    Combining sustainable design education with research on pathway to zero energy historic buildings’

    This paper describes an attempt to bring together the Teaching & Learning experience of students of architecture, a live research project, and the collaboration between leaders of separate modules in order to provide a more integrated educational experience. Cork Centre for Architectural Education (CCAE) provides professional degrees which are recognised under the terms of the EU Directive on Professional Qualifications (Architecture). One of its distinguishing features is the inclusion of a Building Conservation module in the third year of its five-year programme. This module is delivered mostly by architects, engineers and historians who also provide professional services in this specialised area. The Third Year Design Studio usually responds to this specialist resource by setting students a design project which involves design for re-use, adaptation, or extension to an existing, and often historic, building. In 2019, CCAE became the Lead Partner in a Northern Periphery & Artic Region Programme project entitled “Energy Pathfinder – Towards Zero Energy Standards in Historic Buildings”, supported by the European Regional Development Fund. Partners in five countries are studying techniques to reduce energy demands and supply renewable energy to meet the balance of energy requirements in Historic Buildings, and to disseminate guidance. The new, stricter Zero-Energy-Building regulations that have been introduced throughout Europe present considerable challenges and opportunities to product manufacturers, builders, engineers and architects. However, the difficulties are multiplied when dealing with historical buildings, which are often granted exceptions from the strictest energy regulations. The design for retrofitting insulation, for example requires an understanding of the priorities for conservation in different buildings, as well as the skills required to prevent retrofitted insulation and draft stripping from causing problems of condensation and subsequent visual discoloration of finishes, and fabric deterioration. This requires a degree of sophistication in hygrothermal modelling, not always required in new-build design projects. Furthermore, the deployment of renewable energy systems such as photovoltaics, aerogenerators and heat pumps usually demands the installation of large equipment that might detract from the historical environments that society wishes to maintain. The visual impact of these technologies on historic buildings often leads to their rejection. The research programme requires the creation of demonstration projects in different countries. The building selected as a demonstrator in Cork, the re-use of a large, building used most recently by a religious order. This site was also selected as a conservation-related project for Year Three students. This paper examines the range of student proposed solutions, and the potential benefits of meshing a live research project into a teaching curriculum, which itself merged five distinct taught modules

    Good intentions in universal design: A global challenge for higher education

    It is not often that a high-level edict requires higher education centres to promote universal design through their programmes; however the recent United Nations Beijing Declaration and Action Plan (UNESCAP 2017) expressly states that, “academic institutions should provide training programmes on universal design for policymakers, building inspectors and contractors, as well as integrating universal design and accessibility into curricula related to architecture, urban planning, transport, civil engineering and other relevant academic branches”. This is particularly timely in the Asia-Pacific region, where economies continue to show massive expansion of their built environments. This imperative to future-proof any development therefore is vital, especially considering the growing percentile of older people with their needs for safe and accessible living. Achieving these ends clearly implies a need both to educate professionals and to enact appropriate codes and standards, which in turn require the training of personnel to carry them out. Anticipating this need, Goal 3 of the United Nations Incheon Strategy (UNESCAP 2012) optimistically calls for “civil society involvement in conducting accessibility audits, creating guidelines and advocacy work to promote universal design” and “to enhance mechanisms for tracking its progress”. While such good intentions are admirable, they will require radical steps to be achieved. The paper describes examples, including those from the writers' own experiences, outlining a range of practical methods which academics and teachers involved in inculcating universal design principles in both European and Asian centres, through their teaching, training and technology transfer, can positively support continued cooperation towards a more inclusive World for everyone

    Development of an in-house ELISA to detect anti-HPV16-L1 antibodies in serum and dried blood spots

