Erfassung von Vorstellungen von der Zelle und der Zellteilung mit dem Own-Word-Mapping Verfahren
In dieser Studie wurde das Own-Word-Mapping (OWM) im Biologieunterricht mit 75 Schülern dreier 9. Realschulklassen zur Erhebung von Schülervorstellungen von der Zelle und der Zellteilung eingesetzt. Das OWM stellt ein geeignetes Verfahren zur Erhebung von Schülervorstellungen für Forschungszwecke dar. Aus 27% aller OW-Maps konnten 20 Konzepte zum Thema identifi ziert werden, von denen 17 bereits aus anderen Untersuchungen bekannt waren. Zusätzlich wurden drei neue Konzepte zur Zelle und der Zellteilung gefunden. Berücksichtigt man, dass jeweils nur Abschnitte der Verknüpfungsketten in den OW-Maps ausgewertet werden konnten, stellt das hier genutzte OWM wohl kein effi zientes Verfahren dar, um ein Sprechen über Vorstellungen zur Zelle und Zellteilung im Unterricht anzuregen. Im untersuchten Fall erwiesen sich mikroskopische Aufnahmen von Zellen gegenüber Schemazeichnungen als wesentlich günstigere Anfangsund Zielbilder, um spezifi sche und fachnahe Vorstellungen von der Zelle und der Zellteilung bei den Schülern anzuregen
SPSB1-mediated inhibition of TGF-β receptor-II impairs myogenesis in inflammation
BACKGROUND: Sepsis-induced intensive care unit-acquired weakness (ICUAW) features profound muscle atrophy and attenuated muscle regeneration related to malfunctioning satellite cells. Transforming growth factor beta (TGF-β) is involved in both processes. We uncovered an increased expression of the TGF-β receptor II (TβRII)-inhibitor SPRY domain-containing and SOCS-box protein 1 (SPSB1) in skeletal muscle of septic mice. We hypothesized that SPSB1-mediated inhibition of TβRII signalling impairs myogenic differentiation in response to inflammation. METHODS: We performed gene expression analyses in skeletal muscle of cecal ligation and puncture- (CLP) and sham-operated mice, as well as vastus lateralis of critically ill and control patients. Pro-inflammatory cytokines and specific pathway inhibitors were used to quantitate Spsb1 expression in myocytes. Retroviral expression plasmids were used to investigate the effects of SPSB1 on TGF-β/TβRII signalling and myogenesis in primary and immortalized myoblasts and differentiated myotubes. For mechanistical analyses we used coimmunoprecipitation, ubiquitination, protein half-life, and protein synthesis assays. Differentiation and fusion indices were determined by immunocytochemistry, and differentiation factors were quantified by qRT-PCR and Western blot analyses. RESULTS: SPSB1 expression was increased in skeletal muscle of ICUAW patients and septic mice. Tumour necrosis factor (TNF), interleukin-1β (IL-1β), and IL-6 increased the Spsb1 expression in C2C12 myotubes. TNF- and IL-1β-induced Spsb1 expression was mediated by NF-κB, whereas IL-6 increased the Spsb1 expression via the glycoprotein 130/JAK2/STAT3 pathway. All cytokines reduced myogenic differentiation. SPSB1 avidly interacted with TβRII, resulting in TβRII ubiquitination and destabilization. SPSB1 impaired TβRII-Akt-Myogenin signalling and diminished protein synthesis in myocytes. Overexpression of SPSB1 decreased the expression of early (Myog, Mymk, Mymx) and late (Myh1, 3, 7) differentiation-markers. As a result, myoblast fusion and myogenic differentiation were impaired. These effects were mediated by the SPRY- and SOCS-box domains of SPSB1. Co-expression of SPSB1 with Akt or Myogenin reversed the inhibitory effects of SPSB1 on protein synthesis and myogenic differentiation. Downregulation of Spsb1 by AAV9-mediated shRNA attenuated muscle weight loss and atrophy gene expression in skeletal muscle of septic mice. CONCLUSIONS: Inflammatory cytokines via their respective signalling pathways cause an increase in SPSB1 expression in myocytes and attenuate myogenic differentiation. SPSB1-mediated inhibition of TβRII-Akt-Myogenin signalling and protein synthesis contributes to a disturbed myocyte homeostasis and myogenic differentiation that occurs during inflammation
Mesopontine rostromedial tegmental nucleus neurons projecting to the dorsal raphe and pedunculopontine tegmental nucleus: psychostimulant-elicited Fos expression and collateralization
