscale fermentation systems are important high throughput tools in clone selection, and bioprocess set up and optimization, since they provide several parallel experiments in controlled conditions of pH, temperature, agitation, and gas flow rate. In this work we evaluated the performance of biotechnologically relevant strains with different respiratory requirements in the micro-Matrix microbioreactor. In particular Escherichia coli K4 requires well aerated fermentation conditions to improve its native production of chondroitin-like capsular polysaccharide, a biomedically attractive polymer. Results from batch and fed-batch experiments demonstrated high reproducibility with those obtained on 2 L reactors, although highlighting a pronounced volume loss for longer-term experiments. Basfia succiniciproducens and Actinobacillus succinogenes need CO2 addition for the production of succinic acid, a building block with several industrial applications. Different CO2 supply modes were tested for the two strains in 24 h batch experiments and results well compared with those obtained on lab-scale bioreactors. Overall, it was demonstrated that the micro-Matrix is a useful scale-down tool that is suitable for growing metabolically different strains in simple batch process, however, a series of issues should still be addressed in order to fully exploit its potential

    Abstract Purpose Nipple-areola reconstruction represents an important step for final mammary reconstruction. Many techniques have been described. The drawback is the progressive nipple projection loss with time from 50% to over 70% of the initial projection. In this report, we evaluated the effect of injectable poly-lactic acid (PLLA) to improve projection of reconstructed nipples. Results We selected 12 patients with a residual nipple projection between 0.1 and 2 mm. The patients were injected locally inside the nipple with 0.5 ml of PLLA (dilution 1:4) every 4 weeks for 4 times. At the study end, patients were satisfied with results. No adverse effects were observed. After one year, an increase of nipple projection ranging from 0.5 to 3.5 mm was obtained with an average increase of 2.3 mm (282%) and this variation was statistically significant ( p Conclusion The use of injectable PLLA is a simple and effective procedure to improve projection of reconstructed nipple

    The frail world of haemodialysis patients in the COVID-19 pandemic era: a systematic scoping review

    Background: Patients undergoing in-centre haemodialysis (HD) are particularly exposed to the dire consequences of COVID-19. The present systematic scoping review aims to identify the extent, range, and nature of articles related to COVID-19 and maintenance HD: it reports specifically the prevalence of the COVID-19 pandemic in the HD population, implementation of strategies for the prevention, mitigation and containment of the COVID-19 pandemic in HD centres, demographic and clinical characteristics, and outcomes of the pediatric and adult HD patients. Methods: A multi-step systematic search of the literature in Pubmed, Scopus, Ovid Medline, Embase and Web of Science, published between December 1, 2019, and January 30, 2021 was performed. Two authors separately screened the titles and abstracts of the documents and ruled out irrelevant articles. A report of the papers that met inclusion criteria was performed; then, a descriptive analysis of the characteristics of the included articles and a narrative synthesis of the results were performed. Results: The review process ended with the inclusion of 145 articles. Most of them were based on single-centre experiences, which spontaneously developed best practices. Most studies were conducted in high-income countries (69.7%) and a part of them (9.6%) were not in English. Prevalence of COVID-19 among dialysis patients accounted for 0%-37.6%. Preventive measures were reported in 54% of the included articles, with particular emphasis on education, triage, hygiene, and containment measures. Patients experienced a heterogeneous spectrum of symptoms that led 35%-88.2% of them to hospital admission. Median and mean hospital length of stay ranged from 8 to 28.5 and 16.2 to 22 days, respectively. Admission to intensive care units varied widely across studies (from 2.6% to 70.5%) and was associated with high mortality (42.8%–100%). Overall, prognosis was poor in 0%–47% of the hospitalized patients. Conclusions: This systematic scoping review provides an overview of the current knowledge on the impact of COVID-19 on the frail world of HD patients. Furthermore, it may help to implement the existing strategies of COVID-19 prevention and provide a list of unmet needs (safe transport, testing, shelter). Finally, it may be a stimulus for performing systematic reviews and meta-analyses which will form the basis for evidence-based guidelines. Graphic abstract: [Figure not available: see fulltext.

    The Static Failure of Adhesively Bonded Metal Laminate Structures: A Cohesive Zone Approach

    Data on distribution, ecology, biomass, recruitment, growth, mortality and productivity of the West African bloody cockle Anadara senilis were collected at the Banc d'Aguuin, Mauritania, in early 1985 and 1986. Ash-free dry weight appeared to be correlated best with shell height. A. senilis was abundant on the tidal flats of landlocked coastal bays, but nearly absent on the tidal flats bordering the open sea. The average biomass for the entire area of tidal flats was estimated at 5.5 g·m−2 ash-free dry weight. The A. senilis population appeared to consist mainly of 10 to 20-year-old individuals, showing a very slow growth and a production: biomass ratio of about 0.02 y−1. Recruitment appeared negligible and mortality was estimated to be about 10% per year. Oystercatchers (Haematopus ostralegus), the gastropod Cymbium cymbium and unknown fish species were responsible for a large share of this. The distinction of annual growth marks permitted the assessment of year-class strength, which appeared to be correlated with the average discharge of the river Senegal. This may be explained by assuming that year-class strength and river discharge both are correlated with rainfall at the Banc d'Arguin.

