    Co-culture of microalgae, cyanobacteria, and macromycetes for exopolysaccharides production: process preliminary optimization and partial characterization.

    In this study, the biomass and exopolysaccharides (EPS) production in co-cultures of microalgae/cyanobacteria and macromycetes was evaluated as a technology for producing new polysaccharides for medical and/or industrial application. Based on biomass and EPS productivity of monocultures, two algae and two fungi were selected and cultured in different co-culture arrangements. The hydrosoluble EPS fractions from mono- and cocultures were characterized by ¹³C NMR spectroscopy and gas chromatography coupled to mass spectrometry and compared. It was found that co-cultures resulted in the production of an EPS different from those produced by monocultures, showing fungal predominance with microalgal/cyanobacterial traces. Co-cultures conditions were screened (temperature, agitation speed, fungal and microalgae inoculation rate, initial pH, illumination rate, and glucose concentration) in order to achieve maximum biomass and EPS production, resulting in an increase of 33 and 61% in exopolysaccharides and biomass productions, respectively (patent pending)

    Styrene degradation in perlite biofilter: the overall performance characteristics and dynamic response

    [Abstract] Styrene’s degradation in a perlite biofilter including the long-term operation, dynamic response to step-changes in inlet concentration and non-use periods were tested. The study was performed in a bench-scale biofilter with ID 100 mm and a bed height of 1 m. Perlite with a particle size of 2 – 4 mm was used as a packing material. An enrichment mixed culture was immobilized on the packing. The inoculum was obtained from a styrene biofilter. Two different loading conditions were tested: (1) Loading with a high inlet concentration and a high residence time. (2) Loading with a low inlet concentration and the low residence time. Both conditions are common in industrial practice. The dynamic response to a repeated step-change in the inlet concentration (from 50 to 200 mg.m-3) was tested. The dynamic behaviour of the restarting period after varying periods of non-use was also investigated. The results demonstrate a high biofilter stability under extreme loading conditions and also during the step-changes of the inlet concentration. The non-use periods tested had almost no effect on the biofilter performance. The maximum outlet concentration after the restarting of the load was 4 mg.m-3, when a 95 hours idle period was used. After shorter idle periods, the outlet styrene concentrations did not exceed 0.6 mg.m-3

    OCCURRENCE OF CRYPTOSPORIDIUM SP. IN DOGS AND CATS FROM CURITIBA AND ITS METROPOLITAN AREA

    The present study was carried out with the aim of assess the occurrence of Cryptosporidium sp. infection in dogs and cats in Curitiba and its metropolitan area, state of Paraná, Brazil. Techniques employed to detect the protozoan in fecal samples were: staining by Ziehl-Neelsen for oocysts search and nested polymerase chain reaction (nPCR) targeting the 18SSU rDNA gene. To attempt the proposed aim, 91 feces samples of dogs and 25 of cats were collected and analyzed. Ziehl-Neelsen technique was unable to detect any oocyst in both groups analyzed, showing a very low sensitivity. Results of nPCR showed an infection rate of 13.2% (12/91) and 4% (1/25) in dogs and cats respectively.  The implications of these epidemiological data are discussed in this work

    Milk kefir: Composition, microbial cultures, biological activities, and related products

    In recent years, there has been a strong focus on beneficial foods with probiotic microorganisms and functional organic substances. In this context, there is an increasing interest in the commercial use of kefir, since it can be marketed as a natural beverage that has health promoting bacteria. There are numerous commercially available kefir based-products. Kefir may act as a matrix in the effective delivery of probiotic microorganisms in different types of products. Also, the presence of kefir's exopolysaccharides, known as kefiran, which has biological activity, certainly adds value to products. Kefiran can also be used separately in other food products and as a coating film for various food and pharmaceutical products. This article aims to update the information about kefir and its microbiological composition, biological activity of the kefir's microflora and the importance of kefiran as a beneficial health substance.Facultad de Ciencias Exacta

    Unintentional staining of lens posterior capsule with trypan blue during extracapsular cataract extraction: case report

    Report of a complication related to the use of 0.1% trypan blue during capsular staining of the anterior crystalline lens capsule in an extracapsular cataract extraction with intraocular lens implantation. The capsular dye was injected using an air-bubble technique, inadvertently, it was done using high pressure dispersing the dye through the zonules leading to a complete losts of red reflex, the following steps of the procedure becoming more difficult after the cataract extraction and causing a temporary disturbance of visual acuity in the postoperative period. In order to avoid this complication, the authors also describe the use of Modified air-bubble technique where dispersive viscoelastic is placed into the iridolenticular space 360° to create a protection barrier.Relato de complicação relacionada ao uso do azul de tripano 0,1% ocorrida ao corar a cápsula anterior do cristalino em facectomia extracapsular com implante de lente intra-ocular. O corante foi injetado sob alta pressão utilizando a técnica de bolha de ar ocasionando a migração deste através da zônula e conseqüente perda do reflexo vermelho no peroperatório dificultando os passos subseqüentes à extração da catarata e comprometendo temporariamente a acuidade visual no pós-operatório.
