Práca sa postupne zaoberá jednotlivými prvkami a vysvetľuje základne pojmy z oblasti mainframov a tvorby incidentov na týchto systémoch. Teoretická časť slúži k vysvetleniu základných pojmov z oblasti ITIL-u a IBM Mainframe. Následná praktická časť diplomovej práce najprv analyzuje súčasný stav vytvárania incidentov na systémoch IBM Mainframe, analýzu incidentov, u ktorých je možná optimalizácia a následná optimalizácia vybraného incidentu. V závere diplomovej práce je štatistické vyjadrenie optimalizácie a ekonomický prínos pre firmu.Abstract The aim of the diploma work was the rationalization process of incident management on IBM Mainframe at IT company. This work gradually deals with various elements and explains the basic concepts of the mainframe and the creation of incidents on those systems. The theoretical section is to explain the basic concepts of ITIL-in and IBM Mainframe. Follow the practical part of the diploma work firstly analyzes the current situation of creating incidents on the IBM mainframe system, analysis of incidents in which it is possible to optimize and subsequent optimization of the selected incident.     Failure of noninvasive prediction of pulmonary hypertension in patients with idiopathic pulmonary fibrosis

    Background. Pulmonary hypertension (PH) in patients with advanced idiopathic pulmonary fibrosis (IPF) is a complication connected with unfavorable prognosis. Great efforts have been made in attempting to establish a reliable non-invasive method which would enable detection of this complication. In this context a formula using pulmonary function parameters was published with outstanding results. Methods. We tested the formula in 27 IPF patients who underwent a lung function examination, cardiac ultrasonography and catheterisation on the same day. Results. Pulmonary hypertension was detected by catheterisation in 17 patients (63%). In our group, contrary to the published data, the aforementioned formula was neither useful for detecting patients with a high probability of PH nor as a means of calculating the mean pulmonary artery pressure in individual patients (p=0.502 and p=0.833, respectively). Ultrasound examination reached borderline correlation with the values measured by catheterisation when we compare patients with relevant results (r=0.531, p=0.051). However, the examination gave no usable results in 13 patients (48%). Conclusion. Our data suggests that no reliable, noninvasive method is currently available for detecting and confirming PH in IPF patients. We did not confirm the usefulness of the published formula. Further carefully organised studies will be necessary to verify or refute it

    Nutritional cognitive neuroscience of aging: Focus on carotenoids and cognitive frailty

    The term „nutritional cognitive neuroscience” was recently established to define a research field focusing on the impact of nutrition on cognition and brain health across the life span. In this overview, we summarize the robust evidence on the role of carotenoids as micronutrients with different biological properties in persons with cognitive (pre)frailty. As neurodegenerative processes during aging occur in a continuum from brain aging to dementia, we propose the name „nutritional cognitive neuroscience of aging“ to define research on the role of nutrition and micronutrients in cognitive frailty. Further studies are warranted which integrate carotenoid interventions in multidomain, personalized lifestyle strategies

    The relation between amyotrophic lateral sclerosis and inorganic selenium in drinking water: a population-based case-control study

    <p>Abstract</p> <p>Background</p> <p>A community in northern Italy was previously reported to have an excess incidence of amyotrophic lateral sclerosis among residents exposed to high levels of inorganic selenium in their drinking water.</p> <p>Methods</p> <p>To assess the extent to which such association persisted in the decade following its initial observation, we conducted a population-based case-control study encompassing forty-one newly-diagnosed cases of amyotrophic lateral sclerosis and eighty-two age- and sex-matched controls. We measured long-term intake of inorganic selenium along with other potentially neurotoxic trace elements.</p> <p>Results</p> <p>We found that consumption of drinking water containing ≥ 1 μg/l of inorganic selenium was associated with a relative risk for amyotrophic lateral sclerosis of 5.4 (95% confidence interval 1.1-26) after adjustment for confounding factors. Greater amounts of cumulative inorganic selenium intake were associated with progressively increasing effects, with a relative risk of 2.1 (95% confidence interval 0.5-9.1) for intermediate levels of cumulative intake and 6.4 (95% confidence interval 1.3-31) for high intake.</p> <p>Conclusion</p> <p>Based on these results, coupled with other epidemiologic data and with findings from animal studies that show specific toxicity of the trace element on motor neurons, we hypothesize that dietary intake of inorganic selenium through drinking water increases the risk for amyotrophic lateral sclerosis.</p

