    Analysis of Oct4-dependent transcriptional networks regulating self-renewal and pluripotency in human embryonic stem cells

    The POU domain transcription factor OCT4 is a key regulator of pluripotency in the early mammalian embryo and is highly expressed in the inner cell mass of the blastocyst. Consistent with its essential role in maintaining pluripotency, Oct4 expression is rapidly downregulated during formation of the trophoblast lineage. To enhance our understanding of the molecular basis of this differentiation event in humans, we used a functional genomics approach involving RNA interference-mediated suppression of OCT4 function in a human ESC line and analysis of the resulting transcriptional profiles to identify OCT4-dependent genes in human cells. We detected altered expression of >1,000 genes, including targets regulated directly by OCT4 either positively (NANOG, SOX2, REX1, LEFTB, LEFTA/EBAF DPPA4, THY1, and TDGF1) or negatively (CDX2, EOMES, BMP4, TBX18, Brachyury [T], DKK1, HLX1, GATA6, ID2, and DLX5), as well as targets for the OCT4-associated stem cell regulators SOX2 and NANOG. Our data set includes regulators of ACTIVIN, BMP, fibroblast growth factor, and WNT signaling. These pathways are implicated in regulating human ESC differentiation and therefore further validate the results of our analysis. In addition, we identified a number of differentially expressed genes that are involved in epigenetics, chromatin remodeling, apoptosis, and metabolism that may point to underlying molecular mechanisms that regulate pluripotency and trophoblast differentiation in humans. Significant concordance between this data set and previous comparisons between inner cell mass and trophectoderm in human embryos indicates that the study of human ESC differentiation in vitro represents a useful model of early embryonic differentiation in humans

    Oil and gas infrastructure decommissioning in marine protected areas: System complexity, analysis and challenges

    Many offshore oil and gas production facilities are nearing the end of their operational life, with decommissioning now becoming a global challenge. The compatibility of decommissioning operations to marine protected areas (MPAs) creates further challenges. The recently-developed DAPSI(W)R(M) problem structuring framework (covering Drivers, Activities, Pressures, State changes, Impacts (on Welfare) and Responses (as Measures)) was applied here to interrogate the complexity of decommissioning oil and gas infrastructure within MPAs, with outputs feeding into the development of a novel database tool for Screening Potential Impacts of Decommissioning Activities (SPIDA). In meeting the current requirements of the marine regulatory regime, SPIDA provides a more streamlined, evidence-based process which can be applied by industry, statutory nature conservation bodies and regulators for identifying and evaluating evidence that supports the implications of decommissioning alternatives on the condition of MPAs. SPIDA has been developed to be adapted for other activities and sectors, including offshore renewables

    What are the costs and benefits of biodiversity recovery in a highly polluted estuary?

    Biodiversity recovery measures have often been ignored when dealing with the restoration of degraded aquatic systems. Furthermore, biological valuation methods have been applied only spatially in previous studies, and not jointly on a temporal and spatial scale. The intense monitoring efforts carried out in a highly polluted estuary, in northern Spain (Nervión estuary), allowed for the economic valuation of the costs and the biological valuation of the benefits associated with a 21 years sewage scheme application. The analysis show that the total amount of money invested into the sewage scheme has contributed to the estuary's improvement of both environmental and biological features, as well as to an increase in the uses and services provided by the estuary. However, the inner and outer parts of the estuary showed different responses. An understanding of the costs and trajectories of the environmental recovery of degraded aquatic systems is increasingly necessary to allow policy makers and regulators to formulate robust, cost-efficient and feasible management decisions

    The evolution of ocean literacy: A new framework for the United Nations Ocean Decade and beyond

    First introduced in the early 2000s, the concept of ocean literacy has evolved in recent years, not least since its inclusion as a mechanism for change within the United Nations Ocean Decade's goals. Building on early definitions of ocean literacy, there has been increasing recognition of a range of additional dimensions which contribute to an individual or collective sense of ‘ocean literacy’. Drawing on existing research, and parallel and supporting concepts, e.g., marine citizenship, ocean connectedness, and public perceptions research, this paper proposes ten dimensions of ocean literacy: knowledge, communication, behaviour, awareness, attitudes, activism, emotional connection, access and experience, adaptive capacity and trust and transparency, and recommends expanding previously recognised dimensions, in a bid to ensure that ocean literacy encompasses diverse knowledges, values and experiences. The paper provides a useful framework for ongoing ocean literacy research, and highlights aspects of ocean literacy which have received limited focus to date