    Measuring anti-HPV antibody levels is important for surveillance of the immunological response to both natural infection and vaccination. Here, an ELISA test for measurement of HPV-16L1 antibodies was developed and validated in sera and dried blood spots. An in-house ELISA was developed for measuring anti-HPV-16L1 IgA and IgG levels. The assay was standardized against WHO international standard serum and validated on serum, dried blood spots and cervical liquid based cytology samples from women attending colposcopy clinics in Scotland. Antibody avidity index was also measured in serum samples. The average HPV 16-L1 specific IgG and IgA levels measured in sera, in women attending a routine colposcopy service were 7.3 units/ml and 8.1 units/ml respectively. Significant correlations between serum and dried blood spot eluates for both IgG and IgA were observed indicating that the latter serve as a credible proxy for antibody levels. Average IgG Avidity Index was 35% (95% CI 25%-45%) suggesting previous, historical challenge with natural infection. This ELISA has potential for use in epidemiological and field studies of antibody prevalence and if coupled with avidity measurement may be of use in individual case monitoring of vaccine responses and failures

    A rapid assay for affinity and kinetics of molecular interactions with nucleic acids

    The Differential Radial Capillary Action of Ligand Assay (DRaCALA) allows detection of protein interactions with low-molecular weight ligands based on separation of the protein–ligand complex by differential capillary action. Here, we present an application of DRaCALA to the study of nucleic acid–protein interactions using the Escherichia coli cyclic AMP receptor protein (CRP). CRP bound in DRaCALA specifically to 32P-labeled oligonucleotides containing the consensus CRP binding site, but not to oligonucleotides with point mutations known to abrogate binding. Affinity and kinetic studies using DRaCALA yielded a dissociation constant and dissociation rate similar to previously reported values. Because DRaCALA is not subject to ligand size restrictions, whole plasmids with a single CRP-binding site were used as probes, yielding similar results. DNA can also function as an easily labeled carrier molecule for a conjugated ligand. Sequestration of biotinylated nucleic acids by streptavidin allowed nucleic acids to take the place of the protein as the immobile binding partner. Therefore, any molecular interactions involving nucleic acids can be tested. We demonstrate this principle utilizing a bacterial riboswitch that binds cyclic-di-guanosine monophosphate. DRaCALA is a flexible and complementary approach to other biochemical methods for rapid and accurate measurements of affinity and kinetics at near-equilibrium conditions

    Computer-facilitated Review of Electronic Medical Records Reliably Identifies Emergency Department Interventions in Older Adults

    An estimated 14% to 25% of all scientific studies in peer-reviewed emergency medicine (EM) journals are medical records reviews. The majority of the chart reviews in these studies are performed manually, a process that is both time-consuming and error-prone. Computer-based text search engines have the potential to enhance chart reviews of electronic emergency department (ED) medical records. The authors compared the efficiency and accuracy of a computer-facilitated medical record review of ED clinical records of geriatric patients with a traditional manual review of the same data and describe the process by which this computer-facilitated review was completed. Clinical data from consecutive ED patients age 65 years or older were collected retrospectively by manual and computer-facilitated medical record review. The frequency of three significant ED interventions in older adults was determined using each method. Performance characteristics of each search method, including sensitivity and positive predictive value, were determined, and the overall sensitivities of the two search methods were compared using McNemar's test. For 665 patient visits, there were 49 (7.4%) Foley catheters placed, 36 (5.4%) sedative medications administered, and 15 (2.3%) patients who received positive pressure ventilation. The computer-facilitated review identified more of the targeted procedures (99 of 100, 99%), compared to manual review (74 of 100 procedures, 74%; p < 0.0001). A practical, non-resource-intensive, computer-facilitated free-text medical record review was completed and was more efficient and accurate than manually reviewing ED records

