    A sanitation technology demonstration centre to enhance decision making in South Africa

    Limited understanding of the characteristics of different sanitation technology options may affect acceptance of certain technologies in certain circumstances, as well as lead to little appreciation by the users of the long-term financial, environmental and institutional implications of operating and maintaining the various sanitation systems. In South Africa, this has resulted in disturbing increases in the numbers of poorly operated and maintained sanitation technologies across South Africa. The need to establish a facility where people could acquaint themselves with the various sanitation systems was recognised by the CSIR and WRC who made funding available for the establishment of the Sanitation Technology Demonstration Centre at the CSIR. This Centre is aimed at informing local, provincial and national authority officials, NGO’s, CBO’s, consultants, schools, universities, and importantly, communities themselves in order for them to make informed and educated decisions and choices regarding sanitation technologies

    Research in South Africa showed that in most cases the sanitation technology implemented in rural and peri-urban areas seemed to be adequate in providing sanitation services, but that these technologies are under strain due to their incorrect use, misuse/non-use and lack of proper maintenance, therefore placing the sustainability of these sanitation systems in question. Even if the technology was designed and built well, the use of the technology and its acceptance by the user proved to be the most critical elements - a technology is only as good as its user. Spot check assessments in South Africa concluded that water sector services and projects should not be viewed one-dimensionally, but holistically. The interaction between, and integration of, technical aspects and social dynamics are the cornerstones for the long-term operation of facilities and the sustainability of services delivery. This paper shows some results of spot check assessments and will draw the attention to the user issues and their impact on providing sustainable sanitation services in addressing the sanitation backlogs in South Africa

    Prejudices and attitudes toward reuse of nutrients from urine diversion toilets in South Africa

    More than 60 000 urine diversion toilets have been built in South Africa in the last decade, but the use of human excreta for food production is not generally being promoted. This paper discusses the study conducted by The Council For Scientific and Industrial Research (CSIR), investigating users’ perceptions and attitudes towards urine diversion sanitation (UDS) in South Africa for the past seven years. The research method focused on semi-structured household interviews while validating and cross-checking the responses with physical observation and small focus group discussions. The main findings of this research were that people were aware of the fertiliser value of human faeces but not of human urine and that only some users were willing to use only human faeces in their gardens. In South Africa, many communities rely on subsistence agriculture, often in poor soils. Therefore, it is important to change people’s views and attitudes (how they think about and act) towards human excreta in order to achieve ecological sanitation solutions for food production

    Dietary supplementation with Bifidobacterium lactis NCC2818 from weaning reduces local immunoglobulin production in lymphoid-associated tissues but increases systemic antibodies in healthy neonates

    Weaning is associated with a major shift in the microbial community of the intestine, and this instability may make it more acquiescent than the adult microbiota to long-term changes. Modulation achieved through dietary interventions may have potentially beneficial effects on the developing immune system, which is driven primarily by the microbiota. The specific aim of the present study was to determine whether immune development could be modified by dietary supplementation with the human probiotic Bifidobacterium lactis NCC2818 in a tractable model of weaning in infants. Piglets were reared by their mothers before being weaned onto a solid diet supplemented with B. lactis NCC2818, while sibling controls did not receive supplementation. Probiotic supplementation resulted in a reduction in IgA (P0·05). Probiotic-supplemented pigs had more mast cells than unsupplemented littermates (P<0·0001), although numbers in both groups were low. In addition, the supplemented piglets made stronger serum IgG responses to fed and injected antigens (P<0·05). The present findings are consistent with B. lactis NCC2818 reducing intestinal permeability induced by weaning, and suggest that the piglet is a valuable intermediate between rodent models and human infants. The results also strongly suggest that measures of the effect of probiotic supplementation on the immune system need to be interpreted carefully as proxy measures of health benefit.     RATIONALE: Neovascularization stimulated by local or recruited stem cells after ischemia is a key process that salvages damaged tissue and shows similarities with embryonic vascularization. Apelin receptor (Aplnr) and its endogenous ligand apelin play an important role in cardiovascular development. However, the role of apelin signaling in stem cell recruitment after ischemia is unknown. OBJECTIVE: To investigate the role of apelin signaling in recruitment after ischemia. METHODS AND RESULTS: Aplnr was specifically expressed in circulating cKit+/Flk1+ cells but not in circulating Sca1+/Flk1+ and Lin+ cells. cKit+/Flk1+/Aplnr+ cells increased significantly early after myocardial ischemia but not after hind limb ischemia, indicative of an important role for apelin/Aplnr in cell recruitment during the nascent biological repair response after myocardial damage. In line with this finding, apelin expression was upregulated in the infarcted myocardium. Injection of apelin into the ischemic myocardium resulted in accelerated and increased recruitment of cKit+/Flk1+/Aplnr+ cells to the heart. Recruited Aplnr+/cKit+/Flk1+ cells promoted neovascularization in the peri-infarct area by paracrine activity rather than active transdifferentiation, resulting into cardioprotection as indicated by diminished scar formation and improved residual cardiac function. Aplnr knockdown in the bone marrow resulted in aggravation of myocardial ischemia-associated damage, which could not be rescued by apelin. CONCLUSIONS: We conclude that apelin functions as a new and potent chemoattractant for circulating cKit+/Flk1+/Aplnr+ cells during early myocardial repair, providing myocardial protection against ischemic damage by improving neovascularization via paracine action

