    Smashing back Doors in: Negative Attitudes toward Bottoms within the Gay Community

    In the current study, we examined the relationship between hegemonic masculinity and negative attitudes toward effeminacy with prejudice toward sex-role identities in the gay community. Participants recruited from LGBTQA+ student groups from across the United States, completed measures related to their adherence toward hegemonic masculinity, attitudes toward effeminacy, and opinions about bottoms (men who prefer to be penetrated during anal intercourse). The results showed, first, established evidence that a prejudice toward bottoms does exist. Second, anti-effeminacy attitudes, hostile and benevolent sexism, and male toughness norms predicted prejudice toward bottoms. Taken together, the results illuminate both the importance of sex-role identities within the gay community, and marginalization within the community directed toward men who identify as bottoms

    Moving Forward on the Atlantic Gateway: Context and Further Key Questions

    This paper begins by exploring the meaning of the word gateway, and the role of ‘gateways and corridors’ in the current global trading environment. It examines the integrated nature of today’s economic environment, to provide context for the discussion about the players and their interests—what they seek to gain from gateway development. It then steps back to look at the lessons identified in various research activities undertaken in the Atlantic Gateway Research Initiative, developing four questions for those seeking to participate in gateway development. In answering these four questions, the remainder of the paper proposes strategies for the various players to consider. The paper closes with reflections on the vision for the Gateway and the future research agenda that the current research has identified. The concept of Gateway support discussed here takes a much larger view of the Atlantic Gateway in the world trading environment, and sees roles for all players, including universities, in developing the economic opportunities presented by the Atlantic Gateway

    Diversity of Chironomidae (Diptera) breeding in the Great Stour, Kent: baseline results from the Westgate Parks non-biting midge project

    Chalk rivers and streams are of conservation importance due their ecological diversity, historical relevance and economic value. With more than 200 chalk watercourses, England is considered unusual in having the most chalk rivers in the world. However, due to increasing anthropogenic activities, many English chalk rivers and streams are becoming badly degraded. The non-biting midges or chironomids (Diptera, Chironomidae) are considered key-stone taxa in aquatic food webs, and have been used as ecological indicators of freshwater quality and environmental stress. Here we determined the generic richness, diversity, and community structure of Chironomidae across six sites in the mid-section of the Great Stour in Kent, a chalk river for which concern has been expressed regarding both water and habitat quality. Based on the morphological identification of 1336 insect larvae from the six sites (four in Westgate Parks, Canterbury, and two at nearby locations upstream and downstream from Canterbury City), a total of 20 genera of Chironomidae were identified, including some taxa indicative of freshwater habitats with low levels of organic pollution. There were different levels of generic richness and diversity among sites, and while there was little variation in the community composition among the sites within Westgate Parks, there were noticeable generic differences among Westgate Parks sites compared with those upstream and downstream, showing the highest complementarity and Beta diversity values. Overall, the results were comparable with other studies on chironomids in chalk rivers and other river systems. Although spatially limited to a small stretch of river, this represents the first study on chironomids in the Great Stour and provides baseline information on the diversity and structure of this important insect group with aquatic larvae, useful for the objective interpretation of any future biological assessments and monitoring programmes on the Kentish Stour, and also for comparisons with other chalk rivers

    Developmental Acquisition of Regulomes Underlies Innate Lymphoid Cell Functionality

    Innate lymphoid cells (ILCs) play key roles in host defense, barrier integrity, and homeostasis and mirror adaptive CD4(+) T helper (Th) cell subtypes in both usage of effector molecules and transcription factors. To better understand the relationship between ILC subsets and their Th cell counterparts, we measured genome-wide chromatin accessibility. We find that chromatin in proximity to effector genes is selectively accessible in ILCs prior to high-level transcription upon activation. Accessibility of these regions is acquired in a stepwise manner during development and changes little after in vitro or in vivo activation. Conversely, dramatic chromatin remodeling occurs in naive CD4(+) T cells during Th cell differentiation using a type-2-infection model. This alteration results in a substantial convergence of Th2 cells toward ILC2 regulomes. Our data indicate extensive sharing of regulatory circuitry across the innate and adaptive compartments of the immune system, in spite of their divergent developing pathways

    Climate Change and Biosphere Response: Unlocking the Collections Vault

    Natural history collections (NHCs) are an important source of the long-term data needed to understand how biota respond to ongoing anthropogenic climate change. These include taxon occurrence data for ecological modeling, as well as information that can be used to reconstruct mechanisms through which biota respond to changing climates. The full potential of NHCs for climate change research cannot be fully realized until high-quality data sets are conveniently accessible for research, but this requires that higher priority be placed on digitizing the holdings most useful for climate change research (e.g., whole-biota studies, time series, records of intensively sampled common taxa). Natural history collections must not neglect the proliferation of new information from efforts to understand how present-day ecosystems are responding to environmental change. These new directions require a strategic realignment for many NHC holders to complement their existing focus on taxonomy and systematics. To set these new priorities, we need strong partnerships between NHC holders and global change biologists