    Izu-Bonin-Mariana Rear Arc: The Missing Half of the Subduction Factory

    4GT) lies in the western part of the Izu fore-arc basin, ~60 km east of the arc-front volcano Aogashima, ~170 km west of the axis of the Izu-Bonin Trench, 1.5 km west of Ocean Drilling Program (ODP) Site 792, and at 1776 meters below sea level (mbsl). It was drilled as a 150 m deep geotechnical test hole for potential future deep drilling (5500 meters below seafloor [mbsf]) at proposed Site IBM-4 using the D/V Chikyu. Core from Site U1436 yielded a rich record of Late Pleistocene explosive volcanism, including distinctive black glassy mafic ash layers that may record large-volume eruptions on the Izu arc front. Because of the importance of this discovery, Site U1436 was drilled in three additional holes (U1436B, U1436C, and U1436D), as part of a contingency operation, in an attempt to get better recovery on the black glassy mafic ash layers and enclosing sediments and to better constrain the thickness of the mafic ash layers. IODP Site U1437 is located in the Izu rear arc, ~330 km west of the axis of the IzuBonin Trench and ~90 km west of the arc-front volcanoes Myojinsho and Myojin Knoll, at 2117 mbsl. The primary scientific objective for Site U1437 was to characterize “the missing half of the subduction factory”; this was because numerous ODP/Integrated Ocean Drilling Program sites had been drilled in the arc to fore-arc region (i.e., ODP Site 782A Leg 126), but this was the first site to be drilled in the rear part of the Izu arc. A complete view of the arc system is needed to understand the formation of oceanic arc crust and its evolution into continental crust. Site U1437 on the rear arc had excellent core recovery in Holes U1437B and U1437D, and we succeeded in hanging the longest casing ever in the history of R/V JOIDES Resolution scientific drilling (1085.6 m) in Hole U1437E and cored to 1806.5 mbsf

    Fasting and surgery timing (FaST) audit

    Background & aimsInternational guidance advocates the avoidance of prolonged preoperative fasting due to its negative impact on perioperative hydration. This study aimed to assess the adherence to these guidelines for fasting in patients undergoing elective and emergency surgery in the East Midlands region of the UK.MethodsThis prospective audit was performed over a two-month period at five National Health Service (NHS) Trusts across the East Midlands region of the UK. Demographic data, admission and operative details, and length of preoperative fasting were collected on adult patients listed for emergency and elective surgery.ResultsOf the 343 surgical patients included within the study, 50% (n = 172) were male, 78% (n = 266) had elective surgery and 22% (n = 77) underwent emergency surgery. Overall median fasting times (Q1, Q3) were 16.1 (13.0, 19.4) hours for food and 5.8 (3.5, 10.7) hours for clear fluids. Prolonged fasting >12 h was documented in 73% (n = 250) for food, and 21% (n = 71) for clear fluids. Median fasting times from clear fluids and food were longer in the those undergoing emergency surgery when compared with those undergoing elective surgery: 13.0 (6.4, 22.6) vs. 4.9 (3.3, 7.8) hours, and 22.0 (14.0, 37.4) vs. 15.6 (12.9, 17.8) hours respectively, p < 0.0001.ConclusionsDespite international consensus on the duration of preoperative fasting, patients continue to fast from clear fluids and food for prolonged lengths of time. Patients admitted for emergency surgery were more likely to fast for longer than those having elective surgery

    Expanding the diversity of mycobacteriophages: insights into genome architecture and evolution.

    Mycobacteriophages are viruses that infect mycobacterial hosts such as Mycobacterium smegmatis and Mycobacterium tuberculosis. All mycobacteriophages characterized to date are dsDNA tailed phages, and have either siphoviral or myoviral morphotypes. However, their genetic diversity is considerable, and although sixty-two genomes have been sequenced and comparatively analyzed, these likely represent only a small portion of the diversity of the mycobacteriophage population at large. Here we report the isolation, sequencing and comparative genomic analysis of 18 new mycobacteriophages isolated from geographically distinct locations within the United States. Although no clear correlation between location and genome type can be discerned, these genomes expand our knowledge of mycobacteriophage diversity and enhance our understanding of the roles of mobile elements in viral evolution. Expansion of the number of mycobacteriophages grouped within Cluster A provides insights into the basis of immune specificity in these temperate phages, and we also describe a novel example of apparent immunity theft. The isolation and genomic analysis of bacteriophages by freshman college students provides an example of an authentic research experience for novice scientists