The mesopontine rostromedial tegmental nucleus (RMTg) is a GABAergic structure in the ventral midbrain and rostral pons that, when activated, inhibits dopaminergic neurons in the ventral tegmental area and substantia nigra compacta. Additional strong outputs from the RMTg to the pedunculopontine tegmental nucleus pars dissipata, dorsal raphe nucleus, and the pontomedullary gigantocellular reticular formation were identified by anterograde tracing. RMTg neurons projecting to the ventral tegmental area express the immediate early gene Fos upon psychostimulant administration. The present study was undertaken to determine if neurons in the RMTg that project to the additional structures listed above also express Fos upon psychostimulant administration and, if so, whether single neurons in the RMTg project to more than one of these structures. We found that about 50% of RMTg neurons exhibiting retrograde labeling after injections of retrograde tracer in the dorsal raphe or pars dissipata of the pedunculopontine tegmental nucleus express Fos after acute methamphetamine exposure. Also, we observed that a significant number of RMTg neurons project both to the ventral tegmental area and one of these structures. In contrast, methamphetamine-elicited Fos expression was not observed in RMTg neurons labeled with retrograde tracer following injections into the pontomedullary reticular formation. The findings suggest that the RMTg is an integrative modulator of multiple rostrally projecting structures
Neurodevelopmental Disruption of Cortico-Striatal Function Caused by Degeneration of Habenula Neurons
The habenula plays an important role on cognitive and affective functions by regulating monoamines transmission such as the dopamine and serotonin, such that its dysfunction is thought to underlie a number of psychiatric conditions. Given that the monoamine systems are highly vulnerable to neurodevelopmental insults, damages in the habenula during early neurodevelopment may cause devastating effects on the wide-spread brain areas targeted by monoamine innervations.Using a battery of behavioral, anatomical, and biochemical assays, we examined the impacts of neonatal damage in the habenula on neurodevelopmental sequelae of the prefrontal cortex (PFC) and nucleus accumbens (NAcc) and associated behavioral deficits in rodents. Neonatal lesion of the medial and lateral habenula by ibotenic acid produced an assortment of behavioral manifestations consisting of hyper-locomotion, impulsivity, and attention deficit, with hyper-locomotion and impulsivity being observed only in the juvenile period, whereas attention deficit was sustained up until adulthood. Moreover, these behavioral alterations were also improved by amphetamine. Our study further revealed that impulsivity and attention deficit were associated with disruption of PFC volume and dopamine (DA) receptor expression, respectively. In contrast, hyper-locomotion was associated with decreased DA transporter expression in the NAcc. We also found that neonatal administration of nicotine into the habenula of neonatal brains produced selective lesion of the medial habenula. Behavioral deficits with neonatal nicotine administration were similar to those caused by ibotenic acid lesion of both medial and lateral habenula during the juvenile period, whereas they were different in adulthood.Because of similarity between behavioral and brain alterations caused by neonatal insults in the habenula and the symptoms and suggested neuropathology in attention deficit/hyperactivity disorder (ADHD), these results suggest that neurodevelopmental deficits in the habenula and the consequent cortico-striatal dysfunctions may be involved in the pathogenesis and pathophysiology of ADHD
Glutamaterge Fasern der lateralen Habenula projizieren hauptsächlich auf GABAerge Neuronen der VTA
The concept of cortical-subcortical loops emphasizes the importance of the
basal gan-glia for motor, psychomotor, and emotional cortical functions. These
loops are bidirec-tionally controlled by the midbrain dopaminergic system,
predominantly but not exclu-sively at the level of the striatum, including the
accumbens nucleus. Successful behaviors increase the activities of the
mesostriatal (arising in the complex part of the substantia nigra) and
mesolimbic (arising in the ventral tegmental area, VTA) neurons, thereby
reinforcing the corresponding actions. In contrast, unsuccessful behaviors
result in an increased activation of the lateral habenular complex (LHb),
thereby decreasing the activities of mesolimbic neurons. Correspondingly,
electrical stimulation of the LHb effectively blocks neuronal activity in the
VTA. Whether this block is due to an inhibitory projection from the LHb to the
VTA, or whether axons from excitatory LHb neurons tar-get inhibitory neurons
within the VTA, is currently not known. Here we show, using in-situ-
hybridization and immunocytochemical double labelling at the light and
electron microscopic level, that GABAergic neurons are scarce in the LHb and
that glutamatergic axons from the LHb mostly target GABAergic neurons in the
VTA and the mesopontine rostromedial tegmental nucleus. These data explain the
inhibitory effect of LHb activation on the VTA. In addition, however, a small
number of LHb termi-nals in the VTA actually contacts dopaminergic neurons.
