    Development of relations between the Union of Czechoslovak writers and the Communist Party of Czechoslovakia in the 1960s

    The objective of this bachelor thesis is to reflect key moments of the development of relations between the Communist Party of Czechoslovakia and the Union of Czechoslovak Writers in the 1960s when the cultural sphere was no more understood as autonomous but should represent ideological superstructure of the regime. The following work tends to describe the development of this uneven relation in view of the fact that yet in 1962 the party leadership considered this creative union along with its magazines to be the "gear-lever" of the regime. However, in the course of next few years the conflicts between the monopole incumbent and the creative union were escalating and eventually in 1970, after the invasion by the armies of the Warsaw Pact in 1968, led to the demise of UCW. Thus, following bachelor thesis raises questions such as; if and when, from the perspective of CPC, the Union of Czechoslovak Writers terminated to play the role of the "gear-lever" and which methods were consequently introduced by the party apparatus.Cílem bakalářské práce je reflektovat klíčové momenty ve vývoji vztahů mezi Komunistickou stranou Československa a Svazem československých spisovatelů v průběhu 60. let, kdy kulturní sféra nepředstavovala autonomní oblast, ale měla sloužit jako ideologická nadstavba režimu. Práce se pokusí zachytit, jakým způsobem se tento nerovný vztah vyvíjel s ohledem na fakt, že stranické vedení ještě v roce 1962 počítalo s tímto tvůrčím svazem a jeho periodiky jako s jednou z "převodových pák" režimu. V období následujících několika let však spory mezi reprezentantem mocenského monopolu a tvůrčím svazem postupně gradovaly, a nakonec vedly roku 1970, po invazi vojsk Varšavské smlouvy v roce 1968, k zániku SČSS.     Charakterizace buněčného prionového proteinu krevních destiček

    Souhrn Centrálním dějem při vzniku transmisivních neurodegenerativních prionových chorob je konformační změna buněčného prionového proteinu (PrPc) na patologickou isoformu (PrPsc). V současnosti jsou už známé čtyři případy přenosu jedné z prionových chorob, variantní Creutzfeldt-Jakobovy choroby, krevní transfúzí. Porozumění vlastnostem a chování PrPc na krevních elementech je proto velmi důležité. Tato práce se zabývá buněčným prionovým proteinem na krevních destičkách. V první části práce jsme charakterizovali PrPc krevních destiček jako protein ukotvený glykosylfosfatidylinositolovou kotvou, který je schopný vázat měďnaté ionty prostřednictvím N-koncové oktapeptidové repetice. Zjistili jsme, že ze tří možných glykoforem PrPc převládá na destičkách diglykosylovaná forma proteinu. Prokázali jsme, že PrPc krevních destiček není resistentní k proteinase K, čímž se odlišuje od patologické formy proteinu. Potvrdili jsme, že krevní destičky i erytrocyty nesou nezanedbatelné množství PrPc, a mohou se tak stát zdrojem infektivity při přenosu prionových chorob krevní transfúzí. V druhé části práce jsme detailně popsali lokalizaci PrPc v krevních destičkách. Pomocí fluorescenčního značení jsme ukázali, že se PrPc nachází jak v -granulích tak na cytoplasmatické membráně. Většina PrPc krevních destiček je součástí...The conformational conversion of the cellular prion protein (PrPc) to the misfolded isoform (PrPsc) is the central pathogenic event in the transmissible neurodegenerative prion diseases. The recently shown transmissibility of variant Creutzfeldt-Jakob disease by blood transfusion emphasizes the need for better understanding of the PrPc in blood. In the current thesis, we focused on blood platelet PrPc, which has not been very well described so far. In the first part of the thesis, platelet PrPc was characterized as glycosylphosphatidylinositol- anchored glycoprotein with dominant diglycosylated form. Platelet PrPc was shown to be sensitive to cleavage with proteinase K, which is a feature discriminating between cellular and pathological prion protein. We have confirmed that platelet PrPc binds copper ions by its N- terminal octapeptide repeat region. Regarding quantity of PrPc molecules expressed on blood elements we have proved that both platelets and red blood cells express considerable amount of PrPc and thus can not be neglected in the problematic of prions transmission by blood transfusion. The detailed study regarding PrPc localization in blood platelets is presented in the second part of the thesis.     PIV and LIF study of flow and thermal fields of twine plumes in water

    Flow and thermal fields of a pair of plane plumes in water are investigated by means of PIV and LIF experiments. The plumes are generated from thermal line sources, which are made out of electrically heated cylinders with a diameter of D = 1.21 mm. A cylinder-to-cylinder distance was 17.9 D. Either continuous or pulsating heating were used with the same heating input power. Because the cylinder-to-cylinder distance is moderately small, deflections of plumes from a vertical direction occur and the plumes are inclined together. This behavior is caused by a confined entrainment from a space between the both plumes. For a continuous heating, low frequency oscillations were identified and the natural frequency was evaluated as 0.5 Hz. Based on this finding, pulsating heating was used at the subharmonic frequency of 0.25 Hz. The maximum time-mean velocity magnitude at the continuous and pulsating heating were commensurable, approximately 0.007 m/s. On the other hand, pulsating heating achieves by 36 % higher velocity peaks. A very strong velocity oscillations were generated by pulsating heating at the distance approximately 8.3 D above the cylinders, where the velocity maxima oscillate along the time-mean value of 0.0057 m/s from −30% to +70 %. Temperature fields reasonably agree with this findings, despite a relatively fast equalization of the temperature field was concluded.     Existují značné interkulturní rozdíly v pojetí nemoci a péče o seniory s demencí. Tyto přístupy rozšiřují a v mnohém upevňují pohled na tuto problematiku. Pro-rodinná orientace afroamerických národů významně redukuje pečovatelskou zátěž. Z hlediska kvality a úrovně poskytované péče v rodině se ukazuje jako relevantní samotné chápání podstaty nemoci demence. Rodinní pečující národnostních menšin u nás jen zřídka využívají služeb konzultační činnosti a dalších.There are considerable intercultural differences in the concept of the disease and care for the elderly with dementia. These approaches are expanding and in many ways strengthening the view of this issue. The pro-family orientation of African-American nations significantly reduces the caregiver burden. From the point of view of the quality of care provided in the family, the understanding of the essence of dementia is relevant. Family caregivers of national minorities only use at least consulting services and others.495