    The valence-dominance model applies to body perception

    First impressions of a person, including social judgements, are often based on appearance. The widely accepted valence-dominance model of face perception (Oosterhof and Todorov 2008 Proc. Natl Acad. Sci. USA 105, 11 087-11 092 (doi:10.1073/pnas.0805664105)) posits that social judgements of faces fall along two orthogonal dimensions: trustworthiness (valence) and dominance. The current study aimed to establish the principal components of social judgements based on the perception of bodies, hypothesizing that these would follow the same dimensions as face perception. Stimuli were black and white photographs showing bodies dressed in grey clothing, standing in their natural posture, in left profile. Raters (N = 237) judged the stimuli on the 14 traits used in Oosterhof and Todorov's original study (Oosterhof and Todorov 2008 Proc. Natl Acad. Sci. USA 105, 11 087-11 092 (doi:10.1073/pnas.0805664105)). Data were analysed using principal component analysis (PCA), as in the original study, with an additional exploratory factor analysis (EFA) using oblique rotation. While PCA produced a third dimension in line with several replications of the original study, results from the EFA produced two dimensions, representing trustworthiness and dominance, providing support for the hypothesis that social perceptions of bodies can be summarized using the valence-dominance model. These two factors could represent universal perceptions we have about people. Future research could explore social judgements of humans based on other stimuli, such as voices or body odour, to evaluate whether the trustworthiness and dominance dimensions are consistent across modalities

    While the neural mechanisms underpinning the perception of muscularity are poorly understood, recent progress has been made using the psychophysical technique of visual adaptation. Prolonged visual exposure to high (low) muscularity bodies causes subsequently viewed bodies to appear less (more) muscular, revealing a recalibration of the neural populations encoding muscularity. Here, we use visual adaptation to further elucidate the tuning properties of the neural processes underpinning muscle perception for the upper and lower halves of the body. Participants manipulated the apparent muscularity of upper and lower bodies until they appeared ‘normal’, prior to and following exposure to a series of top/bottom halves of bodies that were either high or low in muscularity. In Experiment 1, participants were adapted to isolated own-gender body halves from one of four conditions; increased (muscularity) upper (body half), increased lower, decreased upper, or decreased lower. Despite the presence of muscle aftereffects when the body halves the participants viewed and manipulated were congruent, there was only weak evidence of muscle aftereffect transfer between the upper and lower halves of the body. Aftereffects were significantly weaker when body halves were incongruent, implying minimal overlap in the neural mechanisms encoding muscularity for body half. Experiment 2 examined the generalisability of Experiment 1's findings in a more ecologically valid context using whole-body stimuli, producing a similar pattern of results as Experiment 1, but with no evidence of cross-adaptation. Taken together, the findings are most consistent with muscle-encoding neural populations that are body-half selective. As visual adaptation has been implicated in cases of body size and shape misperception, the present study furthers our current understanding of how these perceptual inaccuracies, particularly those involving muscularity, are developed, maintained, and may potentially be treated

    Colonization Density of the Upper Respiratory Tract as a Predictor of Pneumonia-Haemophilus influenzae, Moraxella catarrhalis, Staphylococcus aureus, and Pneumocystis jirovecii.

    Background. There is limited information on the association between colonization density of upper respiratory tract colonizers and pathogen-specific pneumonia. We assessed this association for Haemophilus influenzae, Moraxella catarrhalis, Staphylococcus aureus, and Pneumocystis jirovecii. Methods. In 7 low- and middle-income countries, nasopharyngeal/oropharyngeal swabs from children with severe pneumonia and age-frequency matched community controls were tested using quantitative polymerase chain reaction (PCR). Differences in median colonization density were evaluated using the Wilcoxon rank-sum test. Density cutoffs were determined using receiver operating characteristic curves. Cases with a pathogen identified from lung aspirate culture or PCR, pleural fluid culture or PCR, blood culture, and immunofluorescence for P. jirovecii defined microbiologically confirmed cases for the given pathogens. Results. Higher densities of H. influenzae were observed in both microbiologically confirmed cases and chest radiograph (CXR)–positive cases compared to controls. Staphylococcus aureus and P. jirovecii had higher densities in CXR-positive cases vs controls. A 5.9 log10 copies/mL density cutoff for H. influenzae yielded 86% sensitivity and 77% specificity for detecting microbiologically confirmed cases; however, densities overlapped between cases and controls and positive predictive values were poor (\u3c3%). Informative density cutoffs were not found for S. aureus and M. catarrhalis, and a lack of confirmed case data limited the cutoff identification for P. jirovecii. Conclusions. There is evidence for an association between H. influenzaecolonization density and H. influenzae–confirmed pneumonia in children; the association may be particularly informative in epidemiologic studies. Colonization densities of M. catarrhalis, S. aureus, and P. jirovecii are unlikely to be of diagnostic value in clinical settings

    Huntingtin subcellular localisation is regulated by Kinase signalling activity in the StHdhQ111 model of HD

    Huntington’s disease is a neurodegenerative disorder characterised primarily by motor abnormalities, and is caused by an expanded polyglutamine repeat in the huntingtin protein. Huntingtin dynamically shuttles between subcellular compartments, and the mutant huntingtin protein is mislocalised to cell nuclei, where it may interfere with nuclear functions, such as transcription. However, the mechanism by which mislocalisation of mutant huntingtin occurs is currently unknown. An immortalised embryonic striatal cell model of HD (StHdhQ111) was stimulated with epidermal growth factor in order to determine whether the subcellular localisation of huntingtin is dependent on kinase signalling pathway activation. Aberrant phosphorylation of AKT and MEK signalling pathways was identified in cells carrying mutant huntingtin. Activity within these pathways was found to contribute to the regulation of huntingtin and mutant huntingtin localisation, as well as to the expression of immediate-early genes. We propose that altered kinase signalling is a phenotype of Huntington’s disease that occurs prior to cell death; specifically, that altered kinase signalling may influence huntingtin localisation, which in turn may impact upon nuclear processes such as transcriptional regulation. Aiming to restore the balance of activity between kinase signalling networks may therefore prove to be an effective approach to delaying Huntington’s disease symptom development and progression
