    Six solutions for more reliable infant research

    Infant research is often underpowered, undermining the robustness and replicability of our findings. Improving the reliability of infant studies offers a solution for increasing statistical power independent of sample size. Here, we discuss two senses of the term reliability in the context of infant research: reliable (large) effects and reliable measures. We examine the circumstances under which effects are strongest and measures are most reliable and use synthetic datasets to illustrate the relationship between effect size, measurement reliability, and statistical power. We then present six concrete solutions for more reliable infant research: (a) routinely estimating and reporting the effect size and measurement reliability of infant tasks, (b) selecting the best measurement tool, (c) developing better infant paradigms, (d) collecting more data points per infant, (e) excluding unreliable data from the analysis, and (f) conducting more sophisticated data analyses. Deeper consideration of measurement in infant research will improve our ability to study infant development

    Potential fire behavior is reduced following forest restoration treatments

    Potential fire behavior was compared under dry, windy weather conditions in 12 ponderosa pine stands treated with alternative thinning prescriptions in the wildland/urban interface of Flagstaff, Arizona. Prior to thinning, stands averaged 474 trees/acre, 158 ft2/acre basal area, crown bulk density 0.0045 lb/ft3, and crown base height 19.2 ft. Three thinning treatments differing in residual tree density were applied to each of three stands (total of nine treated, three control). Treatments were based on historic forest structure prior to Euro-American settlement and disruption of the frequent fire regime (circa 1876). Thinning reduced stand densities 7788 percent, basal areas 3566 percent, crown bulk densities 2448 percent, and raised crown base height an average of 11 ft. Before thinning, simulated fire behavior under the 97th percentile of June fire weather conditions was predicted to be intense but controllable (5.4 ft flame lengths). However, active or passive crownfires were simulated using crown base heights in the lowest quintile (20 percent) or winds gusting to 30 mph, representing the fuel ladders and wind gusts that are important for initiating crown burning. Under the identical conditions after thinning, all three treatments resisted crown burning. The degree of resistance was related to thinning intensity. It is crucial to remove thinning slash fuels through prescribed burning or other means. If not removed, slash fuels can cause crownfire behavior in the thinned stands under severe wildfire conditions. Finally, the crownfire resistance achieved through thinning will deteriorate over time unless maintenance burning and/or thinning is continued

    Case report: Recurrent cervical spinal stenosis masquerading as myalgic encephalomyelitis/chronic fatigue syndrome with orthostatic intolerance

    IntroductionMyalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a complex, chronic, multi-system disorder that is characterized by a substantial impairment in the activities that were well tolerated before the illness. In an earlier report, we had described three adult women who met criteria for ME/CFS and orthostatic intolerance, and had congenital or acquired cervical spinal stenosis. All three experienced substantial global improvements in their ME/CFS and orthostatic intolerance symptoms after recognition and surgical treatment of the cervical stenosis. After a several year period of improvement, one of the individuals in that series experienced a return of ME/CFS and orthostatic intolerance symptoms.Main symptoms and clinical findingsRadiologic investigation confirmed a recurrence of the ventral compression of the spinal cord due to a shift of the disc replacement implant at the involved cervical spinal level.Therapeutic interventionDecompression of the spinal cord with removal of the implant and fusion at the original C5-C6 level was once again followed by a similar degree of improvement in function as had been observed after the first operation.ConclusionThis recapitulation of the outcomes after surgical management of cervical stenosis provides further evidence in support of the hypothesis that cervical spinal stenosis can exacerbate pre-existing or cause new orthostatic intolerance and ME/CFS. Especially for those with refractory symptoms and neurological signs, surgical interventions may offer relief for selected patients with this complex condition

    The roots of bilingualism in newborns

    The first steps towards bilingual language acquisition have already begun at birth. When tested on their preference for English versus Tagalog, “monolingual” newborns, whose mothers spoke only English during pregnancy, showed a robust preference for English. In contrast, “bilingual” newborns, whose mothers spoke both English and Tagalog regularly during pregnancy, showed equal preference for both languages. A group of Chinese-English bilinguals showed an intermediate pattern of preference. Preference for two languages does not suggest confusion between them, however. Study 2 showed that both English monolinguals and Tagalog-English bilinguals could discriminate English from Tagalog. The same perceptual and learning mechanisms that support acquisition in a monolingual environment thus also naturally support bilingual acquisition

    Building a collaborative Psychological Science: Lessons Learned from ManyBabies 1