    The effect of body weight on altered expression of nuclear receptors and cyclooxygenase-2 in human colorectal cancers

    <p>Abstract</p> <p>Background</p> <p>Epidemiological studies on risk factors for colorectal cancer (CRC) have mainly focused on diet, and being overweight is now recognized to contribute significantly to CRC risk. Overweight and obesity are defined as an excess of adipose tissue mass and are associated with disorders in lipid metabolism. Peroxisome proliferator-activated receptors (PPARs) and retinoid-activated receptors (RARs and RXRs) are important modulators of lipid metabolism and cellular homeostasis. Alterations in expression and activity of these ligand-activated transcription factors might be involved in obesity-associated diseases, which include CRC. Cyclooxygenase-2 (COX-2) also plays a critical role in lipid metabolism and alterations in COX-2 expression have already been associated with unfavourable clinical outcomes in epithelial tumors. The objective of this study is to examine the hypothesis questioning the relationship between alterations in the expression of nuclear receptors and COX-2 and the weight status among male subjects with CRC.</p> <p>Method</p> <p>The mRNA expression of the different nuclear receptor subtypes and of COX-2 was measured in 20 resected samples of CRC and paired non-tumor tissues. The association between expression patterns and weight status defined as a body mass index (BMI) was statistically analyzed.</p> <p>Results</p> <p>No changes were observed in PPARγ mRNA expression while the expression of PPARδ, retinoid-activated receptors and COX-2 were significantly increased in cancer tissues compared to normal colon mucosa (<it>P </it>≤ 0.001). The weight status appeared to be an independent factor, although we detected an increased level of COX-2 expression in the normal mucosa from overweight patients (BMI ≥ 25) compared to subjects with healthy BMI (<it>P </it>= 0.002).</p> <p>Conclusion</p> <p>Our findings show that alterations in the pattern of nuclear receptor expression observed in CRC do not appear to be correlated with patient weight status. However, the analysis of COX-2 expression in normal colon mucosa from subjects with a high BMI suggests that COX-2 deregulation might be driven by excess weight during the colon carcinogenesis process.</p

    DNA methylation patterns in bladder cancer and washing cell sediments: a perspective for tumor recurrence detection

    <p>Abstract</p> <p>Background</p> <p>Epigenetic alterations are a hallmark of human cancer. In this study, we aimed to investigate whether aberrant DNA methylation of cancer-associated genes is related to urinary bladder cancer recurrence.</p> <p>Methods</p> <p>A set of 4 genes, including <it>CDH1 </it>(E-cadherin), <it>SFN </it>(stratifin), <it>RARB </it>(retinoic acid receptor, beta) and <it>RASSF1A </it>(Ras association (RalGDS/AF-6) domain family 1), had their methylation patterns evaluated by MSP (Methylation-Specific Polymerase Chain Reaction) analysis in 49 fresh urinary bladder carcinoma tissues (including 14 cases paired with adjacent normal bladder epithelium, 3 squamous cell carcinomas and 2 adenocarcinomas) and 24 cell sediment samples from bladder washings of patients classified as cancer-free by cytological analysis (control group). A third set of samples included 39 archived tumor fragments and 23 matched washouts from 20 urinary bladder cancer patients in post-surgical monitoring. After genomic DNA isolation and sodium bisulfite modification, methylation patterns were determined and correlated with standard clinic-histopathological parameters.</p> <p>Results</p> <p><it>CDH1 </it>and <it>SFN </it>genes were methylated at high frequencies in bladder cancer as well as in paired normal adjacent tissue and exfoliated cells from cancer-free patients. Although no statistically significant differences were found between <it>RARB </it>and <it>RASSF1A </it>methylation and the clinical and histopathological parameters in bladder cancer, a sensitivity of 95% and a specificity of 71% were observed for <it>RARB </it>methylation (Fisher's Exact test (p < 0.0001; OR = 48.89) and, 58% and 17% (p < 0.05; OR = 0.29) for <it>RASSF1A </it>gene, respectively, in relation to the control group.</p> <p>Conclusion</p> <p>Indistinct DNA hypermethylation of <it>CDH1 </it>and <it>SFN </it>genes between tumoral and normal urinary bladder samples suggests that these epigenetic features are not suitable biomarkers for urinary bladder cancer. However, <it>RARB </it>and <it>RASSF1A </it>gene methylation appears to be an initial event in urinary bladder carcinogenesis and should be considered as defining a panel of differentially methylated genes in this neoplasia in order to maximize the diagnostic coverage of epigenetic markers, especially in studies aiming at early recurrence detection.</p

