Effective arithmetic for multivariate algebraic series

We explain how to encode an algebraic series by a finite amount of data, namely its minimal polynomial, a total order and its first terms with respect to this order, and how to do effective arithmetic on the level of these encodings. The reasoning is based on the Newton-Puiseux algorithm and an effective equality test for algebraic series. We also explain how the latter allows to derive information about the support of an algebraic series, e.g. how to compute the (finitely many) vertices and bounded faces of the convex hull of its support.Comment: 8 pages, comes with a Mathematica implementation of the Newton-Puiseux algorithm and a Mathematica noteboo

    Handicapped people in China

(DIPF/Orig.)This article deals with the situation and living conditions of handicapped people in China. It describes the school and educational system, training and working conditions, rehabilitative and preventive activities as well as the public view and the view of the Chinese government on disabled persons. All in all it can be noticed that living conditions of handicapped people have improved a lot in China by, amongst others, national models of integration and local support. Nevertheless, there is still a big difference between rural and urban areas. (DIPF/Orig.

Dielectric constants of bulk ferroelectric PZTmeasured by terahertz time-domain spectroscopy

The complex permittivity of bulk ceramic ferroelectric of nominal composition PbZr0.4Ti0.6O3 was measured in the range 0.2–2 THz using transmission time-domain spectroscopy. The results indicate strong absorption and dispersion in this frequency range as often seen in highly disordered and polar materials. The results are compared to equivalent thin film data in the literature, and significant differences in the real and imaginary permittivity suggest that substrate clamping and degree of polarisation of the ferroelectric thin film materials affect dielectric properties even at these high frequencies

Vascular Morphogenesis in the Context of Inflammation: Self-Organization in a Fibrin-Based 3D Culture System

Introduction: New vessel formation requires a continuous and tightly regulated interplay between endothelial cells with cells of the perivascular microenvironment supported by mechanic-physical and chemical cues from the extracellular matrix.Aim: Here we investigated the potential of small fragments of synovial tissue to form de novo vascular structures in the context of inflammation within three dimensional (3D) fibrin-based matrices in vitro, and assessed the contribution of mesenchymal stromal cell (MSC)-immune cell cross-talk to neovascularization considering paracrine signals in a fibrin-based co-culture model.Material and Methods: Synovial tissue fragments from patients with rheumatoid arthritis (RA) and inflammatory osteoarthritis (OA) were cultivated within 3D fibrin matrices for up to 4 weeks. Cellular and structural re-arrangement of the initially acellular matrix were documented by phase contrast microscopy and characterized by confocal laser-scanning microscopy of topographically intact 3D cultures and by immunohistochemistry. MSC-peripheral blood mononuclear cell (PBMC) co-cultures in the 3D fibrin system specifically addressed the influence of perivascular cell interactions to neo-vessel formation in a pro-inflammatory microenvironment. Cytokine levels in the supernatants of cultured explant tissues and co-cultures were evaluated by the Bio-Plex cytokine assay and ELISA.Results: Vascular outgrowth from the embedded tissue into the fibrin matrix was preceded by leukocyte egress from the tissue fragments. Neo-vessels originating from both the embedded sample and from clusters locally formed by emigrated mononuclear cells were consistently associated with CD45(+) leukocytes. MSC and PBMC in co-culture formed vasculogenic clusters. Clusters and cells with endothelial phenotype emerging from them, were surrounded by a collagen IV scaffold. No vascular structures were observed in control 3D monocultures of PBMC or MSC. Paracrine signals released by cultured OA tissue fragments corresponded with elevated levels of granulocyte-colony stimulating factor, vascular endothelial growth factor and interleukin-6 secreted by MSC-PBMC co-cultures.Conclusion: Our results show that synovial tissue fragments with immune cell infiltrates have the potential to form new vessels in initially avascular 3D fibrin-based matrices. Cross-talk and cluster formation of MSC with immune cells within the 3D fibrin environment through self-organization and secretion of pro-angiogenic paracrine factors can support neo-vessel growth

Rab27a controls HIV-1 assembly by regulating plasma membrane levels of phosphatidylinositol 4,5-bisphosphate

During the late stages of the HIV-1 replication cycle, the viral polyprotein Pr55Gag is recruited to the plasma membrane (PM), where it binds phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2) and directs HIV-1 assembly. We show that Rab27a controls the trafficking of late endosomes carrying phosphatidylinositol 4-kinase type 2 α (PI4KIIα) toward the PM of CD4+ T cells. Hence, Rab27a promotes high levels of PM phosphatidylinositol 4-phosphate and the localized production of PI(4,5)P2, therefore controlling Pr55Gag membrane association. Rab27a also controls PI(4,5)P2 levels at the virus-containing compartments of macrophages. By screening Rab27a effectors, we identified that Slp2a, Slp3, and Slac2b are required for the association of Pr55Gag with the PM and that Slp2a cooperates with Rab27a in the recruitment of PI4KIIα to the PM. We conclude that by directing the trafficking of PI4KIIα-positive endosomes toward the PM, Rab27a controls PI(4,5)P2 production and, consequently, HIV-1 replication.Fil: Pereyra Gerber, Federico Pehuén.