    The field of infancy research faces a difficult challenge: Some questions require samples that are simply too large for any 1 lab to recruit and test. ManyBabies aims to address this problem by forming large-scale collaborations on key theoretical questions in developmental science, while promoting the uptake of Open Science practices. Here, we look back on the first project completed under the ManyBabies umbrella—ManyBabies 1—which tested the development of infant-directed speech preference. Our goal is to share the lessons learned over the course of the project and to articulate our vision for the role of large-scale collaborations in the field. First, we consider the decisions made in scaling up experimental research for a collaboration involving 100+ researchers and 70+ labs. Next, we discuss successes and challenges over the course of the project, including the following: protocol design and implementation, data analysis, organisational structures and collaborative workflows, securing funding, and encouraging broad participation in the project. Finally, we discuss the benefits we see both in ongoing ManyBabies projects and in future large-scale collaborations in general, with a particular eye toward developing best practices and increasing growth and diversity in infancy research and psychological science in general. Throughout the article, we include first-hand narrative experiences to illustrate the perspectives of researchers playing different roles within the project. Although this project focused on the unique challenges of infant research, many of the insights we gained can be applied to large-scale collaborations across the broader field of psychology

    Arabidopsis KCBP interacts with AIR9 but stays in the cortical division zone throughout mitosis via its MyTH4-FERM domain

    The preprophase band of microtubules performs the crucial function of marking the plane of cell division. Although the preprophase band depolymerises at the onset of mitosis, the division plane is 'memorized' by a cortical division zone to which the phragmoplast is attracted during cytokinesis. Proteins have been discovered that are part of the molecular memory but little is known about how they contribute to phragmoplast guidance. Previously, we found that the microtubule-associated protein AIR9 is found in the cortical division zone at preprophase and returns during cell plate insertion but is absent from the cortex during the intervening mitosis. To identify new components of the preprophase memory, we searched for proteins that interact with AIR9. We detected the kinesin-like calmodulin-binding protein, KCBP, which can be visualized at the predicted cortical site throughout division. A truncation study of KCBP indicates that its MyTH4-FERM domain is required for linking the motor domain to the cortex. These results suggest a mechanism by which minus-end-directed KCBP helps guide the centrifugally expanding phragmoplast to the cortical division site.Instituto de Biotecnologia y Biologia Molecula

    Dual roles for LUBAC signaling in thymic epithelial cell development and survival

    Thymic epithelial cells (TECs) form a unique microenvironment that orchestrates T cell differentiation and immunological tolerance. Despite the importance of TECs for adaptive immunity, there is an incomplete understanding of the signalling networks that support their differentiation and survival. We report that the linear ubiquitin chain assembly complex (LUBAC) is essential for medullary TEC (mTEC) differentiation, cortical TEC survival and prevention of premature thymic atrophy. TEC-specific loss of LUBAC proteins, HOIL-1 or HOIP, severely impaired expansion of the thymic medulla and AIRE-expressing cells. Furthermore, HOIL-1-deficiency caused early thymic atrophy due to Caspase-8/MLKL-dependent apoptosis/necroptosis of cortical TECs. By contrast, deficiency in the LUBAC component, SHARPIN, caused relatively mild defects only in mTECs. These distinct roles for LUBAC components in TECs correlate with their function in linear ubiquitination, NFÎşB activation and cell survival.     ManyBabies 3: A multi-lab study of infant algebraic rule learning

    The ability to learn and apply rules lies at the heart of cognition. In a seminal study, Marcus, Vijayan, Rao, and Vishton (1999) reported that seven-month-old infants learned abstract rules over syllable sequences and were able to generalize those rules to novel syllable sequences. Dozens of studies have since extended on that research using different rules, modalities, stimuli, participants (human adults and non-human animals) and experimental procedures. Yet questions remain about the robustness of Marcus et al.’s (1999) core findings, as the presence of significant learning effects has been mixed.     The interest in air-water flows has not diminished in recent years, but it is accompanied by frequent citations of early, sometimes outdated articles. A basic issue is the inadequate, incomplete interpretation of air-water flow instrumentation by hydraulic engineers and researchers. This article comments on high-velocity air-water flow measurements by means of intrusive phase detection probes. This article focus on the bubbly flow structure of high-velocity air-water flow based upon measurements by means of intrusive phase detection probes. It is shown that some advanced post-processing techniques may yields expanded information on the air-water turbulent flow properties and bubbly flow structures. The outcomes demonstrate simple techniques in high-velocity air-water flow analysis