    Gaius’ Concept of The Law of Nations ( Ius Gentium ) and Natural Law ( Ius Naturale )

    The paper, after a brief presentation of the classical Roman jurist Gaius and his mostimportant and best known work, Gai Institutionum commentarii quattuor, presentsand analyzes his legal texts concerning mainly ius gentium and its relationship to iusnaturale. At the beginning of his textbook, this Roman jurist distinguishes only twotypes of law: civil law – ius civile and the law of nations – ius gentium (Gai Inst. 1, 1).Gaius mentions the law of nations (ius gentium) explicitly in the above mentionedfragment in a way that he relates it to “natural ratio” – natural order or natural reason.Gaius defined the concept of ius gentium two centuries after Cicero. Unlike Cicerowho was more a philosopher than a lawyer and perceived “ius gentium” mainly inan abstract sense, Gaius came up with the concrete concept of ius gentium, whichwas clearly separated from the concept of ius civile. Ius gentium is the law which iscommon for all nations. On the other hand ius civile is the law which concerns onlyRoman citizens. Ius gentium is the law which obliges both Romans and foreigners,because its basis is “naturalis ratio”.In addition, Gaius, in characterizing ius gentium, gives attention to two elements:a) any standards apply to all nations; b) or standards come out of natural reason(naturalis ratio). Gaius in this way presented two perspectives on one and the sameius gentium which are intrinsically and therefore necessarily linked. The first point ofview is concrete and the second one is abstract. In other words Gaius presented somelegal institutions which belong to the area of ius gentium from the concrete point ofview, the other ones from the abstract point of view, which is based on the so-called“naturalis ratio”.As far as the category of ius naturale is concerned, Gaius, in terms of content, identifiedit with ius gentium. So ius gentium differs from ius naturale only on the basis ofa point of view, which individual standards or legal institutions are expressed through.If legal standards are expressed from the concrete point of view (i.e. in standardsthere is an explicit mention that they apply to all nations), then they can be includedin the category of ius gentium. Na początku swojego podręcznika ten rzymski jurysta wyróżnił tylko dwa rodzaje prawa: prawo cywilne – ius civile i prawo narodów – ius gentium(Gai Inst. 1, 1). Gaius wyraźnie przywołał prawo narodów (ius gentium) we wspomnianym fragmencie w taki sposób, że odnosiło się do naturalis ratio – porządkunaturalnego. W ten sposób przedstawił dwie perspektywy na jedno i to samo ius gentium, które są wewnętrznie ze sobą powiązane.Pierwszy punkt widzenia jest konkretny, a drugi abstrakcyjny. Innymi słowy, Gaiusprzedstawił pewne instytucje prawne, które należą do obszaru ius gentium z konkretnego punktu widzenia, pozostałe z abstrakcyjnego punktu widzenia, który opierał sięna tak zwanym naturalis ratio. Jeśli chodzi o kategorię ius naturale, Gaius pod względem treści zidentyfikował ją zapomocą ius gentium. Więc ius gentium różni się od ius naturale tylko na podstawiepunktu widzenia, przez który wyrażane są poszczególne standardy lub instytucjeprawne. Jeśli normy prawne są wyrażone z konkretnego punktu widzenia (czyli w normach istnieje wyraźna wzmianka, że mają one zastosowanie do wszystkich narodów),wówczas mogą one zostać włączone do kategorii ius gentium. Druhá kapitola sa zaoberá analázou súčasneho stavu webových stránok firmy TMI. Tretia kapitola potom popisuje návrh a realizáciu webového portálu a vysvetluje funkčnosť jednotlivých prvkov.The bacherol work describes creation of an website focused on a software company. The aim of this thesis is to create a fully working website which provides all necesary information. Piece of this work is also a multimedia content. The second chapter deals with analysis of the present situation in the scopre of the web presentation.