    CECR1-mediated cross talk between macrophages and vascular mural cells promotes neovascularization in malignant glioma

    Glioblastomas (glioblastoma multiforme, GBM) are most malignant brain tumors characterized by profound vascularization. The activation of macrophages strongly contributes to tumor angiogenesis during GBM development. Previously, we showed that extracellular adenosine deaminase protein Cat Eye Syndrome Critical Region Protein 1 (CECR1) is highly expressed by M2-like macrophages in GBM where it defines macrophage M2 polarization and contributes to tumor expansion. In this study, the effect of CECR1 in macrophages on tumor angiogenesis was investigated. Immunohistochemical evaluation of GBM tissue samples showed that the expression of CECR1 correlates with microvascular density in the tumors, confirming data from the TCGA set. In a three-dimensional co-culture system consisting of human pericytes, human umbilical vein endothelial cells and THP1-derived macrophages, CECR1 knockdown by siRNA and CECR1 stimulation of macrophages inhibited and promoted new vessel formation, respectively. Loss and gain of function studies demonstrated that PDGFB mRNA and protein levels in macrophages are modulated by CECR1. The proangiogenic properties of CECR1 in macrophages were partially mediated via paracrine activation of pericytes by PDGFB–PDGFRβ signaling. CECR1–PDGFB–PDGFRβ cross-activation between macrophages and pericytes promoted pericyte migration, shown by transwell migration assay, and enhanced expression and deposition of periostin, a matrix component with proangiogenic properties. CECR1 function in (M2-like) macrophages mediates cross talk between macrophages and pericytes in GBM via paracrine PDGFB–PDGFRβ signaling, promoting pericyte recruitment and migration, and tumor angiogenesis.     Cube law, condition factor and weight-length relationships: history, meta-analysis and recommendations

    This study presents a historical review, a meta-analysis, and recommendations for users about weight–length relationships, condition factors and relative weight equations. The historical review traces the developments of the respective concepts. The meta-analysis explores 3929 weight–length relationships of the type W = aLb for 1773 species of fishes. It shows that 82% of the variance in a plot of log a over b can be explained by allometric versus isometric growth patterns and by different body shapes of the respective species. Across species median b = 3.03 is significantly larger than 3.0, thus indicating a tendency towards slightly positive-allometric growth (increase in relative body thickness or plumpness) in most fishes. The expected range of 2.5 < b < 3.5 is confirmed. Mean estimates of b outside this range are often based on only one or two weight–length relationships per species. However, true cases of strong allometric growth do exist and three examples are given. Within species, a plot of log a vs b can be used to detect outliers in weight–length relationships. An equation to calculate mean condition factors from weight–length relationships is given as Kmean = 100aLb−3. Relative weight Wrm = 100W/(amLbm) can be used for comparing the condition of individuals across populations, where am is the geometric mean of a and bm is the mean of b across all available weight–length relationships for a given species. Twelve recommendations for proper use and presentation of weight–length relationships, condition factors and relative weight are given