The biological importance of these terminals requires further investigation. .Einleitung: Dopaminerge Neurone in der ventralen tegmentalen Area (VTA)
modulieren die neuronale Aktivität der Basalganglienschleifen. Die
Ausschüttung von Dopamin in den Zielarealen wie den Nucleus accumbens führt
dabei zu positiven Emotionen, die in ihrer Summe als Belohnung wahrgenommen
werden. Im Gegenzug führt eine Hem-mung der Dopaminsekretion zu negativen
Emotionen (negative reward). Die VTA ist somit unabdingbar für Lernprozesse
und daraus resultierendes, zielgerichtetes Han-deln. Die Neurone der VTA
integrieren dafür stimulierende und inhibitorische Einflüsse aus verschiedenen
kortikalen und subkortikalen Zentren. Ein wichtiger Zufluss von In-formationen
erwächst der VTA aus dem lateralen Habenularkomplex (LHb). In früheren Studien
konnte bereits gezeigt werden, dass eine elektrische Stimulation von Neuronen
in der LHb zu einer Hemmung der Dopaminausschüttung in der VTA führt. Die
Charak-terisierung dieser Projektionen wurde bisher nur unzureichend
untersucht. Insbesonde-re die beteiligten Neurotransmitter und die Zielneurone
der in der VTA waren nicht be-kannt. Ziel der vorliegenden Arbeit war es,
durch eine Kombination verschiedener anatomischer Techniken eine genaue
Charakterisierung der Projektionen aus der LHb und der angesteuerten
Zielneurone in der VTA zu erreichen. Resultate: Die Existenz von Projektionen
aus der LHb in die VTA wurde durch ver-schiedenen Tracingtechniken
nachgewiesen. Dabei führte die Injektion von Tetra-methylrodamin-markiertem
Dextranamin (TMR-DA) in die LHb zu einer Anreicherung des Tracers in
Axonterminalen in der VTA (anterogrades Tracing). Im Gegenzug konnte nach
Injektion von Gold-markiertem Weizenkeim Agglutinin (ApoHRP-WGA-gold) in die
VTA ein retrograder Transport der Substanz in die Neuronen der LHb dargestellt
wer-den (retrogrades Tracing). Durch nicht radioaktive In-Situ-Hybridisierung
wurde dann gezeigt, dass die Mehrzahl der WGA-markierten Neurone in der LHb
mRNA für den vesikulären Glutamattransporter vGluT2 exprimieren. Dieser Befund
wurde durch den immuncytochemischen Nachweis von vGluT2 in TMR-DA-positiven
Axonterminalen in der VTA ergänzt. GAD-immunreaktive GABAerge Axonterminalen
in der VTA enthielten dagegen keine Tracersubstanz. Die Ursprungsneurone für
die Projektionen aus der LHb in die VTA konnten damit als exzitatorische,
glutamaterge Neurone identifiziert werden. Die Charakterisierung der
Zielzellen erfolgte mittels Pre- und Postembedding-techniken in der
Elektronenmikroskopie. Dazu wurden dopaminerge Zellen mit Antikörpern gegen
Tyrosinhydroxylase, GABAerge Zellen mit Antikörpern gegen GABA und GAD, sowie
glutamaterge Neurone mit Antikörpern gegen vesikuläre Glutamattransporter
(vGluT1 und vGluT2) markiert. TMR-DA-positive Axonterminalen, mittels DAB
visualisiert, fan-den sich dabei hauptsächlich an GABAergen Dendriten, während
direkte Projektionen auf dopaminerge Neurone selten waren. Schlussfolgerung:
In der vorliegenden Arbeit konnte gezeigt werden, dass exzitatori-sche,
glutamaterge Neurone der LHb direkt auf GABAerge Interneurone in der VTA pro-
jizieren. Diese Projektionen bilden das morphologische Korrelat für die
hemmende Wir-kung der LHb auf die Dopaminfreisetzung aus den Neuronen der VTA
und ihren Axonterminalen. Die neuen Erkenntnisse verbessern unser Verständnis
der Funktions-weise der VTA und können so vielleicht zur Entwicklung neuer
Behandlungsstrategien für Erkrankungen wie Depression und Schizophrenie

Die Verfasserin zeichnet die Entwicklung der Frauenerwerbsarbeit nach und arbeitet Grundlagen und Auswirkungen geschlechtshierarchischer Arbeitsteilung heraus. Sie diskutiert die Position der Frauen zwischen Hausarbeit und Erwerbsarbeit und analysiert anhand von Zeitverwendungsanalysen die geschlechtsspezifische Logik von Zeitordnungen. Exemplarische Umverteilungsansaetze (Bayerisch-saechsische Zukunftskommission, Club of Rome, VW-Modell) werden auf ihre Tauglichkeit im Hinblick auf eine Ueberwindung geschlechtsspezifischer Arbeitsteilung hin untersucht. Auf dieses Basis wird die Vision einer humanen, frauenfreundlichen Gesellschaft entworfen, eine Perspektive, fuer die dem Handlungsansatz Arbeitszeitpolitik nach Ansicht der Verfasserin eine zentrale Bedeutung zukommt. Eine Ueberwindung der 'herrschenden Zeitlogik' muss, so das Fazit der Untersuchung, mit einer Ueberwindung der 'herrschenden Produktionsweise' einhergehen.