    Several studies suggest that inflammation has a pivotal role during the progression of osteoarthritis (OA) and cytokines have been identified as the main process mediators. This study aimed to explore the ability to modulate the main OA pro-inflammatory biomarkers of novel gels (H-HA/BC) based on high molecular weight hyaluronan (H-HA) and unsulfated biotechnological chondroitin (BC). For the first time, BC was tested also in combination with H-HA on human primary cells isolated from pathological knee joints. Specifically, the experiments were performed using an OA in vitro model based on human chondrocytes and synoviocytes. To evaluate the anti-inflammatory effects of H-HA/BC in comparison with H-HA and BC single gels, NF-kB, COMP-2, MyD88, MMP-13 and a wide range of cytokines, known to be specific biomarkers in OA (e.g., IL-6, IL-8, and TNF-α), were evaluated. In addition, cell morphology and proliferation occurring in the presence of either H-HA/BC or single components were assessed using time-lapse video microscopy. It was shown that synovial fluids and cells isolated from OA suffering patients, presented a cytokine pattern respondent to an ongoing inflammation status. H-HA and BC significantly reduced the levels of 23 biomarkers associated with cartilage damage. However, H-HA/BC decreased significantly 24 biological mediators and downregulated 19 of them more efficiently than the single components. In synoviocytes cultures, cytokine analyses proved that H-HA/BC gels re-established an extracellular environment more similar to a healthy condition reducing considerably the concentration of 11 analytes. Instead, H-HA and BC significantly modulated 7 (5 only with a longer treatment) and 8 biological cytokines, respectively. Our results suggest that H-HA/BC beyond the viscosupplementation effect typical for HA-based gels, can improve the inflammation status in joints and thus could be introduced as a valid protective and anti-inflammatory intraarticular device in the field of Class III medical devices for OA treatments

    Implementation of Telehealth in Radiation Oncology: Rapid Integration During COVID-19 and its Future Role in our Practice.

    Introduction: The widespread coronavirus disease 2019 (COVID-19) has resulted in significant changes in care delivery among radiation oncology practices and demanded the rapid incorporation of telehealth. However, the impact of a large-scale transition to telehealth in radiation oncology on patient access to care and the viability of care delivery are largely unknown. In this manuscript, we review our implementation and report data on patient access to care and billing implications. As telehealth is likely to continue after COVID-19, we propose a radiation oncology-specific algorithm for telehealth. Material and Methods: In March 2020, our department began to use telehealth for all new consults, post-treatment encounters, and follow-up appointments. Billable encounters from January to April 2020 were reviewed and categorized into one of the following visit types: in-person, telephonic, or two-way audio-video. Logistic regression models tested whether visit type differed by patient age, income, or provider. Results: There was a 35% decrease in billable activity from January to April. In-person visits decreased from 100% to 21%. Sixty percent of telehealth appointments in April were performed with two-way audio-video, and 40% by telephonic only. In-person consultation visits were associated with higher billing codes compared to two-way audio-video telehealth visits (p Conclusions: Since the onset of COVID-19 pandemic, we were able to move the majority of patient visits to telehealth but observed inconsistent utilization of the audio-video telehealth platform.     Midinfrared intersubband absorption in ZnxCd1−xSe/Znx′Cdy′Mg1−x′−y′Se multiple quantum well structures

    The authors report the observation of intersubband absorption in ZnxCd(1−x)Se/Znx′Cdy′Mg(1−x′−y′)Semultiple quantum wells. Lattice-matched samples were grown by molecular beam epitaxy on InP (001) substrates. Photoluminescence measurements indicate that the samples have excellent material quality. The peak absorption wavelengths measured by Fourier transform infrared spectroscopy are 3.99 and 5.35μm for two samples with ZnxCd(1−x)Se well widths of 28 and 42Å, respectively. These values fall within the 3–5μm wavelength range, which is of interest for midinfrared intersubband devices, such as quantum cascade lasers and quantum well infrared photodetectors. Their experimental results fit well with theoretical predictions based on the envelope function approximation. The results indicate that these wide band gap II-VI materials are very promising for midinfrared intersubband device applications

    Influence of the state of light on the optically induced interparticle interaction

    A general expression for the energy of interparticle interaction induced by an arbitrary mode of light is determined using quantum electrodynamics, and it is shown that the Casimir-Polder potential is included within this quantum result. Equations are also derived for the corresponding coupling induced by multimode number states of light, and the dependence of the pair energy on the Poynting vector and polarization state is determined. Attention is then focused on the interactions between particles trapped in counterpropagating coherent beams, both with and without interference, and it is shown that the results afford insights into the multiparticle structures that can be optically fabricated with counterpropagating input. Brief consideration is also given to the effect of squeezing the optical coherent state. Extending previous studies of optical binding in Laguerre-Gaussian beams, results are given for the case of particles trapped at radially different locations within the beam structure. Finally, consideration is given to interparticle interactions induced by broadband light, and it is shown how the length of optically fabricated particle chains can be controlled by the use of wavelength filters
