    Poloxamer 338 affects cell adhesion and biofilm formation in escherichia coli: Potential applications in the management of catheter-associated urinary tract infections

    Poloxamers are nontoxic, amphiphilic copolymers used in different formulations. Due to its surfactant properties, Poloxamer 338 (P388) is herein proposed as a strategy to avoid biofilm formation often causing recalcitrant catheter-associated urinary tract infections (CAUTI). The aim is to evaluate the ability of P388 coatings to affect the adhesion of Ec5FSL and Ec9FSL Escherichia coli strains on silicone urinary catheters. Attenuated total reflection infrared spectroscopy, atomic force microscopy, and static water contact angle measurement were employed to characterize the P388-coated silicone catheter in terms of amount of P388 layered, coating thickness, homogeneity, and hydrophilicity. In static conditions, the antifouling power of P388 was defined by comparing the E. coli cells adherent on a hydrophilic P388-adsorbed catheter segment with those on an uncoated one. A P388-coated catheter, having a homogeneous coverage of 35 nm in thickness, reduced of 0.83 log10 and 0.51 log10 the biofilm of Ec5FSL and Ec9FSL, respectively. In dynamic conditions, the percentage of cell adhesion on P388-adsorbed silicone channels was investigated by a microfluidic system, simulating the in vivo conditions of catheterized patients. As a result, both E. coli isolates were undetected. The strong and stable antifouling property against E. coli biofilm lead us to consider P388 as a promising anti-biofilm agent for CAUTIs control

    Carbon isotope discrimination and water use efficiency in interspecific Prunus hybrids subjected to drought stress

    In C3 plants, carbon isotope composition (δ13C) is influenced by isotopic effects during diffusion from the atmosphere to the chloroplasts and carboxylation reactions. This work aimed to demonstrate if δ13C of leaf soluble carbohydrates (δ13Cleaves) and of dry matter from new-growth shoots (δ13Cshoots) of Prunus plants subjected to a period of water deficit was related to water use efficiency (WUE). For this purpose, three interspecific Prunus hybrids rootstocks (6–5, 7-7 and G × N) were gradually subjected to drought and then rewatered. Soil water content (SWC) decreased from 26.1 to 9.4% after 70 days of water shortage, when plants reached values of predawn leaf water potential (LWP) ranging from −3.12 to −4.00 MPa. Gas exchange, particularly net photosynthetic and transpiration rates, differed among the three hybrids, leading to different values of WUE. After 70 days of drought, a significant δ13C increase of 5.86, 4.28 and 4.99‰ was observed in 6–5, 7-7 and G × N, respectively. Significant correlations between δ13C and other parameters (substomatal CO2/atmospheric CO2 ratio, stomatal conductance and stem water potential) were found in all hybrids. The rewatering phase caused a recovery of the physiological status of the plants. The isotope composition of δ13Cshoots was correlated with the average WUE measured during the whole experiment. δ13Cleaves and δ13Cshoots were positively related (r = 0.87; p < 0.001). The isotopic signature was a reliable screening tool to identify Prunus genotypes tolerant to drought stress. The results suggest the possibility of using δ13C as an integrated indicator of level of drought stress in plants subjected to prolonged stress conditions

    Heterosis, and expressivity of apospory in tetraploid Bahiagrass hybrids

    Breeding procedures developed for apomictic species are based on the idea of fixing superior hybrids by apomixis. However, scarce information is available about the occurrence of heterosis in apomictic hybrids. The objective was to generate a group of apomixis-segregating tetraploid bahiagrass families, evaluate the occurrence of heterosis for a series of agronomic and morphological traits, determine the level of apospory expressivity among hybrids, and estimate the genetic distance among parents and its relationship to heterosis and apospory expressivity. In total, 11 tetraploid families were generated by crossing sexual and apomitic genotypes. The segregation for mode of reproduction was analyzed using a RAPD marker linked to apospory in bahiagrass, and the level of apospory expressivity was determined using embryo sac observations. The genetic distances between parents were determined using ISSR markers. The ratio between sexual and aposporic hybrids varied from 1:1 to 7:1 among families. Discontinuous variation for apospory expressivity was observed in the hybrids, with either low or high levels being exhibited. Mid-parent, high-parent, and standard-heterosis was observed for all evaluated agronomic characteristics. The level of heterosis was dependent on the combination of parents involved, and also on the specific trait. There was a low correlation between genetic distances among parents and initial growth and the level of apospory expressivity. The occurrence of heterosis, and the segregation and expressivity for apospory were highly dependent on the combination of sexual and apomictic parents.Fil: Zilli, Alex Leonel.     The increased use of the MATRICS Consensus Cognitive Battery (MCCB) to investigate cognitive dysfunctions in schizophrenia fostered interest in its sensitivity in the context of family studies. As various measures of the same cognitive domains may have different power to distinguish between unaffected relatives of patients and controls, the relative sensitivity of MCCB tests for relative-control differences has to be established. We compared MCCB scores of 852 outpatients with schizophrenia (SCZ) with those of 342 unaffected relatives (REL) and a normative Italian sample of 774 healthy subjects (HCS).     Teaching precision farming and entrepreneurship for European students: Sparkle online course

