    Cross-study analysis of genomic data defines the ciliate multigenic epiplasmin family: strategies for functional analysis in Paramecium tetraurelia

    <p>Abstract</p> <p>Background</p> <p>The sub-membranous skeleton of the ciliate <it>Paramecium</it>, the epiplasm, is composed of hundreds of epiplasmic scales centered on basal bodies, and presents a complex set of proteins, epiplasmins, which belong to a multigenic family. The repeated duplications observed in the <it>P. tetraurelia </it>genome present an interesting model of the organization and evolution of a multigenic family within a single cell.</p> <p>Results</p> <p>To study this multigenic family, we used phylogenetic, structural, and analytical transcriptional approaches. The phylogenetic method defines 5 groups of epiplasmins in the multigenic family. A refined analysis by Hydrophobic Cluster Analysis (HCA) identifies structural characteristics of 51 epiplasmins, defining five separate groups, and three classes. Depending on the sequential arrangement of their structural domains, the epiplasmins are defined as symmetric, asymmetric or atypical. The EST data aid in this classification, in the identification of putative regulating sequences such as TATA or CAAT boxes. When specific RNAi experiments were conducted using sequences from either symmetric or asymmetric classes, phenotypes were drastic. Local effects show either disrupted or ill-shaped epiplasmic scales. In either case, this results in aborted cell division.</p> <p>Using structural features, we show that 4 epiplasmins are also present in another ciliate, <it>Tetrahymena </it><it>thermophila</it>. Their affiliation with the distinctive structural groups of <it>Paramecium </it>epiplasmins demonstrates an interspecific multigenic family.</p> <p>Conclusion</p> <p>The epiplasmin multigenic family illustrates the history of genomic duplication in <it>Paramecium</it>. This study provides a framework which can guide functional analysis of epiplasmins, the major components of the membrane skeleton in ciliates. We show that this set of proteins handles an important developmental information in <it>Paramecium </it>since maintenance of epiplasm organization is crucial for cell morphogenesis.</p

    Correlation between some environmental variables and abundance of Almophrya mediovacuolata (Ciliophora: Anoplophryidae) endocommensal ciliate of an anecic earthworms (Oligochaeta: Annelida) in Bambui (North-West Cameroon)

    This study was devoted to accessing the influence of some soil physico-chemical parameters on the abundance of Almophrya mediovacuolata Ngassam, 1983, astome ciliate of the digestive tract of earthworms (EW) of the species Alma nilotica collected along “Fa’a ntsa” stream in Bambui. The survey primarily involved soil samples collection from the same spots of EW collection and preparation for physico-chemical analysis; evaluation in situ of the volumic density (VD) of worms (number /dm3 soil), their dissection, isolation and counting of ciliates with respect to different portion of EW’s gut (fore, mid and hindgut). Furthermore, correlation analysis between soil physico-chemical parameters and biological responses (EW volumic density and ciliate abundance) were performed. The results reveal that EW abundance was positively and significantly correlated with the following physico-chemical parameters: Cation Exchange Capacity CEC (p &lt;0.01) and Mg2+ (p &lt;0.05). A positive and significant correlation was found between Almophrya mediovacuolata and the pH of KCl in the foregut and midgut (p &lt;0.01) while a negative and significant correlation was found between the abundance of Almophrya mediovacuolata and Ca2+ in the foregut (p &lt;0.05). Almophrya mediovacuolata were found mostly in the foregut. This result shows that each portion of the digestive tract of Alma nilotica can be considered as a set of natural microhabitat in which a number of physico-chemical factors generate ecological niches suitable for the survival of different species of microorganisms among which ciliated protozoa. The fore and midgut was noticed to be the preferential zones of Almophrya mediovacuolata.Keywords: Biotic parameters, ciliated protozoa, microhabitat, soil physico-chemical parameters

    Description of five new species of hysterocinetian ciliates (Ciliophora, Hysterocinetidae), endocommensal in a terrestrial oligochaete of the genus Alma from Cameroon

