    How does the pitch and pattern of a signal affect auditory arousal thresholds?

    How arousal thresholds vary with different sounds is a critical issue for emergency awakenings, especially as sleepers are dying in fires despite having a working smoke alarm. Previous research shows that the current high pitched (3000+ Hz) smoke alarm signal is significantly less effective than an alternative signal, the 520 Hz square wave, in all populations tested. However, as the number of sounds tested has been small further research is needed. Here we measured auditory arousal thresholds (AATs) across signals with a range of characteristics to determine the most effective waking signal. Thirty nine young adults participated over three nights. In Part A, nine signals were presented in stage 4 sleep with ascending decibel levels. Signals were short beeps in the low to mid frequency range with different spectral complexities: square waves, pure tones, whoops and white noise. Part B manipulated temporal patterns, inserting silences of 0, 10 and 21 seconds after each 12 seconds of beeps. It was found that the low frequency (400 and 520 Hz) square waves yielded significantly lower AATs than the alternatives. A trend was found across the three temporal manipulations, with a 10 second intervening silence showing some advantage. These findings support earlier research indicating that the best sound for awakening from deep sleep is a low frequency square wave. It is argued that the signal with the lowest response threshold when awake may be the same as the most arousing signal when asleep, especially where the sleeper processes the signal as meaningful

    Efficient algorithms for reconfiguration in VLSI/WSI arrays

    The issue of developing efficient algorithms for reconfiguring processor arrays in the presence of faulty processors and fixed hardware resources is discussed. The models discussed consist of a set of identical processors embedded in a flexible interconnection structure that is configured in the form of a rectangular grid. An array grid model based on single-track switches is considered. An efficient polynomial time algorithm is proposed for determining feasible reconfigurations for an array with a given distribution of faulty processors. In the process, it is shown that the set of conditions in the reconfigurability theorem is not necessary. A polynomial time algorithm is developed for finding feasible reconfigurations in an augmented single-track model and in array grid models with multiple-track switche

    Constraints on Late Time Violations of the Equivalence Principle in the Dark Sector

    If dark energy is dynamical due to the evolution of a scalar field, then in general it is expected that the scalar is coupled to matter. While couplings to the standard model particles are highly constrained by local experiments, bounds on couplings to dark matter (DM) are only obtained from cosmological observations and they are consequently weaker. It has recently been pointed out that the coupling itself can become non-zero only at the time of dark energy domination, due to the evolution of dark energy itself, leading to a violation of the equivalence principle (EP) in the dark sector at late times. In this paper we study a specific model and show that such late-time violations of the EP in the DM sector are not strongly constrained by the evolution of the cosmological background and by observables in the linear regime (e.g. from the cosmic microwave background radiation). A study of perturbations in non-linear regime is necessary to constrain late--time violations of the equivalence principle much more strongly.Comment: 8 pages, 4 figures, 2 tables.     The Arg389Gly Beta1-Adrenoceptor Polymorphism and Catecholamine Effects on Plasma-Renin Activity

    ObjectivesThe purpose of this research was to find out whether, in humans, dobutamine-induced hemodynamic effects and increase in plasma-renin activity (PRA) might be beta1-adrenoceptor (β1AR) genotype-dependent.BackgroundIn vitro Arg389Gly-β1AR polymorphism exhibits decreased receptor signaling.MethodsWe studied 10 male homozygous Arg389-β1AR subjects and 8 male homozygous Gly389β1AR subjects; to avoid influences of codon 49 polymorphism, all were homozygous Ser49-β1AR. Subjects were infused with dobutamine (1 to 6 Οg/kg/min) with or without bisoprolol (10 mg orally) pretreatment, and PRA, heart rate, contractility, and blood pressure were assessed.ResultsWith regard to PRA, dobutamine increased PRA more potently in Arg389-β1AR versus Gly389-β1AR subjects. Bisoprolol markedly suppressed the dobutamine-induced PRA increase in Arg389- but only marginally in Gly389-β1AR subjects. With regard to hemodynamics, dobutamine caused larger heart rate and contractility increases and diastolic blood pressure decreases in Arg389- versus Gly389-β1AR subjects. Bisoprolol reduced dobutamine-induced heart rate and contractility increases and diastolic blood pressure decreases more potently in Arg389- versus Gly389-β1AR subjects.ConclusionsCodon 389 β1AR polymorphism is a determinant not only of hemodynamic effects but also of PRA. Thus, β1AR polymorphisms may be useful for predicting therapeutic responses to βAR-blocker treatment

    Slow roll inflation in the presence of a dark energy coupling

    In models of coupled dark energy, in which a dark energy scalar field couples to other matter components, it is natural to expect a coupling to the inflaton as well. We explore the consequences of such a coupling in the context of single-field slow-roll inflation. Assuming an exponential potential for the quintessence field we show that the coupling to the inflaton causes the quintessence field to be attracted toward the minimum of the effective potential. If the coupling is large enough, the field is heavy and is located at the minimum. We show how this affects the expansion rate and the slow-roll of the inflaton field, and therefore the primordial perturbations generated during inflation. We further show that the coupling has an important impact on the processes of reheating and preheating

    Simultaneous determination of seven β-lactam antibiotics in human plasma for therapeutic drug monitoring and pharmacokinetic studies

    There is strong evidence in literature supporting the benefit of monitoring plasma concentrations of β-lactam antibiotics in the critically ill to ensure appropriateness of dosing. The objective of this work was to develop a method for the simultaneous determination of total concentrations piperacillin, benzylpenicillin, flucloxacillin, meropenem, ertapenem, cephazolin and ceftazidime in human plasma. Sample preparation involved protein precipitation with acetonitrile containing 0.1% formic acid and subsequent dilution of supernatant with 0.1% formic acid in water. Chromatographic separation was achieved on a reversed phase column (C18, 2.6. μm, 2.1. ×. 50. mm) via gradient elution using water and acetonitrile, each containing 0.1% formic acid, as mobile phase. Tandem mass spectrometry (MSMS) analysis was performed, after electrospray ionization in the positive mode, with multiple reaction monitoring (MRM). The method is accurate with the inter-day and intra-day accuracies of quality control samples (QCs) ranging from 95 to 107% and 95 to 108%, respectively. It is also precise with intra-day and inter-day coefficient of variations ranging from 4 to 12% and 5 to 14%, respectively. The lower limit of quantification was 0.1. μg/mL for each antibiotic except flucloxacillin (0.25. μg/mL). Recovery was greater than 96% for all analytes except for ertapenem (78%). Coefficients of variation for the matrix effect were less than 10% over the six batches of plasma. Analytes were stable over three freeze-thaw cycles, and for reasonable hours on the bench top as well as post-preparation. This novel liquid chromatography tandem mass spectrometry method proved accurate, precise and applicable for therapeutic drug monitoring and pharmacokinetic studies of the selected β-lactam antibiotics

    Understanding the competing pathways leading to hydropyrene and isoelisabethatriene

    Terpene synthases are responsible for the biosynthesis of terpenes, the largest family of natural products. Hydropyrene synthase generates hydropyrene and hydropyrenol as its main products along with two byproducts, isoelisabethatrienes A and B. Fascinatingly, a single active site mutation (M75L) diverts the product distribution towards isoelisabethatrienes A and B. In the current work, we study the competing pathways leading to these products using quantum chemical calculations in the gas phase. We show that there is a great thermodynamic preference for hydropyrene and hydropyrenol formation, and hence most likely in the synthesis of the isoelisabethatriene products kinetic control is at play