    Annular impinging jet controlled by radial synthetic jets

An annular nozzle was designed with an active flow control system using twelve radial synthetic jets issuing from the central body of the nozzle. Measurements of the wall pressure and wall mass transfer were performed with air as the working fluid. The control action causes modification of the flowfield resulting in changes of the corresponding heat/mass transfer distributions. The convective transfer rate on the stagnation circle can be demonstrably enhanced by 20% at a moderate nozzle-to-wall distance, equal to 0.6 of the nozzle outer diameter.dc201

    Active control of the wake behind the cylinder

    Active control of the flow around a circular cylinder using a synthetic jet (SJ) array was experimentally investigated in a small wind tunnel. The prevailing method was flow visualization. Modification of the wake was investigated at two values of a dimensionless momentum coefficient (Cμ = 0.21 and 1.02). Various α and Cf parameters were tested (where α is the angle between SJs and the main flow direction and Cf is the frequency ratio). It was found that the control moves a separation point more downstream, i.e. the width of the wake can be reduced. Moreover, increasing frequency ratio Cf causes refining of the vortex structure. When the SJs are placed at the front or rear stagnation points, the wake tends towards the symmetrical pattern

    Utilising the Best Practices of Alternative Management Structures in the Gaming Industry

    This paper covers the study conducted on the topic of alternative management structures. The business environment is changing at a faster pace than ever before, especially due to the accelerating technological development, which makes it difficult for companies to stay relevant. Despite this, the majority of companies still utilise traditional management practices which may not be ideal. They are said to restrict creativity, grant decision making power only to some individuals, and negatively impact employee engagement. However, this study explores the rivalling alternative management approaches which could provide a solution to the problem. The aim of the research was to examine two different alternative management structures of Spotify and Valve Corporation and to determine what the best practices and the pitfalls of such models are. These best practices were then to be compiled into an appliable action plan for the beneficiaries, medium to large European game development companies. The research included a theoretical background focusing on the modern business context, where technological inventions such as AI, big data, and business analytics were examined alongside other trends impacting the modern business environment such as globalisation and the COVID-19 pandemic. Moreover, both traditional and alternative management theory was explored. The traditional management section introduced hierarchy and bureaucracy, their role in the business world and the issues often connected with them. The alternative management section, on the other hand, focused on the characterisation of the topic itself, and the introduction of concepts such as humanocracy, polyarchy, and agile method tailoring. A holistic case study approach with multiple units of analysis was used to then tackle the research question of ”How can game development companies utilise the best practices of alternative management structures to gain competitive advantage?”. The unique management structures and practices of Spotify and Valve Corporation were analysed. Each of the company analysis included four main areas of focus: an introduction where the company was set into context, the management structure exploration, where the approaches were described, employee satisfaction, where workplace reviews were examined, and the conclusion. The data was then coded and organised in a table for the two companies to be compared. Following this, another table was compiled assessing the relevance of the practices utilised by the organisations through connection to the issues and strengths of traditional management and the impact of the modern business environment introduced in the theory. Lastly, the best practices and pitfalls were drawn from the results and an appliable action plan was constructed for the beneficiaries. The results generally confirmed that alternative management structures may be better suitable for the current business environment thanks to their focus on innovation, employee empowerment, and adaptability which was demonstrated by both of the studied companies, despite their significantly different organisational structures

    Glabalization of czech tv entertainment: comparison in using global tv formats in ČT and private tv channel (Nova, Prima)

    This diploma thesis deals with the development of using global TV formats on Czech TV channels during 2000-2013. Development is studied in comparison between Czech Television as a public medium and private channels TV Nova and FTV Prima. Global TV formats recently became basic building blocks for production of their own programs in TV entertainment. Thesis focuses on the approach of the TV channels in using global TV formats. It outlines the development of TV genres of global TV formats on Czech TVs and shows how Czech TV stations have dealt with their production and how they have interested the Czech TV spectators. The thesis is based on theoretical background of media globalization supposing that business with global formats has a strong influence over current TV entertainment. TV market offers many various TV formats and TV stations buy them because they have information about success in other countries. It shows that in recent years the offer of global TV formats is so heterogeneous and wide that viewer rating and success of single formats is not so high as in the beginning of the trend in TV entertainment. In the followed period the Czech TV market has been changed by digitalization of Czech TV broadcasting and there is also a phenomenon of media convergence