    International audienceStudies on five new species of hysterocinetian ciliates, endocommensal within a terrestrial oligochaete from the genus Alma (Cameroon), reveal the existence of three distinct types of cytoskeletal topography within their adhesive apparatus. In the first topographical type, four superimposed antagonistic strata of skeletal fibres that correspond to the arrangement observed in the genus Ptychostomum, Stein 1860 are found. The second topographical type is characterized by the existence of an unorganized field of fibrillar structures forming a single dense mass in the central part of the sucker. For this, a new genus is created: Ptychostomoides, Nana, Fokam, Ntoungwa et al. n. g. The third type present in the genus Proptychostomoides, Nana, Fokam, Ntoungwa et al. n. g. differs from the latter by the existence of a bare patch within the central region of the adhesive apparatus. This new evidence of morphological diversification within the Hysterocinetidae supports a new approach to the systematics of this group

    Women’s childbearing experiences vary with pregnancy intentional nature and outcome. An Associative Network study targeted 129 women pregnant &gt;1 year ago and their experiences at pregnancy start and post-pregnancy. Word-associations formed 15 themes and 5 metathemes. The main pregnancy discovery themes were “Affect” (39%), “Relationships with others” (11%), and “Logistics” (7%). The main post-pregnancy themes were “Affect” (18%), “Relationship with the child” (13%), and “Personal progress” (12%). The overall polarity index was higher in intended vs. unintended pregnancies. Whatever pregnancy outcome, women expressed impressions of constructive experience. Discovering pregnancy and deciding about it led anyway to personal and social progress

    International audienceThe paper demonstrates the influence of physico-chemical parameters on the distribution of endocommensal ciliates through the gut of the earthworm Alma emini. We measured physico-chemical parameters of the intestinal liquid extracted with the vacuum aspiration technique and concomitantly recorded biological parameter (species abundance). Furthermore, correlation analysis between physico-chemical parameters and biological parameter was performed in different compartments. In the foregut, among the eleven species of Astomatia recorded, correlation was significant between Metaracoelophrya intermedia, Coelophrya roquei and Water Content (WC = 46.94 Âą 7.77%). In the midgut, among the nine species of Hysterocinetidae recorded, a significant correlation was observed between Metaptychostomum ebebdae, Ptychostomum macrostomum and Electric Conductivity (EC = 84.55 Âą 12.94 ÎźS/ cm). In the same compartment, a significant correlation was also observed between Ptychostomum macrostomum and Total Dissolved Substance (TDS = 16.20 Âą 3.46%). In the hindgut, eight species of Astomatia were found, among which significant correlation was obtained between Coelophrya roquei and Hydrogen potential (pH = 7.35 Âą 0.16). In the same compartment, taking into account the eleven species of Hysterocinetidae recorded, a significant correlation was also obtained between Ptychostomum macrostomum and pH; Ptychostomum commune and WC (28.84 Âą 3.97%). These results suggest that each part of the digestive tract of A. emini can be considered as a set of natural microhabitats in which certain physico-chemical factors generate ecological niches suitable for one or another group of species

    International audienceThe performances of two rice species of the genus Oryza were evaluated by considering parameters such as the number of panicle per rice plant, the number of primary branches of the panicles, panicle length, the plant height, the number of grains and empty balls per panicle. It appears that O. sativa from Japan has a number of empty balls less than NERICA’s varieties from Africa. We also noted that O. sativa has the highest number of panicles per rice plant (a mean of 14.05 ± 6.14). However, the number of grains per panicle NERICA’s varieties was very high (a mean of 164 ± 38.82) and therefore they shows more attractive characters than Orysa sativa lines. The differences were statistically significant above all the parameters considered in both African and Japan rice studied. A significant correlation that was negative has been observed between the number of panicles per rice plant and the number of primary branches per panicle within the NERICA 9 variety (P <0.05). The correlation was highly significant and positive (P <0.001) between the number of primary branches per panicle and the number of grain per panicle. These results suggest another demonstration of the hybrid vigor of NERICA varieties

    Assessment of the potential effect of some streams properties on the isolated Aeromonas hydrophila strains susceptibility against some β-Lactams and Sulfamids