    Expanding the role of participatory mapping to assess ecosystem service provision in local coastal environments

    There has been increasing international effort to better understand the diversity and quality of marine natural capital, ecosystem services and their associated societal benefits. However, there is an evidence gap as to how these benefits are identified at the local scale, where benefits are provided and to whom, trade-offs in development decisions, and understanding how benefits support well-being. Often the benefits of conservation are poorly understood at the local scale, are not effectively integrated into policy and are rarely included meaningfully in public discourse. This paper addresses this disjuncture and responds to the demand for improving dialogue with local communities and stakeholders. Participatory GIS mapping is used as a direct means of co-producing knowledge with stakeholder and community interests. This paper drives a shift from development of participatory approaches to adaptive applications in real-world case studies of local, national and international policy relevance. The results from four sites along the UK North Sea coast are presented. This paper showcases a robust stakeholder-driven approach that can be used to inform marine planning, conservation management and coastal development. Although the demonstration sites are UK-focused, the methodology presented is of global significance and can be applied across spatial and temporal scales

    Marine ecosystem services: Linking indicators to their classification

    © 2014 Elsevier Ltd. All rights reserved. There is a multitude of ecosystem service classifications available within the literature, each with its own advantages and drawbacks. Elements of them have been used to tailor a generic ecosystem service classification for the marine environment and then for a case study site within the North Sea: the Dogger Bank. Indicators for each of the ecosystem services, deemed relevant to the case study site, were identified. Each indicator was then assessed against a set of agreed criteria to ensure its relevance and applicability to environmental management. This paper identifies the need to distinguish between indicators of ecosystem services that are entirely ecological in nature (and largely reveal the potential of an ecosystem to provide ecosystem services), indicators for the ecological processes contributing to the delivery of these services, and indicators of benefits that reveal the realized human use or enjoyment of an ecosystem service. It highlights some of the difficulties faced in selecting meaningful indicators, such as problems of specificity, spatial disconnect and the considerable uncertainty about marine species, habitats and the processes, functions and services they contribute to

    The Interplay among Organisational Learning Culture, Agility, Growth and Big Data Capabilities

    This paper examines how an organisational learning culture impacts organisational agility by developing a model based on dynamic capabilities. The model treats agility as a dynamic capability and explains how an organisational learning culture (OLC) triggers a chain reaction through its influence on organisational agility (OA) that ultimately results in company growth. This paper also investigates the role of big data capabilities in transferring learning outcomes into dynamic capabilities. The model is tested through data collected from a survey of 138 Australian companies. Partial least squares structural equation modeling is adopted to empirically demonstrate how agility fully mediates the impact of the learning culture on growth. In addition, this paper further sheds light on the moderating role of big data competencies on the effects of OLC on OA. After presenting the results with implications to theory and practice, the paper ends with suggestions for future studies

    A rabies lesson improves rabies knowledge amongst primary school children in Zomba, Malawi

    <div><p>Rabies is an important neglected disease, which kills around 59,000 people a year. Over a third of these deaths are in children less than 15 years of age. Almost all human rabies deaths in Africa and Asia are due to bites from infected dogs. Despite the high efficacy of current rabies vaccines, awareness about rabies preventive healthcare is often low in endemic areas. It is therefore common for educational initiatives to be conducted in conjunction with other rabies control activities such as mass dog vaccination, however there are few examples where the efficacy of education activities has been assessed. Here, primary school children in Zomba, Malawi, were given a lesson on rabies biology and preventive healthcare. Subsequently, a mass dog vaccination programme was delivered in the same region. Knowledge and attitudes towards rabies were assessed by a questionnaire before the lesson, immediately after the lesson and 9 weeks later to assess the impact the lesson had on school children’s knowledge and attitudes. This assessment was also undertaken in children who were exposed to the mass dog vaccination programme but did not receive the lesson. Knowledge of rabies and how to be safe around dogs increased following the lesson (both p<0.001), and knowledge remained higher than baseline 9 weeks after the lesson (both p<0.001). Knowledge of rabies and how to be safe around dogs was greater amongst school children who had received the lesson compared to school children who had not received the lesson, but had been exposed to a rabies vaccination campaign in their community (both p<0.001) indicating that the lesson itself was critical in improving knowledge. In summary, we have shown that a short, focused classroom-based lesson on rabies can improve short and medium-term rabies knowledge and attitudes of Malawian schoolchildren.</p></div