    Within the framework of the European project named ‘SPARKLE’, an online course was created after studying educational needs on precision agriculture (PA), state of the art of technologies and a prospective study of the commercial sector. Five educational and research institutions, high-tech farms and enterprises specializing in technology transfer created the syllabus of the course and the platform contents. The course was designed to provide 30 h of student dedication, via online presentations, documents and videos for each topic. A free pilot course started in April 2020 and 385 students from Italy, Portugal, Greece & Spain enrolled. To trace performance and acquisition of competences, questionnaires were completed by students for each topic and a final overall test. Students’ opinions about the course were also registered using anonymous polls, and results evaluated, to be able to enhance the Sparkle course for subsequent editions. Students also took part in a business model competition, to solve real challenges proposed by farms, related to the use of these technologies

    Regulation of Human Formyl Peptide Receptor 1 Synthesis: Role of Single Nucleotide Polymorphisms, Transcription Factors, and Inflammatory Mediators

    The gene encoding the human formyl peptide receptor 1 (FPR1) is heterogeneous, containing numerous single nucleotide polymorphisms (SNPs). Here, we examine the effect of these SNPs on gene transcription and protein translation. We also identify gene promoter sequences and putative FPR1 transcription factors. To test the effect of codon bias and codon pair bias on FPR1 expression, four FPR1 genetic variants were expressed in human myeloid U937 cells fused to a reporter gene encoding firefly luciferase. No significant differences in luciferase activity were detected, suggesting that the translational regulation and protein stability of FPR1 are modulated by factors other than the SNP codon bias and the variant amino acid properties. Deletion and mutagenesis analysis of the FPR1 promoter showed that a CCAAT box is not required for gene transcription. A −88/41 promoter construct resulted in the strongest transcriptional activity, whereas a −72/41 construct showed large reduction in activity. The region between −88 and −72 contains a consensus binding site for the transcription factor PU.1. Mutagenesis of this site caused significant reduction in reporter gene expression. The PU.1 binding was confirmed in vivo by chromatin immunoprecipitation, and the binding to nucleotides −84 to −76 (TTCCTATTT) was confirmed in vitro by an electrophoretic mobility shift assay. Thus, similar to many other myeloid genes, FPR1 promoter activity requires PU.1. Two single nucleotide polymorphisms at −56 and −54 did not significantly affect FPR1 gene expression, despite differences in binding of transcription factor IRF1 in vitro. Inflammatory mediators such as interferon-γ, tumor necrosis factor-α, and lipopolysaccharide did not increase FPR1 promoter activity in myeloid cells, whereas differentiation induced by DMSO and retinoic acid enhanced the activity. This implies that the expression of FPR1 in myeloid cells is developmentally regulated, and that the differentiated cells are equipped for immediate response to microbial infections

    Unsupervised assessment of microarray data quality using a Gaussian mixture model

    <p>Abstract</p> <p>Background</p> <p>Quality assessment of microarray data is an important and often challenging aspect of gene expression analysis. This task frequently involves the examination of a variety of summary statistics and diagnostic plots. The interpretation of these diagnostics is often subjective, and generally requires careful expert scrutiny.</p> <p>Results</p> <p>We show how an unsupervised classification technique based on the Expectation-Maximization (EM) algorithm and the naïve Bayes model can be used to automate microarray quality assessment. The method is flexible and can be easily adapted to accommodate alternate quality statistics and platforms. We evaluate our approach using Affymetrix 3' gene expression and exon arrays and compare the performance of this method to a similar supervised approach.</p> <p>Conclusion</p> <p>This research illustrates the efficacy of an unsupervised classification approach for the purpose of automated microarray data quality assessment. Since our approach requires only unannotated training data, it is easy to customize and to keep up-to-date as technology evolves. In contrast to other "black box" classification systems, this method also allows for intuitive explanations.</p

    Poly (diglycerol adipate) variants as enhanced nanocarrier replacements in drug delivery applications

    Sustainably derived poly(glycerol adipate) (PGA) has been deemed to deliver all the desirable features expected in a polymeric scaffold for drug-delivery, including biodegradability, biocompatibility, self-assembly into nanoparticles (NPs) and a functionalisable pendant group. Despite showing these advantages over commercial alkyl polyesters, PGA suffers from a series of key drawbacks caused by poor amphiphilic balance. This leads to weak drug-polymer interactions and subsequent low drug-loading in NPs, as well as low NPs stability. To overcome this, in the present work, we applied a more significant variation of the polyester backbone while maintaining mild and sustainable polymerisation conditions. We have investigated the effect of the variation of both hydrophilic and hydrophobic segments upon physical properties and drug interactions as well as self-assembly and NPs stability. For the first time we have replaced glycerol with the more hydrophilic diglycerol, as well as adjusting the final amphiphilic balance of the polyester repetitive units by incorporating the more hydrophobic 1,6-n-hexanediol (Hex). The properties of the novel poly(diglycerol adipate) (PDGA) variants have been compared against known polyglycerol-based polyesters. Interestingly, while the bare PDGA showed improved water solubility and diminished self-assembling ability, the Hex variation demonstrated enhanced features as a nanocarrier. In this regard, PDGAHex NPs were tested for their stability in different environments and for their ability to encode enhanced drug loading. Moreover, the novel materials have shown good biocompatibility in both in vitro and in vivo (whole organism) experiments