    The isolation and characterization of A. hydrophila strains were carried out a given stream located in the equatorial zone in Cameroon (Central Africa), during the mild rainy season and the mild dry season. The water physicochemical parameters measured were temperature, pH, electrical conductivity, color, turbidity and suspended solids. It has been noted that bacterial abundances as well as the values of physicochemical parameters undergoes temporal variations. Antimicrobial susceptibility tests showed that the means diameters of inhibition with Oxacillin, Ceftriazone, Penicellin, Sulfamethoxazol-trimethoprim, Chloramphenicol, Imipenem, Amoxicillin-Clavulanic acid and Cefazolin were 6.44, 21, 6.63, 9.58, 15.37, 22.18, 14.29 and 10.26 mm respectively. 100% of strains tested were resistant to Penicillin, 89.47 % resistant to Oxacillin, 68.42% resistant to Sulphamethoxazole-trimetroprim and 63.15% were resistant to Cefazolin. There were 54.60% cases of antibiotic resistance, 24.34 % of intermediate sensitivity and 21.05% of sensitive strains against the tested antibiotics. The encoded redundancy analysis indicates that the percentage of the variation explained on the canonical axes is of 47.8% for the F1 axis and 25.9% for the F2 axis, for an accumulated percentage of 73.7%. The suspended solids, the Ceftriazon and Chloramphenicol are correlated to the F1 axis in positive coordinates, and the Temperature and Amoxicillin-Clavulanic acid in negative coordinates. The correlation test between the water physicochemical characteristics and the inhibition diameters of antibiotics showed some significant correlations (P&lt;0.05). The regulation of bacterial genes would be regulated by complex mechanisms. Although many factors implied are linked to the bacterial cell, others may belong to the closed environment

    Actin-interacting and flagellar proteins in Leishmania spp.: Bioinformatics predictions to functional assignments in phagosome formation

    Several motile processes are responsible for the movement of proteins into and within the flagellar membrane, but little is known about the process by which specific proteins (either actin-associated or not) are targeted to protozoan flagellar membranes. Actin is a major cytoskeleton protein, while polymerization and depolymerization of parasite actin and actin-interacting proteins (AIPs) during both processes of motility and host cell entry might be key events for successful infection. For a better understanding the eukaryotic flagellar dynamics, we have surveyed genomes, transcriptomes and proteomes of pathogenic Leishmania spp. to identify pertinent genes/proteins and to build in silico models to properly address their putative roles in trypanosomatid virulence. In a search for AIPs involved in flagellar activities, we applied computational biology and proteomic tools to infer from the biological meaning of coronins and Arp2/3, two important elements in phagosome formation after parasite phagocytosis by macrophages. Results presented here provide the first report of Leishmania coronin and Arp2/3 as flagellar proteins that also might be involved in phagosome formation through actin polymerization within the flagellar environment. This is an issue worthy of further in vitro examination that remains now as a direct, positive bioinformatics-derived inference to be presented

    Molecular Phylogeny and Evolution of Parabasalia with Improved Taxon Sampling and New Protein Markers of Actin and Elongation Factor-1Îą

    BACKGROUND: Inferring the evolutionary history of phylogenetically isolated, deep-branching groups of taxa-in particular determining the root-is often extraordinarily difficult because their close relatives are unavailable as suitable outgroups. One of these taxonomic groups is the phylum Parabasalia, which comprises morphologically diverse species of flagellated protists of ecological, medical, and evolutionary significance. Indeed, previous molecular phylogenetic analyses of members of this phylum have yielded conflicting and possibly erroneous inferences. Furthermore, many species of Parabasalia are symbionts in the gut of termites and cockroaches or parasites and therefore formidably difficult to cultivate, rendering available data insufficient. Increasing the numbers of examined taxa and informative characters (e.g., genes) is likely to produce more reliable inferences. PRINCIPAL FINDINGS: Actin and elongation factor-1Îą genes were identified newly from 22 species of termite-gut symbionts through careful manipulations and seven cultured species, which covered major lineages of Parabasalia. Their protein sequences were concatenated and analyzed with sequences of previously and newly identified glyceraldehyde-3-phosphate dehydrogenase and the small-subunit rRNA gene. This concatenated dataset provided more robust phylogenetic relationships among major groups of Parabasalia and a more plausible new root position than those previously reported. CONCLUSIONS/SIGNIFICANCE: We conclude that increasing the number of sampled taxa as well as the addition of new sequences greatly improves the accuracy and robustness of the phylogenetic inference. A morphologically simple cell is likely the ancient form in Parabasalia as opposed to a cell with elaborate flagellar and cytoskeletal structures, which was defined as most basal in previous inferences. Nevertheless, the evolution of Parabasalia is complex owing to several independent multiplication and simplification events in these structures. Therefore, systematics based solely on morphology does not reflect the evolutionary history of parabasalids
